Repressed mTORC1 signaling and transient dendritic pruning support axonal regeneration in adult zebrafish.

Dendrite degeneration is an early pathological feature following axonal damage, yet its role during successful axonal regeneration remains poorly understood. Using sparse labeling of retinal ganglion cells in adult zebrafish, we show that axonal regeneration is accompanied by a transient phase of dendritic pruning following optic nerve crush. Dendritic pruning occurs during axon elongation and is reversed after brain reinnervation. Although insulin-mechanistic target of rapamycin (mTOR) signaling is rapidly upregulated immediately after injury, it is subsequently suppressed, coinciding with de

The mechanosensitive channel TMEM63B contributes to noxious heat sensation in the dorsal root ganglia.

In mammals, heat sensation is primarily mediated by thermosensitive channels, particularly transient receptor potential (TRP) channels, in dorsal root ganglion (DRG) neurons. However, whether additional channels contribute to noxious heat detection remains unclear. Here, we investigated the role of the mechanosensitive cation channel TMEM63B in DRG neurons and unexpectedly discovered its involvement in noxious thermosensation rather than mechanosensation. TMEM63B was broadly expressed in DRG neurons, and its global knockout or DRG-specific conditional knockout markedly reduced heat sensitivity

Internal Carotid Artery Ligation: A Murine Model for Acute Retinal Ischemic Injury.

Accumulating evidence indicates that retinal ischemia constitutes a pivotal pathological trigger of visual impairment. The cerebral arterial circle (circle of Willis), formed by the vertebral and internal carotid arteries, provides cerebral blood perfusion. As a direct branch originating from the internal carotid artery (ICA), the ophthalmic artery supplies blood to the ocular globe. Based on the anatomical features of the ocular vasculature, a retinal ischemic injury model was successfully generated in C57BL/6 mice via selective left ICA ligation. Morphological and vascular analyses demonstra

Comparison of raw-signal, feature-based and hybrid representations for machine-learning classification of retinal diseases from pattern electroretinography.

Pattern electroretinography (PERG) is a non-invasive test of macular and retinal ganglion cell function, but its low amplitude and inter-subject variability limit its diagnostic use. This study compares three representations of the transient PERG waveform for the detection of six retinal and optic nerve conditions - Birdshot chorioretinopathy, cone-rod dystrophy, hereditary optic atrophy, macular dystrophy, retinitis pigmentosa and Stargardt disease - against healthy controls, using the publicly available PERG-IOBA dataset. A total of 672 recordings from 336 examinations of 304 participants we

Exome-wide association study reveals common and rare coding variants shaping chronic pain in 327,642 UK biobank participants.

BACKGROUND: Chronic pain is a major global health burden with a substantial but incompletely understood genetic basis. Although genome-wide association studies have identified multiple common-variant loci for pain-related traits, the contribution of rare coding variants to chronic pain susceptibility, biological mechanisms, and shared architecture across pain phenotypes remains unclear. METHODS: We performed an exome-wide association study across ten chronic pain phenotypes using whole-exome sequencing data from 327,642 European participants in the UK Biobank. Variant-level and gene-level ass

Ocular Hypertension and Glaucoma in Graves' Orbitopathy: A Single-Center Retrospective Cross-Sectional Study.

Background/Objectives: Elevated intraocular pressure (IOP) in Graves' orbitopathy (GO) is a recognized risk factor for glaucomatous optic neuropathy, but the extent to which it translates into measurable structural and functional damage remains unclear. This study investigated structural and functional correlations in patients with GO stratified by IOP and glaucoma status. Methods: In this retrospective, cross-sectional study, 165 adult patients with GO were stratified into three groups of 55 each: normal IOP, ocular hypertension, and clinically diagnosed glaucoma. All patients underwent clini

Stellate ganglion block for ventricular arrhythmia disease states: Emerging treatment options.

Autonomic modulation has emerged as an effective therapeutic target in the management of ventricular arrhythmias and electrical storm (e.g., refractory ventricular arrhythmias), in a background of growing evidence of the link between the autonomic nervous system and arrhythmias. Stellate ganglion blockade (SGB) is a minimally invasive procedure targeting the stellate ganglion with an anesthetic agent that has emerged as a temporizing treatment option for management of refractory and hemodynamically unstable ventricular arrhythmias and electrical storm (ES). SGB which can be performed with fluo

Robot-Assisted Gasless Trans-Hairline Excision of Submandibular Gland Tumors.

Benign submandibular gland tumors are usually resected through a transcervical incision, which provides direct exposure but leaves visible cervical scarring. Remote-access endoscopic techniques improve scar concealment but can be limited by two-dimensional visualization, assistant-dependent camera control, and rigid instrument collision in the narrow submandibular gland space. This article presents a standardized video protocol for robot-assisted submandibular gland excision via a gasless trans-hairline approach for benign tumors. The procedure begins with postauricular trans-hairline incision

Physiological Postnatal Neurogenesis, Development, and Maturation in the Regenerable Retina.

The retina enlarges postnatally by ∼440% in axolotls and ∼150% in humans, predicting a reduction in neuron densities by 81.5% and 60%, respectively, if neurogenesis is absent. The concurrent retinal remodeling is poorly characterized. We studied retinas of 2-15 cm axolotls. Results showed that hatchling (∼2 cm) retinas, particularly the peripheral (Peri) had 1-2 more soma rows in the ganglion cell (GCL) layer and outer (ONL) and inner nuclear (INL) layers. Cell density reduced in GCL (GCs, -49%) in juveniles (4-12 cm) but increased in INL (74%) and ONL (55.5%) in young juveniles (4-6 cm), demo

Retinal layer thickness as a potential marker for future cardiovascular events - a UK Biobank cohort study using optical coherence tomography.

AIMS: Cardiovascular disease (CVD) is the leading cause of death worldwide. For CVD prevention, effective screening is needed to identify high-risk individuals. Optical coherence tomography (OCT) provides a noninvasive, low-cost "quick look" into the brain and has recently shown thinning of certain retinal layers in patients with previous CVD. We explored if OCT-derived retinal layer thicknesses could predict future major adverse cardiovascular events (MACE: stroke/myocardial infarction/cardiovascular death). METHODS: In this retrospective cohort study, data of 51,293 participants from the UK

Sacral Neural Crest-Derived Extracellular Vesicles Promote Pelvic Ganglion Repair Through Anti-Pyroptotic Effects Involving ANXA1-FPR2 Signaling.

Pelvic autonomic nerve injury is a major cause of postoperative bowel dysfunction, yet effective strategies for restoring damaged pelvic ganglia remain lacking. Here, we demonstrate that extracellular vesicles derived from human pluripotent stem cell-derived sacral neural crest cells (Sacral NC-EVs) promote pelvic neuroregeneration by suppressing oxidative stress-induced pyroptosis. In a rat model of major pelvic ganglion (MPG) injury, local administration of Sacral NC-EVs improved gastrointestinal motility and partially restored rectal neural innervation and epithelial barrier integrity. Peripheral nerve injury alters metabolic function and neuronal excitability through the Lin28/let-7 pathway.

Sensory neurons require efficient metabolic processes to reliably transmit information including touch, temperature, and noxious stimuli from peripheral to central nervous systems. Peripheral nerve injury is known to disrupt metabolic function resulting in aberrant sensory nervous system excitation, but molecular pathways linking nerve injury to persistent alterations in metabolism and neuronal activity have been incompletely understood. We find that a conserved metabolic regulator, RNA binding protein Lin28, is elevated in the somas of dorsal root ganglion (DRG) nociceptive neurons following

Early-life high-fat diet accelerates the development of vagal satiation signaling and predisposes females to overconsumption.

Overconsumption of foods rich in fats, sugars, and calories is a major contributing factor to increased risk for cardiometabolic disease. Ingestive experience with these foods can begin early in children, yet there is limited understanding of the impact of early life nutrition on the development of vagal afferent neurons necessary for coordinating appetitive and satiating behaviors. To address this gap, mice reared on a chow diet (control) were compared to those reared on a high-fat diet (HFDEARLY). We demonstrate that the vagally mediated satiation response to cholecystokinin (CCK) does not m

Mitochondrial Genetic Background Shapes Baseline Retinal Function, Age-Related Bioenergetic Remodeling, and Microglial STING Phenotypes.

Mitochondria are central regulators of retinal bioenergetics and innate immune signaling, yet how mitochondrial genetic background and aging jointly influence immunometabolic responses in the retina remains poorly defined. Here, we investigated the effects of mitochondrial DNA (mtDNA) variation on retinal function and the combined effects of mtDNA variation and age on bioenergetic composition, mtDNA damage, and the microglial expression and intracellular localization of stimulator of interferon genes (STING) protein. Using mitochondrial-nuclear eXchange (MNX) mice harboring identical C57BL/6J

Aspiration of wrist ganglion cysts: a scoping review of aspiration alone versus aspiration with intralesional corticosteroid injection and implications for contemporary ultrasound-guided practice.

OBJECTIVES: To map the extent, nature and quality of the available comparative literature evaluating aspiration alone versus aspiration with intralesional corticosteroid injection for wrist ganglion cysts, and to identify evidence gaps relevant to contemporary ultrasound-guided practice. Given the anticipated scarcity of comparative studies, the review aimed to characterise the evidence base rather than determine treatment superiority. MATERIALS AND METHODS: Given the anticipated scarcity and heterogeneity of comparative studies, a scoping review was conducted in accordance with the PRISMA-Sc
