Severe fever with thrombocytopenia syndrome-associated encephalitis and intracranial hypotension: A case report.

Severe fever with thrombocytopenia syndrome (SFTS) is a natural infectious disease caused by severe fever with thrombocytopenia syndrome virus (SFTSV). Central nervous system (CNS) involvement is not rare in SFTS. In this work, we report a case of SFTS with CNS symptom onset and the metagenomic next-generation sequencing of cerebrospinal fluid (CSF), confirming invasion of SFTSV into the CNS. The case's lumbar puncture indicated an exceptionally low CSF opening pressure-an uncommon finding in other CNS infections and in case reports of SFTS. A spontaneous subdural effusion was observed on the

Recent advances in protein extraction methods from bone tissue for proteomics analysis.

Proteomic analysis is a fundamental tool for obtaining new information about biological functions that occur at the molecular level in the body, allowing the understanding of physiological and pathological processes, as well as contributing to the diagnosis and prevention of diseases. However, a critical step for the success of these analyses is the protein extraction from the tissue of interest, with well-described and reproducible protocols still scarce in the current literature, especially for bone tissues. Therefore, the main objective of this study was to present a reliable and standardiz

Insights into the Osteoprotective Potential of Chemically Modified Natural Polysaccharides for Animal Skeletal Health: A Narrative Review.

In intensive livestock production, the imbalance between rapid muscle accretion and insufficient skeletal mineralization predisposes animals to skeletal disorders, threatening production efficiency and animal welfare. Natural polysaccharides demonstrate osteoprotective potential, yet their in vivo application is largely limited by high molecular weight, low solubility, and poor bone-targeting capacity. Strategies involving chemical modification can overcome these limitations by enhancing endogenous osteogenic bioactivity and, in some cases, introducing functional groups that confer osteoinduct

Osteoporosis and Alzheimer's Disease: The Role of Sclerostin.

PURPOSE OF REVIEW: Little is known about the underlying cause contributing to the association between Osteoporosis (OP) and Alzheimer's disease (AD). This review highlights the emerging role of sclerostin, a protein mainly produced by osteocytes, in the newly discovered crosstalk between bone and brain, with a focus on its potential contribution to the relationship between OP and AD. RECENT FINDINGS: Circulating sclerostin levels increase with aging and are positively associated with brain amyloid-β burden in older adults. Studies demonstrate that elevated sclerostin can cross the blood-brain

Denosumab Therapy Duration and Osteonecrosis of the Jaw and Skeletal Events in Bone-Metastatic Prostate Cancer.

IMPORTANCE: Denosumab reduces skeletal-related events in bone-metastatic prostate cancer but carries a cumulative risk of medication-related osteonecrosis of the jaw (ONJ). The treatment duration that best balances these risks has not been established in clinical settings. OBJECTIVE: To evaluate the association between duration of denosumab treatment and the 3-year cumulative incidence of ONJ and skeletal events. DESIGN, SETTING, AND PARTICIPANTS: This nationwide retrospective cohort study emulated a target trial using Korean Health Insurance Review and Assessment Service data from January 1

Graft-versus-host disease prophylaxis shapes T cell biology and immune reconstitution after hematopoietic cell transplant.

Successful hematopoietic cell transplant requires immunosuppression to prevent graft-versus-host disease (GVHD), a lethal, T cell-mediated complication. The phase 3 BMT CTN 1703 trial demonstrated superior GVHD-free/relapse-free survival for posttransplant cyclophosphamide (PT-Cy)-based GVHD prophylaxis versus tacrolimus/methotrexate (Tac/MTX), but the T cell biology underpinning these outcomes remains unknown. To determine this, 324 patients were coenrolled onto BMT CTN 1801 to investigate the distinct T cell immune reconstitution pathways of these regimens, including extensive evaluation of

[EARLY DETECTION OF BONE METASTASIS BY WHOLE-BODY MRI AND TRIPLET THERAPY AFTER RADICAL PROSTATECTOMY: A CASE REPORT].

(Background) Although radical prostatectomy is a curative treatment for localized prostate cancer, some high-risk patients experience early recurrence due to occult metastases undetected by conventional imaging. (Case presentation) A 53-year-old man with high-risk prostate cancer (cT2bN0M0, Gleason score 4+4, initial PSA 10.6 ng/mL) underwent robot-assisted radical prostatectomy with pelvic lymph node dissection. Pathology revealed Gleason score 4+5, pT3bN1 disease. At 1 month postoperatively, PSA increased to 13.6 ng/mL, and whole-body MRI detected a solitary right iliac bone metastasis that

Association of rs3736228 polymorphism in LRP5 gene with bone fracture and osteoporosis: a systematic review and updated meta-analysis.

BACKGROUND: The low-density lipoprotein receptor-related protein 5 (LRP5) gene is a key regulator of bone mineral density and skeletal metabolism. The rs3736228 (Ala1330Val) polymorphism in LRP5 has been firmly established as a genome-wide significant locus for bone mineral density and fracture risk in large-scale GWAS. However, demographic and clinical subgroups that are frequently underrepresented in GWAS cohorts, such as sex, menopausal status and ethnicity, may have different impact sizes and ideal genetic models. Previous candidate-gene meta-analyses, while informative, did not systematic

Ferro-aging: a potential driver of degenerative bone and joint diseases.

Degenerative bone and joint diseases (DBJDs), including osteoporosis (OP), osteoarthritis (OA), and intervertebral disc degeneration (IVDD), are chronic age-related disorders characterized by progressive destruction of bone and joint tissues and loss of function, posing a heavy global health burden. Emerging evidence indicates that iron dyshomeostasis, chronic lipid peroxidation, and cellular senescence are common pathological features shared by these diseases. The newly discovered concept of ferro-aging-a non-lethal, persistent cellular state driven by sub-lethal lipid peroxidation that leads

Cadmium exposure triggers a functional switch of APP to drive joint degeneration.

BACKGROUND: Cadmium (Cd) is a pervasive environmental heavy metal linked to various chronic pathologies; however, its specific role and underlying molecular mechanisms in osteoarthritis (OA) pathogenesis remain poorly characterized. METHODS: We performed a cross-sectional analysis using the NHANES database (2001-2018) to evaluate the association between blood Cd levels and OA risk. Hub targets were identified by integrating the CTD and GEO databases through network toxicology and machine learning algorithms. Single-cell RNA sequencing (scRNA-seq) was employed to resolve the cellular localizat

Intrinsic Antimicrobial Properties of a Novel Silorane-Based Material for Orthopedic Applications.

One of many drawbacks of PMMA bone cement used to secure orthopedic implants is that its surface is highly susceptible to bacterial attachment and biofilm formation. In an effort to find alternative materials, we focused on silorane-based polymers previously developed for use in dental composites. Compared to PMMA, this composite has low shrinkage, low exothermic polymerization, low toxicity, and the potential for lower bacterial susceptibility. Building on this dental material's advantages, a novel silorane-based material for orthopedic use was developed utilizing a dual initiation system inc

[Molecular diagnosis of non-syndromic craniosynostosis: new challenges for real clinical practice].

Craniosynostoses (CS) are a congenital malformation caused by absence or premature closure of skull sutures. This leads to deformation and impaired growth of skull. Traditionally, syndromic and non-syndromic forms are distinguished. For a long time, it was believed that molecular genetic mechanisms are significant exclusively for syndromic CS. However, clear data on involvement of somatic and germinal mutations in pathogenesis of non-syndromic forms were accumulated in recent years. Currently, there are no unified clinical guidelines defining the strategy for molecular genetic diagnosis for no

Addressing unilateral lambdoid craniosynostosis using endoscopic suturectomy and barrel-stave osteotomies: surgical technique and literature review.

PURPOSE: Unilateral lambdoid craniosynostosis (ULC) is a rare cranial deformity requiring intervention to correct asymmetry of the posterior skull. Conventional open remodeling and distraction osteogenesis offer robust expansion but carry risks of higher blood loss, hardware complications, or reoperation. We report a novel, minimally invasive technique combining endoscopic lambdoid suturectomy with barrel-stave osteotomies, supplemented by a literature review. CASE DESCRIPTION: A 2-month-old healthy male presented with classic left ULC features, including occipitoparietal flattening, ipsilate

Bone-Targeting Antibodies for Skeletal Precision Therapy.

Therapeutic antibodies have transformed the treatment of cancer, inflammation, infection, and disorders of bone biology by recognizing disease-associated molecular targets with high specificity. However, achieving therapeutically effective antibody concentrations at the site of disease remains a major challenge, as systemic administration does not inherently result in efficient tissue-specific delivery. Bone is a particularly demanding destination because mineralized matrix, limited vascular access, marrow complexity, and active remodeling can restrict local antibody exposure. Recent advances

High genetic predisposition and vertebral fracture burden in pregnancy and lactation-associated osteoporosis: clinical insights from a Taiwanese cohort.

BACKGROUND: Pregnancy and lactation-associated osteoporosis (PLO) is a rare disorder causing rapid bone loss and severe fragility fractures. While familial clusterings suggest genetic susceptibility, the specific clinical features, genetic landscape, and skeletal recovery trajectory of PLO remain poorly characterized in non-Caucasian populations. This study aimed to evaluate a Taiwanese PLO cohort to optimize diagnostic awareness. METHODS: Eleven postpartum women with PLO were evaluated using bone densitometry and spinal imaging. Whole-exome sequencing was performed utilizing a targeted 398-g

Central nervous system, cranial, and cerebrovascular conditions beyond the Kimonis criteria in patients with Gorlin syndrome: a retrospective cohort study.

BACKGROUND: Gorlin syndrome is a multisystem developmental and cancer-predisposing disorder commonly caused by germline PTCH1 variants. The Kimonis clinical criteria capture the canonical phenotype; however, central nervous system (CNS), cranial, and cerebrovascular conditions documented beyond these criteria remain poorly defined. METHODS: We conducted a retrospective descriptive cohort study of 32 patients who fulfilled the Kimonis clinical criteria and received care at either or both hospitals in Japan. Clinical information through March 31, 2026, including sufficiently documented care els

SIRT6 Activation Improves Intervertebral Disc Health in the Aging Spine.

Aging is a critical risk factor for intervertebral disc degeneration, a major contributor to chronic low back pain. Loss-of-function studies showed that SIRT6, a nuclear NAD+-dependent deacetylase, maintains disc health. We therefore investigated whether long-term SIRT6 activation via systemic delivery of the small-molecular agonist MDL-800 improves disc health in 24-month-old mice. Studies revealed healthy disc morphology, enhanced cell viability, and lower degeneration scores following MDL-800 treatment. SIRT6 activation decreased H3K9ac levels, improved extracellular matrix quality, and red
