Integrating Behavioral Interventions into the Routine Immunization System in Chad: Implementation Lessons from a Fragile Setting.

Despite global progress, COVID-19 pandemic disruptions increased immunization inequities, particularly in fragile contexts like Chad. In 2022-2023, Chad reported 124,544 zero-dose children, in addition to 122,935 children who were classified as under-vaccinated. Low immunization uptake reflects persistent barriers related to access, knowledge, misinformation, and hesitancy. Behavioral interventions, adapted to low-resource settings, can help address these challenges. Anchored in the WHO Behavioral and Social Drivers (BeSD) framework and aligned with the Immunization Agenda 2030 (IA2030), we de

Cell DiffErential Expression by Pooling (CellDEEP) highlights issues in differential gene expression in scRNA-seq.

Accurate identification of differentially expressed genes (DEGs) in single-cell RNA sequencing data remains challenging. Single-cell-specific statistical models often report large numbers of candidate genes but can exhibit inflated false positive rates, whereas pseudobulk approaches improve false discovery control at the cost of reduced sensitivity. To overcome the noise and bias that other tools have, and allow the user to have more control of the DEG process, we present CellDEEP, which uses a cell aggregation (metacell) approach. This tool provides a framework for flexible selection of pooli

Viral Infection-Induced Epilepsy and the Gut Microbiome Crosstalk.

Viral infections of the central nervous system (CNS) pose a significant risk for seizures, and in some cases, long-term epileptogenesis. Seizure development following viral infection involves diverse CNS mechanisms, with emerging evidence that peripheral factors, including gut microbiome composition and immune-inflammatory responses, also contribute. The gut microbiome is a multifaceted ecosystem with important roles in CNS health, and advances in sequencing have enabled investigation of host and microbial genomic changes associated with seizure development. While both animal and human studies

Effects of prolonged SARS-CoV2 spike protein exposure in long COVID and a mouse model of the disease.

We addressed the question: can the SARS-CoV2 spike protein persist in ACE2+ endothelia in the CNS/other sites and induce a prolonged microvasculature injury syndrome? People who died from COVID-19, with age-matched controls, and people with long COVID with symptoms persisting from 1-5 years, with controls of people who had recovered from COVID-19, were studied. Also compared were WT mice injected with the spike protein sacrificed at day 5 versus 1.5 months. The ACE2+ microvasculature density in the CNS increased 19-fold in the mice exposed to spike for 1.5 months. Human fatal COVID-19 cases sh

Association between herpes simplex virus infection and cardiometabolic diseases: protocol for a systematic review and meta-analysis.

INTRODUCTION: Herpes simplex virus (HSV) infection is a lifelong viral infection with high global prevalence. Emerging evidence suggests that chronic viral infections, including HSV, may contribute to the development of cardiometabolic diseases through persistent inflammation, endothelial dysfunction and immune dysregulation. However, findings on the association between HSV infection and cardiometabolic outcomes remain inconsistent. This systematic review and meta-analysis will synthesise the available evidence on the relationship between HSV infection and cardiometabolic diseases. METHODS AN

Live imaging and interactome analysis of Zika and chikungunya viral RNAs via dual-action aptamer tag.

Arthropod-borne, positive-sense RNA viruses encompass many human pathogens posing significant health threats worldwide. Understanding intracellular dynamics and host-factor interactions of the viral genome is essential for devising effective antiviral strategies. Here, we engineer dual-labeled Zika and chikungunya viruses incorporating a Mango-II aptamer for RNA pulldown and real-time imaging, achieving high-resolution imaging of flavivirus RNA in live cells. Tandem imaging with viral replication proteins revealed intracellular dynamics of replication complexes and RNA molecules of these virus

Integrative structure of norovirus NS3 suggests a role in RNA transport.

Human noroviruses (HuNoVs) are the leading global cause of acute gastroenteritis, yet no vaccines or antiviral therapies are currently approved. The non-structural protein NS3 is a membrane-bound AAA+ ATPase of superfamily 3 (SF3) with multiple proposed roles in the norovirus replication cycle. However, the structure of NS3, and the mechanisms by which it contributes to genome replication and membrane remodeling, have remained unknown. We engineered a soluble, hexameric, and catalytically active form of NS3 and determined its cryo-EM structure in the presence of a nucleotide analogue at 2.9 Å

Prevalence of adeno-associated virus type 3B neutralizing antibodies in cohorts of healthy donors and Wilson's disease adults.

Adeno-associated virus (AAV) is widely used for liver-directed gene therapy, with AAV serotype 3B (AAV3B) showing particularly high tropism for human hepatocytes. However, pre-existing neutralizing antibodies (NAbs) against AAV represent a major barrier to efficient gene transfer and patient eligibility. We have recently demonstrated that subjects with intermediate NAb levels (1:5-1:45), that would be excluded from clinical trials, can be converted in eligible patients by the administration of IgG-cleaving enzymes. It is thus crucial to assess not only the seroprevalence but also titer distrib

Genome-wide identification and characterization of the PE/PPE gene family in Mycobacterium abscessus.

Mycobacterium abscessus (Mab) is a rapidly growing non-tuberculous mycobacterium that causes opportunistic infections and exhibits extensive intrinsic antibiotic resistance. The proline-glutamate (PE) and proline-proline-glutamate (PPE) protein families are mycobacteria-specific virulence factors implicated in immune modulation, antigenic variation, and persistence. While Mycobacterium tuberculosis (Mtb) harbors approximately 170 PE/PPE members, the repertoire of PE/PPE genes in Mab remains largely uncharacterized. Here, we performed a genome-wide survey of PE/PPE genes in Mab ATCC 19,977 and

Hantaviruses as Emerging Zoonotic Pathogens: Molecular Biology, Epidemiology, Therapeutics, vaccine, and Pathogenesis.

Hantaviruses are globally distributed zoonotic pathogens belonging to the family Hantaviridae and are responsible for two major human diseases, hemorrhagic fever with renal syndrome (HFRS) and hantavirus cardiopulmonary syndrome (HCPS). Despite substantial progress in hantavirus research, important challenges remain in understanding the interactions among viral evolution, reservoir ecology, environmental change, transmission dynamics, and host immunopathology. This review provides an integrated One Health perspective that connects molecular virology and host-virus interactions with transmissio

Retrospective serological evidence of exposure to selected equine viruses in horses from Indigenous communities of Costa Rica, 2011.

Horses are essential for transportation and livelihoods in Indigenous communities of Costa Rica. However, information on their exposure to viral pathogens remains scarce. We conducted a retrospective cross-sectional serosurvey using samples collected from horses from four Indigenous communities (Alto Pacuare, Amubre, Paso Marcos, and Vereh) sampled between March and October 2011 (n = 278). Sera were tested in 2014 using commercial immunodiffusion or ELISA assays for antibodies against West Nile virus (WNV), equine herpesviruses 1 and 4 (EHV-1 and EHV-4), influenza A virus, equine arteritis vir

The effect of artificial intelligence-driven, computed tomography-based assessment of body composition parameters on 28-day mortality of patients with severe fever with thrombocytopenia syndrome: a retrospective cohort study.

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) is a life-threatening infectious disease with high mortality. Although sarcopenia and myosteatosis are associated with adverse outcomes in multiple acute illnesses, their prognostic impact on SFTS remains unknown. This study aimed to evaluate the association between body composition parameters and 28-day mortality in SFTS patients, and to identify independent prognostic factors. METHODS: In this retrospective observational single-center study, 234 SFTS patients with complete clinical data and available computed tomography (CT) ima

StitchR-based dual-AAV delivery strategies enable efficient transduction of defined neural cell populations.

Dual adeno-associated virus (AAV) vector reconstitution has become an indispensable technique for delivering large genes in neuroscience and gene therapy. A novel technology termed StitchR addresses the challenge of low efficiency in dual-AAV reconstruction, yet related tools enabling targeted gene delivery to specific neural cell populations remain insufficiently developed. Herein, we developed four StitchR-based dual-viral reconstitution strategies by combining high-efficiency AAV serotypes with neuron- or glia-specific promoters. Specifically, the AAV11-hSyn-driven StitchR strategy achieved

Passive acquisition of hepatitis B virus antibodies after allogeneic hematopoietic cell transplantation.

BACKGROUND: Newly positive hepatitis B virus (HBV) serologic results after allogeneic hematopoietic cell transplantation (HCT) may reflect passive antibody transfer from blood-derived products rather than HBV infection. We investigated the incidence and temporal patterns of hepatitis B surface antibody (anti-HBs) and antibody to hepatitis B core antibody (anti-HBc) acquisition after allogeneic HCT. STUDY DESIGN AND METHODS: We retrospectively evaluated 193 adults who underwent allogeneic HCT for hematologic diseases between May 2012 and March 2025 at a single institution in Japan. Anti-HBs, a

Climate-driven redistribution of Hyalomma marginatum (Ixodida: Ixodidae): robust ecological niche modeling reveals future expansion and quantifies projection uncertainty.

Hyalomma marginatum (Ixodida: Ixodidae) is the principal vector of Crimean-Congo hemorrhagic fever virus (CCHFV) and an emerging public health concern because climate change may facilitate its establishment in regions where it is currently absent. We used ecological niche modeling to characterize the current climatic suitability of H. marginatum and project its future redistribution across the Old World under CMIP6 climate scenarios. Models were calibrated using a comprehensive dataset of spatially filtered occurrence records and bioclimatic predictors within a biologically informed accessible
