Sublytic C5b-9 induces S100A8/A9 expression via SMYD4-dependent SOX7-K159 methylation in the glomerular mesangial cells of Thy-1 nephritis rats.

Rat Thy-1 nephritis (Thy-1N) is a model for human mesangioproliferative glomerulonephritis (MsPGN). Although sublytic C5b-9 has been reported to affect pro-inflammatory cytokine expression in glomerular mesangial cells (GMCs), the induction of other pro-inflammatory mediators remains unclear. In this study, we demonstrated that SET and MYND domain-containing protein 4 (SMYD4), SRY-related HMG-box gene 7 (SOX7), S100 calcium-binding protein A8 (S100A8), and S100A9 were overexpressed in the renal tissues of Thy-1N rats and in the GMCs stimulated by sublytic C5b-9. Mechanistically, SOX7 triggered

Transcriptional factor KLF9 in pulmonary fibrosis: from molecular mechanisms to precision intervention-a narrative review.

BACKGROUND AND OBJECTIVE: Pulmonary fibrosis (PF) is a devastating and progressive respiratory disease characterized by extracellular matrix (ECM) remodelling and functional decline, yet current therapeutic options remain limited. Recent studies have highlighted the critical, albeit complex, role of Krüppel-like factor 9 (KLF9) in the pathogenesis of PF. This review aims to comprehensively integrate the existing fragmented mechanistic evidence, delineate the differential regulatory networks of KLF9, and outline forward-looking strategies to promote the transition of KLF9 from a mechanistic can

Cancer-Associated Fibroblast Exosomes in Breast Cancer: Mechanisms of Tumor-Stroma Crosstalk and Clinical Implications.

Breast cancer progression is shaped by a continuous exchange of signals between tumor cells and the surrounding stroma, in which cancer-associated fibroblasts (CAFs) are among the most influential partners. This review concentrates on CAF-derived exosomes as the principal vehicles of that exchange, and on how their cargo and mode of delivery differ across functionally distinct CAF subtypes. Exosomes are nanoscale vesicles that carry microRNAs (miRNAs), long noncoding RNAs, proteins, and metabolites between cells, and CAF-derived vesicles have been linked to epithelial-mesenchymal transition (E

LGR5-positive fibroblasts orchestrate skin mechanoadaptation through JAK1 modulation.

Mechanical adaptation is a fundamental yet poorly understood process in stromal tissue biology. Here, we identify a population of LGR5+ mechanically induced regulatory fibroblasts (LGR5+ MIRFs) in mouse and human dermis, whose induction requires a YAP/AP-1-dependent transcriptional program. LGR5+ MIRFs channel mechanical input toward adaptive tissue remodeling by coordinating extracellular matrix reorganization while restraining inflammation. Their ablation or fibroblast-specific deletion of Lgr5 converts adaptive stretching into pathological fibro-inflammation, while dermal Lgr5 overexpressio

Atrial fibroblast metabolic reprogramming in atrial fibrillation: a candidate mechanism within multicellular atrial cardiomyopathy.

Atrial fibrillation (AF) commonly coexists with obesity, diabetes, and other cardiometabolic disorders. However, how systemic metabolic stress creates an arrhythmogenic atrial substrate remains unclear. This narrative review evaluates atrial fibroblast metabolic reprogramming as a testable mechanism within multicellular atrial cardiomyopathy, not as an established causal pathway. We organize the evidence by source: human atrial fibroblasts, human atrial tissue, atrial animal models, non-atrial cardiac fibroblasts, extracardiac fibroblasts, and reduced experimental systems. Studies of non-atria

Sanguinarine ameliorates rheumatoid arthritis by activating the Hippo-YAP/TAZ pathway to induce apoptosis and suppress inflammation in fibroblast-like synoviocytes.

INTRODUCTION: Rheumatoid arthritis (RA) is a chronic autoimmune disease with progressive joint damage, functional disability and systemic comorbidity. Available therapies are not satisfactory, including low rates of remission and about 30% of patients with poor response to first-line treatment. Thus, more effective therapeutic strategies are urgently needed. Sanguinarine, a benzophenanthridine alkaloid primarily isolated from Sanguinaria canadensis and other members of the Papaveraceae family, has been documented to possess notable anti-bacterial, anti-inflammatory, and anti-neoplastic propert

Integrated transcriptomic and experimental analyses identify SRPK1/SRSF1 as a therapeutically relevant splicing regulatory Axis in pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is characterized by progressive extracellular matrix deposition and irreversible distortion of lung architecture. We hypothesized that aberrant kinase-dependent splicing may contribute to IPF pathogenesis and offer an avenue for therapeutic intervention. We therefore analyzed multiple independent transcriptomic cohorts and found that SRPK1, compared with other SRPK family members, was most consistently elevated in fibrotic lung tissue. Higher SRPK1 expression was linked to poorer lung function and was associated with mortality after adjustment for age and se

Sporadic Multiple Meningiomas Presenting With Drug-Resistant Seizures in a Young Adult: A Case Report.

Meningiomas represent the most common benign primary intracranial tumors. However, multiple meningiomas (MMs) are relatively rare and can be challenging to manage, often requiring repeated surgical interventions and long-term follow-up. Here, we present the case of a 28-year-old woman who was operated on in 2019 for a right parasagittal MM, WHO grade 1. Her initial presentation was characterized by seizures and left-sided hemiplegia. Seven years later, she presented with recurrent seizures that were resistant to medical treatment. Neurological examination revealed residual left-sided spastic p

Pleuroparenchymal fibroelastosis in a monogenic mitochondrial DNA depletion syndrome.

BACKGROUND: Pleuroparenchymal fibroelastosis (PPFE) is a rare, most often idiopathic fibrotic lung disease with poor prognosis, mainly affecting adults. Biallelic pathogenic variants in the deoxyguanosine kinase gene (DGUOK) cause a mitochondrial DNA (mtDNA) depletion syndrome, predominantly affecting infants with severe hepatic and neurological disease. Neither detailed pulmonary manifestations nor a pulmonary-dominant phenotype has been reported. METHODS: We describe nine patients with PPFE and DGUOK-associated mitochondriopathy. Clinical, radiological, histopathological, and genetic data w

Adipocytokines as Pathogenic Mediators and Therapeutic Targets in Metabolic Syndrome-Related Cardiocerebrovascular Events.

Metabolic syndrome (MS), characterized by central obesity, hyperglycemia, hypertension, and dyslipidemia, substantially increases the risk of cardiovascular and cerebrovascular events. Adipose tissue is an active endocrine organ, and dysregulated adipokine secretion may contribute to the link between MS and vascular injury. In this narrative review, we synthesize evidence on adipokines implicated in MS-related cardiocerebrovascular disease and discuss their therapeutic potential. Adiponectin, fibroblast growth factor 21 (FGF21), and omentin are generally associated with improved insulin sensit

Mathematical modeling of in vitro replication dynamics for multiple highly pathogenic avian influenza clade 2.3.4.4 viruses in chicken and duck cells.

The introduction, spread, and subsequent detection of highly pathogenic avian influenza viruses (HPAIVs) in poultry are influenced by the virus replication fitness, transmission fitness, and virulence. Quantifying these fitness parameters is important for implementing surveillance and control measures for poultry. This study employs an avian in vitro model using primary chicken embryo fibroblast and duck embryo fibroblast to identify cellular correlates of fitness for a reference panel of eight dominant, highly pathogenic avian influenza clade 2.3.4.4 virus genotypes (H5N1, H5N8, and H5N6) iso

Human Epididymis Protein 4 in Cardiovascular Disease: A Fibrosis-Driven Mediator of Tissue Remodeling and Inter-Organ Communication.

Human epididymis protein 4 (HE4), originally identified as a tumor biomarker, has emerged as a regulator of tissue remodeling, with roles that extend beyond oncology. Recent findings suggest that HE4 is involved in extracellular matrix turnover, immune system modulation, and maintenance of epithelial balance, indicating its complex role in fibrosis and tissue remodeling. In cardiovascular diseases, the circulating HE4 level is consistently associated with disease severity, myocardial fibrosis, and adverse clinical outcomes in conditions such as heart failure, aortic stenosis, and ischemic hear

Fibroblast Activation Protein Inhibitor Positron Emission Tomography Imaging in Atrial Fibrillation: Current Evidence and Future Perspectives.

Fibroblast activation protein inhibitor (FAPI) positron emission tomography/computed tomography (PET/CT) has emerged as a promising molecular imaging modality for visualizing fibroblast activation and extracellular matrix remodeling. In addition to its established role in oncology imaging, FAPI PET/CT is gaining recognition in the evaluation of cardiovascular disease. Emerging evidence suggests that this technique may provide molecular insights into atrial fibroblast activation and the development of a fibrotic substrate in atrial fibrillation. Meanwhile, related findings from other arrhythmia

Therapeutic landscape evolution for steatotic liver disease: from lifestyle interventions to precision pharmacotherapy.

Steatotic liver disease (SLD) is a growing global epidemic, with metabolic dysfunction-associated steatotic liver disease now the most common chronic liver disease worldwide and closely linked to adverse cardiovascular outcomes. Lifestyle modification remains the cornerstone of management, with the Mediterranean diet as the preferred dietary intervention and emerging evidence supporting additional dietary and exercise strategies. Pharmacotherapy has advanced rapidly in recent years, with two U.S. Food and Drug Administration-approved options, resmetirom and semaglutide, for noncirrhotic metabo

O-GlcNAcylation in diabetic cardiac remodeling and a proposed endothelial-macrophage-fibroblast network.

Diabetic cardiac remodeling involves microvascular dysfunction, inflammatory myeloid responses and fibroblast activation. O-linked beta-N-acetylglucosamine (O-GlcNAc) modification connects nutrient metabolism to protein function within each compartment, but its role in coordinating communication between them remains a hypothesis. This Mini Review evaluates that hypothesis against the available evidence. Diabetic models support causal effects of endothelial O-GlcNAc perturbation on coronary dysfunction, and an arterial endothelial CaMKIIalpha-small-extracellular-vesicle circuit provides evidenc

Human trilayer engineered blood vessel reveals influence of fibroblasts on disease progression in model of Hutchinson-Gilford progeria.

In vitro models of vascular disease have focused primarily on the pathophysiology of smooth muscle cell (SMC) or endothelial cell (EC). Adventitial fibroblasts can contribute to disease progression, but their specific influence is not well understood in different disease contexts. To elucidate fibroblast's impact on vascular pathology, we developed trilayer tissue-engineered blood vessels (TEBVs) with SMCs, ECs, and fibroblasts. We modeled atherosclerosis in the accelerated aging disease, Hutchinson-Gilford progeria syndrome (HGPS). HGPS fibroblasts substantially elevated several features of t

Integrated single-cell and bulk-tissue transcriptomic analysis reveals an epithelial-stromal inflammatory microenvironment in biliary atresia.

BACKGROUND: Biliary atresia is a severe fibroinflammatory cholangiopathy of infancy. Its pathological process involves biliary epithelial injury, immune activation, and intrahepatic stromal remodeling, but the cellular interactions linking these processes remain incompletely defined. Using public single-cell and bulk-tissue transcriptomic cohorts, this study systematically characterized disease-associated alterations across liver cell populations in biliary atresia and further evaluated their consistency in independent cohorts. METHODS: Public single-cell liver data (GSE176189) and three inde
