Increased Expression of Serca2b in the Adipose Tissue of a Cancer Cachexia Model.

BACKGROUND/AIM: Cancer cachexia is a complication that emerges in approximately 50-80% of patients with advanced cancer, characterized by symptoms such as lipoatrophy, skeletal muscle loss, metabolic abnormalities, and anorexia. While UCP1, a mitochondrial uncoupling protein, is implicated in lipolysis associated with cancer cachexia, the involvement of other thermogenic proteins remains unclear. In this exploratory study, we examined the expression of thermogenic genes in a mouse model of cancer cachexia. MATERIALS AND METHODS: Tumor-bearing mice were generated by injecting Colon-26 cells (C

Immune Cell Mitochondrial Phenotypes Are Largely Preserved in Mitochondrial Diseases and Do Not Reflect Disease Severity.

BACKGROUND AND OBJECTIVES: The aim of this study was to profile immune cell mitochondrial phenotypes in mitochondrial diseases (MitoD) and evaluate how these phenotypes relate to disease manifestations or biomarkers. METHODS: We profiled mitochondrial content and oxidative phosphorylation (OxPhos) enzymatic activities in isolated monocytes, lymphocytes, neutrophils, platelets, and mixed peripheral blood mononuclear cells (PBMCs) from 37 individuals with MitoD (m.3243A > G, n = 23; single, large-scale mitochondrial DNA (mtDNA) deletions, n = 14) and 68 healthy women and men from the Mitochondr

The Baraitser-Winter Cerebrofrontofacial Syndrome Recurrent R196H Variant in Cytoplasmic β-Actin Impairs Its Cellular Polymerization and Stability.

Variants in cytoskeletal actin encoding genes are associated with a broad spectrum of disorders, called non-muscle actinopathies. Among them, the Baraitser-Winter cerebrofrontofacial syndrome (BWCFF) displays the most severe symptoms, such as intellectual disability and epilepsy. We found that the BWCFF-associated R196H mutation results in reduced proliferation and migration of patient-derived fibroblast cells, and the latter is likely related to decreased fibronectin expression. The mutation causes a 50% drop in filamentous (F-) actin content, which is correlated with an approximately fourfol

Increased vein wall thickness in Behçet's disease: from intravascular mechanical stress to vascular remodelling and venous thrombosis.

Behçet’s disease (BD) is a chronic, multisystem inflammatory disorder characterized by recurrent oral ulcers, genital ulcers, and uveitis, together with various manifestations. Vascular involvement occurs in 25–30% of BD patients. Recent studies on BD have remarkably claimed that even Behçet patients without vascular problems exhibit increased vein wall thickness (VWT). Researchers suggested that this increase in VWT may have a diagnostic value for BD. However, the mechanisms responsible for this process and its clinical significance remain unclear. In this Perspective article, we describe how

Failure of Allogeneic Transplant to Correct Sialidosis Despite Early Diagnosis and Full Donor Engraftment of Non-Carrier Leucocytes.

Sialidosis, also known as Mucolipidosis Type I, is a rare condition caused by defects in the NEU1 gene which causes the accumulation of sialylated peptides, oligosaccharides, and glycoproteins leading to neurological decline. Haematopoetic stem cell transplantation has been performed in the symptomatic phase twice in the literature but has failed to prevent deterioration. We report on a case where a 4-year-old child was diagnosed with pre-symptomatic sialidosis due to investigation following the incidental detection of a cherry-red spot prior to the onset of neurological symptoms. We performed

Decoding collagen cues: the interplay of integrins and discoidin domain receptors in health and disease.

The extracellular matrix (ECM) provides critical biochemical and biophysical cues that regulate cell behavior in health and disease. Collagens dominate in abundance and structural importance, shaping tissue-specific ECM signatures that guide cellular behavior. Two major and distinct transmembrane receptor families, integrins and discoidin domain receptors (DDRs), serve as primary sensors for collagens, yet they employ fundamentally distinct binding mechanisms and signaling kinetics. While both can activate shared downstream pathways, their functional interplay remains complex and context-depen

Ultramicronized palmitoylethanolamide restores astrocyte-neuron metabolic coupling and Klotho/FGF21 signaling in a triple-transgenic mouse model of Alzheimer's disease.

Alzheimer's disease (AD), a multifactorial neurodegenerative disorder, is characterized by metabolic deficiency, neuroinflammation, and synaptic impairment. Astrocyte-neuron metabolic coupling regulates cerebral energy homeostasis through key metabolites such as lactate, glutamate, and taurine. We investigated the therapeutic potential of ultramicronized-palmitoylethanolamide (um-PEA) in restoring the homeostasis of these metabolites in the triple transgenic (3 ×Tg-AD) mouse model of AD. Using in vivo magnetic resonance imaging and spectroscopy (MRI/MRS) combined with Western blot, we evaluate

Bidirectional CRISPR screens decode a GLIS3-dependent fibrotic cell circuit.

The stromal cell compartment plays a central part in the maintenance of tissue homeostasis by coordinating with the immune system throughout inception, amplification and resolution of inflammation1. Chronic inflammation can impede the phased regulation of tissue restitution, resulting in the scarring complication of fibrosis. In inflammatory bowel disease, stromal fibroblasts have been implicated in treatment-refractory disease and fibrosis2,3; however, their mechanisms of activation have remained undefined. Through integrative single-cell and spatial profiling of intestinal tissues from patie

A novel potential toxic target of PBDEs in cetaceans: Impaired wound healing demonstrated in Indo-Pacific finless porpoise fibroblasts.

Cetaceans, particularly those inhabiting coastal areas, face compounding threats from diverse anthropogenic activities, including vessel strikes, entanglement, and pollution. However, these stressors have predominantly been assessed in isolation, overlooking the potential synergistic effects arising from their frequent co-occurrence. Polybrominated diphenyl ethers (PBDEs), a class of toxic flame retardants ubiquitous in marine environments, bioaccumulate in cetaceans posing significant health risks. This study provides the first integrated mechanistic insight into how PBDE exposure not only di

Automating Vascular Biology: An End-to-End Automated Workflow for High-Throughput Blood Vessel-on-a-Chip Production and Multi-Site Validation.

There is a growing demand for automated organ-on-a-chip platforms that are compatible with off-the-shelf robotic liquid-handling systems and plate readers to improve reproducibility and scalable analysis. In this work, we present an end-to-end automated method for fabricating tubular blood vessel models at scale using a custom 384-well open-top platform (AngioPlate384), designed to support integration with liquid-handling systems and large-scale analysis. Our approach enables the generation of over 100 perfusable blood vessels fully embedded in hydrogel and supported by stromal cells (fibrobla

STL1267 inhibits myofibroblast differentiation in a TGFβ1-driven human lung fibroblast model.

Pulmonary fibrosis is a progressive interstitial lung disease characterized by excessive fibroblast-to-myofibroblast transition (FMT) and extracellular matrix (ECM) deposition, largely driven by transforming growth factor-beta 1 (TGFβ1). Existing therapies offer limited efficacy, particularly in advanced disease. Circadian rhythms have recently emerged as key modulators of lung inflammation and fibrosis. In this study, we developed an in vitro model of chronic fibrotic signaling using adenovirus-mediated TGFβ1 overexpression (Ad-TGFβ1) or human recombinant protein TGFβ1 in primary normal human

Genomic landscape of metastatic breast cancers in young adults: a liquid biopsy analysis of women aged 20-40 years.

INTRODUCTION: Breast cancer in young adults (YA) aged 20-40 years has distinct clinical and biological traits compared with older patients. This study evaluated the genomic landscape of metastatic breast cancers (MBC) among YA. METHODS: Patients with MBC enrolled in the STING molecular profile platform (NCT04932525) between 2021 and May 2023 were included. Clinical and genomic features were analyzed by age (≤40 vs > 40 years). Tumor profiling used the FoundationOne Liquid CDx assay (324 genes) at baseline or later in the disease course. Variant frequencies were compared across age groups. RE

Oncostatin M promotes versican expression in cardiac fibroblasts to facilitate extracellular matrix remodeling.

Myocardial injury is accompanied by an acute inflammatory response. Our previous work identified Oncostatin M as a key cytokine promoting heart regeneration through stimulating cardiomyocyte proliferation, while its effects on fibroblasts cannot be ignored. Cytokines are generally recognized to activate profibrotic gene programs in cardiac fibroblasts, a process traditionally regarded as detrimental to functional repair. Here, we showed that OSM enhanced cardiac fibroblasts to express versican, a pro-regenerative ECM component in the heart, through activating gp130/JAK/STAT3 signaling pathway.

Integrated human and mouse single-cell profiling reveals immune-stromal niche driving silicosis.

Silicosis is an inflammation-driven pulmonary fibrosis caused by occupational inhalation of silica particles. Macrophages are crucial in silicosis pathology, yet their interaction with stromal cells in orchestrating fibrosis progression remains poorly understood. Single-cell RNA sequencing (scRNAseq) of whole-lung lavages from silicosis patients identified the expansion of an intermediate CCL2-hi monocyte-like macrophage (MLM) cluster that further differentiated into inflammatory IL1B-hi and pre-fibrotic SPP1-hi (osteopontin) subsets. SPP1-hi MLMs showed enrichment for tissue remodelling (SPP1

Mitochondrial proteomics reveals reductive metabolism dependent on glutamine in fibroblasts of idiopathic pulmonary fibrosis under hypoxia.

Pulmonary fibrosis, particularly idiopathic pulmonary fibrosis (IPF), is a chronic disease characterized not only by a transcriptionally active signature associated with hypoxia but also by feedback loops that may underlie disease progression. IPF fibroblasts are known to contribute to disease by the pronounced differentiation of fibroblasts into myofibroblasts, which results in the accumulation of excessive extracellular matrix, creating a hypoxic microenvironment that supports the characteristic phenotype observed in the fibroblasts of these patients. Although several changes have been linke

Targeting osteopontin with gold nanoparticles for enhanced molecular imaging in Kawasaki disease: in-depth mechanistic study of STAT3 signaling in Col1 regulation.

BACKGROUND: Kawasaki disease (KD) is a primary cause of acquired heart disease in children, often leading to significant complications like coronary artery lesions and vasculitis. Despite the administration of standard treatments, 10–20% of patients remain unresponsive, emphasizing the necessity for the development of improved diagnostic and treatment modalities. Given the limitations of conventional imaging techniques, this study investigates the potential of molecular imaging using targeted nanoparticles. Here, the role of osteopontin (OPN) in macrophage-dependent vascular inflammation and c

TGFβ/Smad2/3-Mediated Crosstalk Between Vocal Fold Fibroblasts and Myoblasts In Vitro.

OBJECTIVES: Traditionally, disorders of the vocal fold (VF) mucosa and underlying musculature have been regarded as mutually exclusive entities. However, emerging evidence from other organ systems suggests mucosal and muscle compartments engage in reciprocal interactions with functional consequences. We hypothesized that similar crosstalk exists in the VF, whereby fibrotic mucosa influences adjacent muscle. To model this process, we stimulated human VF fibroblasts (HVOX) with TGF-β1, a central mediator of fibrosis, and examined the effects on rat VF myoblasts (rVF-Mbs), as well as reciprocal i

Targeting activated kidney fibroblasts via ferroptosis: a potential antifibrotic strategy.

Kidney fibrosis is characterized by excessive deposition of extracellular matrix, which is ultimately disrupting normal renal architecture. Despite its clinical relevance, no targeted antifibrotic therapies are currently available. Myofibroblasts, primarily derived from pericytes and resident fibroblasts, are key effectors of fibrosis due to their high extracellular matrix production. Here, we tested the hypothesis that ferroptosis induction would enable the targeted elimination of activated kidney fibroblasts. We found that kidney fibroblasts exhibit marked sensitivity to ferroptotic cell dea

Cellular and molecular signals of cardiac wound healing after myocardial infarction.

Wound healing after myocardial infarction (MI) is a dynamic and multifaceted process that links the molecular alterations induced by or in response to prolonged ischemia with structural and physiological changes to the damaged myocardium. Changes, at the tissue level, are driven by a complex intersection of cellular and molecular mechanisms that operate along a classic wound healing paradigm as an attempt to repair the damaged myocardium and restore cardiac physiology. Maladaptive healing prevents a return to the original homeostasis, rather yielding a myocardium reset to a new homeostatic sta

A partial differential equation model of a novel treatment for chronic wound biofilm infections.

BACKGROUND: Chronic wounds, such as pressure ulcers, venous leg ulcers, and diabetic foot ulcers, present a significant healthcare challenge due to their prolonged healing times and association with biofilm infections. This study introduces a mathematical model to investigate wound healing dynamics during the proliferative stage, focusing on chronic wound conditions characterised by poor vascularisation, clotting deficiencies, cellular senescence, and bacterial biofilms. METHODS: The model examines the interactions between fibroblasts, keratinocytes, granulation tissue, nutrients, and signall
