Prediction of fluoride contamination in groundwater of Nigeria using a weighted ensemble machine-learning framework.

Fluoride (F-) contamination of groundwater remains a significant health risk in Nigeria, affecting rural and urban communities. This study examines the spatial distribution and factors influencing F- levels using a weighted ensemble machine learning model to forecast high-risk areas and estimate exposed populations. The primary factor controlling geogenic F- is the aquifer lithology, with the highest F- levels found in groundwater from crystalline basement complexes and sedimentary sandstones. The problem is exacerbated in dry regions, including semiarid and arid zones, where evaporation rates

Chitosan‑selenium nanoparticles mitigate fluoride-induced bone injury by regulating OPG/RANKL/NF-κB pathway through the gut-bone axis.

Excessive fluoride intake can lead to skeletal fluorosis, which is a global public health concern. In recent years, selenium-loaded chitosan nanoparticles (CS-SeNPs) have attracted considerable attention due to their high bioactivity and favourable adhesion to the intestinal mucosa, and have demonstrated significant potential in mitigating skeletal damage. Nevertheless, it remains unclear whether CS-SeNPs can improve fluoride-induced bone damage and what the underlying regulatory mechanisms are. In this study, we found that CS-SeNPs alleviated fluoride-induced intestinal barrier disruption and

Spatiotemporal distribution, health risk assessment, and preventable disease burden of dental and skeletal fluorosis via defluoridation scenarios in Fars Province, Iran.

Fluoride contamination in groundwater poses a persistent public health challenge in arid regions, where elevated concentrations lead to dental and skeletal fluorosis. Current defluoridation policy evaluations have overlooked the population health benefits of interventions, the variability in technology performance, and the direct link between treatment efficacy and health outcomes. Hence, this study employed a comprehensive spatiotemporal probabilistic health risk assessment with disease burden estimation. It integrated disability-adjusted life years (DALY)-based intervention policy modeling a

Application of musculoskeletal ultrasound in endemic skeletal fluorosis: a comparative study of water-borne and brick-tea-borne subtypes.

BACKGROUND: Endemic skeletal fluorosis is a chronic disorder caused by prolonged excessive fluoride intake and is characterized by skeletal hyperostosis, periarticular soft-tissue degeneration, and joint dysfunction. Digital radiography (DR) is the conventional diagnostic modality, but it has limited sensitivity for early soft-tissue changes and provides little dynamic information. Musculoskeletal ultrasound (MSKUS) may address these limitations, yet comparative evidence across major fluorosis subtypes remains limited. OBJECTIVE: To characterize MSKUS findings in endemic skeletal fluorosis an

The potential of urinary miR-200c-3p as a biomarker of fluorosis in rats.

Fluorine is a strong oxidizing element and excessive intake can have harmful effects, particularly on the body's calcified tissues. Recent studies have demonstrated a link between miRNA and fluorosis. This study aimed to evaluate the time-dose-effect relationship of miR-200c-3p in plasma, urine and cartilage of rats with drinking water fluorosis, and to explore its potential as a biomarker. Analyses were conducted using Generalised linear models, Restricted cubic spline, Spearman correlation analysis, and Receiver operating characteristic curve (ROC). Results indicated that both fluoride expos

Association between ESR1 and COL1A1 gene polymorphisms and skeletal fluorosis in Tibetan, Kazakh, Mongolian and Russian populations, China.

BACKGROUND: Skeletal fluorosis is a chronic metabolic bone disease caused by excessive accumulation of fluoride in the bones. Previous studies have found that when the intake of tea fluoride is similar, the prevalence of skeletal fluorosis varies greatly among different ethnic groups, which may be related to different genetic backgrounds. Single nucleotide polymorphisms (SNPs) of estrogen receptor 1 (ESR1) and collagen type 1 α1 (COL1A1) were strongly associated with bone metabolism as well as bone growth and development, but their association with the risk of skeletal fluorosis has not been r

Fluoride Exposure Modulates Skeletal Development and Mineralization in Zebrafish Larvae.

The presence of high levels of fluoride (F) in groundwater is a major issue worldwide. Although F is essential for healthy teeth and bones, excessive exposure can cause fluorosis or F toxicity. This condition primarily affects the hard tissues due to their high F retention capacity. F accumulation alters bone formation and resorption mechanisms interfering with mineral homeostasis and eventually manifests as skeletal fluorosis. Albeit the numerous studies on skeletal fluorosis, the effect of F on developmental osteogenesis is inconclusive. In light of this, we studied the effect of F on osteog

Curcumin's protective role against fluoride-induced bone damage: Implications for pullet pathology and skeletal biomechanics.

This study aimed to investigate the effects of dietary fluoride (F) and curcumin (Cur) supplementation on the tibial biomechanical performance, histopathology, and behavior of pullets. Four dietary F levels (0, 400, 800, 1200 mg/kg) supplemented with CUR (0, 200 mg/kg) were used to create 8 experimental groups in Hyline Brown pullets. Behavioral study results showed that supplements of 1200 mg/kg F reduced the percentages of feeding, walking, standing, and preening behaviors while increasing the percentage of lying behavior (P < 0.05). This is associated with F-induced tibial elastic modulus,

BACKGROUND: Fluorosis caused by excess intake of fluoride can affects various soft tissues of the body, such as the gastrointestinal tract, blood, brain tissues and thyroid gland apart from dental fluorosis and skeletal fluorosis. Nonskeletal fluorosis is considered reversible if diagnosed early and treated promptly. Therefore, diagnostic methods that can be easily performed even by primary health care workers and depict any ongoing health problems, should be validated. Dental fluorosis, assessment of fluoride in urine and water are tests that fulfill these requirements. To date, no study has

Interaction of environmental fluoride exposure and gut microbes: Potential implication in the development of fluorosis in human subjects.

Fluoride exposure primarily occurs through contaminated water and leads to fluorosis, which is a global health concern. After ingestion, fluoride is absorbed via gastrointestinal tract, where it interacts with the gut microbiota. While animal studies have explored fluoride's effects on gut microbiota, no human studies have yet been conducted. Most research emphasizes metagenomic diversity, neglecting isolation and characterization of pure cultures for further applications. Additionally, the association between gut microbiota with fluorosis outcomes in fluoride-exposed populations is unexplored

Quercetin Simultaneously Treats Skeletal Fluorosis and Kashin-Beck Disease by Modulating HIF-1 and Ferroptosis Signaling Pathways.

Excessive fluoride exposure can lead to skeletal fluorosis (SF), and selenium deficiency is one of the important pathogenic factors of Kashin-Beck disease (KBD). Although the pathogenic factors of these two diseases vary, there are many similarities in their pathogenic mechanisms on skeletal and articular cartilage lesions. There are currently no specific drugs for either disease, and investigating their shared pathogenic mechanisms may facilitate the development of new drugs for the treatment. This study found through bioinformatics technology that the HIF-1 signaling pathway and ferroptosis

Zebrafish larvae as a model for investigating dual effects of fluoride on bone development.

Long-term excessive ingestion of fluoride is a severe public health threat globally. Skeletal fluorosis, a significant manifestation of prolonged fluoride exposure, is characterized by aberrant bone structure and alterations in bone function. However, there is currently a shortage of an efficient, fast, and easy-to-operate biological model for application in the field of fluorosis research. Zebrafish larvae, with human - like skeletal traits, high reproduction, rapid development, and transparency, are commonly used in bone disease studies. This study evaluates the potential of zebrafish larvae
