A series of novel C-28 amino acid/amide conjugates and oligo(ethylene glycol)-modified C-2, C-3 pyrazole-fused betulinic acid derivatives were designed, synthesized, and evaluated for their inhibitory effects on RANKL-induced osteoclastogenesis. Among these, compound 18, bearing a C-28 oligo(ethylene glycol) amino amide ester linkage, exhibited the most potent inhibitory activity. It demonstrated an IC50 of 7.96 nM against RANKL-induced osteoclastogenesis in RAW264.7 cells, representing a 10-fold increase in potency compared to the XJ13 (IC50 = 0.08 μM) and achieved >50 % inhibition at 0.01 μM

OBJECTIVE: To evaluate the relationship between dietary patterns and bone health in the elderly, and to guide the elderly to establish a reasonable dietary pattern and improve bone health.
METHODS: This cross-sectional study included 544 elderly individuals aged 60 and above in rural areas of Qingdao. Dietary patterns were analyzed using factor analysis. A logistic regression model was employed to assess the relationship between dietary patterns and osteoporosis. The correlation between body composition and T-score was analyzed using Spearman's rank correlation method in the R software.

Romosozumab, a sclerostin inhibitor, exerts a dual effect of increasing bone formation while decreasing bone resorption to rapidly increase bone mineral density (BMD) and reduce fracture risk in postmenopausal women. Iterative Convolution OptimizatioN (ICON) allows accurate calculation of deconvolved cortical thickness from high-resolution quantitative computed tomography (HR-QCT) scanning. Pregnancy- and lactation-related osteoporosis (PLO) is a rare condition characterized by reduced bone density and increased fracture risk during late pregnancy or early postpartum. Because of its rarity, no well-established treatment exists for PLO. Here, we report a case demonstrating significant improvements in bone density and the absence of new fractures after 12 mo of romosozumab (ROMO) treatment following weaning. This report suggests that ROMO could effectively increase bone density and reduce fracture risk in patients with PLO.
