Bone marrow neutrophil density regulates myelopoiesis during obesity and weight loss.

The bone marrow (BM) is altered in obesity to promote myeloid cell generation, but the mechanisms driving these changes remain unclear. Here, we show that obesogenic stimuli promote adipose tissue macrophages to recruit neutrophils from the BM in mice. Recruitment of BM neutrophils activates hematopoietic stem cells, which produce myeloid cells that accumulate in the circulation and drive inflammation. This recruitment is not resolved by weight loss, leading to sustained myelopoiesis in previously obese mice. Inhibiting neutrophil recruitment out of the BM in obese mice or during weight loss r

Co-occurrence of endometriosis with psoriasis and psoriatic arthritis: Genetic insights (Review).

Immunological factors appear to play an important role in the development of endometriosis, as evidenced by the aberrant functioning of immune cells often observed in affected women. Although endometriosis is not classified as an autoimmune disease, individuals with the condition show a higher tendency to develop various autoimmune disorders as comorbidities. There appears to be a bidirectional association between laparoscopically confirmed endometriosis and diagnosed psoriasis (PS) and psoriatic arthritis (PsA). Endometriosis is a chronic, complex, estrogen-dependent and progressive gynecolog

The influence of soluble epoxide hydrolase inhibition and their PUFA-derived epoxides in osteoblast bone metabolism: an in vitro study.

EpFAs are crucial mediators in resolving inflammation and regulating various biological processes. However, their activity is constrained by the rapid metabolism mediated by the soluble epoxide hydrolase (sEH), which converts EpFAs into inactive or even pro-inflammatory diols. Nevertheless, the specific effects of soluble epoxide hydrolase inhibition (sEHI) and EpFAs on osteogenic cell metabolism remain unclear. Cultures of the human immortalized osteoblast-like cell line (SAOS-2) were treated with varying concentrations (0.1-10 μM) of the potent sEHI TPPU or EpFAs (epoxyeicosatrienoic acids [

Molecular Basis of Fracture Pseudarthrosis Associated with Neurofibromatosis Type 1.

UNLABELLED: Neurofibromatosis Type 1 (NF1) is a tumor-predisposition syndrome caused by heterozygous mutations in the NF1 gene. In addition to oncologic manifestations, individuals with NF1 face a heightened risk of developing secondary skeletal complications associated with somatic loss-of-heterozygosity. Long bone dysplasia and post-fracture pseudarthrosis are among the earliest skeletal manifestations observed in children with NF1. Most commonly occurring in the tibia, pseudarthroses are often treated with a combination of resection, fixation, bone grafting, and rhBMP2. Although rates of in

Dual effects of bisphosphonates on bone metabolism and implant osseointegration: mechanisms, risks and clinical strategies.

Osseointegration is the fundamental mechanism facilitating the direct bonding between implants and bone tissue, which significantly influences the success and long-term stability of the implant. Bisphosphonates, potent antiresorptive agents, are widely utilized in the treatment of osteoporosis and other bone metabolic disorders due to their capacity to inhibit osteoclast activity. However, their effects on implant osseointegration present a 'double-edged sword.' Bisphosphonates inhibit mitochondrial ADP/ATP translocase through an interaction with class II aminoacyl-tRNA synthetase, leading to

Role of TGF-β3 in Regulating Neural Crest Cell Fate and Craniofacial Development: Insights From Zebrafish Models.

BACKGROUND: Craniofacial malformations, caused by dysregulated neural crest cell (NCC) differentiation, affect approximately one-third of newborns worldwide. Although TGFB3 mutations were recently associated with human coronoid process hypertrophy (CPH) and other craniofacial disorders, the mechanisms by which TGF-β3 regulates NCC fate determination through cell-cell communication remains unknown. METHODS: A zebrafish model was used to investigate the impact of tgfb3 on craniofacial cartilage and bone development. Additionally, Tgfb3 was knocked down in neural crest stem cells (NCSCs) in vitr

Osteoporosis and other bone-loss-related disorders remain major health concerns, underscoring the demand for innovative regenerative therapies. Propolis, a natural resinous substance rich in bioactive compounds such as caffeic acid phenethyl ester (CAPE), has emerged as a promising candidate for modulating bone regeneration. However, the cellular and molecular mechanisms underlying its effects on osteoblast function are not fully elucidated. In this study, we evaluated the impact of an aqueous extract of propolis on pre-osteoblast adhesion, proliferation, differentiation, extracellular matrix

Muscle-bone biochemical crosstalk in osteosarcopenia: focusing on mechanisms and potential therapeutic strategies.

Osteosarcopenia (OS) is a syndrome defined by the concurrent presence of sarcopenia and osteoporosis in the elderly population, which markedly elevates the risk of falls, fractures, and mortality. Recent studies demonstrate that disruption of muscle-bone biochemical crosstalk emerges as a key driver of OS pathogenesis, and that targeting pivotal mediators and pathways can concurrently restore musculoskeletal homeostasis. However, the precise molecular mechanisms and targeted therapeutic strategies remain inadequately explored. This review systematically summarizes the epidemiological risk fact

Exome Sequencing Reveals Novel Variants in Genetic Skeletal Disorders: Insights From a Cohort in Southwest Iran.

Genetic skeletal disorders (GSDs) comprise a diverse group of disorders that affect bone development and homeostasis. In some areas of Iran, GSD occurs more frequently than in other places for still unknown reasons. The aim of this study was to characterize the genetic landscape of GSDs in a cohort from southwestern Iran using Exome sequencing (ES), with a focus on identifying pathogenic and likely pathogenic variants. Osteogenesis Imperfecta (OI) was the most prevalent disorder, with an unexpectedly high frequency of autosomal recessive subtypes, likely due to a high consanguinity rate (61.3%

Visible Light Induced DLP-Printed Oxygen-Releasing TPMS Scaffolds Mitigate Early Hypoxia in Bone Defects.

Oxygen deprivation within large or poorly vascularized bone defects remains a key barrier to successful regeneration, especially during the early postimplantation period before vascular ingrowth. Here, the development of COSnPPOD (CaO2-Silica NP Platform for Osteogenic Development) is reported, a visible light digital light processing-printed hydrogel scaffold that integrates oxygen-releasing nanoparticles (NPs) within a Primitive-type triply periodic minimal surface architecture. The scaffold combines a gelatin methacrylate-poly(ethylene glycol) diacrylate matrix with calcium peroxide (CaO2)-

Notch signaling modulates Fgf23 expression through crosstalk with hypoxia and PTH pathways in osteogenic cells.

Fibroblast growth factor 23 (Fgf23) is produced by bone and functions primarily as a phosphaturia hormone. We previously reported that overexpression of the Notch intracellular domain (NICD) in osteogenic cells enhances Fgf23 expression in association with osteomalacia in vivo. Here, we investigated the underlying mechanisms using osteogenic cell lines UMR-106 and IDG-SW3 cells. NICD overexpression increased Fgf23 levels in both cell types. Manipulating RBPJ-κ activity, either a dominant-negative or constitutively active form, revealed that Notch-mediated Fgf23 expression is dependent on RBPJ-

Development of a leucine-rich repeat-containing protein 15-targeted radio-immunotheranostic approach to deplete pro-tumorigenic mechanisms and immunotherapy resistance.

Leucine-rich repeat containing 15 (LRRC15) has emerged as an attractive biomarker and target for cancer therapy. Transforming growth factor-β (TGFβ) induces the expression of this plasma membrane protein specifically in aggressive and treatment resistant tumor cells derived from mesenchymal stem cells, with minimal expression observed in non-neoplastic tissues. We have developed a humanized monoclonal antibody, DUNP19, that specifically binds with high affinity to a phylogenetically conserved LRRC15 epitope and is rapidly internalized upon LRRC15 binding. In multiple subcutaneous and orthotopi

Introduction to cancer biology and current research trends.

Cancer is a serious illness caused by changes in the genome that lead to many mutations. These mutations can cause harmful tumor growth that impacts individuals physically and mentally. Cancer cells grow uncontrollably, spread to other body parts, change genetically, form new blood vessels, and evade detection by the immune system. They develop ways to avoid destruction by the immune system, making it important for researchers to understand these mechanisms to enhance immune responses against cancer. Treatments like immune checkpoint inhibitors help the immune system recognize and attack cance
