The dose-time effects of fluoride on the expression and DNA methylation level of the promoter region of BMP-2 and BMP-7 in rats.

Skeletal fluorosis is a chronic metabolic bone disease caused by excessive exposed to fluoride. Recent studies have shown that fluoride causes abnormal bone metabolism through disrupting the expression of Bone Morphogenetic Proteins (BMPs). However, the relationship between fluoride and BMPs is not fully understood, and the mechanism of fluoride on BMPs expression is still unclear. This study investigated the dose-time effects of fluoride on BMP-2 and BMP-7 levels and DNA methylation status of the promoter regions of these two genes in peripheral blood of rats. Eighty Wistar male rats were ran

Effect of fluoride on osteocyte-driven osteoclastic differentiation.

Excessive systemic uptake of inorganic fluorides causes disturbances of bone homeostasis. The mechanism of skeletal fluorosis is still uncertain. This study aimed to study the effect of fluoride on osteocyte-driven osteoclastogenesis and probe into the role of PTH in this process. IDG-SW3 cells seeded in collagen-coated constructs were developed into osteocyte-like cells through induction of mineral agents. Then, osteocyte-like cells were exposed to fluoride in the presence or absence of parathyroid hormone (PTH). Cell viability and their capacity to produce receptor activator of nuclear facto

Evaluating the Association Between Dental Fluorosis and Polymorphisms in Bone Development and Mineralization Genes Among Population from a Fluoride Endemic Region of Eastern India.

Close to 12 million people in India are affected by more than the desirable level of fluoride in drinking water that could lead to dental, skeletal, and non-skeletal fluorosis. Dental fluorosis is a developmental defect that results in hypo-mineralization and pronounced porosity of enamel in the affected individuals. As estrogen receptor 1 (ESR1), collagen type 1 alpha 2 (COL1A2), bone γ-carboxyglutamic acid protein (BGLAP), and secreted protein acidic and cysteine rich (SPARC) genes are involved in bone development and mineralization, polymorphisms in these genes could be determining factors

Fluoride in Drinking Water and Skeletal Fluorosis: a Review of the Global Impact.

When safe and adequate exposure of an essential trace element is exceeded it becomes potentially toxic. Fluoride is one classic example of such a double edged sword which both plays a fundamental role in the normal growth and development of the body for example the consumption of levels between 0.5-1.0 ppm via drinking water is beneficial for prevention of dental caries but its excessive consumption leads to development of fluorosis. PURPOSE OF REVIEW: The abundance of fluorine in the environment as well as in drinking water sources are the major contributors to fluorosis. It is a serious publ

Two distinct mechanisms of fluoride enrichment and associated health risk in springs' water near an inactive volcano, southeast Iran.

Groundwater fluoride contamination is a major issue of water pollution in the world with health hazards such as dental and skeletal fluorosis. This research focused on exposure to the high concentration of fluoride in the springs water in the Bazman volcanic area, southeast Iran. The combination of chemical/isotopic analysis, geochemical modeling, health risk assessment and multivariate statistical methods were applied to investigate the contamination and sources of fluoride in the samples. Groundwater samples were collected from cold and thermal springs. Major ions, fluoride, trace elements a

Spatial distribution of endemic fluorosis caused by drinking water in a high-fluorine area in Ningxia, China.

Endemic fluorosis is widespread in China, especially in the arid and semi-arid areas of northwest China, where endemic fluorosis caused by consumption of drinking water high in fluorine content is very common. We analyzed data on endemic fluorosis collected in Ningxia, a typical high-fluorine area in the north of China. Fluorosis cases were identified in 539 villages in 1981, in 4449 villages in 2010, and in 3269 villages in 2017. These were located in 19 administrative counties. In 2017, a total of 1.07 million individuals suffered from fluorosis in Ningxia, with more children suffering from

Increasing fluoride content deteriorates rat bone mechanical properties.

Elevation of bone fluoride levels due to drinking beverages with high fluoride content or other means such as inhalation can result in skeletal fluorosis and lead to increased joint pain, skeletal deformities, and fracture. Because skeletal fluorosis alters bone's mineral composition, it is likely to affect bone's tissue-level mechanical properties with consequent effects on whole bone mechanical behavior. To investigate this, we determined whether incubation with in vitro sodium fluoride (NaF) altered bone's mechanical behavior at both the tissue- and whole bone-levels using cyclic reference

[Etiology, diagnosis, prevention and treatment of dental fluorosis].

Dental fluorosis is the most common and prominent symptom in the early stage of chronic fluorosis, which is caused by excessive fluorine intake during tooth development. In severe cases, it may be accompanied by skeletal fluorosis. There are also systemic damages to the nervous system, cardiovascular system, endocrine system and so on. The pathogenesis of dental fluorosis is not totally clear, which may be a complex pathological process involving both genetic and environmental factors. The prevalence of dental fluorosis has an upward trend arround the world, thus certain public prevention and

Citric acid modified granular activated carbon for enhanced defluoridation.

Excess fluoride (F-, >1.5 mg F-/L) in drinking water affects >260 million people across the globe and leads to dental and skeletal fluorosis. In this study, commercially available granular activated carbon (GAC) was modified with 0.3 M citric acid to get citric acid modified GAC (CAGAC). Over 70% of fluoride was removed in the first 60 min by CAGAC, whereas unmodified GAC removed only 30%. There were negligible interferences by co-existing ions (NO3-, Cl-, HCO3-, SO42-, PO43-) and organic matters. Maximum adsorption capacity of CAGAC was two times (1.65 mg/g) that of unmodified GAC (0.88 mg/g)

A review on the potential sources and health implications of fluoride in groundwater of Sub-Saharan Africa.

Groundwater is a major source of drinking water for millions of people around the world. Over 400 million people in Africa depend solely on it as their main source of water supply. Fluoride is a common contaminant in groundwater. In low concentration (0.5-1.0 mg/L), fluoride is needed by humans for healthy development of bones and teeth, however, a concentration >1.5 mg/L has been linked with several fluorosis and non-fluorosis diseases. Dental and skeletal fluorosis are the major fluorosis diseases commonly reported with the consumption of fluoride-rich water. Although fluoride intake through

Prevalence and causes of elevated bone mass.

INTRODUCTION: Reports of elevated bone mass (EBM) on routine DXA scanning are not infrequent. However, epidemiological studies of EBM are few in number and definition thresholds variable. The purpose of this study was to assess the prevalence and causes of EBM in the general population referred to a single university hospital - catering for a population of 4 million inhabitants - for DXA scanning. MATERIAL AND METHODS: DXA databases were initially searched for individuals with a bone mineral density (BMD) Z-score ≥+4 at any site in the lumbar spine or hip from April 1st, 2008 to April 30st, 2

Removal of fluoride from water by using a coagulant (inorganic polymeric coagulant).

Excess fluoride (F) ion of drinking water is a major problem in many areas of India and causes harmful effects such as dental and skeletal fluorosis. The World Health Organization (WHO 2004) recommends an upper limit of 1.5 mg/L fluoride in drinking water, and the concentration of fluoride in groundwater has been found 10-20 times higher in many of the States in India. In this study, the performance of inorganic polymeric coagulant (IPC) named as IPC-23, IPC-13, IPC-17, and alum for fluoride removal from drinking water was investigated. The amount of IPC was decided according to the Al2O3 amou

miR-122-5p Mediates Fluoride-Induced Osteoblast Activation by Targeting CDK4.

Chronic intake of fluoride, existing in the environment, may cause endemic fluorosis, which is characterized by the occurrence of skeletal and dental fluorosis. However, the pathogenesis of fluorosis has not yet been elucidated. Abnormal osteoblast proliferation and activation have a pivotal role in bone turnover disorders which are linked to skeletal fluorosis. MicroRNAs are involved in fundamental cellular processes, including cell proliferation. Based on our previous study, population study and in vitro experiments were designed to understand the effect of miR-122-5p on osteoblast activatio

sKlotho is associated with the severity of brick tea-type skeletal fluorosis in China.

The change of serum soluble Klotho (sKlotho) content is related to a variety of osteoarthropathy. However, its association with the severity of skeletal fluorosis (SF) is not clear. Here, the association of tea fluoride exposure with serum sKlotho levels and the severity of SF were investigated and further verified in a rat model of fluorosis. A cross sectional case control study was conducted in residents over 50 years old from brick-tea drinking areas in Qinghai and Xinjiang Provinces, China. Concentrations of fluoride in brick tea water and urine were determined by ion selective electrode m

Downregulation of miR-4755-5p promotes fluoride-induced osteoblast activation via tageting Cyclin D1.

BACKGROUND: Endemic fluorosis remains a major public health issue in many countries. Fluoride can cause abnormalities in osteoblast proliferation and activation, leading to skeletal fluorosis. However, its detailed molecular mechanism remains unclear. Based on a previous study, the aim of this study is to explore the role of miRNA in osteoblast activation of skeletal fluorosis via targeting of Cyclin D1. METHODS: A population study of coal-burning fluorosis and in vitro experiments were performed in this study. Urine fluoride (UF) concentrations of the participants were determined using a nat

Involvement of Bmal1 and circadian clock signaling in chondrogenic differentiation of ATDC5 cells by fluoride.

Skeletal fluorosis causes growth plate impairment and growth retardation during bone development. However, the mechanism of how fluoride impairs chondrocyte is unclear. To explore the effect of fluoride on chondrocyte differentiation and the regulation of circadian clock signaling pathway during chondrogenesis, we treated ATDC5 cells with fluoride and carried out a series of experiments. 10-3 M fluoride inhibited cell viability and significantly decreased the expression of Sox9 and Col2a1 (P < 0.05). Fluoride inhibited proteoglycan synthesis and decreased significantly the expression of Aggrec
