3D minichannel-morphed adipose stem cell-cartilage granule reconstituted tough grafts for repairing segmental tracheal defects in preclinical models.

Segmental tracheal reconstruction requires biomimetic substitutes that replicate both the rigid stenting of cartilage rings and the nutritional support of vascularized fascia. However, conventional tissue engineering is hampered by scarce donor chondrocytes and lengthy cell expansion. Here, we show a rapid tracheal reconstruction strategy using three-dimensional printed porous minichannel scaffolds filled with cartilage granules, platelet-rich plasma, and adipose-derived stem cells. Short-term ectopic incubation enables the grafts to acquire native-like mechanical resilience and transmural vas

Copper single-atom carbon dots hydrogel resets oxidative and immune homeostasis for rheumatoid arthritis therapy.

Rheumatoid arthritis pathogenesis is perpetuated by a self-sustaining oxidative stress-immunodysregulation circuit. To address the limitations of conventional antioxidants, including unclear catalytic structures and single catalytic activity, we engineered copper single-atom carbon dots via topological transformation of sodium copper chlorophyllin. Encapsulated within a pH-responsive injectable hydrogel, this system achieves targeted delivery to inflamed joints, where it simultaneously scavenges superoxide anions and hydrogen peroxide, breaking the reactive oxygen species-inflammation cycle. ENTR1 ameliorates osteoarthritis through decreasing FAS expression by binding PTPN13.

PURPOSE: This study aimed to explore the effects and mechanisms of endosome-associated trafficking regulator 1 (ENTR1) in osteoarthritis (OA), thereby providing a novel therapeutic target. METHODS: We established OA rat model via the destabilization of the medial meniscus (DMM) surgery and OA cell model via IL-1β-stimulated ATDC5 cells. HE and safranin O/fast green staining were employed to assess the pathological changes of cartilage tissues in rats. Moreover, TUNEL staining was applied to examine chondrocyte apoptosis in rat cartilage tissues. Protein and mRNA expressions were measured empl

Imaging in Rheumatic Diseases of the Head and Neck.

Multimodality imaging is essential for evaluating head and neck rheumatologic disease. Computed tomography (CT) can demonstrate osseous erosions, mineralization, and structural complications, including temporomandibular joint and cervical spine involvement in JIA/RA, crowned dens in pseudogout, and sinonasal or airway disease in giant cell arteritis (GCA) and relapsing polychondritis (RP). Ultrasound is useful for detecting synovitis in JIA/RA, auricular chondritis in RP, salivary gland abnormalities in Sjögren's syndrome, and vascular changes in GCA. FDG-PET/CT can identify metabolically acti

Surface marker-defined functional subpopulations of mesenchymal stromal cells: mechanisms, applications, and clinical translation.

Mesenchymal stromal cells (MSCs) hold considerable promise in tissue regeneration due to their multilineage differentiation, paracrine signaling, and immunomodulatory capabilities, as evidenced by thousands of clinical trials. However, clinical translation is constrained by inconsistent and unpredictable efficacy. This primarily stems from MSC heterogeneity, including differences in tissue origin, functional subpopulations, donor age and disease status, and phenotypic drift and functional decline during in vitro expansion. Traditional markers recommended by the International Society for Cell T

Malignant neoplasm of bone and articular cartilage mortality trends in the United States: A 25-year population-based serial cross-sectional study.

Malignant neoplasms of bone and articular cartilage (MNBAC) carry a disproportionately high mortality burden. Long-term mortality patterns may inform targeted public health interventions. This study examines MNBAC-related mortality among adults aged 25 years and older from 1999 to 2023. This study analyzed MNBAC-related mortality from 1999 to 2023 using the CDC WONDER Multiple Cause of Death database and International Classification of Diseases, Tenth Revision codes. Age-adjusted mortality rates (AAMRs) per 100,000 population were standardized to the 2000 US population. Trends were assessed us

Chondroitin sulfate in inflammatory joint diseases: How structural variability shapes its therapeutic effects.

Inflammatory joint diseases, including rheumatoid arthritis and osteoarthritis (OA), are characterized by pain, inflammation, and progressive cartilage degeneration. Chondroitin sulfate (CS) is widely used as a symptomatic treatment, but the influence of structural variability on its biological activity remains unclear. We compared two pharmaceutical CS preparations with distinct structural features: CS-A, predominantly 4-sulfated, and CS-C, predominantly 6-sulfated. Disaccharide composition was determined by chondroitinase AC digestion followed by SAX-HPLC and compared with previously reporte

Shear Strain Stiffening in Cartilage is Governed by a Mechanical Phase Transition.

UNLABELLED: Large shear deformation in cartilage can lead to tissue failure and injury, which are often precursors to diseases such as post-traumatic osteoarthritis. Under shear, cartilage strain-stiffens prior to failure, which makes stiffening an important indicator of changes to the loading of the extracellular matrix that gives rise to this tissue's properties. While a variety of stiffening mechanisms ranging from uncrimping to fiber-alignment have been explored, whether such mechanisms apply to the depth dependent shear strain stiffening in articular cartilage remains poorly understood. Soluble Nanomedicine Against Soluble Neurosensitizers to Effectively Relieve Osteoarthritis Pain and Joint Damage.

Targeting soluble neurosensitizers in the osteoarthritis (OA) joint cavity is a key strategy for alleviating OA pain. Soluble neurosensitizers are ubiquitous in the joint cavity and cause uncontrollable growth and sensitization of nociceptor axons therein. However, eliminating these soluble nerve growth factors (NGFs) remains a considerable challenge due to the diversity of neurosensitizers and the complexity of OA pathology. This study introduces an innovative soluble nanodrug (LAC)-based strategy to eliminate three key neurosensitizers (H+, NGF, and netrin-1) to simultaneously relieve OA-ass

Immune-metabolic convergence in skeletal remodeling disorders: shared osteoimmune, senescent and lipid mechanisms across aging-related and immune-mediated bone disease.

Bone and joint disorders of the aging musculoskeletal system are conventionally treated as separate clinical entities. This disease-centered view aids diagnosis but obscures the immune, inflammatory, senescent and metabolic programs they share. This Review compares five of them, grouped by etiology rather than by age: two aging-driven remodeling disorders, osteoporosis (OP) and osteoarthritis (OA); two immune-mediated arthritides in which aging modifies rather than causes the skeletal outcome, rheumatoid arthritis (RA) and psoriatic arthritis (PsA); and one focal, largely monogenic disorder in

Poly(hexamethylenebiguanide) hydrochloride induces craniofacial cartilage defects and alters Wnt signaling related gene expression in zebrafish larvae.

Poly(hexamethylenebiguanide) hydrochloride (PHMB) is widely used as a broad spectrum antimicrobial preservative, but its effects on craniofacial cartilage development require further characterization. Here, zebrafish were used to evaluate the developmental effects of PHMB following embryonic exposure to 0.15, 0.35, or 0.55 mg/L from 6 to 72 hours post fertilization (hpf). PHMB exposed larvae exhibited craniofacial cartilage malformations alongside general developmental abnormalities and altered locomotor behavior. These phenotypic changes were accompanied by increased reactive oxygen species (

Autoamputation of pelvic adnexa: two case reports.

BACKGROUND: Parasitic ovarian teratoma is a rare consequence of chronic torsion or auto-amputation of the adnexa. Delayed diagnosis may result in a free-floating pelvic mass without identifiable adnexal tissue. CASE DESCRIPTION: Respectively, two young women (23 and 29 years) presented with ignored pelvic pain a few months ago. Both had previously self-medicated with oral analgesics without further treatment. The ultrasound revealed an isolated solid-cystic pelvic mass with absent colour Doppler signals. No normal ipsilateral ovarian or tubal tissue was visualised. Both patients had no positi

Strengthening health systems to prioritise, provide care, and prevent osteoarthritis: a roadmap for global health policy.

By 2050, almost 1 billion people are expected to live with osteoarthritis-a disabling musculoskeletal condition with currently no cure. Despite its burden and costs, osteoarthritis remains largely invisible in national and global health agendas, is poorly integrated within service models for other chronic health conditions, and is frequently perceived as inevitable with ageing and associated with limited therapeutic options. This context drives an osteoarthritis identity problem that fuels frustration among those living with the disease and health-care professionals, perpetuates stigma and ine

Distinct biological and biomechanical features in jaw and knee cartilage contribute to their degenerative susceptibility.

The temporomandibular joint (TMJ) and knee joint are both prone to degenerative diseases such as osteoarthritis (OA); however, TMJ-OA generally manifests earlier than knee OA, with underlying mechanisms largely unknown. In this study, we explored potential explanations through comprehensive biological and biomechanical analyses in mice. Structurally, the mandibular condylar cartilage (MCC) exhibits prominent differences from tibial articular cartilage (AC) at occlusion establishment, evidenced by distinct extracellular matrix organization and enriched transcription factor expression toward the

Anatomy of the Head and Neck in Rheumatic Disease.

This article examines immune-mediated inflammatory diseases (IMIDs) and rheumatological diseases that affect the head and neck, with a focus on how immunopathogenesis drives anatomic and clinical manifestations. Advances in cytokine-targeted therapies have revealed disease-specific immune pathways that underlie tissue inflammation, cartilage destruction, and aberrant bone remodeling. These mechanisms explain characteristic changes in the temporomandibular joint, craniovertebral joints, larynx, salivary glands, and upper airway. Integrating molecular immunology with anatomy improves diagnosis,

Histologically Confirmed Adult Tracheomalacia at Autopsy After Single-Lung Transplantation.

A man in his 50s developed persistent ventilator dependence, recurrent pneumonia, and lung abscess after right single-lung transplantation, and later died following complications from native lung pneumonectomy. Autopsy confirmed the diagnosis of tracheomalacia, allowing detailed histological examination that revealed extensive cartilage degeneration, disruption, and fibrosis of the trachea. Because histopathological studies of adult tracheomalacia are extremely rare, this case provides valuable insight into its pathology.

A novel mouse trauma model for osteonecrosis of the femoral head.

Osteonecrosis of the femoral head (ONFH) is a devastating condition characterized by the disruption of vascular supply, progressive bone degeneration, and eventual collapse of the femoral head. While corticosteroid- and alcohol-induced ONFH models are well established, a reliable trauma-induced ONFH model in mice has not been previously described. However, traumatic ONFH is commonly seen in clinical settings-such as following FN fracture, dislocation, or hip trauma-but remains poorly understood at the mechanistic level due to a lack of appropriate small-animal models. To address this gap, we e

Sinensetin ameliorates osteoarthritis progression by modulating the NF-κB/NLRP3 signaling pathway and suppressing chondrocyte pyroptosis.

Sinensetin (SIN) is a natural flavonoid with potential anti‑inflammatory effects. This study investigated whether SIN ameliorates osteoarthritis (OA) and explored its underlying mechanisms. In vitro, SIN (0.05-1 μg/mL) protected human OA chondrocytes from IL‑1β‑induced viability loss. Transcriptome sequencing and molecular docking identified SERPINA3 as a candidate target of SIN. SIN reduced the expression of NLRP3, GSDMD, and ASC, inhibited pyroptotic morphology, decreased TUNEL‑positive cells and LDH release, and lowered IL‑1β and IL‑18 secretion in IL‑1β‑treated chondrocytes. Knockdown of S

Laryngeal Manifestations of Rheumatic Disease.

Rheumatologic diseases are systemic, immune-mediated disorders that frequently involve the larynx, producing manifestations that range from subtle dysphonia to life-threatening airway obstruction. Laryngeal involvement may precede systemic disease, progress despite adequate systemic control, or be the sole presenting feature of active disease. Diagnosis relies on flexible laryngoscopy with disease-specific serology and biopsy when indicated. Corticosteroids, immunosuppressants, and biologics such as rituximab form the foundation of medical therapy, while surgery ranges from serial endoscopic d
