Enumeration of Benign and Malignant Plasmacytoid Dendritic Cells in Peripheral Blood and Bone Marrow by Flow-Cytometric Analysis.

Dendritic cells (DCs) are powerful antigen-presenting cells involved in regulating immune response. Plasmacytoid dendritic cells (pDCs) are a subtype of DCs present in small quantities in the bone marrow, peripheral blood, and lymph nodes. They are an important component of our immune system's regulation and response to diseases. They activate T cells and play a critical role in immune tolerance. In this chapter, we review the immunophenotypic features of pDCs and provide a practical protocol for pDCs enumeration in the peripheral blood and in bone marrow samples.

Updated Protocols for 3D and 2D Culture and Histological Analysis of Intestinal Organoids.

The intestinal epithelium maintains continuous self-renewal and multi-differentiation capacities through the integrated regulation of multiple signaling pathways, including Wnt, bone morphogenetic protein, epidermal growth factor (EGF), and Notch signaling pathways. Based on this principle, a defined combination of EGF, Noggin, and the Wnt agonist R-spondin has been shown to support long-term expansion of mouse intestinal stem cells and the formation of organoids with indefinite self-renewal and full differentiation capacity. In human intestinal organoid cultures, the addition of small molecul

Diagnostic value of integrated 18 F-fluorodeoxyglucose PET/MRI in initial staging and recurrence detection of Ewing sarcoma.

OBJECTIVES: Ewing sarcoma (EwS) is the second most common malignant bone tumour in children and adolescents. Due to its highly aggressive nature, rapid and accurate initial diagnosis as well as reliable recurrence detection are essential for treatment planning. Standard diagnostic work-up includes MRI for local tumour assessment and whole-body MRI or 18 F-fluorodeoxyglucose-PET-computed tomography ( 18 F-FDG-PET/CT) for metastatic evaluation. 18 F-FDG-PET/MRI combines the soft-tissue contrast of MRI with the metabolic information of PET while reducing radiation exposure. As data on PET/MRI in

Solitary Skull Metastasis as Initial Presentation of Hepatocellular Carcinoma: A Rare Case Report and Management Review.

Hepatocellular carcinoma (HCC) rarely metastasizes to the skull, often leading to diagnostic challenges. This case report describes a 64-year-old man presenting with right-sided limb weakness and a scalp mass in the left parietal region. Initial surgery for the scalp swelling was performed without a diagnostic workup, and the lesion recurred with neurological impairment. Imaging revealed a skull lesion eroding the bone and extending intracranially, with a solitary liver lesion identified as the primary tumour. Surgical resection of the skull metastasis was performed, followed by chemotherapy.

Personalizing Bone Tumor Ablation: Decision Models from the University Hospital of Strasbourg.

PURPOSE: To describe the clinical decision-making underlying the selection of the percutaneous ablation modality for bone tumors. MATERIALS AND METHODS: Two experienced interventional radiologists designed in consensus a model for ablation modality selection. Consecutive patients with primary and metastatic bone tumors were retrospectively identified. Demographic, clinical, and imaging data were collected for each tumor. The dataset was randomly divided into a training set (618/926 tumors; 66.7%) and a test set (308/926 tumors; 33.3%). A decision-tree model derived from the training set and t

Efficacy and safety of low-temperature plasma ablation combined with collagenase in the treatment of lumbar disc herniation.

BACKGROUND: Lumbar disc herniation (LDH) is a common cause of radicular pain and disability. Evidence regarding the incremental effectiveness of combining plasma-based intradiscal decompression with collagenase chemonucleolysis remains limited. METHODS: This single-center retrospective cohort study included consecutive hospitalized adults with imaging-confirmed LDH treated between July 2021 and July 2025. Patients underwent low-temperature plasma ablation alone (control group, n = 108) or plasma ablation plus intradiscal collagenase (observation group, n = 116). The primary outcomes were long

A Rare Case of Malignant Transformation in Bilateral Phyllodes Tumor Leading to Osteosarcomatous Metastases.

Phyllodes tumors are rare fibroepithelial neoplasms of the breast that typically exhibit benign or borderline behavior, with malignant transformation being an uncommon event. Even more exceptional is the occurrence of sarcomatous differentiation, particularly into osteosarcoma. We report the strikingly rare case of a 35-year-old female who initially presented with bilateral benign phyllodes tumors. Despite surgical excision and close follow-up, she developed a malignant transformation with osteosarcomatous features and widespread metastatic disease, including pulmonary and subcutaneous involve

68Ga-citrate visualization study on ferroptosis through transferrin receptor 1 in periodontitis.

BACKGROUND: The loss of alveolar bone in periodontitis is linked to ferroptosis mediated by Transferrin Receptor 1 (TfR1). This study employed a multimethod approach integrating in vitro and in vivo experiments to investigate the utility of gallium-68 citrate (68Ga-citrate), a high-affinity iron-mimetic tracer, for the non-invasive, dynamic positron emission tomography/computed tomography (PET/CT) imaging of ferroptosis and periodontal remodeling in vivo. METHODS: In vitro, MC3T3-E1 were stimulated with Porphyromonas gingivalis lipopolysaccharide (LPS) to model the inflammatory microenvironme

Prospective feasibility study of diagnostic CT-based simulation-free palliative radiotherapy.

Conventional radiotherapy (RT) workflows require planning computed tomography (pCT) and treatment planning. However, these processes often take several days, leading to delays in RT initiation. Simulation-free radiotherapy (SFRT), which uses diagnostic CT (dCT) instead of pCT for treatment planning, may reduce the time from consultation-to-treatment delivery. However, the number of prospective studies conducted in Japan remains limited. This study prospectively evaluated the feasibility of SFRT in Japan. A single-center, single-arm prospective study (UMIN000054721) was performed in June 2024.

The Many Faces of Acromegaly: Understanding Its Clinical and Biological Heterogeneity.

BACKGROUND: Acromegaly, in 95% of cases resulting from a PitNET that hypersecretes growth hormone (GH), is constantly revisited in the literature due to the repercussions on morbidity and mortality and difficulties that persist in early diagnosis. SUMMARY: To shorten the interval between the onset of acromegaly manifestations and the diagnosis, our goal was to summarize the main situations that deviate from the typical presentation of the disease, highlighting the variability in phenotypic features, biochemical findings, and GH tumor expression. These non-classic findings contribute to the di

Pyroptotic cell-derived factors promote osteoclast differentiation.

The NLRP3 D301N substitution, the murine ortholog of the human D303N variant, causes NLRP3 constitutive activation (NLRP3CA) and inflammasome assembly. Here, we show that priming signals induced by lipopolysaccharide (LPS) are sufficient to trigger GSDMD-dependent pyroptosis in NLRP3CA-expressing bone marrow-derived macrophages (BMDMs), but not in wild-type (NLRP3WT) cells. NLRP3CA mice exhibit elevated IL-1β secretion and LDH release in bone marrow supernatants under both basal and LPS-challenged conditions. Other cytokines (e.g., TNF-α) increased comparably between genotypes, indicating that

Longitudinally Monitoring Prostate Cancer Bone Metastasis and Treatment Response via Circulating Extracellular Vesicles.

Lethal prostate cancer (PCa) is dominated by treatment-refractory progression in bone. In this study, we used plasma-derived extracellular vesicles (EVs) for monitoring the tumour and therapy-induced changes in the bone tumour microenvironment (bone-TME) in men with bone metastatic PCa (M1b PCa). Transcriptome analysis of differentially expressed genes in plasma EVs from patients with M1b and localized (M0) PCa, identified a 'bone metastasis signature' (BMS) that reflects tumour-induced bone formation. In preclinical studies, the BMS was enriched in plasma EVs from mice inoculated with bone-me

[Expert consensus on composite co-ablation for pain caused by bone metastases].

Pain caused by bone metastases from malignant tumors is mediated by multiple mechanisms, including direct tumor invasion or nerve compression, release of inflammatory mediators, and peripheral/central sensitization, which severely compromise patients' quality of life and functional status. Composite co-ablation, through alternating cycles of deep cryogenic temperature (-196 ℃) and hyperthermia (80 ℃), offers advantages such as a large ablation volume per probe, lower tissue temperature than conventional cryoablation, a visible ice ball on imaging, and synergistic cryo-thermal effects. This tec

[Expert consensus on co-ablation therapy for the perioperative management of bone and soft tissue tumors].

Co-ablation therapy is playing an increasingly important role in the comprehensive treatment of bone and soft tissue tumors. However, these tumors can originate in various sites and organs throughout the body, exhibiting numerous histological types and complex classifications. To standardize the perioperative management of co-ablation therapy for bone and soft tissue tumors at different anatomical locations, the Expert Committee on Radiological Interventional Therapy, Chinese Society of Clinical Oncology (CSCO), the Expert Committee on Ablation Therapy, Chinese Society of Clinical Oncology (CS

New insights into bone pathology through a transcriptomic profiling of an osteoblast model of Gaucher disease.

Although up to 90% of Gaucher Disease (GD) patients report bone symptoms, the molecular basis underlying the contribution of osteoblasts in bone pathology remains poorly understood. To identify potential molecular players in this process, we developed and characterized a new model of GD osteoblasts by knocking out (KO) the GBA1 gene in the SaOS cell line and subsequently studied its transcriptional profile. SaOS GBA1 KO cells showed reduced matrix deposition compared with wild-type cells and enhanced induction of osteoclastogenesis, likely through increased release of the receptor activator of

Thermoregulated photothermal therapy for bone defects using graphene oxide hydrogel with mesoporous silica-derived phase-change nanocapsules.

The clinical translation of photothermal therapy for bone repair is hindered by the risk of thermal damage to surrounding healthy tissues due to uncontrolled high temperatures. We addressed this by integrating phase change materials (PCMs) into a photothermal platform, developing a hydrogel scaffold with temperature regulation. Polyethylene glycol (PEG) was encapsulated in mesoporous nanosilica (MSNs) to form nanocapsules, achieving 82.5% efficiency. This encapsulation prevents PEG leakage and enhances thermal regulation and bioactivity. Graphene oxide (GO) was integrated into an alginate-chit
