Role of FXR/PPARγ pathway mediated fatty acid oxidation in acute kidney injury induced by cadmium in rats.

Cadmium is a common heavy metal pollutant in the environment. Kidney is the main target organ of cadmium toxicity, which can cause acute kidney injury. Fatty acid oxidation is the main source of energy produced by proximal renal tubular epithelial cells, and FXR is a bile acid receptor involved in lipogenesis. However, the regulatory mechanisms by which FXR on FAO remain unclear. In this study, both in vivo (SD rats) and in vitro (rPT cells) cadmium poisoning models were established to determine whether fatty acid oxidation is involved in cadmium-induced acute kidney injury. The results of in

Iron-quercetin complex ameliorates chronic kidney disease via inhibiting the renal TGF-β1/Smad3/Egr1 axis-mediated kidney injury and fibrosis.

BACKGROUND: Chronic kidney disease (CKD) remains a significant global health burden due to the lack of effective interventions. Here, we evaluated whether the metal-flavonoid compound, Iron-Quercetin complex (IronQ) can ameliorate CKD and examine its effect on the TGF-β1/Smad3/Egr1 signaling cascade. METHODS: To assess the IronQ's therapeutic efficacy in renal fibrosis, two well-recognized in vivo models were employed: the unilateral ureteral obstruction (UUO) model and the adenine (Ade)-elicited renal fibrosis systems. The efficacy of IronQ was assessed through comprehensive analyses of rena

KDIGO 2026 Clinical Practice Guideline for Anemia in Chronic Kidney Disease (CKD): a commentary from the European Renal Best Practice (ERBP).

The 2026 Kidney Disease: Improving Global Outcomes (KDIGO) Clinical Practice Guideline for Anemia in Chronic Kidney Disease represents a substantial update, coming more than a decade since the previous KDIGO guideline on the topic. This time lapse was necessary in order to accumulate new scientific evidence that was meaningful for an updated guideline. The new guideline now includes updated recommendations and practice points for anaemia management, incorporating recent evidence on intravenous iron therapies and hypoxia-inducible factor prolyl hydroxylase inhibitors (HIF-PHIs). SLC7A11-FUNDC1 axis drives Cr(VI)-Induced renal injury through mitophagy-ferroptosis crosstalk.

Hexavalent chromium [Cr(VI)] is a widespread environmental contaminant known to cause severe organ damage, with acute exposure leading to significant nephrotoxicity. To elucidate the underlying mechanisms, this study investigated the role of the mitophagy-ferroptosis axis in Cr(VI)-induced renal injury using mouse models and renal tubular epithelial cells (mRTECs). We found that Cr(VI) exposure disrupted mitochondrial iron homeostasis in mRTECs, leading to Mito-Fe2+ accumulation and mitochondrial damage. Consequently, this triggered an overproduction of mitochondrial and total reactive oxygen

Dual Therapeutic Effects of Ultra-Small Platinum-Selenium Nanoparticles on Oxalate-Induced Acute Kidney Injury.

Serum oxalate levels can rapidly increase due to specific dietary factors or ethylene glycol exposure, triggering acute kidney injury (AKI). Disorders like nephrocalcinosis and calcium oxalate (CaOx) nephropathy cause inflammation and renal failure without effective therapy. To address this challenge, ultra-small platinum-selenium (Pt-Se) nanoparticles (NPs) were synthesized to inhibit CaOxcrystallization by adsorbing C2O4 2- to block nucleation and by binding to crystal growth sites. in vitro and in vivo studies were performed to assess the inhibition of CaOx crystallization and oxalate-induc

Evaluation of the protective effects of selenium against iron overload-induced toxicity in rats using a multiple-markers approach.

BACKGROUND: Iron overload can promote the generation of reactive oxygen species (ROS), leading to oxidative stress and different human diseases. The trace element selenium has biological functions and can act as both an antioxidant and a prooxidant. This study aimed to evaluate the protective effects of selenium against iron overload-induced toxicity in rats. Adult Wistar rats were exposed to three increasing concentrations of iron (25, 50, and 100 mg/kg body weight [b.w.]), either alone or in combination with selenium (0.5 mg/kg b.w.). METHODS: The biological interactions between these two c

Metabolic Reprogramming and Ferroptosis: Unlocking New Therapeutic Frontiers in Cancer and Diabetes.

SIGNIFICANCE: Ferroptosis, an iron- and lipid peroxidation-dependent mode of programmed cell death, is presently realized as a converging mediator that bridges redox imbalance and metabolic dysfunction. Differing from apoptosis and necroptosis, ferroptosis involves iron homeostasis, glutathione depletion, and redox lipid damage. Thus, it becomes the intersection of metabolic reprogramming and redox signaling. Ferroptosis is a double-edged metabolic vulnerability and adaptive resistance pathway in malignancy. RECENT ADVANCES: Oncogenic signaling cascades such as PI3K/Akt/mTOR and AMPK restruct

Metal mixture inflammatory index and mortality risk in chronic kidney disease patients: A nationally representative cohort study from the NHANES 1999 to 2018.

Patients with chronic kidney disease (CKD) are frequently exposed to environmental heavy metals, which can easily aggravate the condition or even accelerate death. The association between the metal mixture inflammatory index (MMII) and mortality in CKD patients has not been investigated yet. The MMII was developed using reduced rank regression models to gauge the systemic inflammatory potential of a multi-metal mixture. We analyzed data from 2569 CKD patients who participated in the National Health and Nutrition Examination Survey (1999-2018) with follow-up until December 31, 2019. The associa

Cadmium exposure induces renal fibrosis by inhibiting hsa_circ_0075684/miR-363-3p/KLF4 signaling pathway.

To date, no effective chelation therapy exists to remove cadmium (Cd) from the kidneys, a condition that increases the risk of cadmium-induced chronic kidney disease among humans. consequently, it is vital to prevent kidney damage due to cadmium exposure. However, it has been challenging to identify an early diagnostic marker of cadmium-induced kidney damage through mechanism studies. Interestingly, our previous study revealed that the expression of the microRNA miR-363-3p was upregulated in workers who had been diagnosed with chronic occupational cadmium toxicity. Thus, we aimed to investigat

Ferritinophagy and organ injury.

Ferritinophagy is a selective form of macroautophagy/autophagy that mediates the degradation of ferritin complexes, releasing stored iron, and maintaining intracellular iron homeostasis. Proper regulation of ferritinophagy is essential for cellular adaptation to metabolic stress, whereas dysregulation disrupts iron balance and contributes to pathological processes. Excessive ferritinophagy leads to iron overload and reactive oxygen species accumulation, driving oxidative stress, ferroptosis, and inflammation, which are key contributors to cellular injury and progressive organ dysfunction. Beyond Thiol-Enzyme Inhibition: Sterically Bulky NHC-Au(I) Complexes are Catalytically Active Anticancer Agents with Reprogrammed Immunomodulatory Function.

Metallodrug-induced immunogenicity offers significant potential for improving the efficacy of cancer immunotherapy with metals such as gold serving as a notable example. However, heavy metals may concurrently disrupt immune cell function, and a few of them can augment immunogenicity without compromising immunological integrity. Here, we report sterically bulky Au(I) complexes as catalytically active anticancer agents with the capability to reprogram immunomodulatory function. Our initial studies identified complex IPr-Au-Cl, characterized by bulky substituents, which catalytically activated an

Resveratrol regulates nickel oxide-induced kidney damage by targeting the PERK/ATF-6/IRE1 and Nrf2/HO1 pathways.

Nickel oxide (NiO) is one of the most toxic heavy metals and poses a danger to human and animal health by causing serious kidney damage. Resveratrol (RES) is a flavonoid with antioxidant and anti-inflammatory properties, abundant in grapes and fruits. The current study is a first to investigate how RES affects oxidative stress, apoptosis, endoplasmic reticulum (ER) stress, inflammation, and histopathology against NiO-induced nephrotoxicity in rats using molecular, histological, immunohistochemical, and biochemical methods. The study used 24 Sprague-Dawley rats, randomly distributed into 4 grou

Accelerating Small Molecule Ferroptosis Regulator Development With Advances in Biological Functions.

Ferroptosis is an iron-dependent cell death characterized by the disorder of iron homeostasis and the accumulation of lipid peroxidation, which trigger a cascade of events resulting in cell death. This process has a dual regulatory effect in the evolution of diseases, which can not only attenuate the progress of diseases, but also become a promoting force to accelerate pathological alterations, and widely participate in the occurrence and development of multiple diseases. In recent years, there has been a growing body of research identifying regulators-both inducers and inhibitors-that target

Environmental Pollution and Its Impact on Kidney Diseases: A Comprehensive Review of Current Evidence.

Kidney disease is a growing global public health challenge that accounts for substantial morbidity, premature mortality, and rising healthcare costs. Although diabetes mellitus and hypertension remain the principal clinical risk factors for renal injury, accumulating evidence indicates that environmental pollution represents an independent and globally pervasive contributor to kidney disease burden. Long-term exposure to environmental toxicants, including heavy metals, ambient air pollutants, persistent organic pollutants, and endocrine-disrupting chemicals, has been consistently associated wi

An exploratory study of sex differences in thallium-induced nephrotoxicity in rats.

Sexual dimorphism is recognized in nephrotoxic acute kidney injury, with females being less susceptible than males. Thallium (Tl), a highly toxic heavy metal, induces severe systemic disorders following exposure, including gastrointestinal and neurological disorders and renal failure. However, sex-related differences in Tl-induced nephrotoxicity remain poorly understood. Previous studies have shown that Tl preferentially accumulates in the outer medulla of the kidney of male rats, resulting in mitochondrial dysfunction and medullary thick ascending limb (mTAL) injury, characterized by calcium

Macrophage ferritin heavy chain/α-synuclein regulatory axis modulates ferroptosis during kidney injury.

Macrophages, endowed with remarkable phenotypic plasticity, are essential for orchestrating injury responses and regulating iron homeostasis. Given the central role of ferritin heavy chain (FtH) as a molecular rheostat linking iron sequestration to redox-dependent signaling, we examined how myeloid FtH governs renal iron trafficking and ensuing oxidative stress pathways during acute kidney injury (AKI). Transcriptome analysis revealed coupling of FtH deficiency in monocytes and macrophages with activation of ferroptosis, a regulated cell death associated with iron accumulation. Moreover, myelo

Human umbilical cord-derived mesenchymal stem cell exosomes ameliorate hepatocyte ferroptosis and sepsis-associated liver injury by restoring iron homeostasis in hepatocytes.

Mesenchymal stem cells-derived exosomes (MSCs-Exo) constitute critical mediators of intercellular communication between stem cells and other cell types. Among these, exosomes derived from human umbilical cord mesenchymal stem cells (HUMSCs-Exo) have attracted considerable attention owing to their potent immunomodulatory capacity, low surface antigenicity, and favorable biosafety profile. Accumulating evidence indicates that HUMSCs-Exo can attenuate ferroptosis in target tissues across a spectrum of pathological conditions, including inflammatory bowel disease and viral cardiomyopathy, thereby
