INTRODUCTION: Although the etiology of osteoporosis is different between men and women, the underlying pathophysiological mechanism is similar, namely an absolute or relative increase in bone resorption, leading to progressive bone loss. Transforming growth factor (TGF)-beta1 is a growth factor in human bone, which is produced by osteoblasts, and which has various effects on osteoclasts and osteoblasts. The aim of our study was to determine serum TGF-beta1 levels in male patients with idiopathic osteoporosis.
METHODS: Twenty five males with idiopathic osteoporosis and 25 age-matched controls

BACKGROUND: Primary osteoporosis can be caused by pathogenic variants in multiple genes. Recently, rare heterozygous variants in SGMS2, encoding SGMS2, have been identified to cause early-onset osteoporosis or more severe skeletal dysplasia. The incidence of pathogenic SGMS2 variants and their consequent clinical features, however, remain limited.
METHODS: This study aimed to identify the prevalence and nature of SGMS2 variants in Finnish patients with genetically undiagnosed idiopathic early-onset osteoporosis. All eleven exons and exon-intron boundaries of SGMS2 were sequenced.
RESULTS: In

The melatonin receptor 1 A (MTNR1A), a highly conserved G protein-coupled receptor (GPCR), mediates crucial physiological functions. Its structure features an N-terminus responsible for melatonin binding and a C-terminus that initiates downstream signaling pathways to modulate target gene expression. Given MTNR1A's ability to form homodimers or heterodimers with other GPCRs, its expression levels are critical for the precise control of cellular signaling. This review article provides a comprehensive update on MTNR1A, highlighting recent developments concerning its expression distribution, gene

BACKGROUND: European cities are placing a larger emphasis on urban data consolidation and analysis for optimizing public transport in response to changing urban mobility dynamics. Despite the existing efforts, traffic data analysis often disregards vital situational context, including large-scale events, weather factors, traffic generation poles, social distancing norms, or traffic interdictions. Some of these sources of context data are still private, dispersed, or unavailable for the purpose of planning or managing urban mobility. Addressing the above observation, the Lisbon city Council has

BACKGROUND: Brain tumors present a challenge in oncology, necessitating comprehensive treatment strategies. Despite treatment, recurrence rates, especially in malignant tumors, remain high. Novel therapies have emerged, but treatment-related effects such as pseudo-progression (PsP) and radiation necrosis (RN) complicate diagnosis. Imaging techniques such as perfusion imaging and susceptibility-weighted imaging (SWI) show promise in distinguishing tumor progression from treatment effects. This retrospective study evaluated the effectiveness of these modalities in detecting brain tumours, their

Osteoporosis (OP), a systemic bone disease, is characterized by degeneration of bone microstructure and susceptibility to fracture. Teriparatide (TPD) is an active fragment of human endogenous parathyroid hormone which has been revealed to promote osteogenesis of mesenchymal stem cells (hMSCs) to alleviate osteoporosis. Currently, the underlying cellular and molecular mechanisms of TPD in treating OP were not fully understood. This study aimed to investigate the roles of non-coding RNA-regulated osteogenic differentiation of hMSCs under TPD treatments. Circular RNA FNDC3B was significantly dow

Premenopausal osteoporosis is often overlooked because fragility fractures and low bone mass are uncommon in premenopausal women. The definition and diagnostic criteria for premenopausal osteoporosis are less well defined than for postmenopausal women. Diagnostic procedures should be initiated in premenopausal women with existing fragility fractures or diseases and drug therapies that cause bone loss. Recent studies have shown that lifestyle and dietary habits influence bone mass in the premenopausal phase. Bone mass can be improved by an adequate intake of calcium and vitamin D in combination

The development of osteoporosis and fractures in young premenopausal women is infrequent and is usually associated with secondary causes of osteoporosis. Therefore, it is recommendable to perform a clinical evaluation and a thorough laboratory study searching for possible causes of bone loss in these patients. Treatment depends on the cause of osteoporosis and the associated processes as well as the future gestational desire of the patient, all of which should be taken into account when evaluating the most adequate diagnostic and the therapeutic approach in these patients.

UNLABELLED: In premenopausal IOP, BMD gains on teriparatide-denosumab were maintained on bisphosphonate over 1 year. In 19 followed after bisphosphonate, there was a significant bone loss over 6M, leading to retreatment in some. BMD remained significantly above pretreatment baseline. In premenopausal IOP, the optimal duration of bisphosphonate after denosumab requires further study. We have previously reported that sequential teriparatide-denosumab resulted in large BMD increases in premenopausal women with idiopathic osteoporosis (IOP), and that BMD remained stable with bisphosphonate (alendr

Osteoporosis is a metabolic bone disorder which increases fragility fracture risk. Elderly individuals, especially postmenopausal women, are particularly susceptible to osteoporosis. Although rare, osteoporosis in children and young adults is becoming increasingly evident, highlighting the need for timely diagnosis, management and follow-up. Early-onset osteoporosis is defined as the presence of a low BMD (Z-score of ≤ -2.0 in individuals aged < 20 years; T-score of ≤ -2.5 in those aged between 20 to 50 years) accompanied by a clinically significant fracture history, or the presence of low-ene

OBJECTIVE: The goal of this study was to characterize the microRNA (miRNA) expression signatures in patients with Primary hyperparathyroidism (PHPT) and identify miRNA biomarkers of bone homeostasis.
BACKGROUND: PHPT is associated with increased bone turnover and decreased bone mass. miRNA are markers of bone remodeling.
METHODS: We performed a prospective case-control study of postmenopausal females with PHPT and control subjects matched for race, age, and bone mineral density (BMD). We collected clinical and biochemical data, assessed BMD by dual-energy x-ray absorptiometry, and measured 2

Idiopathic osteoporosis (IOP) is a rare form of early-onset osteoporosis diagnosed in patients with no known metabolic or hormonal cause of bone loss and unknown pathogenesis. Patients with IOP commonly report both childhood fractures and family history of osteoporosis, raising the possibility of genetic etiologies of IOP. Whole-exome sequencing analyses of different IOP cohorts identified multiple variants in melatonin receptor 1A (MTNR1A) with a potential pathogenic outcome. A rare MTNR1A variant (rs374152717) was found in members of an Ashkenazi Jewish family with IOP, and an MTNR1A variant

SUMMARY: A 44-year-old athletic man presented in 2009 with severe low back pain. Dual-energy x-ray absorptiometry revealed severe osteoporosis; serum testosterone was 189 ng/dL while serum estradiol (E2) measured by liquid chromatography/mass spectrometry was 8 pg/mL. DNA was extracted and sequenced from a blood sample from the patient since his maternal first cousin also had low bone mass and both patients were screened for aromatase dysfunction by PCR analysis for the CYP19A1 gene, which encodes aromatase. No known pathologic mutations were observed in the coding exons, but novel single nucl

This case describes a young man with an unusual cause of severe osteoporosis and markedly deranged bone microarchitecture resulting in multiple fractures. A potentially pathogenic germline variant in the runt-related transcription factor 1 (RUNX1) gene was discovered by a focused 51-gene myeloid malignancy panel during investigation for his unexplained normochromic normocytic anemia. Further bone-specific genetic testing and a pedigree analysis were declined by the patient. Recent experimental evidence demonstrates that RUNX1 plays a key role in the regulation of osteogenesis and bone homeosta

PURPOSE: To understand the pathophysiology of idiopathic osteoporosis (IOP) better, we conducted a systematic review and meta-analysis of bone mineral density (BMD), hormones, and bone turnover markers (BTMs) between IOP patients and healthy controls.
METHODS: Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, an appropriate search query was created, and three databases, including PubMed, ScienceDirect, and Google Scholar, were searched for screening relevant original articles. Feasible information, both qualitative and quantitative, was extr

CONTEXT: We have previously reported that teriparatide is associated with substantial increases in bone mineral density (BMD) at the lumbar spine (LS), total hip (TH), and femoral neck (FN) and small declines at the distal radius in 41 premenopausal women with idiopathic osteoporosis (IOP), all severely affected with low trauma fractures and/or very low BMD. Effects of teriparatide dissipate if not followed by antiresorptives.
OBJECTIVE: To assess the effects of 12 and 24 months of denosumab in premenopausal women with IOP completing 24 months of teriparatide.
