Necrotic Bone Fluid Suppresses Energy Metabolism of Porcine PBMC-Derived Macrophages In Vitro.

Legg-Calvé-Perthes disease is a juvenile ischemic osteonecrosis (ON) of the femoral head. A disruption of blood supply to the femoral head produces extensive cell death and necrotic debris. Macrophages are innate immune cells recruited to the necrotic bone to orchestrate the repair process. However, the role macrophages play in the ON repair process is still not elucidated. The purpose of this study was to determine the effect of artificial necrotic bone fluid (NBF) on porcine peripheral blood mononuclear cell (PBMC)-derived macrophages. Monocytes were positively selected by CD14 MicroBeads fr

Development of an iPSC-based screening platform identifying enhancers of chondrogenesis.

OBJECTIVE: There is currently no long-term treatment for the repair of damaged cartilage and osteoarthritis (OA). Induced pluripotent stem cells (iPSCs) are an ideal cell source for screening platforms due to their ability to self-renew and differentiate to cell types that would otherwise require invasive surgeries to obtain, such as chondrocytes and mesenchymal stromal cells (MSCs). Here, we developed an iPSC-based screening platform and tested previously described pro-chondrogenic small molecule compounds, to determine their potential to identify hits. DESIGN: iPSC derived chondroprogenitor

The Periosteum in Health and Disease.

PURPOSE OF REVIEW: The periosteum has long been recognized as being essential for bone growth and repair. More recently, its involvement has been observed in a range of pathological conditions, including cancer progression and invasion, premature closure of the growth plate, bone pain, pseudarthrosis, and craniosynostosis. Given that the periosteal reaction often serves as an early indicator of skeletal abnormalities, the periosteum likely plays a pivotal role at the interface of bone health and disease. In this review, recent advances in our understanding of periosteal biology are highlighted

Intensive weight loss intervention versus usual care in adults with obesity: a protocol for the LightCARE randomised clinical trial.

INTRODUCTION: Total diet replacements (TDRs) and weight loss medications (WLMs) have proven effective in producing substantial weight loss for individuals with obesity. Evidence is lacking on whether combining these treatments is effective and cost-effective in primary care for adults with obesity class I (body mass index (BMI) 30-34.9) or uncomplicated obesity class II or higher (BMI≥35 without obesity-related disease). METHODS AND ANALYSIS: LightCARE is a 2-year 1:1 randomised, parallel-group, clinical superiority trial with blinded outcome assessment evaluating the benefits and harms of an

Occipital Bone Defect With Meningoencephalocele and Plexiform Neurofibroma in Neurofibromatosis-1.

Neurofibromatosis type 1 (NF-1) is the most common phakomatosis with autosomal dominant inheritance, caused by mutations in the NF-1 gene on chromosome 17q11.2, which codes for neurofibromin protein, a negative regulator of the RAS/MAPK pathway. A defect in this can lead to altered cellular growth and tumor development. NF-1 is a complex multi-system neurocutaneous disorder with some of the common manifestations being café au lait macules, axillary/inguinal freckling, and neurofibromas. Musculoskeletal manifestations include scoliosis, sphenoid wing dysplasia, tibial pseudoarthrosis, and, rare

Bone morphogenetic protein-2-independent bone formation by human iPSC-derived megakaryocytes and platelets.

Bone regeneration (BR) is an ongoing challenge in biomedical research. While recombinant human bone morphogenetic protein-2 (rhBMP-2) is available for clinical use, its side effects remain a concern. Patient-derived platelet-rich plasma has been proposed as an alternative, but its heterogeneity poses a major hurdle to standardization. Here, we evaluated the effectiveness of induced pluripotent stem cell (iPSC)-derived megakaryocytes and platelets (iMPs) and recombinant human bone morphogenetic protein-2 (rhBMP-2) as potential BR therapy in a rat lumbar bone graft model. We implanted artificial

Mapping the murine TMJ glenoid fossa over development, homeostasis and in response to disease.

The temporomandibular joint (TMJ) plays a key role in facilitating complex mammalian jaw movements required for daily life. It is formed between the condylar process of the mandible (or dentary bone) of the lower jaw, the glenoid (or mandibular) fossa of the squamosal/temporal bone in the upper jaw and an interposed fibrocartilage disc. Structural defects in any component of the TMJ can disrupt the entire joint, contributing to TMJ disorders. Embryonic defects in the condyle in mice have been shown to have an impact on the shape and development of the glenoid fossa, highlighting the importance

Particulate matter increases bone morphogenetic protein 2 in lung fibroblasts.

Particulate matter (PM) < 2.5 µm (PM2.5) contributes to many chronic respiratory disorders, but the mechanisms for this are not fully understood. The actions of PM2.5 on lung epithelial cells have been well studied, but their effect on lung fibroblasts has not been as extensively reported. Bone morphogenetic protein 2 (BMP2), part of the transforming growth factor cytokine family, plays crucial roles in the development, morphogenesis, repair, and functions as a critical mediator in the pathogenesis of lung diseases such as pulmonary fibrosis and chronic obstructive lung disease. Here, we inves

Inflammation impairs post-hospital discharge growth among children hospitalised with acute illness in sub-Saharan Africa and south Asia.

In resource-limited settings, children often experience poor growth following illness, but the mechanisms are poorly understood. This cohort study in six countries in sub-Saharan Africa and south Asia investigates pathways linking inflammation and post-discharge weight gain among children hospitalised with acute illness. We determine associations between inflammation, enteropathy, growth mediators and other exposures at hospital discharge and weight gain during 90 days and explain how these exposures influence growth. Here, we show that systemic inflammation impacts mediators of linear growth

[IL-6/STAT3 Signaling Pathways: Molecular Insights and Emerging Thereapeutic Targets].

The cytokine interleukin 6 (IL-6) has redundant biological activities in regulating the proliferation, differentiation, and function of various cell types. IL-6 regulates inflammation, immune responses, and hematopoiesis, as well as homeostasis of the nervous, renal, hormonal, and bone systems. IL-6 is also involved in tumorigenesis, including myeloma cell growth. The janus kinase (JAK)-signal transducer and activator of transcription (STAT) pathway is one of the key IL-6 signals both in normal and pathological conditions. In particular, STAT3 plays essential roles in mediating IL-6 signals, a

Peripheral Precocious Puberty Due to Autonomous Gonadal Activation: A Multicenter Experience.

INTRODUCTION: Peripheral precocious puberty (PP) is far less commonly encountered compared to central precocious puberty (CPP) in pediatric endocrine practice. Long-standing non-diagnosis may cause rapidly advancing bone age, leading to CPP and resultant short stature. We aimed to report the clinical profile and current Indian experience in the management of children with peripheral PP due to autonomous gonadal activation. METHODS: This multicentric retrospective study reports data on 23 children (20 girls) presenting with peripheral PP as a result of autonomous gonadal activation from eight

Structurally and functionally optimized silk fibroin-alginate-based biomimetic scaffolds reinforced with nanobioceramics for bone tissue engineering applications.

In this study, we prepared chemically crosslinked silk fibroin (SF) and sodium alginate (Alg) biomimetic scaffolds reinforced with bioceramic nanohydroxyapatite (nHAp) for bone tissue engineering (BTE) applications. The pore sizes of these scaffolds were effectively controlled by varying the composition of the SF/nHAp/Alg biocomposites. The scaffold prepared with a 40:20:40 SF: nHAp: Alg ratio exhibited excellent swelling properties, reaching over 1700% within 70 min. In vitro degradation studies demonstrated that these biomimetic scaffolds exhibited controlled degradation, taking over 30 days

Genetic diversity, population structure and selective sweeps in Italian Leccese chickens.

Italy hosts a rich heritage of poultry genetic resources, leading to the development of several unique native chicken breeds. Among them, the Leccese (LEC) chicken breed requires urgent genetic characterization to support conservation, development, and sustainable use. This study assessed the genetic diversity and population structure of the LEC chicken breed using the 600 K Affymetrix SNP array, comparing it with 25 Italian indigenous breeds and four commercial lines. In addition, selection signatures were explored through runs of homozygosity and integrated haplotype score analyses. The obse

Deep learning-based micro-CT grayscale analysis for early detection and staging of osteoporosis in rats.

BACKGROUND: Rat models are widely used in preclinical osteoporosis research to study disease mechanisms and evaluate therapies. Current Micro-CT studies mostly rely on cross-sectional comparisons at a single time point, and there is a lack of standardized reference data across multiple time points. To address this gap, the present study provides standardized reference data from multiple time points using a deep learning-based Micro-CT grayscale analysis, enabling early detection and precise staging of osteoporosis. METHODS: A standardized osteoporosis model was established in ovariectomized S

In vivo human embryonic spinal cord atlas validates stem cell-derived human dorsal interneurons and reveals ASD spinal signatures.

Spinal cord injuries (SCI) result in the loss of motor and sensory function. We are working towards restoring sensation by developing directed differentiation protocols to generate dorsal spinal interneurons (dIs; dI1-dI6) from human embryonic stem cells (hESCs). Here, we present an improved method that produces human dIs via a neuromesodermal progenitor state, the physiological intermediate for spinal cord development. We show that retinoic acid (RA), bone morphogenetic protein 4 (BMP4) and growth differentiation factor (GDF) 11 direct dI identity, while GDF11 and extended time in culture pro
