Excellent survivorship and functional outcomes following lateral unicompartmental knee arthroplasty in patients aged 55 years or younger.

PURPOSE: Fixed-bearing lateral unicompartmental knee arthroplasty (UKA) is a bone- and ligament-preserving treatment option for isolated lateral-compartment osteoarthritis (OA), but evidence in younger patients remains limited. The primary purpose of this study was to assess lateral component revision-free survival and reoperation-free survival following primary fixed-bearing lateral UKA in patients aged 55 years or younger. Secondary objectives were to evaluate functional and radiological outcomes. METHODS: This retrospective case series was derived from a prospectively maintained institutio

Protocol for standardized minimally invasive mouse models of bisphosphonate-related and radiation-induced jaw osteonecrosis.

We present a protocol for establishing murine models of bisphosphonate-related osteonecrosis of the jaw (BRONJ) and osteoradionecrosis of the jaw (ORNJ). We describe steps for a uniform, low-trauma tooth extraction technique with disease-specific induction, systemic zoledronic acid for BRONJ and precision-targeted irradiation for ORNJ, to generate robust and clinically relevant models of jaw osteonecrosis, approaching minimizes procedural variability and off-target effects, yielding robust, clinically relevant phenotypes. This protocol provides a standard and minimally invasive framework for i

Computed Tomographic Findings of Ischemic Necrosis of the Accessory Carpal Bone in a Dog.

A 1-year-and-9-month-old Golden Retriever presented with a 9-month history of right thoracic limb lameness. Radiographs revealed a monostotic, expansile lesion of the right accessory carpal bone (ACB). A computed tomography (CT) confirmed the aggressive nature of the lesion, demonstrating multifocal moth-eaten osteolysis, sclerosis, periosteal reaction, and flexor carpi ulnaris tendinopathy. Differential diagnoses included the ischemic necrosis of the ACB, osteomyelitis, and primary bone neoplasia. Histopathology confirmed osteonecrosis. This case highlights the value of CT in evaluating aggre

Novel bioactive compounds in the treatment of osteoporosis: chemistry-driven discovery from preclinical validation to clinical translation.

Osteoporosis affects an estimated 200 million people worldwide. Fragility fractures of the hip, spine, and wrist carry high mortality and impose a heavy socioeconomic burden. Bisphosphonates, denosumab, selective estrogen receptor modulators (SERMs), teriparatide, abaloparatide, and the recent sclerostin antibody romosozumab have improved clinical outcomes, but unmet needs remain. Long-term antiresorptive use is complicated by atypical femoral fractures and osteonecrosis of the jaw; denosumab discontinuation causes rebound bone loss; romosozumab carries a cardiovascular caution. Persistence on

Safety of alternating denosumab and zoledronic acid for osteonecrosis of the femoral head in children.

Osteonecrosis, or avascular necrosis, of the femoral head (ONFH) results from an avascular insult of traumatic or non-traumatic origin and can lead to significant morbidity, including pain, arthritis and the need for joint replacement at a young age. Non-surgical management of ONFH focuses on preservation of femoral head shape to reduce the need for arthroplasty or delay the age at which it is performed. Thirty patients diagnosed with ONFH on bone scan were prospectively studied to determine the safety of alternating denosumab and zoledronic acid over a 66-week treatment period. Patients under

Identifying incident medication-related osteonecrosis of the jaw and antiresorptive drug holidays using natural language processing on clinical notes in men and women prescribed bisphosphonates or denosumab for fracture prevention.

OBJECTIVE: Medication-related osteonecrosis of the jaw (MRONJ) is a rare complication of antiresorptive therapy for osteoporosis, with risk potentially influenced by "drug holidays" around dentoalveolar procedures. MRONJ risk is higher in rheumatic diseases. Epidemiologic studies are limited by poor administrative code performance for MRONJ identification and lack of structured pharmacy data capturing drug holidays. We developed and validated a natural language processing (NLP) algorithm to classify MRONJ and detect drug holidays from free-text clinical notes in electronic health records (EHR)

Evaluation of Salter and Pemberton osteotomies in developmental dysplasia of the hip: the role of the congruity index as a prognostic factor.

BACKGROUND: The Salter osteotomy (SO) and the Pemberton osteotomy (PO) are common surgical methods for treating paediatric developmental dysplasia of the hip (DDH). Although they each have their own advantages, complications resulting from open surgeries are still relatively common. This study aimed to evaluate the clinical outcomes of SO and PO in the treatment of DDH and to identify prognostic factors associated with postoperative outcomes. METHODS: Between April 2011 and May 2018, we retrospectively reviewed 72 DDH patients who underwent open reduction combined with pelvic osteotomy (SO/PO

Evaluating the Readability of Online Patient Education Materials for Avascular Necrosis.

PURPOSE: Avascular necrosis (AVN) is a progressive pathology affecting pediatric and adult patients. Given the risks of joint collapse and surgery, patients frequently research their condition online. The average reading level in the United States is approximately the eighth-grade level. The National Institutes of Health (NIH) recommend that patient materials be written at or below a sixth-grade level. This study aimed to assess the readability and quality of online AVN resources. METHODS: A Google search using the terms "avascular necrosis" and "osteonecrosis" was conducted, and first-page r

Generation and Characterization of an Osteoclastic Cell Model of Gaucher's Bone Disease.

Gaucher disease (GD) is caused by recessive mutations in the GBA1 gene (Glucosylceramidase beta 1) generating lysosomal accumulation of glucosylceramides. Bone phenotypes related to GD include Erlenmeyer flask deformity, osteosclerosis, osteonecrosis, osteopenia, and bone pain. Lysosomes modulate osteoclast differentiation, playing a fundamental role in osteoclastogenesis and bone resorption. However, the role of GBA1 mutations in the osteoclast function remains poorly understood. We aimed to generate and characterize an in vitro osteoclast cell model of GD. The Gba1 gene was knocked out with
