Urinary cadmium exposure and biomarkers of inflammation, lipid peroxidation, and kidney injury in apparently healthy adults from the Central Region of Mexico.

Human exposure to cadmium remains a major global public health concern because of its potential to induce multiple adverse health effects. This study investigated the associations between cadmium exposure and effect biomarkers of oxidative stress, inflammation, and renal injury in a population from the Central Region of Mexico. The mean urinary cadmium concentration was 0.55 ± 1.15 µg/g creatinine. Higher urinary cadmium concentrations were significantly associated with increased levels of biomarkers of oxidative stress, inflammation, and kidney injury. These findings indicate that cadmium exp

Allium polyrhizum Turcz. flavonoids extract alleviates cadmium-induced renal injury through differential regulation of autophagy mediated by mTORC1 and mTORC2.

Cadmium (Cd)-induced nephrotoxicity remains a global health challenge, with dysregulated mechanistic target of rapamycin kinase (mTOR) signaling implicated in its pathogenesis. Allium polyrhizum Turcz. flavonoids (APTF), a traditional Mongolian medicinal plant extract, exhibits potent antioxidant and anti-inflammatory properties, but its protective effects against Cd nephrotoxicity via mTOR pathway modulation have not been evaluated. This study investigated the protective role of APTF against Cd-induced kidney injury, focusing on its dual regulation of mTOR complex (mTORC)1 and mTORC2 pathways

MHY@MO@PPP inhibits ferroptosis positive feedback and delays the acute kidney injury-to-chronic kidney disease transition by remodeling the Fe2+-mTORC1-GPX4 axis.

AKI-to-CKD progression is shaped by sustained tubular stress, ferroptosis-related iron imbalance, and oxidative injury. To target this process, we developed an MHY1485-loaded molybdenum oxide (MoOx) nanozyme coated with a pH-responsive phenylboronic acid-polyethylene glycol-phenylboronic acid (PBA-PEG-PBA) polymer (MHY@MO@PPP). We tested whether this platform could weaken the ferroptosis-associated injury loop and delay AKI-to-CKD transition through regulation of the Fe2+-mechanistic target of rapamycin complex 1 (mTORC1)-glutathione peroxidase 4 (GPX4) axis. Material properties were examined

Single-subject detection of speech network activation using functional near-infrared spectroscopy.

SIGNIFICANCE: Most neuroimaging studies rely on group-level analyses, yet many clinical and translational applications require reliable inference at the level of individual participants. AIM: We evaluated whether functional near-infrared spectroscopy (fNIRS) can detect expected speech network related activation patterns at the single-subject level. APPROACH: Twenty-seven adults completed simple listening and speaking tasks while fNIRS data were recorded over bilateral frontal, temporal, and parietal regions. Data were analyzed using general linear models with false discovery rate correction

Acute copper sulfate poisoning in yearling beef cattle in Brazil.

In a herd of 70 crossbred beef calves (10-12-mo-old) managed extensively in Santa Catarina, Brazil, 14 animals developed clinical signs; 9 of the 14 died within 7 d of accidental ingestion of a copper sulfate (CuSO4) product. Affected calves were lethargic, anorectic, and had green diarrhea. A bag containing remnants of blue-green product was found near the carcasses; this product was later determined to contain 53% metallic copper (Cu; dry weight). Postmortem examinations were performed on 3 calves found dead; tissue samples were collected for histopathology and mineral analysis. Gross findin

The role of ferroptosis in renal cell carcinoma: molecular mechanisms and therapeutic implications.

Of the numerous regulatory cell death mechanisms, ferroptosis has emerged as a pivotal player in the pathobiology of renal cell carcinoma (RCC), particularly the clear-cell subtype (ccRCC). This iron-dependent form of cell death, characterized by the catastrophic accumulation of lipid peroxides, is intimately linked to the unique metabolic landscape of ccRCC, including its lipid-rich environment and reliance on specific antioxidant pathways. This review comprehensively synthesizes the current understanding of the molecular mechanisms governing ferroptosis in RCC. We delineate the core regulato

Insights into the cell type-specific susceptibility of kidney cells to ferroptosis.

Ferroptosis is a regulated form of cell death driven by iron-dependent lipid peroxidation. Emerging evidence implicates ferroptosis in the pathogenesis of various kidney diseases by inducing death in renal tubular epithelial cells (RTECs), podocytes, mesangial cells (MCs), and endothelial cells (ECs), which exhibit distinct structural and functional traits that might determine their differential susceptibility to ferroptosis. Despite its importance, the precise mechanism of ferroptosis in kidney pathology remains unclear, limiting its therapeutic potential and drug development. Understanding c

The Protective Effect of Sodium Molybdate on Folic Acid-Induced Chronic Kidney Injury in Rat Model: A Preclinical Investigation.

Chronic kidney injury is a cosmopolitan concern of health as it is related to high morbidity and mortality. The current study was a preclinical investigation that used a rat model to study the renal protective effect of sodium molybdate (SM) against folic acid (FA)-induced nephropathy. Chronic kidney disease (CKD) was induced in rats by a single intraperitoneal (i.p.) injection of 250 mg/kg FA. SM orally administered at doses of 0.1 and 0.2 g/kg/day for 35 days. SM attenuated FA-induced chronic kidney injury that demonstrated by a significant decrease in urine total proteins/24 h, serum creati

New insights in the chronic kidney disease-associated osteoarthritis progression: Role of uremic toxin indoxyl sulfate-induced chondrocyte senescence and ferroptosis.

AIMS: Epidemiological data have shown a notably high co-occurrence of osteoarthritis (OA) and chronic kidney disease (CKD). The pathological mechanisms for CKD-associated OA still remain to be clarified. Uremic toxin Indoxyl Sulfate (IS), accumulated in CKD, may play a role in OA pathology. This study aimed to clarify the role of IS in CKD-associated OA and investigated the involvement of chondrocyte senescence and ferroptosis through the crosstalk between calcium and iron metabolism. MATERIALS AND METHODS: Human chondrocytes were treated with IS and co-treated with either calcium chelator BA

Precision-Engineered Silver Single-Atom Carbon Dot Nanozymes for Theranostic Management of Acute Kidney Injury.

Overproduction of reactive oxygen species (ROS) is a key pathogenic feature in acute kidney injury (AKI), leading to rapid decline in renal function with high mortality rates that call for effective antioxidant therapies. Herein, we present triphenylphosphonium-functionalized carbon dots supported by single-atom silver (T-AgSA-CDs) that integrate fluorescent antioxidant nanozymes for the accurate ROS scavenging and real-time bioimaging of AKI. By anchoring silver in single-atom and sub-nanocluster states on a carbon dot matrix, T-AgSA-CDs exhibit exceptional superoxide dismutase (SOD)- and glu

Preclinical evaluation of 68Ga-labeled acetazolamide derivatives as radiotracers targeting carbonic anhydrase IX in clear cell renal cell carcinoma.

Clear-cell renal cell carcinoma (ccRCC) represents the predominant histological subtype of renal carcinoma and accounts for the majority of kidney cancer-related deaths. Conventional [18F]FDG PET frequently fails to detect ccRCC because of its intrinsically low glucose metabolism. Carbonic anhydrase IX (CAIX), a hypoxia-inducible, tumor-specific cell-surface enzyme that is highly expressed in nearly all ccRCCs but absent from healthy kidney tissue, represents an attractive alternative target. Here, we developed a panel of modularly designed 68Ga-labeled acetazolamide analogues incorporating va

Role of FXR/PPARγ pathway mediated fatty acid oxidation in acute kidney injury induced by cadmium in rats.

Cadmium is a common heavy metal pollutant in the environment. Kidney is the main target organ of cadmium toxicity, which can cause acute kidney injury. Fatty acid oxidation is the main source of energy produced by proximal renal tubular epithelial cells, and FXR is a bile acid receptor involved in lipogenesis. However, the regulatory mechanisms by which FXR on FAO remain unclear. In this study, both in vivo (SD rats) and in vitro (rPT cells) cadmium poisoning models were established to determine whether fatty acid oxidation is involved in cadmium-induced acute kidney injury. The results of in
