The role of integrins in T cell-mediated resistance to Cryptosporidium parvum.

Cryptosporidium is a protozoan that infects epithelial cells of the small intestine and is a cause of diarrhea and death in immunocompromised individuals and malnourished children. Immunity to this parasite is mediated by an intestinal T cell response, which is generated in gut-associated lymphoid tissues and dependent on type 1 conventional dendritic cells (cDC1s). The initial priming of T cells is accompanied by changes in integrin expression and subsequent trafficking to the site of infection. The role of specific integrins in trafficking to the ileum during cryptosporidiosis is largely unk

Case Report: The complete radiological resolution of diffuse cholangitis in a HIV-positive patient with cryptosporidium infection after anti-retroviral therapy.

We present a case of an HIV-positive patient with AIDS cholangiopathy secondary to Cryptosporidium infection. Imaging examination showed intrahepatic and extrahepatic cholangitis without papillary stenosis and extrahepatic bile duct strictures, indicating mild bile duct disease. However, it failed to obtain positive results in fecal microscopy examinations. Alternatively, metagenomic next-generation sequencing (mNGS) of a blood sample identified Cryptosporidium infection. The diagnostic power of mNGS is highly sensitive and can simultaneously identify various pathogens. To avoid irreversible d

Molecular detection of Cryptosporidium spp. in cattle in southern Zhejiang Province, China: Prevalence and genetic characteristics.

Cryptosporidium sp. is a genus of parasitic protozoa that infects the gastrointestinal tracts of humans and animals, potentially leading to zoonotic transmission. A molecular investigation was carried out on Cryptosporidium spp. in cattle reared in southern Zhejiang Province, China, to elucidate its epidemiology. A total of 265 fresh cattle fecal specimens were collected and subjected to detection of Cryptosporidium spp. via polymerase chain reaction (PCR) and Sanger sequencing of its small subunit ribosomal RNA (SSU rRNA) gene. Specimens testing positive for C. parvum were further analyzed us

Systematic review and meta-analysis on the prevalence of cryptosporidiosis among children in India.

UNLABELLED: Diarrhoea is the second leading cause of death in children under five years in India. The overall prevalence of childhood diarrhoea in India stands at 7.3%. Cryptosporidium is the third major contributor to diarrhoea-related morbidity and mortality, associated with growth stunting, malnutrition, and cognitive dysfunction in children. Nevertheless, the contribution of Cryptosporidium to the diarrhoeal burden in India remains unclear, underscoring the need for a systematic review to address the existing knowledge gap. This systematic review aims to determine the overall pooled preval

Coccidian co-infections in immuno-compromised patients: Prevalence, clinical impact, and insights from a tertiary care centre.

Coccidian parasites are significant emerging contributors to the changing aetiological landscape of diarrhoeal diseases, primarily affecting individuals from lower socio-economic backgrounds. They pose a great threat to the immuno-compromised in whom co-infections may lead to considerable morbidity and mortality. Timely detection is essential for improving clinical outcomes; however, comprehensive data on associated risk factors, transmission routes, and socio-demographic patterns remain limited. Modified Ziehl-Neelsen staining confirmed the presence of co-infection of Cryptosporidium and Cycl

Structure-guided design of calcium-dependent protein kinase 1 (CDPK1) inhibitors for cryptosporidiosis.

BACKGROUND: Calcium-dependent protein kinase 1 (CDPK1) has emerged as a protozoan specific target for the treatment of cryptosporidiosis. A previous study identified pyridopyrimidinones as new Cryptosporidium parvum (Cp) CDPK1 inhibitors with potent growth inhibition against C. parvum and C. hominis. Docking analyses suggested the unique positioning of the kinase's αC-helix could present refinement opportunities. METHODS: Compounds designed to optimize the pyridopyrimidinones focused on the back-pocket region predicted to be proximal to the αC-helix, the solvent exposed region and the ATP rib

Evaluation of the effect of auranofin alone and in combination with nitazoxanide on Cryptosporidium parvum in immunosuppressed mice.

Cryptosporidium parvum (C. parvum), a zoonotic protozoan parasite with a worldwide distribution, causes life-threatening diarrhea in children and immunosuppressed individuals. Although Nitazoxanide (NTZ) is the only FDA-approved drug against C. parvum, it has shown unsatisfactory results. Auranofin (AUR), an FDA-approved drug for rheumatoid arthritis, has a robust anti-protozoal effect through thioredoxin reductase (TrxR) inhibition, inducing oxidative stress and reactive oxygen species (ROS) production, thus causing parasites' apoptosis. The purpose of the present study was to evaluate the ef

Downregulation of HNF4α Mediates Microvillus Damage During Cryptosporidium parvum Infection.

Cryptosporidium is a significant zoonotic protozoan parasite that primarily infects intestinal epithelial cells, causing microvillus damage and other gastrointestinal malfunctions. However, the precise molecular mechanisms underlying this process have yet to be elucidated. In this study, IFN-γ knockout mice were infected with a virulent Cryptosporidium parvum IId isolate, and intestinal pathological changes and microvillus alterations were monitored throughout the infection. The results showed that C. parvum invasion induced both intestinal and microvillus damage in the host. Ileum tissues wer

CpCML subverts host PI3K/AKT-NF-κB signaling and autophagic flux via direct interaction with SUGT1 to facilitate Cryptosporidium parvum infection.

Cryptosporidium parvum is a water and foodborne apicomplexan pathogen that vigorously disrupts host signaling networks to enable intracellular colonization. However, the parasite-encoded effectors responsible for this modulation and the specific host signaling cascades they involve is still unexplored. In this study, we identify CpCML, a calmodulin-like protein encoded by C. parvum, as a perilous virulence effector that modulates host PI3K-NF-κB signaling and autophagic flux through direct interaction with the host protein SUGT1. Biochemical and cellular analyses reveal that CpCML binds SUGT1

Zoonotic Cryptosporidium and Giardia infections in pigeons (Columba spp.) at a wildlife hospital in France: Occurrence and molecular identification.

The feral rock pigeon (Columba livia domestica) and wood pigeon (Columba palumbus) are ubiquitous species found in cities worldwide. Its interaction with humans, domestic animals and wild birds makes them potential carriers of transmissible pathogens, including zoonotic parasites. The aim of this study was to identify protozoan parasites belonging to the genera Cryptosporidium spp. and Giardia spp. in these two pigeon species hosted at the Wildlife Animal Hospital of the Veterinary School of Alfort, in France. Fecal samples from 367 pigeons (Columba livia and Columba palumbus) including a wide
