Pathology of Lankesterella sp. infection in a cliff swallow from California, USA.

Lankesterella spp., apicomplexan coccidian protozoa historically associated with amphibians, are increasingly reported in birds. Here, we describe lesions associated with infection from, and depict phylogenetic relationships of, a Lankesterella sp. in a cliff swallow (Petrochelidon pyrrhonota) from California, USA. An 8-wk-old male, hand-raised, cliff swallow housed in an outdoor aviary had progressive nonspecific clinical signs for 12 d. The bird was euthanized because of poor prognosis. Histologically, histiocytic, lymphocytic, and plasmacytic myocarditis and necrotizing pectoral myositis we

Evaluation of a genetically attenuated Cryptosporidium parvum strain in the calf clinical model of cryptosporidiosis.

BACKGROUND: The protozoan parasite Cryptosporidium parvum is a leading cause of diarrhea and mortality in young children and neonatal calves. There are no effective drugs to treat cryptosporidiosis, and no vaccines are available to prevent infection or reduce transmission. The substantial global public health burden of cryptosporidiosis calls for urgent efforts to develop effective treatments and prevention strategies. This includes identifying and functionally characterizing new parasite targets and evaluating vaccine candidates in relevant animal models. Recently, we identified the important

Metagenomic analysis of the gut microbiota in Cryptosporidium-infected Tibetan sheep.

Cryptosporidium are important causative parasitic protozoa that cause gastrointestinal discomfort and diarrhea in humans and animals, posing a huge threat to public health. Ruminants serve as the main source of Cryptosporidium infection. However, the relationship between this intestinal parasite and host gut microbiota in Tibetan sheep remains almost unknown. In the present study, using nested PCR targeting the SSU rRNA gene, we detected Cryptosporidium in 9% (38/420) of fecal samples. The positive rate was significantly higher in 4-7 month-old lambs than in adult sheep. Infection of Cryptospo

Large-scale molecular epidemiological survey of Giardia and Cryptosporidium in Victoria, Australia (2020-2024), reveals novel subtypes and outbreak-associated lineages.

Cryptosporidium and Giardia are major causes of gastrointestinal illness globally. In Australia, cryptosporidiosis is nationally notifiable, yet molecular characterization is not routinely undertaken, limiting outbreak detection, source attribution, and zoonotic risk assessment. This limitation became apparent in 2024, when Australia experienced a 273% increase in cryptosporidiosis notifications, the third-highest rise globally. We conducted a molecular epidemiological investigation of Cryptosporidium and Giardia in fecal samples from humans with gastroenteritis in Victoria, Australia, between

The revised Cryptosporidium life cycle offers opportunities for fundamental and translational advances.

Cryptosporidium is a leading cause of severe diarrheal disease, for which effective treatments and vaccines are lacking. The past decade has seen breakthrough advances in how the parasite and disease can be studied in the field and the laboratory. This has led to an increasingly molecular understanding of how parasite protein secretion orchestrates invasion and host-parasite interactions and to a revision of the Cryptosporidium life cycle. Parasite sex emerges as an engine of genomic innovation, allowing the parasite to adaptively evolve host specificity, virulence, and drug resistance. The un

The triple threat of Cryptosporidium, Giardia, and Entamoeba infections in Nigerian children.

The epidemiological patterns and risk factors of three protozoan parasites, Cryptosporidium, Giardia, and Entamoeba, that cause diarrheal diseases in low-resource settings were investigated among children aged 10 years and below across ten states in Nigeria. Previous studies in Nigeria relied primarily on microscopy, a method with limited sensitivity for parasite detection, which underestimated the true burden of infection. We applied a real-time PCR technique on DNA extracted from 977 stool samples collected from children who answered a structured questionnaire in this cross-sectional survey.

Cryptosporidium within a One health framework: a comprehensive review of public health impact, environmental concerns, and emerging strategies for prevention and treatment.

Cryptosporidium species are globally distributed parasites and are a major cause of cryptosporidiosis, a diarrheal disease that disproportionately affects immunocompromised individuals and young children in low-resource settings. Cryptosporidium is widely regarded as a critical contaminant of drinking water and is strongly associated with an increased risk of waterborne disease, posing a serious threat to public health. Furthermore, agricultural environments can serve as sources of contamination with Cryptosporidium oocysts through fecal material originating from both humans and animals. Despi

First joint report on the prevalence and genotyping of Cryptosporidium spp., Giardia duodenalis, and Enterocytozoon bieneusi among hospitalized children in Inner Mongolia, China.

INTRODUCTION: Cryptosporidium spp., Giardia duodenalis, and Enterocytozoon bieneusi are zoonotic intestinal parasitic protozoa that occur globally and can cause varying degrees of diarrhea, especially in children. In immunocompromised individuals, these protozoa may lead to persistent diarrhea and even death. Most studies worldwide have focused on the occurrence of these protozoa in animals, while few have investigated the prevalence and genotyping of these three protozoans among hospitalized children in Inner Mongolia, China. METHODS: DNA was extracted from fresh fecal samples of hospitalize

Recent advances in Cryptosporidium secreted proteins.

As an apicomplexan parasite, Cryptosporidium possesses the conserved secretory organelles, micronemes, rhoptry and dense granules in addition to the unique small granules. These specialized organelles house various effectors that are secreted upon invasion, helping to establish cryptosporidiosis, a leading cause of severe diarrhea among children. Only recently has its proteome been defined, laying the foundation to unravel host-parasite interactions that are poorly understood in Cryptosporidium. Recent technical advances in genetic manipulation have helped elucidate the role of a few secreted

First identification of Cryptosporidium parvum subtype IIaA15G2R1 and two Eimeria species in the edible dormouse (Glis glis Linnaeus, 1766).

As part of the edible dormouse (Glis glis Linnaeus, 1766) monitoring program, faecal samples of seven animals from Montes do Invernadeiro Natural Park (Galicia, NW Spain) were collected following current guidelines for the ethical use of animals in research. Fresh faecal samples were individually processed using a diphasic concentration method and nucleic acids were extracted. PCR protocols were applied to amplify a fragment of the small-subunit ribosomal RNA (ssu-rRNA) gene of Eimeria as well as fragments of ssu-rRNA and 60-kDa glycoprotein (gp60) loci of Cryptosporidium spp. The hypertransmi

BEZ235 inhibits Cryptosporidium parvum infection in mice by targeting the PI3K-NF-κB-c-MYB/BCL2A1 axis to restore host cell apoptosis.

Cryptosporidium parvum (C. parvum) is a zoonotic protozoan that causes cryptosporidiosis, primarily characterized by watery diarrhea. To clarify the molecular mechanisms of C. parvum-mediated host cell apoptosis regulation and identify potential therapeutics, we examined apoptosis rates, caspase activation, and apoptosis-related protein expression in infected NCM460 cells, along with the involvement of the phosphatidylinositol 3-kinase-nuclear factor kappa B (PI3K-NF-κB) signaling pathway. We further evaluated the efficacy of BEZ235, a dual PI3K/mammalian target of rapamycin (mTOR) inhibitor,

Cryptosporidium parvum outbreak among volunteers of a licensed wildlife rehabilitation center housing raccoons-2024.

Zoonotic transmission of Cryptosporidium parvum (C. parvum) is often associated with ruminants. We report an investigation of cryptosporidiosis among persons who had contact with raccoons. In July 2024, two states identified 18 human cryptosporidiosis cases who volunteered at the same animal facility. One human specimen and two raccoon specimens tested positive for C. parvum. Molecular subtyping of the C. parvum (subtype IIaA15G2R1) suggested zoonotic transmission between raccoons, humans, or their shared environments. These findings can inform public health actions, developed in collaboration

Effective Treatment using Bumped Kinase Inhibitor 1708 in a Calf Clinical Model of Cryptosporidiosis.

Cryptosporidium parvum, an apicomplexan parasite and the causative agent of cryptosporidiosis, contributes to a high burden in mortality and morbidity for humans and livestock. Currently, there are no reliably successful parasite-specific treatments for the debilitating diarrhea associated with the infection. Bumped kinase inhibitors (BKIs), which selectively target parasite calcium-dependent protein kinases (CDPKs), have been shown to decrease infections caused by several medical and veterinary-relevant parasites, including Toxoplasma gondii, Plasmodium falciparum, and Cryptosporidium parvum.

Molecular Characterization and Zoonotic Potential of Cryptosporidium spp. and Enterocytozoon bieneusi in Wild Rodents and Sympatric Livestock in Xinjiang, China.

Cryptosporidium spp. and Enterocytozoon bieneusi are significant enteric pathogens that affect a wide range of animal hosts, posing potential zoonotic risks. Wildlife-livestock interfaces are increasingly recognized as key settings for pathogen spillover, yet molecular epidemiological data from Xinjiang, China, remain limited. In this study, 191 fecal samples were collected from wild rodents and sympatric artiodactyls on the northern slope of the Tianshan Mountains in Bortala Prefecture, Xinjiang. Nested PCR targeting the small subunit (SSU) rRNA gene of Cryptosporidium spp. and the internal t

Proactive access strategies for novel cryptosporidiosis therapeutics.

Cryptosporidiosis is a major cause of parasitic diarrhea in lower income countries disproportionately affecting young children, malnourished populations, and people with compromised immunity. While still underemphasized on the global health agenda, the disease is gradually gaining momentum for therapeutic development. However, the real-world impact of any new medicine will hinge on its effective use in regions that currently face significant access barriers. This Perspective asks what it will take to achieve access to new cryptosporidiosis treatments and outlines strategies to bridge scientifi

Phylogenomics reveals multiple evolutionary lineages of marine crustacean-infecting gregarine apicomplexans.

Apicomplexans are a diverse phylum of single-celled protists that are all obligate animal symbionts. The most well-studied members are severe pathogens of humans and livestock, such as the causative agents of malaria, cryptosporidiosis, toxoplasmosis, and coccidiosis, but comparatively little is known about their invertebrate-infecting counterparts, which are evolutionarily and ecologically important, but often overlooked. One of these groups is the Cephaloidophoroidea gregarine superfamily which is one of the most often-detected apicomplexan groups in molecular surveys of marine environments.
