Quercetin Alleviates Osteoarthritis Pain by Inhibiting Vascular Endothelial Growth Factor A Through Regulating cGAS/STING Pathway.

Knee osteoarthritis (KOA), a common degenerative joint disease marked by pain, inflammation and cartilage degradation, has been increasingly associated with dysregulated innate immune signalling. Among the implicated molecular pathways, cGAS/STING has emerged as a key modulator in both disease pathogenesis and therapeutic intervention. Quercetin, a naturally derived bioflavonoid with well-documented antitumour and antioxidant activities, also exerts notable anti-inflammatory and analgesic effects. This study investigated the mechanistic interaction between quercetin and the cGAS/STING pathway

Cldn11 deficiency aggravates osteoarthritis by inhibiting Notch signalling in a Tspan5-dependent manner.

As a prevalent degenerative condition affecting joints, osteoarthritis (OA) involves the gradual deterioration of articular cartilage. Impaired chondrocyte function is a major driver of osteoarthritis pathogenesis. In this study, we focused on deciphering the mechanistic basis of Tspan5 activity in the absence of the tight junction component Claudin11(Cldn11), utilizing male SD rats as the experimental model, which regulates the process of OA through the Notch signalling pathway. An animal model of OA was successfully established by injecting monosodium iodoacetate (MIA) into the knee joint ca

T-2 toxin induces degradation of chondrocytes extracellular matrix through the methyltransferase-like 3/m6A/Csf1 axis.

T-2 toxin can induce the development of cartilage diseases. N6-methyladenosine (m6A) modification mediated by methyltransferase-like 3 (METTL3) is involved in the occurrence and progression of bone-related diseases. However, the mechanism of action of METTL3 in T-2 toxin-caused cartilage injury has not been fully revealed. It is unclear whether METTL3 regulates cartilage injury by modulating its downstream target macrophage colony-stimulating factor (Csf1). Here, we aimed to investigate the mechanism by which the T-2 toxin induces cartilage damage. We performed histological staining, western b

Amphiphilic hemin-based nanoparticles with hypoxia tolerance and inflammation inhibition for osteoarthritis therapy.

Osteoarthritis (OA) remains a major clinical challenge, driven by persistent cytokine-based inflammation and poor regenerative capacity of hypoxic microenvironment and avascular cartilage. Despite recent advances, strategies that concurrently address inflammation and tissue repair remain elusive. Here, we report a dual-functional, injectable nanoparticle system based on amphiphilic hemin, a naturally derived hemoglobin prosthetic group, for the synergistic treatment of OA. Harnessing its amphiphilic properties, hemin self-assembles into nanocarriers capable of encapsulating hydrophobic anti-in

Tetrahedral Framework Nucleic Acid-Based Cepharanthine Attenuates TMJOA via cGAS-STING-NF-κB Pathway Modulation and Macrophage Reprogramming.

Temporomandibular joint osteoarthritis (TMJOA) is a degenerative joint disease characterized by cartilage degradation, subchondral bone remodeling, and chronic inflammation for which effective therapies remain lacking. In this study, we develop a cepharanthine-loaded tetrahedral framework nucleic acid nanoplatform (tFNA@Cep) and evaluate its physicochemical properties, biological performance, and therapeutic potential in TMJOA. The nanostructure exhibits excellent stability, favorable size distribution, and efficient cellular uptake, with preserved structural integrity under physiological cond

Alveolar distribution of ruxolitinib in pulmonary alveolar proteinosis developing during the treatment of chronic GVHD.

SUMMARY: Osteoarthritis (OA) and rheumatoid arthritis (RA) are chronic diseases characterized by persistent inflammation and autoimmune responses that affect the joints and other organs. Scientific evidence indicates that the therapeutic effects of mesenchymal stem cells (MSCs) are mediated through the release of soluble factors and extracellular vesicles (EVs), particularly exosomes. The release of microRNAs from MSCs holds substantial potential for cell-free treatment in OA and RA. METHODS: A comprehensive search was conducted on Web of Science, PubMed, Scopus, and Google Scholar to identif

Multilayer lipid vesicles loaded with deoxyribonuclease 1 for the degradation of neutrophil extracellular traps in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent immune cell infiltration and synovial inflammation. Inflammatory stimuli recruit and trigger neutrophils to release neutrophil extracellular traps (NETs), which exacerbate joint inflammation and promote cartilage destruction. While deoxyribonuclease 1 (DNase 1) can degrade the DNA backbone of NETs, its therapeutic potential is hindered by rapid systemic clearance and poor bioavailability. To address these limitations, we develop multilayer lipid vesicles (MLVs) encapsulating DNase 1 (DMLVs) for NET degradati

The association between life's crucial 9 and osteoarthritis in US adults: the mediating role of central adiposity.

BACKGROUND: Osteoarthritis (OA), a prevalent degenerative joint disorder characterized by progressive cartilage loss and secondary low-grade inflammation, is recognized to be associated with cardiovascular diseases (CVD). Building upon the cardiovascular assessment framework of Life’s Essential 8 (LE8), emerging guidelines propose the integration of psychological health metrics (Patient Health Questionnaire-9, PHQ-9) into the expanded Life’s Crucial 9 (LC9) model. While LC9 demonstrates cardiovascular predictive capacity, its role in OA pathogenesis remains inadequately substantiated. METHODS:

Leveraging single cell multiomic analyses to identify gene regulatory networks that drive human articular cartilage cell fate.

OBJECTIVE: Compromises to the integrity of articular cartilage, whether genetic, post-traumatic or age-related, lead to debilitating chronic degenerative diseases including osteoarthritis. In this study, our objective was to leverage multiomic and spatial transcriptomic datasets to identify gene regulatory networks that drive human articular cartilage cell fate, and build a foundation from which novel therapeutics to overcome degenerative disease could be developed. DESIGN: We jointly profiled the transcriptome and open chromatin regions in individual nuclei recovered from distal femora at 2

PFHxS exposure causes osteoarthritis through the innate immune signaling pathway mediated by lipid peroxidation.

PFHxS have been widely used and detected in the environment. The toxicological effects of PFHxS exposure on cartilage tissues and cells are not fully clear. The objective of this work was to study the toxicological effects of PFHxS on cartilage. We investigated these effects using both in-vitro and in-vivo models. In the in-vitro model, we exposed chondrocytes to environmentally relevant concentrations of PFHxS. In the in-vivo model, we assessed cartilage damage using various staining methods, including toluidine blue, alcian blue, and safranin-fast green staining. In chondrocytes, PFHxS expos

The role of neprilysin in musculoskeletal diseases.

Neprilysin (NEP) is a membrane-bound endopeptidase that degrades various substrates, including natriuretic peptide, bradykinin, and substance P. It is distributed in various organs such as the heart, kidneys, and brain, regulating multiple physiological processes. This paper elucidates the role of NEP in osteogenic, chondrogenic, and myogenic differentiation processes, along with its time-specific expression during differentiation. We focus on how NEP regulates key pathways in skeletal muscle disorders, explores potential mechanisms, and demonstrates clinical symptom improvement. NEP degrades

Innovative applications and future challenges of shape memory scaffolds for functional reconstruction in diseases of the musculoskeletal system.

Musculoskeletal disorders (MSDs), such as osteoporosis, cartilage degeneration, and tendon injuries, are common worldwide and have a significant impact on patients' quality of life. These conditions are frequently associated with aging, trauma, or chronic diseases. They are typically characterized by slow and limited self-repair, particularly in tissues like cartilage and tendons, which exhibit low regenerative potential. Traditional treatment methods, such as implantable scaffolds, often face challenges related to incomplete tissue integration and inadequate adaptation to dynamic biomechanica

Dynamic Compression System Bracing as a Treatment Option for Chondromanubrial Pectus Carinatum.

IntroductionChondromanubrial pectus carinatum (PC) is a rare type of pectus deformity in which there is protrusion of the manubrium and superior costal cartilage. Studies have shown that orthotic bracing is successful in correcting chondrogladiolar PC, but surgery has been the primary treatment for the chondromanubrial subtype. Due to the rarity of this type of PC, we aimed to describe our experience and outcomes with the dynamic compression system (DCS) bracing in these patients.MethodsThis is retrospective single center review of all patients with chondromanubrial PC treated with DCS from 20

Gαs variant detection discerns skeletal cell types involved in fibrous dysplasia pathogenesis.

Fibrous dysplasia (FD) is a mosaic disorder caused by activating Gαs variants. Skeletal stem cells give rise to abnormal osteoprogenitors, resulting in fibro-osseous lesions with prominent osteoclastogenesis. Gαs variants are reliably demonstrated in osteoprogenitors, however involvement in other skeletal cell types is not well-characterized. We evaluated human specimens to investigate Gαs variant expression in chondrocytes and osteoclasts, two prominent cell types in FD lesions. Seven of 20 bone specimens had cartilage present in addition to typical fibrous tissue. Droplet digital polymerase

Nell-1 regulates fibrocartilage stem cells and alleviates temporomandibular joint osteoarthritis progression by inhibiting hedgehog pathway.

BACKGROUND: Nel-like molecule-1 (Nell-1) could prevent the damage of cartilage in osteoarthritis (OA), including temporomandibular joint osteoarthritis (TMJOA), though, the definite mechanism of the process is unclear. Fibrocartilage stem cells (FCSCs) were discovered in the temporomandibular joint (TMJ), and were associated with the TMJ health and disease. However, little information is available on the effect of Nell-1 on FCSCs fate in TMJOA progression. METHODS: In this study, TMJOA was induced in rabbits through collagenase injection. 8 weeks after injection, the left and right joints of

Preserving scientific integrity in academic publishing: Navigating artificial intelligence, journal policies and the impact factor as a quality indicator.

The integration of artificial intelligence (AI), the rise of mega-journals, and the manipulation of impact factors present challenges to scientific integrity. These trends threaten the core principles of objectivity, reproducibility, and transparency. This editorial highlights two categories of threats: (1) external pressures, such as AI misuse and metric-driven publishing models, and (2) internal systemic flaws, including the 'publish or perish' culture and methodological fragility. Mega-journals, characterized by high-volume publishing and broad interdisciplinary scopes, improve accessibilit
