Stress-induced IL-6 regulation in pediatric bone growth disorders: current insights and therapeutic strategies.

Stress-induced upregulation of interleukin-6 (IL-6) signaling and its downstream pathophysiological consequences have garnered considerable attention in recent years. However, no comprehensive review has specifically examined the association between stress-induced IL-6 and its implications for bone health in children. During childhood, linear growth and peak bone mass acquisition are tightly regulated processes. Hence, this review aims to investigate current evidence on stress-induced IL-6 upregulation and its detrimental effects on pediatric bone health. Findings from animal models, knockout

Metabolic Bone Disease in Captive Flying Foxes: A Conceptual Framework and Future Perspectives.

In Pteropus spp., metabolic bone disease has been consistently associated with fruit-based diets that are deficient in calcium, vitamin D precursors, and protein, as well as limited ultraviolet B (UVB) exposure, as reported in zoological surveys and clinical observations. Comparative mammalian physiology suggests that dysregulation of the endocrine axis involving parathyroid hormone (PTH), fibroblast growth factor 23 (FGF23), calcitonin, and calcitriol may contribute to disease development, although direct species-specific endocrine data in flying foxes remain scarce. This narrative review syn

Overview of matrix metalloproteinases (MMPs) in health and disease.

Matrix metalloproteinases (MMPs) are a family of structurally related, zinc-dependent endopeptidases with broad substrate specificity, extending far beyond fibrillar collagens to include chemokines, cytokines, growth factors, cell-surface receptors and diverse extracellular matrix (ECM) components. In addition to their well-established proteolytic functions, MMPs can exert non-proteolytic effects through scaffolding roles, interactions with other membrane proteins, or modulation of cell-cell and cell-matrix contacts. Through these catalytic and non-catalytic mechanisms, MMPs influence cell sig

Rete ridges form via evolutionarily distinct mechanisms in mammalian skin.

The loss of fur during human evolution has long mystified scientists and the public1-5. Reduced hair density coincides with acquisition of epidermal rete ridges, the developmental timing and molecular mechanisms of which are poorly understood despite their prominence in humans1,6-9. Examination of human and pig skin development has shown that rete ridges form through a mechanism independent from those of hair follicles10,11 and sweat glands3,4,12-15 by establishing interconnected epidermal invaginations. Here we document the occurrence of rete ridges across Mammalia, including in grizzly bears

Nutritional Monitoring and Intervention in Paediatric Inflammatory Bowel Disease.

Paediatric Inflammatory Bowel Disease (PIBD), encompassing Crohn's Disease (CD), Ulcerative Colitis (UC) and IBD-Unclassified (IBDU), onsets during a critical window for growth, pubertal development, and bone accrual, making nutrition a primary, disease-modifying component of care rather than a purely supportive one. This review summarises current evidence and clinical experience on nutritional assessment, monitoring, and dietary treatment in PIBD. Malnutrition, growth faltering and micronutrient deficiency are common at diagnosis and during flares, and the interpretation of biochemical marker

Neurological effects of encapsulated dexamethasone sodium phosphate in children aged 6-9 years with ataxia telangiectasia (NEAT): a multicentre, randomised, double-blind, placebo-controlled, phase 3 trial.

BACKGROUND: Ataxia telangiectasia is a rare, multisystem disorder with progressive cerebellar neurodegeneration and no approved treatments. The efficacy of corticosteroids, including erythrocyte encapsulated dexamethasone sodium phosphate (eDSP), which have been studied for two decades in this disease, has not yet been proven in randomised trials. We aimed to investigate the safety and efficacy of eDSP in children aged 6-9 years with ataxia telangiectasia. METHODS: NEAT was a multicentre, randomised, double-blind, placebo-controlled phase 3 study, conducted at 20 sites across nine countries (

Lesion Expansion in Craniofacial Fibrous Dysplasia: Natural History and Indicators of Progression in the Frontal, Zygomatic, and Sphenoid Bones.

Fibrous dysplasia (FD) is characterized by expansile fibro-osseous lesions that may occur in association with endocrinopathies as part of McCune-Albright syndrome (MAS). Craniofacial FD may result in substantial morbidity, including facial asymmetry and compression of vital neurovascular structures. There is a critical need to understand the natural history and risk factors for craniofacial lesion expansion to develop preventative trials and identify candidates for intervention. The purpose of this study was to evaluate expansion rates in periorbital bones (frontal, zygomatic, and sphenoid). P

Autoimmune osteomalacia: a novel FGF23-related hypophosphatemic osteomalacia.

BACKGROUND: Fibroblast growth factor 23 (FGF23)-related hypophosphatemic rickets/osteomalacia arises from excessive FGF23 activity, with X-linked hypophosphatemia (XLH) and tumor-induced osteomalacia (TIO) as the most common congenital and acquired forms, respectively. However, in a substantial subset of patients with acquired FGF23-related hypophosphatemic osteomalacia, phosphaturic mesenchymal tumors (PMTs) remain undetectable despite extensive imaging studies. A recent study identified autoantibodies against PHEX, the gene responsible for XLH, in 5 of 13 patients with acquired FGF23-related

Renal osteodystrophy in pediatric patients. A systematic review and meta-analysis.

Chronic kidney disease (CKD) in children is associated with significant comorbidities, including mineral and bone disorder (CKD-MBD). A key manifestation of CKD-MBD is renal osteodystrophy (ROD), diagnosed by bone biopsy followed by histomorphometric analysis. In pediatric patients, ROD can lead to bone deformities and impaired growth. However, the limited use of bone biopsy in this population hampers the understanding of ROD patterns and severity. This systematic review and meta-analysis aimed to evaluate histomorphometric findings in children with CKD. We screened 5082 abstracts published up

MECP2 Insufficiency Attenuates RUNX2-Dependent Osteoblast Differentiation via miR-126-3p/DKK1-Mediated Canonical Wnt Signaling Inhibition in Rett Syndrome.

Rett syndrome (RTT) is a rare neurodevelopmental disorder caused by loss-of-function mutations in the gene encoding methyl-CpG-binding protein 2 (MECP2) that is located on the X chromosome. Affected individuals also exhibit a variety of non-neurological symptoms such as kyphoscoliosis and osteoporosis. Thus, MECP2 may play a functional role in bone remodeling and osteoblast differentiation. This study aimed to clarify the molecular mechanisms underlying the deregulation of bone remodeling in RTT. Human deciduous tooth-derived mesenchymal stem cells that exhibit osteoblast plasticity were used

Discoidin Domain Receptor 2 Is Required for Tooth Extraction Socket Healing.

Discoidin Domain Receptor 2 (DDR2) is a collagen-activated tyrosine kinase required for bone and tooth development as well as regeneration of calvarial and long bones. Although DDR2 is expressed in alveolar bone, its functions in this site have not been previously examined. The present study used global and conditional knockout approaches and lineage tracing to examine the functions of DDR2 in alveolar bone formation during tooth socket healing. First molars were extracted from the maxillae of wild-type and globally Ddr2-deficient (Ddr2LacZ/LacZ) mice, and socket healing was measured after 1,

Addressing evidence gaps in haemodialysis care: a critical review of clinical trials, technological innovations, and implementation priorities.

Kidney failure treated with maintenance haemodialysis (HD) carries high risks of premature mortality, hospitalisation and impaired quality of life, yet important evidence gaps persist across routine HD care. The Dialysis Research Innovation Network, part of the UK Kidney Research Consortium, convened the inaugural UK Haemodialysis Research Nexus in Birmingham on 11th May 2026. There were fifty-six delegates: 22 doctors, 13 nurses, six dietitians, one renal scientist, seven people in other roles (including one patient), and seven whose role was not recorded. The meeting reviewed recent trials,

Hepatic estrogen receptor α contributes to stage-specific coupling of liver metabolism and proliferation during pregnancy.

BACKGROUND AND AIMS: Pregnancy requires dynamic, stage-specific adaptations of maternal liver metabolism and growth to sustain fetal development while preserving systemic homeostasis. Estrogen signaling increases markedly during pregnancy and is primarily mediated in hepatocytes by estrogen receptor α (ERα). Although hepatic ERα regulates female liver metabolism under non-pregnant conditions, its contribution to pregnancy-induced hepatic remodeling remains unclear. METHODS: We studied non-pregnant and pregnant control and liver-specific ERα knockout (LERKO) mice across gestational stages usin
