Unbalanced occlusal loading elicited remodeling responses in growing rat temporomandibular joints and Notch1/Hes1 signaling pathway expression.

OBJECTIVE: We aimed to assess the impact of unbalanced occlusal loading on the TMJ and the expression of the Notch1/Hes1 signaling pathway using a unilateral occlusal loss rat model. METHODS: We established a unilateral occlusal defect animal model. At three different times, all female Wistar rats were randomized into three groups (4 weeks, 6 weeks, and 10 weeks): (1) unilateral occlusal loss experiment group (extraction of the left mandibular molars and trimming of the left mandibular incisors every other day), (2) restoring incisal occlusion experiment group (extraction of the left mandibul

Heart Failure and Osteoporosis: Shared Challenges in the Aging Population.

In clinical practice, heart failure (HF) and osteoporosis (OP) are commonly paired conditions. This association is particularly relevant in patients over the age of 50, among whom its prevalence increases dramatically with every decade of life. This can be especially impactful since patient prognosis when facing both conditions is poorer than that of each disease alone. Clinical studies suggest that prior fractures increase the risk for heart failure hospitalization and, conversely, an episode of heart failure increases the risk of subsequent fractures. In other words, the relationship between

Critical roles of miR-21 in promotions angiogenesis: friend or foe?

MiRNAs are small RNA strands that are managed following transcription and are of substantial importance in blood vessel formation. It is essential to oversee the growth, differentiation, death, movement and construction of tubes by angiogenesis-affiliated cells. If miRNAs are not correctly regulated in regard to angiogenesis, it can deteriorate the health and lead to various illnesses, which include cancer, cardiovascular disorder, critical limb ischemia, Crohn's disease, ocular diseases, diabetic microvascular complications, and more. Consequently, it is vital to understand the crucial part t

Identification of a non-inhibitory aptameric ligand to CRL2ZYG11B E3 ligase for targeted protein degradation.

As a crucial element of proteolysis targeting chimeras (PROTACs), the choice of E3 ubiquitin ligase significantly influences degradation efficacy and selectivity. However, the available arsenal of E3 ligases for PROTAC development remains underexplored, severely limiting the scope of targeted protein degradation. In this study, we identify a non-inhibitory aptamer targeting ZYG11B, a substrate receptor of the Cullin 2-RING ligase complex, as an E3 warhead for targeted protein degradation. This aptamer-based PROTAC platform, termed ZATAC, is facilely produced through bioorthogonal chemistry or

VGF and the VGF-derived peptide AQEE30 stimulate osteoblastic bone formation through the C3a receptor.

New therapeutic targets, especially those that stimulate bone formation in cortical bone, are needed to overcome the limitations of current antiosteoporotic drugs. We previously demonstrated that factors secreted from megakaryocytes (MKs) promote bone formation. Here we conducted a proteomic analysis to identify a novel bone-forming factor from MK secretions. We revealed that Vgf, a nerve growth factor-responsive gene, and its derived active peptide AQEE30 in MK-conditioned medium play important roles in osteoblast proliferation and in vitro bone formation. In both Vgf-deficient male and femal

Impact of hematopoietic stem cell transplantation on growth outcomes in mucopolysaccharidosis: a systematic review.

Mucopolysaccharidosis (MPS) is a group of genetic disorders characterized by defective lysosomal enzyme activity that can result in growth abnormalities and other complications. Hematopoietic stem cell transplantation (HSCT), especially bone marrow transplantation (BMT), aims to restore enzyme function and improve growth parameters in patients with MPS. This systematic review evaluates the impact of HSCT on growth outcomes, including height, weight, body mass index (BMI), head circumference, and pubertal development, in pediatric patients with MPS. Using the PRISMA (Preferred Reporting Items f

Network analysis and experimental validation analysis reveal the mechanism by which psoralen improves glucocorticoid-induced growth retardation.

Glucocorticoids (GCs) are widely used, particularly concerning in pediatric patients. GC-induced growth retardation (GIGR) is one of its significant side effects. Endochondral ossification of growth plate chondrocytes is crucial for skeletal growth in children; excessive GCs inhibit growth plate development and longitudinal bone growth. Previous studies have shown that psoralen (PSO) has anti-osteoporotic effects, preserves cartilage homeostasis, and enhances chondrocyte proliferation. However, the specific mechanisms remain unclear. This study used network pharmacology and molecular docking t

Recapitulation of endochondral ossification by hPSC-derived SOX9+ sclerotomal progenitors.

Endochondral ossification generates most of the load-bearing bones, recapitulating it in human cells remains a challenge. Here, we report generation of SOX9+ sclerotomal progenitors (scl-progenitors), a mesenchymal precursor at the pre-condensation stage, from human pluripotent stem cells and development of osteochondral induction methods for these cells. Upon lineage-specific induction, SOX9+ scl-progenitors have not only generated articular cartilage but have also undergone spontaneous condensation, cartilaginous anlagen formation, chondrocyte hypertrophy, vascular invasion, and finally bone

Race-neutral Pediatric Reference Ranges for Bone Mineral Density Predict Prospective Fractures in Childhood.

INTRODUCTION: Race-specific reference ranges for pediatric areal bone mineral density (BMD) are widely used, but the value of race-based clinical algorithms has been questioned. We developed race-neutral pediatric reference ranges for areal BMD and bone mineral apparent density (BMAD) and compared race-specific vs race-neutral Z-scores in their ability to predict prospective fractures. MATERIAL AND METHODS: This secondary analysis of the Bone Mineral Density in Childhood Study used longitudinal BMD data of the spine, hip, forearm, and total body less head and BMAD from dual-energy x-ray absor

Development of Polymer-Based Piezoelectric Materials for the Bone Tissue Regeneration.

With the aging population, fitness issues, poor bone healing, and high infection rates are associated with bone fractures and other bone diseases. Nevertheless, traditional approaches and materials struggle to treat orthopedic diseases by loading exogenous stem cells, growth factors, or merely structurally simulating the bone periosteum. The advancement of biomedical materials has become critical in addressing the challenges associated with bone tissue regeneration, encompassing a range of conditions including bone wounds, inflammation, infections, fractures, and the degenerative effects of ag

The Function of Myostatin in Ameliorating Bone Metabolism Abnormalities in Individuals with Type 2 Diabetes Mellitus by Exercise.

PURPOSE: The molecular mechanisms involved in bone metabolism abnormalities in individuals with type 2 diabetes mellitus (T2DM) are a prominent area of investigation within the life sciences field. Myostatin (MSTN), a member of the TGF-β superfamily, serves as a critical negative regulator of skeletal muscle growth and bone metabolism. Current research on the exercise-mediated regulation of MSTN expression predominantly focuses on its role in skeletal muscle. However, due to the intricate and multifaceted mechanical and biochemical interactions between muscle and bone, the precise mechanisms b

Loss of macrophage fibroblast growth factor 12 attenuates cardiac fibrosis in pressure-overloaded myocardium.

BACKGROUND: Cardiac fibrosis, a leading cause of death worldwide, plays a functional role in the development of heart failure. Unfortunately, there are currently no therapeutic strategies in clinical practice that can specifically attenuate the activation of cardiac fibroblasts, the effector cells of fibrosis in the heart. In this study, we aimed to identify a novel approach to target myocardial fibrosis through the crosstalk between macrophages and fibroblasts. METHODS: We investigated the expression of fibroblast growth factor 12 (FGF12), a novel regulator of macrophage activation, in both

Investigation into biased signaling, glycosylation, and drug vulnerability of acute myeloid leukemia.

Understanding and harnessing biased signaling offers significant potential for developing novel therapeutic strategies or enhancing existing treatments. By managing biased signaling, it is possible to minimize adverse effects, including toxicity, and to optimize therapeutic outcomes by selectively targeting beneficial pathways. In the context of acute myeloid leukemia (AML), a highly aggressive blood cancer characterized by the rapid proliferation of abnormal myeloid cells in the bone marrow and blood, the dysregulation of these signaling pathways, particularly those involving G protein-couple

Tibial adaptations to dietary 25-hydroxyvitamin D3 supplementation under two distinct vitamin regimens in young ducks.

Tibia disorders in modern ducks are frequently associated with rapid weight gain and compromised bone quality, which is defined as the structural and material properties of bone tissue that determines its strength and fracture resistance. These factors significantly increase the risk of fractures and chronic pain. Studies suggest that 25-hydroxycholecalciferol (25-OH-D3), a vitamin D3 metabolite, effectively addresses bone disorders, potentially depending on dietary vitamin regimens, which are determined by the amount and ratio of vitamins in the diet. This study used a 2 × 2 factorial design

Insulin-Like Growth Factor 2 mRNA-Binding Protein 2 Drives Subchondral Bone Damage in Temporomandibular Joint Osteoarthritis through Peroxisome Proliferator-Activated Receptor γ/Cellular FOS Proto-oncogene-Regulated Dual Pathways: Nuclear Factor of Activated T Cells 1 Signaling and Autophagy-Related 16-Like 2-Mediated Autophagy.

Overactivated osteoclastogenesis leading to abnormal subchondral bone loss is the main feature of temporomandibular joint osteoarthritis (TMJOA) deterioration. The role of N6-methyladenosine in osteoclast-mediated subchondral bone loss in TMJOA remains unknown. Here, it was found that an N6-methyladenosine reader insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2) was essential for mature osteoclast induction. In TMJ tissues of patients with TMJOA, the expression of IGF2BP2 was increased. Moreover, IGF2BP2 was augmented in subchondral bone of monosodium iodoacetate (MIA)-induced TMJO

The bone-muscle unit: from mechanical coupling to soluble factors-mediated signaling.

Skeletal muscles (SKM) and bones form a morpho-functional unit, interconnected throughout life primarily through biomechanical coupling. This relationship serves as a key reciprocal stimulus, but they also interact via various hormones, such as sex steroids, growth hormone-insulin-like growth factor 1 (GH-IGF1) axis hormones, and adipokines like leptin and adiponectin. Additionally, myokines (released by muscles) and osteokines (released by bones) facilitate dense crosstalk, influencing each other's activity. Key myokines include interleukin (IL)-6, IL-7, IL-15, and myostatin, while osteocalci

Drugs targeting novel pathways in pulmonary arterial hypertension.

Over the past three decades, several drugs have been developed to target three major dysfunctional pathways in pulmonary arterial hypertension (PAH), including the prostacyclin, endothelin and nitric oxide pathways. Despite these advances, PAH remains incurable, necessitating further drug discovery efforts. New therapies focus on previously untargeted pathways, especially the bone morphogenetic protein (BMP)/transforming growth factor (TGF)-β signalling pathway. Dysregulation of this pathway, involving the Smad2/3 and Smad1/5/8 signalling branches, plays a key role in pulmonary vascular remode
