Association between intraoperative balloon morphology and postoperative sensory dysfunction after percutaneous balloon compression for trigeminal neuralgia: a propensity score-matched cohort study.

BACKGROUND: Percutaneous balloon compression (PBC) is widely used for trigeminal neuralgia, but the association between intraoperative balloon morphology and postoperative sensory outcomes remains unclear. Defining this relationship may improve technique and risk stratification. Prior studies suggest selective injury to large myelinated fibers rather than generalized sensory fiber or ganglion injury, and balloon morphology should be interpreted as a procedural marker rather than a direct measure of neural injury. METHODS: We retrospectively identified patients who underwent PBC for trigeminal

[IGFBP-4 improves synovitis by reducing M1 polarization of macrophages via the Wnt/β-catenin pathway].

Objective To investigate the mechanism by which insulin-like growth factor binding protein 4 (IGFBP-4) ameliorates synovial inflammation in osteoarthritis (OA) by inhibiting M1-type polarization of synovial macrophages via the Wnt/β-catenin signaling pathway. Methods The OA model was established in male C57BL/6 mice by intra-articular injection of sodium iodoacetate. Adenoviral vectors carrying IGFBP-4 overexpression or knockdown sequences were injected into the knee joint cavity for in vivo intervention. Micro-computed tomography (Micro-CT) was used to observe the subchondral bone changes in

Mitochondrial transfer protects against cisplatin-induced hearing loss by fumarate-mediated activation of mitophagy.

Sensorineural hearing loss (SNHL) affects millions of individuals worldwide, with platinum-based chemotherapeutics identified as a significant cause. In this study, we elucidate the mechanism by which mitochondrial transfer from mesenchymal stem/stromal cells (MSCs) protects against cisplatin-induced cochlear injury in a mouse model. Systemically administered MSCs home to the cochlea and transfer mitochondria to damaged spiral ganglion neurons (SGNs) via tunneling nanotubes, leading to elevated intracellular levels of the Krebs cycle intermediate fumarate, which activates mitophagy to eliminat

Cinnamaldehyde protects against early diabetic retinopathy: an integrative In vivo and computational study.

BACKGROUND: Diabetic retinopathy (DR) is a leading cause of vision impairment, and current treatments are limited to advanced stages. Cinnamaldehyde (CA), the primary bioactive constituent of cinnamon, has demonstrated anti-apoptotic and anti-diabetic properties, but its role and mechanism in early DR remain unexplored. This study investigated whether CA protects against early DR and whether the effect is associated with phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT) pathway activation. PURPOSE: This study investigated whether CA exerts anti‑apoptotic effects in early DR and whether

Applications of single-cell transcriptomics in glaucoma research: from the aqueous humor outflow pathway to retinal ganglion cell neurodegeneration.

Glaucoma is the leading cause of irreversible blindness worldwide, driven by progressive retinal ganglion cell (RGC) degeneration. While genetic and environmental risk factors have been identified, the cell-type-specific mechanisms underlying pathogenesis remain incompletely understood. Single-cell RNA sequencing (scRNA-seq) enables high-resolution analysis of cellular heterogeneity, developmental trajectories and intercellular communication networks. This review synthesizes recent scRNA-seq advances across two critical domains in glaucoma research: (1) aqueous humor outflow pathway (AHOP) phy

Protectin DX alleviates lumbar radicular pain in rats by suppressing RIP3/MLKL-associated necroinflammation through restoration of autophagic function.

BACKGROUND: Lumbar radicular pain (LRP) is a prevalent neuropathic pain disorder driven by nucleus pulposus (NP)-induced inflammation. Protectin DX (PDX), a novel specialized pro-resolving lipid mediator (SPM), exerts pro-resolving and analgesic effects in LRP. However, the underlying molecular mechanisms remain unclear. Emerging evidence suggests that crosstalk between necroptosis and autophagy coordinately regulates cell fate and inflammatory responses. This study therefore investigated whether this crosstalk mediates the therapeutic effects of PDX in LRP. METHODS: Rat models of LRP were es

Beyond development: LINX/Islr2 expression and TrkA interaction in the adult nervous system under neuroinflammatory challenge.

INTRODUCTION: LINX, also known as immunoglobulin superfamily containing leucine-rich repeat 2 (Islr2), was identified as a TrkA pathway component and demonstrated to modulate the NGF/TrkA-mediated axonal growth, guidance, and branching during development. However, there is limited information on LINX expression in the adult nervous system, and the possible functions of LINX in mature neurons remain unexplored. These two questions are addressed by the present study, which first investigates LINX expression in the adult spinal cord (SC), dorsal root ganglia (DRG), and cerebral cortex (Cx), and s

Metabolic-Redox Remodeling of Inflammatory Synovium by Mannose-Modified Selenium Nanoparticles for Osteoarthritis Therapy.

Osteoarthritis (OA) progression is driven by persistent synovial inflammation, amplified by oxidative stress and enhanced glycolysis. Our previous study has demonstrated that glucose transporter 1 (GLUT1) plays a key role in initiating OA synovitis via regulation of glycolysis. Single-cell RNA sequencing and tissue-specific fluorescent reporter mice (Prg4GFPCreERt2 and Lyz2-iCre;tdTomato) reveal that GLUT1 marks a hyperinflammatory synovial state coupled to impaired selenoprotein-dependent redox homeostasis. Accordingly, mannose-modified selenium nanoparticles (M-SeNPs) are developed as an int

Transcriptomic Identification and Validation of NDUFS2 in Acute Rat TMJ Synovitis and Its Functional Role in LPS-Induced Synovial Fibroblast Inflammation.

PURPOSE: Temporomandibular joint (TMJ) synovitis persistently drives TMJ osteoarthritis (TMJOA) progression. Aberrant reactive oxygen species (ROS) accumulation triggers chronic oxidative stress, amplifying synovial inflammation. This study aimed to identify candidate ROS-related genes through transcriptomic screening and explore their potential functional involvement in TMJ synovitis. METHODS: Acute TMJ synovitis was induced in male Sprague-Dawley rats via intra-articular injection of Complete Freund's Adjuvant (CFA). RNA sequencing of inflamed and normal synovial tissues was performed to sc

Gustatory Connectivity and Central Projections of the Mouse Petrosal-Nodose-Jugular Ganglia Complex.

The petrosal, nodose, and jugular ganglia (PNJ) complex is a major sensory gateway through which gustatory, visceral, and somatosensory information from the body is transmitted to the brain. Despite its importance, experimental access to the PNJ complex in mice remains technically challenging, limiting strategies for anatomical tracing and functional manipulation of these pseudounipolar neurons. Here, we detail an improved surgical approach for direct viral vector delivery to the mouse PNJ complex and demonstrate utility of the technique by labeling peripheral and brain-wide central projection

Early arthritis signs and symptoms revisited-is synovitis what we are assessing?

Classic signs of joint inflammation at physical examination of patients with rheumatoid arthritis (RA) are joint swelling and joint tenderness. In the hand, additional characteristic signs include a positive squeeze test, reduced grip strength, and incomplete fist closure. Typical symptoms that are presumed to be inflammatory are pain and morning stiffness. Traditionally, these signs and symptoms have been thought to reflect underlying synovitis, ie, inflammation of the intra-articular synovial lining. Over the past 15 years, however, we have recognised the involvement of other inflamed tissue

Insulin Enhances Nitroglycerin-Induced Hyperalgesia Through Insulin-Responsive Receptor Signalling Involving the Trigeminovascular System and Meningeal Mast Cells.

Migraine headache prevalence is lower in patients with diabetes, but the underlying mechanisms remains unclear. This study explored the influences of hyperglycemia, insulin and insulin receptor (IR)/insulin-like growth factor 1 receptor (IGF-1R)-related signalling on migraine-related mechanisms in in vivo and ex vivo rat models. In in vivo experiments, migraine-relevant responses were induced by systemic injection of nitroglycerin (NTG), whereas hyperglycemia was induced by streptozotocin (STZ). The effects of hyperglycemia, insulin and the dual IR/IGF-1R inhibitor BMS-754807 were investigated
