ZFP36 Protects against Abdominal Aortic Aneurysm Formation by Regulating Vascular Smooth Muscle Phenotypic Switch.

Cryptosporidiosis is a zoonotic disease caused by a well-known parasitic protozoan called Cryptosporidium. Infection in livestock causes important economic losses among farm animals and its control has a global concern. In this study, internal white and external red layers were separated from pomegranate peels (Punica granatum) then; they were grinded to reach Nano form. Anticryptosporidial effect of their water extracts was investigated in experimentally infected mice. Also, their antioxidant activity, biochemical and histopathological changes were studied. Briefly, hot aqueous extracts of po

Lysosome-Bursting Calcium Carbonate Nanoparticles Amplify Immunogenic Cell Death via Synergistic Phototherapy and Calcium Overload for Immune Checkpoint Blockade.

OBJECTIVE: An undetectable postoperative serum cortisol has been regarded as a definition of cure in Cushing's disease. However, we noted disease recurrence amongst patients with Cushing's disease despite undetectable postoperative cortisol levels, and this led us to audit our data. We have also previously assessed surgical outcome for acromegaly and microprolactinoma for a single surgeon. The aims of this study were two-fold: (i) to investigate the treatment and surgical outcome of patients with Cushing's disease. In particular, we wished to compare the data with outcome for other pituitary t

Antioxidative Defence Mechanisms in Tomato Plants Induced by Biopriming with Trichoderma virens and Jasmonic Acid Against Fusarium Wilt and Damping-Off.

In plants, pathogenic infection causes oxidative damage to the host plants and eventually leads to the destruction of the plant's metabolism due to the oxidation of essential biomolecules. In this investigation, we monitored the effects of fusarium wilt and damping-off mediated generation of Reactive oxygen species (ROS) and their effective disposal by the antioxidative defence system in tomato plants. The main aim of the current research work was to assess the seed priming remedial effects of Trichoderma virens (Tv) (1 × 107 spores/mL) as a biocontrol agent and jasmonic acid (JA) (1 μM) as a

DNA injection through coleoptile; a choice to overcome species barrier and bring about rapid expansion of gene pool in rice.

INTRODUCTION: Femoral neck fractures are among the most common injuries in the elderly, and total hip arthroplasty is increasingly preferred due to superior functional outcomes and lower reoperation rates; however, instability and dislocation remain major concerns with conventional fixed-bearing total hip arthroplasty. Dual mobility total hip arthroplasty has been introduced to reduce these complications. METHODS: This retrospective analytical study included 335 patients aged over 55 years with AO/OTA type 31B or 31C femoral neck fractures who underwent total hip arthroplasty between January

Cryptosporidium spp. in Mink, Foxes, and Raccoon Dogs in Five Provinces of Northern China.

Cryptosporidium spp. is a common intestinal pathogen that causes diarrhea in both humans and animals. Northern China has a large-scale fur animal farming industry. Although there have been related studies, systematic data remain lacking. In this study, a total of 871 fecal samples were collected from fur animals in Shandong, Hebei, Liaoning, Jilin, and Heilongjiang provinces. Using nested polymerase chain reaction, 75 Cryptosporidium-positive samples were detected, and the overall infection rate was 8.61%. The infection rates ranged from 4.23% to 16.81% across different provinces, with the hig

Cryptosporidium Diagnostic Assays: Microscopy.

Stained microscopy of fecal smears was the cornerstone of Cryptosporidium diagnosis for many years and provides a low-cost method for detecting oocysts. The development and commercialization of improved enzyme immunosorbent assays (EIAs) for coproantigen detection provided an automatable method for mass testing and rapid diagnostics when incorporated onto a cartridge format. Similarly, immunochromatographic lateral flow (ICLF) assays enable rapid diagnostics. Nevertheless, it is important that positive reactions by EIAs or ICLF assays are confirmed. Here, we describe microscopical methods usin

Cryptosporidium Diagnostic Assays: Molecular Detection and Species Determination by Real-Time PCR-Based Assays.

Molecular diagnostic assays for Cryptosporidium are usually based on polymerase chain reaction (PCR) and may detect the entire genus or target-specified species. Of the ever-growing number of species, fewer than half have been reported from humans, and most human cases of cryptosporidiosis are caused by Cryptosporidium parvum or Cryptosporidium hominis. Here, we describe a real-time PCR for the detection of all Cryptosporidium spp. that can then be differentiated by sequencing the PCR amplicons and a duplex real-time PCR for the simultaneous detection and differentiation of C. parvum and C. ho

Genomic Characterization of Cryptosporidium spp. via iNextEra Library Preparation and Hybridization Bait Capture.

Next-generation DNA sequencing (NGS) is used to study the genome sequences of Cryptosporidium spp., but NGS is challenging when pure Cryptosporidium oocysts are limited in number or not available. Varying levels of parasites present in fecal samples, combined with the abundance of host cells, bacterial and other microbial cells, and undigested food particles, often result in fecal DNA samples with ~0.1% Cryptosporidium DNA, making genome-scale sequencing of Cryptosporidium from such samples cost-prohibitive. DNA extractions from fecal samples are, however, widely available and commonly used fo

Preparation of Cryptosporidium for Whole-Genome Sequencing.

The application of whole-genome sequencing (WGS) to Cryptosporidium has been challenged by the relatively small numbers of oocysts present in feces and lack of suitable culture systems to generate sufficient DNA that is required. Here, we provide a method for the preparation of sufficient, high-quality Cryptosporidium DNA for WGS from feces, incorporating parasite purification stages and an optional whole-genome amplification step.

A Real-Time PCR-Based Method for the Sequence Determination of Cryptosporidium hominis and Cryptosporidium parvum gp60 Subtypes During Epidemiological Investigations.

The gp60 gene has been widely used to subtype Cryptosporidium for many years. Predominantly, this has involved nested polymerase chain reaction (PCR) amplification, gel electrophoresis, and Sanger sequencing of the amplicons. However, laboratories may no longer have gel electrophoresis capability, and during outbreak investigations a more timely approach may be required. Here, we describe a real-time PCR assay combined with high-resolution melt curve analysis that streamlines the PCR detection of gp60 amplicons of Cryptosporidium parvum and Cryptosporidium hominis for sequencing during public

Cryptosporidium Genotyping for Epidemiology Tracking.

Cryptosporidium genotyping has made significant contributions to the species structure and population genetics of Cryptosporidium spp. In addition, the standardized method has shown utility in epidemiologic investigations identifying case linkages and contamination sources that could not be determined with traditional epidemiologic tools. The standardized method for Cryptosporidium genotyping from stool specimens described here comprises multiple individual protocols to amplify and sequence regions of the small subunit ribosomal ribonucleic acid (SSU rRNA) and gp60 genes of Cryptosporidium.

A Multilocus Variable-Number Tandem-Repeat Analysis (MLVA) Scheme for Epidemiological Investigations of Cryptosporidium parvum.

A multi-locus variable-number tandem-repeat analysis (MLVA) scheme has been validated and used for subtyping Cryptosporidium parvum, to infer linkage between cases, and suspected sources, in outbreaks and for epidemiological surveillance. Here, we describe the multiplex polymerase chain reaction (PCR) assays, fragment sizing, and bioinformatics to identify alleles and create a seven-locus MLVA profile for each specimen. The PCRs to produce amplicons for Sanger sequencing to confirm new alleles are also described.

Evolutionary Genomic Analyses of Cryptosporidium.

The increasing amount of whole-genome sequencing (WGS) data allows us to study the evolution and transmission of Cryptosporidium spp. at the population level. Population genomics is helping us to understand the genetic structure and adaptive evolution of Cryptosporidium and the mechanism and determinants of emergence of hypertransmissible subtypes and variants. The integrated pipeline described here provides an example of the use of WGS data for evolutionary genomic analysis of Cryptosporidium.

A Novel Method to Silence Genes in Cryptosporidium.

A major bottleneck for developing new drugs and vaccines against cryptosporidiosis has been the lack of methods to study gene function in this parasite. Silencing of genes using small interference RNA (siRNA) is a powerful methodology to investigate gene function in multiple organisms and has been widely used to identify drug targets for multiple pathogens. Cryptosporidium does not possess the enzymes of the classical siRNA pathway. Therefore, its genes cannot be silenced by standard siRNA technology. To circumvent that problem, we developed a strategy to knock down Cryptosporidium mRNA by rec

Genetic Manipulation of Cryptosporidium parvum.

Cryptosporidium parvum can be reliably genetically manipulated using CRISPR/Cas9 driven homologous repair coupled to in vivo propagation in immunodeficient mice. This chapter will guide through foundational procedures for excystation, transfection, infection, collection, and purification of transgenic Cryptosporidium parvum. The genetic tools for Cryptosporidium research were expanded significantly over the last 5 years. While we cannot cover all advances in detail, we will highlight novel selection markers, conditional mutagenesis strategies, and genetic crosses.

Mouse Models for Use in Cryptosporidium Infection Studies, Quantification of Parasite Burden Using Flow Cytometry, qPCR and Histopathology, and Confocal Imaging of Oocysts.

Cryptosporidiosis threatens the lives of young children in developing countries and newborn calves around the world. No vaccine or therapy can prevent or cure this diarrhea-inducing enteric disease caused by Cryptosporidium spp. protozoan parasites. There is an essential need to discover new therapeutic drugs efficient in reducing parasite burden in infected individuals. Research therefore relies on reliable small animal models of cryptosporidiosis. Here, we present excellent mouse models which can efficiently mimic pathogenesis of human and bovine cryptosporidiosis. We also describe methods t

Calf Clinical Model of Cryptosporidiosis for Efficacy Evaluation of Therapeutics.

Cryptosporidiosis, caused by the apicomplexan parasite Cryptosporidium parvum, is a moderate-to-severe diarrheal disease, now recognized as one of the leading causes of morbidity and mortality in livestock globally and in humans living in resource-limited parts of the world, particularly those with AIDS or who are malnourished. This recognition has fueled efforts for the discovery of effective therapeutics. While recent progress in drug discovery has been encouraging, there are presently no acceptably effective parasite-specific drugs for the disease. The urgent need for new drug discovery or

High-Content Screening for Cryptosporidium Drug Discovery.

High-content screening (HCS) is a cell-based type of phenotypic screening that combines multiple simultaneous readouts with a high level of throughput. A particular benefit of this form of screening for drug discovery is the ability to perform the interrogation in a biologically relevant system. This approach has greatly advanced the field of drug discovery for cryptosporidiosis, a diarrheal disease caused by protozoan parasites of Cryptosporidium spp. These parasites are obligate intracellular parasites and cannot be cultured in vitro without the support of a host cell, limiting the options f
