The Coalescence of a "Happy Accident" and an In Vitro Structure-Activity Panel: Increasing the Potency of a Cyclic Peptide Inhibitor to Thrombosis Initiation.

Cyclic peptides for novel drug discovery utilize the functional groups recognized by most biological targets but impose physical constraints as a preventative to degradation. Substituting non-natural, aromatic amino acids in cyclic peptide motifs can improve the ability of such peptides to bind to their protein targets and can further prevent instability. Here, we appended our previously described synthetic head-to-tail cyclized peptide with naphthylalanine (Nap) or biphenylalanine (BiPhe) substitutions in one or two positions. Our first-generation inhibitor targeted the protein-protein intera

En bloc removal of a giant renal calculus in a single sitting via transperitoneal laparoscopic pyelolithotomy: a safe and effective minimally invasive alternative.

Percutaneous nephrolithotomy remains the standard treatment for renal calculi larger than 2 cm; however, giant renal pelvic stones are often associated with the need for multiple tracts or staged procedures and carry risks of bleeding, sepsis, and residual fragments. We report the case of a 60-year-old man who presented with chronic left flank pain and burning micturition. Imaging demonstrated preserved renal function with a 5.2-cm solitary left renal pelvic calculus. The patient underwent transperitoneal laparoscopic pyelolithotomy using a 3-port approach, which enabled complete en bloc stone

The Influenza-Specific Immunoglobulin Repertoire - Nimble Yet Steeped in Tradition.

Influenza virus has circulated in the human population and co-evolved with us for centuries or longer, shaping our immune repertoires through a continuous host-virus arms race. Across individuals, antibodies repeatedly converge on shared solutions for capturing this shape-shifting virus by precisely targeting conserved sites of vulnerability-sites the virus cannot alter without functional cost-through structural features encoded directly by germline immunoglobulin genes. These recurring public antibodies, termed multi-donor class antibodies, have been pivotal for understanding human anti-influ

Understanding Pain Expression Using Visual Art: A Qualitative Study of the Artistic Pain Exploration Tool in Patients with Temporomandibular Disorder.

BACKGROUND: Pain associated with temporomandibular disorders (TMD) is multifaceted, often eluding conventional pain assessment tools that prioritize biomedical metrics over emotional and experiential dimensions. Arts-based research has shown promise in capturing the complexity of chronic pain, offering patients alternative modes of expression and clinicians' richer insight into subjective experiences. OBJECTIVE: This study explores how the Artistic Pain Exploration (APE) tool, a novel, art-based pain assessment method, supports patients with TMD in articulating their pain experiences across e

An Exploratory Study of the Short-Term Association of Resistance Training With Circulating Inflammatory Biomarkers, Plasma Brain-Derived Neurotrophic Factor, and Cognitive Function in Healthy Midlife Adults.

BACKGROUND: Systemic low-grade inflammation and subtle executive-function decline can emerge during midlife, yet short-term lifestyle approaches remain understudied. This single-arm interventional pre-post study examined whether a 4-week supervised resistance-training program was associated with changes in circulating inflammatory biomarkers, plasma brain-derived neurotrophic factor (BDNF), and executive-task performance in healthy midlife adults. METHODS: Five hundred healthy adults aged 50-65 years entered a supervised resistance-training program conducted three times weekly for 4 weeks aft

Research Progress of Transcutaneous Auricular Vagus Nerve Stimulation in Chronic Pain Management: A Narrative Review.

Chronic pain represents a significant global public health burden, affecting millions of individuals and substantially impairing quality of life. However, its clinical management is largely limited by the modest efficacy and considerable adverse effects of current pharmacological interventions, highlighting an urgent need for safe and effective non-pharmacological alternatives. Transcutaneous auricular vagus nerve stimulation (taVNS), a non-invasive neuromodulation technique, has emerged as a promising therapeutic strategy. By multitarget neurotransmitter modulation, activating the cholinergic

Inhibition effects and binding interactions of epigallocatechin and gallocatechin on tyrosinase and their anti-melanogenesis activity.

Tyrosinase is a key enzyme in melanogenesis and food enzymatic browning. This study systematically investigated tyrosinase inhibition effects and binding interactions of two tea catechins epigallocatechin (EGC) and gallocatechin (GC), and their practical efficacy. In vitro enzymatic assays showed that EGC and GC both reversibly inhibited tyrosinase in a mixed-type manner, with IC50 values of 0.059 ± 0.002 mg/ml (192.02 ± 7.19 µM) and 0.036 ± 0.001 mg/ml (118.77 ± 4.11 µM), respectively. Dehydration-induced 4D shape transformation in 3D printed egg yolk gels: Insights into the coupling between water redistribution and network restructuring.

The swift advancement of food 3D/4D printing requires inks with suitable rheological and structural properties. This research used calcium ions to improve the printability of salt-induced egg yolk gels, and subsequent dehydration enabled 4D transformations. Rheological analysis revealed that the viscosity of these gels increased from 107.4 Pa·s (control, without calcium chloride, all samples contained the same NaCl concentration) to 483.8 Pa·s (Ca-4, shear rate = 1 s-1), which enhanced extrusion and structural stability. Complementary low-field nuclear magnetic resonance and scanning electron

Simulating the effects of an alcohol minimum unit price policy on distilled spirits sales in 28 states of the USA.

BACKGROUND AND AIMS: Excessive alcohol use is a leading preventable chronic disease risk factor. Alcohol minimum unit pricing (MUP) policies are not used in the United States despite evidence of associations with reduced drinking and alcohol-related harms. To inform potential population-level chronic disease prevention strategies, we estimated effects of various hypothetical MUPs on alcohol sales. METHOD: Simulation based on observational time-series data. We used weekly off-premises product-specific alcohol retail sales and prices in 28 states of the United States for November 2022-November

Neurovascular Signaling in the Perivascular Space: Biophysical Principles and Disruption in Hypertension.

The brain's vasculature-from pial arteries through penetrating arterioles and the capillary bed to venous drainage-is enveloped by a narrow, compositionally elaborate perivascular space that provides the primary signaling interface between neural tissue and blood vessels. Here, we advance the principle that the geometry and composition of this space are not constant but change systematically along the vascular tree, and that perivascular space composition at these locations imposes biophysical constraints that dictate the fidelity and gain of neurovascular signaling. We argue that its narrow m

Physical frailty and the risk of peripheral vertigo: evidence from the UK Biobank cohort.

BACKGROUND: Peripheral vertigo is a common clinical condition that can substantially impair patients' balance and disrupt their daily and social activities. In some patients, symptoms may persist or recur and may be accompanied by auditory dysfunction or psychological problems, thereby imposing a considerable burden on individuals, families, and healthcare systems. Physical frailty, a prevalent clinical syndrome marked by reduced physiological reserve and increased stress vulnerability, is closely linked to various age-related functional impairments. Nevertheless, the specific association betw

NADES-mediated glycation of casein with mannose and mannose-oligosaccharides: conformation, antioxidant activity, and antimicrobial activity of gastrointestinal digests.

This study aimed to improve the physicochemical and functional properties of casein (CN) via glycation based on natural deep eutectic solvents (NADES). Four glycated CN conjugates (CN-Man1, CN-Man2, CN-Mos1, and CN-Mos2) were prepared with mannose (Man) or mannose-oligosaccharides (Mos) in water and NADES systems under optimized conditions. Structural and physicochemical changes were analyzed. The results indicated that Mos-mediated glycation in NADES was associated with marked conformational changes, and CN-Mos2 exhibited the lowest surface hydrophobicity and the highest thermal stability. The cost of persistent alarm: temporal transcriptional reprogramming of macrophages from acute activation to chronic immune suppression by day 11.

INTRODUCTION: Chronic inflammation has long been associated with cancer initiation, yet the mechanisms linking sustained immune activation to an immune-permissive tumor microenvironment remain incompletely defined. Prevailing explanations such as immune exhaustion (IEX) or free radical mediated tissue damage, fail to account for the active state of immune tolerance, a process driven by potent negative feedback loops that systematically suppress host effector responses. METHODS: To address this gap, we developed an in vitro model of macrophage tolerance driven by sustained Toll-like receptor 4

Serum sCD163 as a potential biomarker for predicting poor functional outcome at 3 months in acute ischemic stroke without reperfusion: a prospective study.

INTRODUCTION: Neuroinflammation plays a critical role in the prognosis of acute ischemic stroke (AIS). Soluble CD163 (sCD163), a marker of monocyte/macrophage activation, has been linked to stroke severity, but its independent prognostic value and clinical utility remain uncertain. This study aimed to evaluate whether serum sCD163 measured within 24 hours of admission predicts 3_month poor functional outcome in AIS patients without reperfusion therapy, and to construct and externally validate a prognostic model incorporating sCD163. METHODS: This prospective, dual_cohort study enrolled 664 fi
