Extra-articular Synovial Chondromatosis of the Subacromial-Subdeltoid Bursa Mimicking Rice Body Synovitis: A Case Report.

Synovial chondromatosis can lead to proliferation of the synovium followed by the appearance of multiple cartilaginous nodules, most commonly in large joints. Extra-articular involvement of the subacromial-subdeltoid bursa could resemble an inflammatory or infectious process, especially if the chondromatosis is present in the form of rice bodies. A 37-year-old right-hand-dominant male presented with left shoulder pain for three months, with restricted shoulder motion and pain during overhead activities, with no past medical history of trauma or surgery. He presented to the orthopedic outpatien

Tissue-specific peripheral sensory remodeling in chronic low back pain: established and emerging nociceptive niches.

Chronic low back pain arises from heterogeneous peripheral and central mechanisms, yet the sensory biology of individual lumbar tissues is rarely considered within a unified framework. This Mini Review compares peripheral sensory remodeling and sensitization across four candidate nociceptive niches: the intervertebral disc, vertebral endplate, thoracolumbar fascia, and paraspinal muscle. Disc pathology is characterized by nociceptive ingrowth, endplate pathology by osteochondral sensory remodeling and afferent sensitization, and thoracolumbar fascia by inflammation-associated remodeling of a p

Identification of TREK channels in mouse intracardiac ganglion neurons.

The intracardiac nervous system is a complex neural network formed by a diffuse plexus, the so-called intracardiac ganglion (ICG). These ganglia locally innervate and control heart function, but little is known about how ICG neuronal activity is regulated. Although various ionic currents have been characterized in cardiac tissue, the expression and role of TWIK-related K+ (TREK) channels in ICG neurons remain unexplored. TREK channels, members of the two-pore domain potassium channel family, are essential regulators of neuronal membrane resting potential and excitability, with recognized neuro

SAPHO-like bone scintigraphy in diffuse idiopathic skeletal hyperostosis: a case report.

BACKGROUND: Bone scintigraphy is sensitive to increased osteoblastic activity but is not disease specific. Diffuse idiopathic skeletal hyperostosis (DISH) may therefore be mistaken for synovitis, acne, pustulosis, hyperostosis, and osteitis (SAPHO) syndrome when uptake involves the anterior chest wall and spine. CASE PRESENTATION: A 50-year-old woman presented with a 1-year history of painless left sternoclavicular prominence. She reported that her daughter had SAPHO syndrome with anterior chest wall pain and palmoplantar pustulosis. Whole-body 99mTc-MDP scintigraphy and single-photon emissio

CYBB regulates synovial fibroblast inflammatory activation and sEV-mediated synovial-cartilage communication in osteoarthritis.

OBJECTIVE: Although synovial inflammation is recognized as a factor in the progression of osteoarthritis (OA), the specific role of fibroblast-like synoviocytes (FLS) in cartilage degradation has yet to be fully elucidated. This study sought to identify key regulators that connect inflammatory FLS to chondrocyte dysfunction. METHODS: An integrated re-analysis of bulk and single-cell RNA sequencing data from OA synovial tissues was conducted to identify consistently upregulated genes. Functional assays, including migration and invasion tests, were performed on inflammatory FLS. FLS with silenc

Orofacial neuropathic pain and enhanced excitability of non-nociceptive-like and nociceptive-like trigeminal Aβ-afferent neurons in mice following infraorbital nerve chronic constrictive injury.

Orofacial neuropathic pain is a debilitating pain condition. It is poorly treated, partially due to an incomplete understanding of its underlying neurophysiological mechanisms. Orofacial neuropathic pain is often caused by injury or dysfunction of trigeminal nerve branches, including the infraorbital nerve (IoN). Clinically, a main symptom of orofacial neuropathic pain is mechanical allodynia, excruciating pain induced by gentle touch. While previous studies on neuropathic pain have mainly focused on the involvement of C- and Aδ-afferents, the potential contribution of Aβ-afferents to orofacia

ABCC1 Deficiency Mediates Susceptibility to Cd-Induced Hearing Loss via the NOX2/ROS Axis.

Cadmium (Cd), a prevalent environmental contaminant, is associated with auditory dysfunction. The cochlear blood-labyrinth barrier (BLB) protects the inner ear from toxic substances, a process in which the ATP-binding cassette subfamily C member 1 protein, ABCC1, plays a critical role in expelling toxins from its cells. To understand the specific contribution of ABCC1 to Cd-induced hearing damage, we employed both an in vivo model (ABCC1 knockout mice, Abcc1 -/- mice) and an in vitro endothelial model (ABCC1-deficient human umbilical vein endothelial cells, ABCC1 -/-HUVECs). These models were

Superior cervical sympathetic ganglion enlargement in idiopathic intracranial hypertension: An exploratory case-control study.

AimIdiopathic intracranial hypertension (IIH) may involve bidirectional interactions between elevated intracranial pressure (ICP) and sympathetic nervous system activity along an ICP-sympathetic axis. We hypothesized that superior cervical sympathetic ganglion (SCG) volume is increased in IIH and investigated whether SCG enlargement on MRI may serve as a potential imaging covariate of disease severity associated with cerebrospinal fluid opening pressure (OP).MethodsIn this retrospective case control study, we selected IIH patients with a brain MRI within six weeks prior to lumbar puncture, and

Therapeutic potential of zinc supplementation in rheumatoid arthritis: clinical evidence, molecular mechanisms, and formulation-specific effects.

Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disorder primarily affecting the synovial joints, characterized by persistent synovitis, progressive joint destruction, and systemic complications that contribute to significant morbidity. Despite advances in disease-modifying anti-rheumatic drugs and biologic therapies, many patients fail to achieve sustained disease control or experience treatment-related adverse effects, highlighting the need for safe adjunctive strategies. Zinc, an essential trace element with critical roles in immune regulation, antioxidant defense, and inflam

Repressed mTORC1 signaling and transient dendritic pruning support axonal regeneration in adult zebrafish.

Dendrite degeneration is an early pathological feature following axonal damage, yet its role during successful axonal regeneration remains poorly understood. Using sparse labeling of retinal ganglion cells in adult zebrafish, we show that axonal regeneration is accompanied by a transient phase of dendritic pruning following optic nerve crush. Dendritic pruning occurs during axon elongation and is reversed after brain reinnervation. Although insulin-mechanistic target of rapamycin (mTOR) signaling is rapidly upregulated immediately after injury, it is subsequently suppressed, coinciding with de

Identification and characterization of neoplastic cells in keratin-positive giant cell-rich tumors.

Keratin-positive giant cell-rich tumor (KPGCT) is a newly described bone and soft tissue tumor. The tumor is characterized by scattered keratin-positive cells and the presence of HMGA2::NCOR2 fusions. It is not known if the HMGA2::NCOR2 fusion is located in the keratin-positive cells, and little is known about how KPGCT develops. KPGCT shares some histologic features with tenosynovial giant cell tumor (TGCT), a soft tissue tumor with CSF1 rearrangements. Single-nuclei RNA sequencing and Xenium spatial transcriptomics were used to elucidate the most likely mechanisms driving KPGCT and compare K

The mechanosensitive channel TMEM63B contributes to noxious heat sensation in the dorsal root ganglia.

In mammals, heat sensation is primarily mediated by thermosensitive channels, particularly transient receptor potential (TRP) channels, in dorsal root ganglion (DRG) neurons. However, whether additional channels contribute to noxious heat detection remains unclear. Here, we investigated the role of the mechanosensitive cation channel TMEM63B in DRG neurons and unexpectedly discovered its involvement in noxious thermosensation rather than mechanosensation. TMEM63B was broadly expressed in DRG neurons, and its global knockout or DRG-specific conditional knockout markedly reduced heat sensitivity

Pstpip2-deficient mouse models in SAPHO syndrome research: Utility, mechanistic insights, and translational limitations.

Synovitis, acne, pustulosis, hyperostosis, and osteitis (SAPHO) syndrome is a rare autoinflammatory disorder characterized by combined cutaneous and osteoarticular involvement. Its etiology and pathogenesis remain incompletely understood, and an ideal animal model capable of fully recapitulating its phenotype is still lacking. Pstpip2-deficient mice exhibit sterile osteitis, inflammatory bone destruction, abnormal bone remodeling, and varying degrees of extraosseous inflammation, accompanied by myeloid immune abnormalities and IL-1β-associated inflammatory dysregulation. These mice have theref

Internal Carotid Artery Ligation: A Murine Model for Acute Retinal Ischemic Injury.

Accumulating evidence indicates that retinal ischemia constitutes a pivotal pathological trigger of visual impairment. The cerebral arterial circle (circle of Willis), formed by the vertebral and internal carotid arteries, provides cerebral blood perfusion. As a direct branch originating from the internal carotid artery (ICA), the ophthalmic artery supplies blood to the ocular globe. Based on the anatomical features of the ocular vasculature, a retinal ischemic injury model was successfully generated in C57BL/6 mice via selective left ICA ligation. Morphological and vascular analyses demonstra

Comparison of raw-signal, feature-based and hybrid representations for machine-learning classification of retinal diseases from pattern electroretinography.

Pattern electroretinography (PERG) is a non-invasive test of macular and retinal ganglion cell function, but its low amplitude and inter-subject variability limit its diagnostic use. This study compares three representations of the transient PERG waveform for the detection of six retinal and optic nerve conditions - Birdshot chorioretinopathy, cone-rod dystrophy, hereditary optic atrophy, macular dystrophy, retinitis pigmentosa and Stargardt disease - against healthy controls, using the publicly available PERG-IOBA dataset.
