Insulin-like growth factor 2 mRNA-binding Protein 2 regulates PINK1 expression through m6A pathway to promote mitophagy in BMSCs alleviating postmenopausal osteoporosis.

The senescence and altered differentiation potential of bone marrow mesenchymal stem cells (BMSCs) contribute to the pathogenesis of postmenopausal osteoporosis (PMOP). Insulin-like growth factor 2 mRNA-binding protein 2 (IMP2) has been demonstrated to regulate BMSCs. However, its specific mechanistic actions remain unclear, particularly due to the lack of concrete evidence within the ovariectomy (OVX) in vivo microenvironment. In this study, we utilized Cre-LoxP technology to achieve BMSC-specific IMP2 knockout. This approach conclusively demonstrated in vivo that IMP2 deficiency induces BMSC

Long-term cannabidiol treatment did not restore bone microstructural defects in skeletally mature ovariectomized Sprague-Dawley rats.

Cannabidiol (CBD) effects on bone metabolism in postmenopausal osteoporosis remain unclear. While endocannabinoids and phytocannabinoids bind to receptors in bone cells, direct evidence of CBD's bone-protective effects is lacking. We evaluated the effects of CBD on bone metabolism in ovariectomized (OVX) rat model of estrogen deficiency. Twelve-week study with treatment initiated 2 wk after the surgery was conducted. Five experimental groups were established: sham-operated with vehicle (SHM/VEH), sham with CBD (SHM/CBD5), OVX with vehicle (OVX/VEH), OVX with 17β-estradiol (OVX/E2), and OVX wit

The Role of Glucose-Dependent Insulinotropic Polypeptide (GIP) in Bone Metabolism.

Glucose-dependent insulinotropic polypeptide (GIP) was the first incretin hormone identified, best known for promoting glucose-stimulated insulin secretion. Increasing evidence has expanded its physiological relevance beyond glucose metabolism, revealing a significant role for GIP in the gut-bone axis. In vitro studies demonstrate that GIP inhibits osteoclast differentiation and activity while promoting osteoblastic bone formation. Findings from genetic animal models and human variant analyses further support the essential role of endogenous GIP signaling in maintaining bone mass and quality.

γ-Mangostin attenuates osteoclastogenesis and bone resorption by suppressing the PI3K/AKT/NF-κB pathway.

BACKGROUND: Postmenopausal osteoporosis (PMOP), driven predominantly by estrogen deficiency-induced hyperactivation of osteoclasts, represents a critical public health burden. The pursuit of naturally sourced inhibitors of osteoclast function with minimized adverse effects remains a pivotal research endeavor. γ-Mangostin (γ-Mag), a natural xanthone derived from the pericarp of mangosteen, possesses broad anti-inflammatory and anti-tumor activities. Nevertheless, its influence on bone metabolic homeostasis, particularly osteoclast biology, remains entirely unexplored. This study aims to elucida

Alendronic acid modified PLGA drug delivery system loaded with 17β-Estradiol and vitamin D3 has anti-osteoporotic effect.

Postmenopausal osteoporosis caused by estrogen deficiency often requires hormone replacement therapy (HRT), but its systemic side effects limit clinical application. Here, we developed a bone-targeted Poly (lactic-co-glycolic acid) (PLGA) nanocarrier modified with Alendronic acid (ADA) to co-deliver 17β-Estradiol (E2) and Vitamin D3 (VitD3), aiming to enhance efficacy and safety. The ADA-functionalized nanoparticles (E2+VD@PLGAIR780ADA) showed high drug loading (7.2 wt% for E2 and 2.3 wt% for VitD3), sustained release (>90 % over 48 h). In ovariectomized (OVX) mice, targeted delivery significa

Valproic Acid Inhibits Ferroptosis and Improves Bone Integration in OVX Rats Through the AMPK/SIRT1 Pathway.

The prevalence of postmenopausal osteoporosis (PMOP) has been steadily increasing. Ferroptosis has been recognized as a critical factor influencing the bone-forming ability of bone marrow mesenchymal stem cells (BMSCs). Valproic acid (VPA), an HDAC inhibitor, has been suggested to play a role in regulating osteoporosis development; however, its underlying mechanism remains unclear. This study aims to explore the impact of VPA on ferroptosis, a process that is triggered by Erastin, and to assess its implications for postmenopausal osteoporosis (PMOP). We evaluated the effects of valproate sodiu

Estradiol regulates osteoclast sialylation via ST3Gal1 in postmenopausal osteoporosis.

Estrogen deficiency after menopause accelerates bone loss by stimulating osteoclast formation and activity, but the molecular pathways that link estrogen signaling to osteoclast regulation remain incompletely defined. Here, we identify the sialyltransferase ST3GAL-I as a key mediator of RANKL-induced osteoclastogenesis. RANKL activates c-FOS to drive ST3GAL1 transcription, whereas estrogen-bound ERα competes with TRAF6 and suppresses this c-FOS-dependent induction. In a clinical cohort of pre-menopausal and post-menopausal women with or without osteoporosis, serum total and α-2,3-linked sialic
