Therapeutic Effect of Ethanol Extract of Propolis Against Osteoarthritis in a Mouse Model via Modulation of NF-κB and Hippo Signaling Pathways.

Osteoarthritis (OA), a chronic degenerative joint disease, leads to disability and increases the health burden. This study identified the chemical components in the ethanol extract of propolis (EEP) and investigated its effects and mechanism on monosodium iodoacetate-induced OA in mice. Three phenolic acid esters, alongside 16 flavonoids and their derivatives, were tentatively characterized in EEP using HPLC-MS, and four main components were quantified. EEP treatment alleviated pathological changes in articular cartilage and reduced serum CTX-II levels in OA mice. Network pharmacology and mole

Clinical relevance of patient-reported outcome measures in the surgical management of focal chondral defects of the knee: a systematic review.

INTRODUCTION: To evaluate clinical outcome following surgical management of focal chondral defects in the knee, patient-reported outcome measures (PROMs) are used. To give these measures meaning, parameters such as the minimal clinically important difference (MCID), patient-acceptable symptom state (PASS), minimally detectable change (MDC), clinically important difference (CID) and substantial clinical benefit (SCB) have been introduced. This systematic review investigated the MCID, SCB, CID, PASS and MDC of the most commonly used PROMs for assessing patients following surgical repair of focal

PARP-1 prevents osteoarthritis pathogenesis by inhibiting apoptosis in chondrocytes: an animal study.

BACKGROUND: Osteoarthritis (OA) is a degenerative joint disorder characterized by cartilage breakdown, synovial inflammation, and subchondral bone changes. Chondrocyte apoptosis is a key contributor to cartilage degradation in OA. Poly(ADP-ribose) polymerase-1 (PARP-1), an enzyme involved in DNA repair and apoptosis regulation, is associated with various degenerative diseases. However, its role in OA pathogenesis remains unclear. Therefore, we aimed to investigate the role of PARP-1 in OA pathogenesis using PARP-1 knockout (KO) mice and primary chondrocytes. METHODS: OA was induced in wild-typ

Skeletal interfaces in health and disease - Where two worlds interact: The Salzburg Leopoldina Symposium 2025 - Bridging disciplines and perspectives.

INTRODUCTION: The Leopoldina Symposium on "Skeletal Interfaces in Health and Disease" at Paracelsus Medical University (Salzburg, Austria) gathered an interdisciplinary community of young and experienced scientists, to explore how crossing disciplinary boundaries can generate new insights and methodologies. While research traditionally focuses on organ- or disease-specific questions, this symposium aimed to connect seemingly distant scientific fields by exploring interfaces, pivotal zones of separation and integration. It intended to foster broader understanding of musculoskeletal biology and

Preserving scientific integrity in academic publishing: Navigating artificial intelligence, journal policies, and the impact factor as a quality indicator.

UNLABELLED: The integration of artificial intelligence (AI), the rise of mega-journals, and the manipulation of impact factors present challenges to scientific integrity. These trends threaten the core principles of objectivity, reproducibility, and transparency. This editorial highlights two categories of threats: (1) external pressures, such as AI misuse and metric-driven publishing models, and (2) internal systemic flaws, including the 'publish or perish' culture and methodological fragility. Mega-journals, characterized by high-volume publishing and broad interdisciplinary scopes, improve

Bone sialoprotein: a multifunctional regulator of bone remodelling and tumour progression.

Bone sialoprotein (BSP) is a major non-collagenous protein of the bone extracellular matrix and an important regulator of bone formation and resorption. BSP is produced by bone cells and chondrocytes and present in the bone matrix, cells, dentin and cartilage. However, its aberrant expression in primary tumour tissues and the sera of cancer patients with metastases implicates BSP in tumour biology and progression. The Arg-Gly-Asp (RGD) motif of BSP may be crucial not only for the attachment of metastasising cells to the bone surface but also for tumour growth, survival and activity. This revie

Missense mutation of BMP1 may cause feline osteogenesis imperfecta without bone deformity.

A 3-mo-old domestic shorthair cat was presented with multiple fractures. Bone morphology was normal radiographically, with no long bone deformity or increased bone translucency. A bone biopsy from the ilium was examined histologically, revealing that bone matrix in the trabeculae extended from the growth plate, but cartilage remained in the distal trabeculae. Osteoblasts were observed at the bone surface via immunohistochemical detection with an anti-RUNX2 antibody. Whole-genome sequencing identified a homozygous missense mutation (valine to methionine) in the zinc-dependent metalloprotease do

Denosumab-functionalized nanoparticles target macrophages to mitigate age-related osteoarthritis and osteoporosis.

Osteoarthritis (OA) and osteoporosis (OP) are prevalent degenerative diseases in the elderly population, often coexisting and sharing a common pathogenic mechanism involving the receptor activator of nuclear factor κB ligand/receptor activator of nuclear factor κB (RANKL/RANK) signaling pathway. Denosumab (DS) has shown efficacy in treating OA and OP. This study aimed to investigate the mechanism through which DS mitigates age-related OA-OP by regulating macrophage polarization. Due to the poor targeting ability of DS to macrophages, we have prepared folate-modified liposomes (Lip-FA) loaded w

Antibody-conjugated mesenchymal stromal cell drug delivery system for the treatment of autoimmune diseases in mice.

Autoimmune diseases (AIDs), such as psoriasis and rheumatoid arthritis, are driven by immune dysfunction, leading to chronic inflammation and tissue damage. Mesenchymal stromal cells (MSCs) possess immunomodulatory and tissue repair properties; however, the therapeutic applicability of MSCs faces limitations due to the low homing efficiency upon systemic infusion. Inspired by antibody-drug conjugates (ADC), here we develop an antibody-conjugated MSC-based drug delivery system (AcM-DDS) that combines CD4 monoclonal antibody-conjugated MSC (CD4-mBMSC) with liposome-encapsulated Cedirogant, a ROR

Signaling network exploration of microRNA140-5p in response to TMJ-OA pathological changes.

MicroRNA140-5p has been implicated in temporomandibular joint osteoarthritis (TMJ-OA), though its precise mechanistic role remains unclear. This study investigates the molecular mechanisms underlying microRNA140-5p-mediated inflammatory responses in TMJ-OA. In vitro, mandibular condylar chondrocytes (MCCs) were treated with IL-1β and transfected with small interfering RNA (siRNA) targeting Smad3, a direct target of microRNA140-5p. Expression of inflammatory cytokines was assessed via Western blotting and RT-qPCR. In vivo, a TMJ-OA model was established in Sprague-Dawley (SD) rats, followed by

FSCN1 induces subchondral bone sclerosis in osteoarthritis via modulating actin cytoskeleton dynamics and YAP signaling.

BACKGROUND: Subchondral bone is a dynamic tissue maintained by bone remodeling and responded rapidly to mechanical loading, which is strongly associated with cartilage degradation in osteoarthritis (OA), but the underlying mechanisms of which is still blurring. METHODS: Experimental OA mice models were generated by mechanical overload with 60 cycles of 14 N axial compressive loads twice a week or anterior cruciate ligament transection surgery. FSCN1 expression was evaluated in subchondral bone from experimental OA mice and human OA. Mice with FSCN1 conditional knockout in pre-osteoblasts were

Small molecule inhibition rescues the skeletal dysplasia phenotype of Trpv4 mutant mice.

The TRPV4 skeletal dysplasias are characterized by short stature, short limbs with prominent large joints, and progressive scoliosis. They result from dominant missense mutations that activate the TRPV4 calcium permeable ion channel. As a platform to understand the mechanism of disease and to test the hypothesis that channel inhibition could treat these disorders, we developed a knock-in mouse that conditionally expresses the p.R594H Trpv4 mutation. Embryonic, chondrocyte-specific induction of the mutation using Col2a1-Cre resulted in a skeletal dysplasia affecting the long bones, spine, and c

Biomimetic Multifunctional Scaffolds for Osteochondral Regeneration: Bridging Material Design and Functional Integration.

In recent years, the increasing prevalence of osteochondral defects has highlighted the critical need for effective repair strategies. The complex anatomical interdependence between bone and cartilage poses significant challenges in achieving synchronized tissue regeneration and functional reconstruction. Osteochondral tissue engineering (OCTE) has emerged as a promising solution, particularly through the development of integrative repair systems. Beyond meeting mechanical requirements and biodegradability standards, contemporary osteochondral scaffolds must achieve seamless biological integra

Macrophage polarization: A bridge connecting osteoarthritis and osteoporosis.

Macrophage polarization plays a pivotal role in maintaining bone homeostasis and serves as a crucial mechanistic bridge in the comorbidity of osteoarthritis (OA) and osteoporosis (OP). Macrophages exhibit functional plasticity, polarizing into pro-inflammatory M1 and anti-inflammatory M2 phenotypes in response to various environmental cues. Recent advances in macrophage biology have highlighted the significant heterogeneity of macrophages, which display a spectrum of phenotypic states beyond the classical M1/M2 dichotomy. Dysregulated macrophage polarization, particularly an excess of M1 macro

Mechanisms and Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles as Nanocarriers in Musculoskeletal Disorders.

The musculoskeletal system consists of bones, joint cartilage, tendons, ligaments, muscles, and their associated nerves and blood vessels. These components work together to maintain human movement and mechanical stability, serving as the critical foundation for sustaining life activities. In recent years, with the acceleration of population aging, the incidence of musculoskeletal-related diseases such as sarcopenia, muscle atrophy, osteoporosis (OP), osteoarthritis (OA), and tendon injuries has continued to rise, significantly impacting patients' quality of life and imposing a heavy social hea

In-situ RhoA editing via heparinylated LNP-microsphere system for rheumatoid arthritis treatment.

Macrophages play a pivotal role in modulating inflammation and osteoclastogenesis during the progression of rheumatoid arthritis (RA). Although local mRNA-mediated gene editing offers high precision and efficiency, there remains a lack of robust strategies tailored for effective editing of synovial macrophages. Here, we identified RhoA as a macrophage target via bioinformatics, validated its therapeutic potential, and built a heparinylated LNP-microsphere system (hLNP-RhoA−/−@MS) to replace PEGylated LNPs for in situ RhoA editing. Specifically, a low-molecular weight heparin-cholesterol conjug

Clinical applications of UTE-T2* in knee MRI.

Many of the tissues of interest in the evaluation of the knee by magnetic resonance imaging (MRI), including subchondral bone, deep calcified layer of cartilage, menisci, tendons, and ligaments, have very short transverse (T2 and T2*) relaxation times related to their intrinsic structure. These tissues appear anechoic on conventional MRI sequences as signal has already decayed to its minimum when image acquisition begins. Only in the setting of significant injury or degeneration is there detectable signal on conventional MRI sequences. Ultrashort echo time (UTE) MRI, which allows for the quali

Knee muscle strength as a mediator of sex differences in incident knee osteoarthritis.

BACKGROUND: Females have a higher risk of incident knee osteoarthritis (KOA) than males, particularly in the lateral compartment; however, the underlying mechanisms remain unclear. We investigated whether knee flexor and extensor strength mediate the association between sex and lateral radiographic and symptomatic KOA risk. METHODS: This cohort study utilized data from the Osteoarthritis Initiative, a longitudinal prospective study of participants aged 45–79 years. We included knees without radiographic or symptomatic KOA at baseline. Knee extensor and flexor strength were measured isometrical

Low ozone concentrations promote in vitro preservation of explanted articular cartilage: an ultrastructural study.

Ozone (O3) is an oxidizing natural gas widely applied as adjunctive therapeutic treatment for a variety of pathological conditions. Currently, O3-based therapies rely on the low-dose concept i.e., the administration of low O3 concentrations able to induce a mild oxidative stress stimulating antioxidant and anti-inflammatory response without causing cell damage. In addition, low O3 concentrations are thought to activate cellular and molecular mechanisms responsible for analgesic and regenerative effects. Due to these properties, in the last decade interest has arisen in the fields of orthopedic
