Metabolic-Epigenetic Reprogramming of Fibroblast-Like Synoviocytes in Rheumatoid Arthritis: Perspectives for Traditional Chinese Medicine.

BACKGROUND: In rheumatoid arthritis (RA), persistent synovial inflammation and clinical relapse cannot be fully explained by immune activation alone, as fibroblast-like synoviocytes (FLS) can acquire stable pathogenic states that sustain synovitis and joint destruction. Metabolic reprogramming and epigenetic remodeling are functionally integrated in RA-FLS, thereby promoting hyperproliferation, invasiveness, apoptosis resistance, and inflammatory memory. METHODS AND RESULTS: In this review, we summarize the interplay between altered cellular metabolism and epigeneticmechanisms, including DNA

Microsphere System Coloading Type I Collagen and Dexamethasone Sodium Phosphate for Osteoarthritis Therapy.

Knee osteoarthritis (KOA) has no effective nonsurgical treatment. Intra-articular therapy constitutes an effective localized intervention; however, persistent lymphatic drainage and synovial vascular exchange lead to rapid drug elimination. Encapsulating bioactive agents into microsphere delivery platforms enables localized intra-articular administration, with their scaffold-based spherical architecture serving as pharmaceutical and cellular carriers or tissue engineering scaffolds, representing a highly promising therapeutic avenue for KOA. Herein, PLGA was employed to prepare microspheres an

Distribution of doublecortin in the developing retina of birds: A comparative analysis.

The avian retina presents an elegant and highly accessible model for investigating the spatial and temporal dynamics of neurogenesis within the central nervous system. In recent years, the microtubule cytoskeleton has emerged not merely as a structural scaffold, but as a dynamic regulator of the transition of a morphologically homogeneous pool of multipotent progenitor cells to postmitotic neurons. Doublecortin (DCX) is a microtubule-associated protein broadly recognized for its fundamental roles in neuronal migration, differentiation, and structural plasticity within the developing nervous sy

Retinal Ganglion Cell Quantification for Evaluating Neurodegeneration in Mouse Models of Multiple Sclerosis.

Quantification of retinal ganglion cell (RGC) loss provides a sensitive and reproducible measure of neurodegeneration in mouse models of neurological  disease and injury. In experimental autoimmune encephalomyelitis (EAE), a widely used mouse model of multiple sclerosis, inflammation of the optic nerve can lead to optic neuritis, axonal injury, and progressive RGC loss. Therefore, quantification of RGC survival provides a valuable readout of inflammation-mediated neurodegeneration and can be used to evaluate disease severity and neuroprotective interventions. Here, we describe a protocol for r

Towards a nonlinear "neuroimmune gate-model": beyond stellate ganglion block and vagus nerve stimulation.

We propose a three-level neuroimmune gate model to characterize the communication between the nervous system and the immune system. The first gate, Gate 1, is the peripheral neuroimmune gate. Here, immune cells, inflammatory mediators, sensory neurons, autonomic nervous system neurons, blood vessels, and specific tissue components interact. The second gate, Gate 2, is the spinal neuroimmune gate and includes spinal afferents, spinal neurons and circuits, preganglionic sympathetic neurons, and microglia. The third gate, Gate 3, comprises supraspinal structures, including the nucleus tractus sol

Whole-Mount Dissection of the Vestibular Complex and Quantification of Vestibular Ganglion Neurons in Mice.

The vestibular system is crucial for our sense of balance, for detecting head movements, and for regulating gaze and posture. There are five vestibular organs located inside the temporal bone, including two otolith organs (utricle and saccule), which detect linear movement, and three cristae (lateral/horizontal, anterior, and posterior), which detect rotational movement. Each organ contains a sensory epithelium with hair cells that detect head movements and supporting cells that provide structural support, release growth factors and cytokines, and phagocytose dying hair cells. The neurons that

Therapeutic Window of AAV-SIRT1 Gene Therapy in Mouse Models of Optic Nerve Degeneration.

PURPOSE: Optic neuropathies involve progressive loss of retinal ganglion cells (RGCs) and their axons, leading to irreversible vision deficits. While pretreatment with drugs or gene therapy that upregulate sirtuin 1 (SIRT1), an NAD⁺-dependent deacetylase, has shown neuroprotective effects, its effectiveness when administered after disease onset remains uncertain. We investigated whether adeno-associated virus (AAV)-mediated SIRT1 overexpression in RGCs can provide post-onset neuroprotection in preclinical models of optic neuropathy. METHODS: RGC degeneration was induced in C57BL/6 mice (8-10/

Targeting the NLRP3 inflammasome in rheumatoid arthritis fibroblast-like synoviocytes: pathogenic mechanisms, therapeutic strategies, and translational perspectives.

Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease characterized by persistent synovitis, pannus formation, cartilage degradation, and bone erosion. Although conventional synthetic, biologic, and targeted synthetic disease-modifying antirheumatic drugs have improved disease control, many patients fail to achieve sustained remission, lose responsiveness over time, or develop treatment-limiting safety concerns. Fibroblast-like synoviocytes (FLSs) are increasingly recognized as long-lived pathogenic stromal cells that sustain synovial inflammation, promote matrix degradation, supp

TYK2-Wnt5a Crosstalk in Psoriatic Arthritis: A Hypothesized Immunopharmacological Axis for Inflammation and Structural Remodelling.

INTRODUCTION: Psoriatic Arthritis (PsA) is a chronic immune-mediated inflammatory disease marked by synovitis, enthesitis, bone erosion, and pathological new bone formation, via the Interleukin-23/T helper cell-17 (IL-23/Th17) axis. Tyrosine Kinase 2 (TYK2) mediates downstream signaling of IL-23, IL-12, and type I Interferons (IFN-I), whereas Wnt5a regulates non-canonical Wnt signaling in inflammation and skeletal remodelling. This review proposes and examines TYK2- Wnt5a crosstalk in PsA as a testable mechanistic hypothesis rather than an established pathway. METHODS: A narrative literature
