A skeletally immature patient with Diamond-Blackfan anemia with Deferoxamine induced bone dysplasia: An analysis on diffusion tensor imaging.

Deferoxamine-induced bone dysplasia is a rare but clinically significant complication of chronic iron chelation therapy, characterized by metaphyseal abnormalities and growth disturbances. While conventional imaging demonstrates abnormal endochondral ossification, the extent of physeal involvement remains poorly understood. Diffusion tensor imaging (DTI) enables microstructural evaluation of physeal architecture and may provide additional insight beyond routine MRI. We report a 9-year-old skeletally immature male with Diamond-Blackfan anemia treated with chronic deferoxamine iron chelation the

Bioenergetic Materials for Tissue Regeneration: Modulating Metabolism to Promote Cellular Anabolism.

Tissue regeneration is an energy-demanding process that requires adequate ATP production to support cellular proliferation, biosynthesis, and tissue remodeling. Under pathological and age-related conditions, bioenergetic deficits impair intrinsic regenerative capacity by disrupting mitochondrial function, redox homeostasis, and anabolic signaling. Although conventional scaffold- and cell-based bioengineering strategies have advanced regenerative medicine, they generally do not directly address the metabolic dysfunction that limits tissue repair. Recently, bioenergetic materials (BEMs) have eme

Prediction of prognosis and treatment effect of cervical cancer by immunogenic cell death-related genes.

OBJECTIVE: Osteoarthritis is one of the chronic diseases with a high incidence. CircRNA is a circular non-coding RNA. Studies show that CircRNA is closely relevant to the pathogenesis of OA chondrocytes. However, the specific principle is still unclear. PATIENTS AND METHODS: 38 patients with OA tissues and 38 patients with normal knee cartilage in our hospital were selected, respectively. The mRNA expression levels of CircVCAN were measured by quantificational real-time polymerase chain reaction (qRT-PCR). Cell proliferation was detected by the Cell Counting Kit (CCK8). Cell cycle and apoptos

Genetic Mutation and Epigenetic Silencing Drive Antigen-Negative Relapse in CD7 CAR T-Treated T-cell Lymphoid Malignancies.

When suffering from osteoarthritis (OA), articular cartilage homeostasis is out of balance and the living quality declines. The treatment of knee OA has always been an unsolved problem in the world. At present, symptomatic treatment is mainly adopted for OA. Drug therapy is mainly used to relieve pain symptoms, but often accompanied with adverse reactions; surgical treatment involves the problem of poor integration between the repaired or transplanted tissues and the natural cartilage, leading to the failure of repair. Biotherapy which aims to promote cartilage in situ regeneration and to rest

Connection Between Mitochondria and Rheumatoid Arthritis and Osteoarthritis: Personalized Treatment Strategies.

This review explores the critical role of mitochondria in the immunometabolic processes underlying rheumatoid arthritis (RA) and osteoarthritis (OA). It examines the interplay between immune cells, metabolic demands, and tissue environments, emphasizing the impact of bioenergetics on immune responses and disease progression. Mitochondrial dysfunction in chondrocytes and immune cells contributes to OA and RA through mechanisms such as oxidative stress, disrupted calcium homeostasis, and inflammasome activation. In OA, mitochondrial dysfunction in chondrocytes results in impaired energy producti

A retrospective study of treatment and outcomes in synchronous systemic and central nervous system large B-cell lymphoma.

Synchronous systemic and de novo secondary central nervous system (CNS) large B-cell lymphoma (LBCL) is an aggressive clinical entity with a historically poor prognosis. Given the rarity of this presentation, prospective studies are limited, and treatment paradigms and outcomes are extrapolated from small, heterogenous retrospective studies. We performed a retrospective study with extended follow-up for 63 consecutive patients with previously untreated synchronous systemic and de novo secondary CNS LBCLs presenting to 2 institutions over 21 years. Most patients had diffuse LBCL (73%) and were

Bone marrow mesenchymal stem cells overexpressing FGF-2 loaded onto a decellularized extracellular matrix hydrogel for the treatment of osteoarthritis.

Osteoarthritis, as one of the major disabling diseases in the elderly, has a long-term impact on patients' quality of life and brings heavy medical and social burden. The pathogenesis of osteoarthritis is still unclear, and the main pathological changes include chondrocyte death and osteochondral damage. Therefore, how to solve the cartilage damage caused by osteoarthritis has become the key and difficult point in the clinical treatment of osteoarthritis. Bone marrow mesenchymal stem cells (MSCs) have the potential for self-renewal and multidirectional differentiation, and their engineering ha

Orthobiologics in 2025 and Beyond.

Orthobiologics are biologic substances intended to enhance healing and treat a range of musculoskeletal pathologies. Commonly well-studied orthobiologics such as platelet-rich plasma, bone marrow aspirate concentrate, and micro-fragmented adipose tissue have more than 2 decades of scientific research and clinical use, although with variable outcomes. It is important to discuss both current utilization and future applications of the aforementioned orthobiologics while elucidating some novel orthobiologics with recent regulatory approval, and finally, those orthobiologics on the horizon and beyo

Depth-Wise Changes in Tibial and Femoral Human Knee Joint Cartilage at Different Severities of Osteoarthritis.

Articular cartilage undergoes structural and compositional changes during osteoarthritis (OA), one of the most common joint diseases. Earlier research shows that these changes are dependent on species and joint site, and they also vary across cartilage depth. In this study, we analyzed the depth-wise proteoglycan and collagen contents, as well as collagen fibril orientation angle and fibril alignment in human tibial and femoral cartilage at different severities of osteoarthritis. Samples were divided into normal cartilage, moderate OA cartilage, and severe OA cartilage based on OARSI grade. Co
