Investigation of acylcarnitine profile in naturally infected calves with Cryptosporidium parvum.

Cryptosporidium parvum (C. parvum) is one of the common infectious and zoonotic agents in neonatal calves. Carnitine is an important molecule of mitochondrial energy metabolism that can lead to metabolic and clinical disorders. This study aims to determine the carnitine and acylcarnitine profiles in calves infected with C. parvum, to evaluate their role in the pathogenesis of the disease, and to investigate their diagnostic effectiveness. A total of 32 calves were included in the study, 20 calves infected with C. parvum and 12 calves determined to be healthy, aged between 1 and 28 days. Blood

Unexpected Cryptosporidium species Identified from New York State Clinical Stool Specimens.

BACKGROUND: Cryptosporidiosis is predominantly considered a waterborne illness that occurs after becoming infected with Cryptosporidium parvum or Cryptosporidium hominis, the two species that typically infect humans. However, identifying non-C. parvum/C. hominis species that infect humans could reveal additional behaviors or exposures that contribute to cryptosporidiosis. Here, we identify species, non-C. parvum/C. hominis species causing infections and determine their distribution within New York State (NYS). METHODS: To do this, real-time Polymerase Chain Reaction (RT-PCR) was used to presc

Unveiling the Silent Regulators: Noncoding RNAs in Parasitic Disease Diagnosis and Therapy.

Parasitic diseases remain a major global health burden, particularly in tropical and low-resource regions, where limitations in early diagnosis and effective therapeutics contribute to high morbidity and mortality. Emerging evidence highlights noncoding RNAs (ncRNAs) as critical regulators of host-parasite interactions, offering novel opportunities for diagnosis, prognosis, and targeted therapy. However, most of these findings are derived from in vitro experiments and preclinical models with limited validation in clinical settings. This review comprehensively examines the roles of microRNAs (m

Status of cryptosporidiosis in cattle raised by marginalized nomadic communities of Southern Punjab, Pakistan: an observational study.

INTRODUCTION: Cryptosporidium (C.) is one of the most important causes of gastrointestinal disorders in various animal species, leading to significant economic losses in the dairy sector. The parasite has also significant zoonotic relevance, with well-documented cross-transmission between animals and humans. However, data on the status of cryptosporidiosis in scattered nomadic populations is scarce. Therefore, this study was conducted to determine the prevalence and associated risk factors of cryptosporidiosis in cattle raised by socioeconomically deprived nomadic communities in an important a

Stool antigen-based enzyme immunoassays: performance evaluations and automation.

Three enzyme immunoassays (EIAs) were evaluated on two platforms each for detecting Cryptosporidium, Giardia, and Campylobacter in stool samples compared to reference methods. The Cryptosporidium EIAs run on stool specimens in three preservative media showed 100% agreement with direct fluorescent antibody (DFA) testing and high correlation with the microscopic method. The Giardia EIAs demonstrated 100% concordance for positive, but only moderate overall correlation with the microscopic method. The Campylobacter EIAs had 93.5% sensitivity (out of 65 positives) and 100% specificity compared to t

Induction of β-Defensin 1 (DEFB1) via synthetic mRNA reduces Cryptosporidium parvum infection in human intestinal cells.

BACKGROUND: Cryptosporidiosis, caused by the protozoan parasite Cryptosporidium, is a leading cause of severe diarrhea, morbidity, and mortality in immunocompromised patients and malnourished children. Because the only FDA-approved treatment, nitazoxanide, shows limited efficacy in malnourished children and is ineffective in the immunodeficient, novel therapeutics for vulnerable populations are urgently needed. Host-directed therapy (HDT) is a promising strategy that utilizes the induction of endogenous protective molecules. For example, intestinal epithelial cells defend the mucosa by upregul

Survey of Cryptosporidium spp. detection in endangered ground geckos, Goniurosaurus spp. (Squamata: Eublepharidae) in Japan, with a preliminary assessment of cloacal swab sampling.

Kuroiwa's ground geckos (Goniurosaurus spp.) are endangered species endemic to the Ryukyu Archipelago, Japan, but the role of infectious diseases in their wild populations remains unclear. This study investigated the occurrence of Cryptosporidium infection in wild Goniurosaurus populations to provide baseline epidemiological information and preliminarily evaluated cloacal swab sampling as an alternative to fecal collection, given the low recovery rate of fecal samples in the field. Cryptosporidium varanii was detected at two of ten sites of G. kuroiwae (2/23 individuals) and at two sites of th

Prevalence, molecular characterization and zoonotic potential of Cryptosporidium spp. in horses from diverse husbandry systems across four provinces of northern and eastern China.

Cryptosporidium is an obligate intracellular zoonotic protozoan causing diarrheal disease in humans and animals globally. Horses have more frequent close contact with humans through equestrian sports, breeding, and cross-regional trade. However, comprehensive molecular epidemiological data on diverse equine husbandry systems in China are limited. We tested 412 fecal samples from horses across four Chinese provinces via small subunit ribosomal RNA (SSU rRNA) nested polymerase chain reaction (PCR). C. parvum and C. equi were subtyped using the 60 kDa glycoprotein (gp60) gene, and C. andersoni wa

Morin inhibits fructose bisphosphate aldolase activity and resists Cryptosporidium parvum infection.

Cryptosporidium parvum is a zoonotic intestinal protozoan that causes severe diarrhea in children and immunocompromised individuals. The lack of effective therapies underscores the urgent need for novel drug targets. As C. parvum primarily relies on glycolysis for energy, fructose-1,6-bisphosphate aldolase (FBA) represents a promising target. Here, we characterized CpFBA and evaluated its potential as a therapeutic target. Sequence analysis revealed that CpFBA contains a signature domain associated with the type I fructose-bisphosphate aldolase superfamily and adopts a tetrameric structure. Cp

Cryptosporidium in chickens during 2000 to 2025: A systematic review and meta-analysis of prevalence, species/genotypes distribution and zoonotic significance.

Cryptosporidium is an important avian pathogen. It can also be transmitted to humans through water sources contaminated with avian faeces or through direct avian-human contact. We conducted a meta-analysis to evaluate the global prevalence of Cryptosporidium in chicken. A total of 74 datasets from 21 countries/regions were used for the quantitative analysis. The prevalence of Cryptosporidium infection in chicken varied from 0.0% to 37.7%, with a summary estimate of 15.7% (3402/25,627; 95% confidence interval 14.0-17.4%). Nine Cryptosporidium species/genotypes were detected in chicken globally.

Mapping Prevalence, Diagnostics, and Evidence Gaps of Cryptosporidium in Southeast Asia Across Human, Animal, and Environmental Domains: Protocol for a One Health Scoping Review.

BACKGROUND: Cryptosporidium is a waterborne and zoonotic protozoan parasite that causes cryptosporidiosis, a diarrheal disease that disproportionately affects young children and immunocompromised individuals in low- and middle-income settings. In Southeast Asia, ecological diversity, agricultural intensification, and uneven sanitation infrastructure create overlapping transmission pathways across human, animal, and environmental domains. Despite a growing body of regional literature, the structure of the evidence base and its utility for surveillance and control have not been comprehensively e

Addition of multi-locus variable number tandem repeat analysis to assist Cryptosporidium parvum foodborne outbreak investigation - France, 2025.

Cryptosporidium spp. is a parasite that causes gastroenteritis in humans and is associated with waterborne and foodborne outbreaks. The international nomenclature for discrimination of Cryptosporidium isolates is based on sequence polymorphisms in the highly variable gp60 gene. In 2025, the number of human cases of cryptosporidiosis caused by infection with Cryptosporidium parvum gp60 genotype IIdA24G1 in France increased significantly and prompted public health authorities to investigate. During January through September 2025, 166 cases of infections with C. parvum genotype IIdA24G1 were iden

Efficacy of combination of Lacticaseibacillus rhamnosus and Bifidobacterium bifidum probiotic strains against mixed infections of bovine Rotavirus A and Cryptosporidium parvum induced diarrhea by modulating host immune response in mouse model.

Neonatal calf diarrhea is a serious health issue in dairy calves with significant economic losses. The immature gut immunity and the inadequate passive transfer of immunity in neonates favour the transmission and replication of the bovine rotavirus A (RVA) and the apicomplexan Cryptosporidium spp., and others, causing significant morbidity and mortality. To prevent such losses against these major pathogens, the efficacy of two-strain probiotics, namely Lacticaseibacillus (L.) rhamnosus strain NCDC 610 and Bifidobacterium (B.) bifidum strain BbVK3, were explored in a laboratory mouse model. Structural basis for the lack of immunogenicity of a Cryptosporidium octapeptide: anchor switching induces MHC-I groove remodeling and instability.

Cryptosporidium parvum is an intracellular protozoan parasite causes severe diarrheal disease, particularly in immunocompromised individuals, underscoring the importance of host immunity in controling infection. Here, we elucidate the structural basis for the poor immunogenicity of the C. parvum MEDLE2-derived octapeptide FLF8 (FLFLFENV) bound to the mouse H-2Kb molecule. X-ray crystallography of the H-2Kb-FLF8 complex revealed a non-canonical anchoring mode: the P1-Phe occupies the B and C pockets, bypassing the A pocket, while the P2-Leu inserts into the A and D pockets without forming hydro

Cryptosporidium hominis and Cryptosporidium parvum subtypes Identified from Patient Specimens in New York State.

BACKGROUND: Cryptosporidium is a pathogenic intestinal parasite that causes cryptosporidiosis, a diarrheal disease common in the US. The predominant species infecting humans are C. hominis and C. parvum. These species can be further classified into families and subtypes by analyzing the sequence of the 60-kDa glycoprotein (gp60) gene. Currently, there are no data available regarding the families and subtypes of Cryptosporidium found in New York State (NYS) or their distribution within the state. METHODS: To address these unknowns the gp60 gene was sequenced to identify the subtypes for C. hom

Azithromycin Mitigates Experimental Cryptosporidiosis-Driven Ileocecal Adenocarcinoma by Modulating Autophagy, Apoptosis, and PI3K/AKT Signaling.

Background/Objectives:Cryptosporidium parvum (C. parvum), a waterborne intestinal parasite, causes severe, persistent infections in immunocompromised hosts and has been linked to the onset of ileocecal adenocarcinoma. However, the molecular pathways linking chronic infection to carcinogenesis remain unclear. Nitazoxanide (NTZ), the only FDA-approved drug for this infection, shows limited efficacy. In contrast, azithromycin (AZM) possesses both antiparasitic and anticancer activity, though conclusive evidence supporting its effectiveness against cryptosporidiosis is still lacking.
