The incidence of type 2 diabetes mellitus is approaching pandemic levels, posing a major global healthcare problem. Diabetic kidney disease (DKD) is the main cause of the chronic kidney disease. Hyperglycemia-dependent renal hypoxia drives the onset and progression of DKD. The adaptive cellular response to hypoxia depends on the zinc signalling, which promotes expression of the hypoxia-inducible factor-1α (HIF-1α). Zinc is further relevant for anti-inflammatory, antioxidant, anti-apoptotic, and anti-fibrotic signalling pathways with nephroprotective effects in DKD. In this review, we focus on

Near-infrared-II (NIR-II) emissive gold nanoclusters (AuNCs) have garnered significant attention for fluorescence imaging owing to their excellent biocompatibility. However, achieving highly specific imaging at lesion sites remains a challenge. Herein, we constructed a peptide-modified NIR-II luminescent AuNCs probe (AuNCs-Q&K) for specific imaging of acute kidney injury by leveraging the covalent cross-linking properties of transglutaminase (TGase), an enzyme significantly upregulated in acute kidney injury (AKI). The TGase enzyme enables the recognition of peptide sequences containing glutam

Nanoparticles show great potential for the management of acute kidney injuries (AKIs). It is well known that nanoparticle size plays a critical role in targeting renal tubules due to the pore structure and kidney filtration threshold (KFT) of the glomerulus. However, AKI can compromise the physiological barriers, which might render the established KFT (∼6 nm) and size laws under healthy conditions not applicable in AKI conditions. Reinvestigating the nanoparticle size effect on targeting injured kidneys is thus essential for developing efficacious AKI nanomedicine. Herein, using different-size

Programmed cell death (PCD) is a central determinant of kidney injury, maladaptive repair, and chronic progression. Beyond classical apoptosis, the identification of ferroptosis, pyroptosis, and cuproptosis has expanded the conceptual framework of renal pathophysiology by linking cell fate decisions to redox imbalance, inflammatory signaling, mitochondrial metabolism, and metal ion homeostasis. Ferroptosis is driven by iron-dependent phospholipid peroxidation and impaired antioxidant defenses; pyroptosis is mediated by inflammasome activation, gasdermin pore formation, and cytokine release; ap

BACKGROUND: Systemic readmissions after hip fracture surgery impose a substantial clinical and economic burden. Hip fracture represents a sentinel orthopedic injury with prolonged postoperative vulnerability extending beyond the index admission. Evidence from the Middle East remains scarce, particularly regarding the incidence, causes, and timing of systemic readmissions in multiethnic populations. This study aimed to determine the one-year incidence, etiologies, and temporal distribution of systemic readmissions following proximal hip fracture surgery, and to explore whether commonly document

Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disorder characterized by chronic inflammation, oxidative stress, and multi-organ damage, with lupus nephritis (LN) constituting a major cause of morbidity and mortality. Dysregulated iron metabolism and insufficient expression of heme oxygenase-1 (HO-1), a pivotal antioxidant enzyme regulating iron homeostasis and immune responses, have been implicated in LN pathogenesis. However, therapeutic strategies targeting HO-1 and iron dysregulation remain underexplored. Here, we demonstrate that HO-1 expression is markedly decreased in

Hepcidin, a 25-amino-acid peptide hormone produced primarily by hepatocytes, is the master regulator of systemic iron homeostasis. By binding the cellular iron exporter ferroportin and inducing its internalization and lysosomal degradation, hepcidin restricts iron entry into plasma from enterocytes, macrophages, and hepatocytes. Its transcription is governed by an intricate molecular network that integrates iron status, erythropoietic demand, oxygen tension, and inflammation, with the BMP-HJV-ALK2/SMAD axis acting as the canonical activating pathway and erythroferrone (ERFE) and matriptase-2 (

Cadmium (Cd) exposure induces renal injury by triggering inflammation, cellular senescence, and fibrosis. BMAL1, a core component of the circadian clock, participates in the regulation of cellular stress responses. Lycium barbarum polysaccharide (LBP) and melatonin (MT) possess anti-ageing and anti-inflammatory activities. However, their effect on Cd-induced nephrotoxicity has not yet been elucidated. In this study, wild-type and Bmal1-deficient mice were exposed to Cd for 8 weeks. For protective effect experiments, mice were treated with Cd exposure alone or in combination with LBP and MT. Ou

The kidney is a primary organ of heavy metal accumulation, and metal toxicity poses a great challenge to kidney health. We conducted a repeated-measure study to examine the longitudinal associations between blood heavy metal concentrations and kidney function biomarkers, including the urinary albumin-to-creatinine ratio (UACR), estimated glomerular filtration rate (eGFR), and N-acetyl-β-d-glucosaminidase (NAG). Using linear mixed models, we found that chromium (Cr) and lead (Pb) were associated with impaired kidney function across multiple biomarkers. Manganese (Mn) levels were linked to lower

BACKGROUND: Aristolochic acid nephropathy (AAN) exhibits direct and irreversible tubular epithelial cell toxicity, posing multiple therapeutic challenges. To address this, we have developed a class of biocompatible nanomachines for active treatment of AAN.
METHODS: PEG@Ni@MoS2 (PNM) nanoparticle-based mobile nanomachines, featuring a composite architecture where nickel (Ni) is deposited on molybdenum disulfide (MoS2) followed by polyethylene glycol (PEG5000) modification, indicate photoelectric response characteristics based on their ion-sputtering construction.

BACKGROUND: Lead (Pb) and cadmium (Cd) remain widespread environmental toxicants, and the kidney is a major target organ. However, reliance on a single biomarker matrix or renal endpoint may incompletely characterize Pb-Cd-associated renal abnormalities. Whether blood- and urine-based Pb/Cd biomarkers exhibit differential association profiles across renal outcomes, and whether outcome-oriented networks converge on shared candidate pathways, remains unclear.
METHODS: We analyzed NHANES 2011-2018 data from 5049 adults. Blood and urinary Pb and Cd biomarkers were evaluated in relation to estimat

Acute kidney injury (AKI) is a serious clinical condition with high morbidity and mortality, yet early detection and effective treatment remain challenging. Herein, we developed ultrasmall quantum dots (QDs) for dual-modal imaging-guided early detection and antioxidant therapy of AKI. Benefiting from the ultrasmall hydrodynamic size, the as‑prepared Ag₂Se@Mn QDs exhibit favorable renal clearance and prolonged retention in injured kidneys. The QDs possess concentration-dependent T1-weighted magnetic resonance imaging (MRI) contrast and near-infrared fluorescence properties, enabling real-time,

Environmental stressors that disrupt redox homeostasis pose a significant threat to metabolic balance, tissue integrity, and organismal development. Increasing evidence identifies N6-methyladenosine (m6A), a dynamic and stress-responsive RNA modification, as a central regulator that translates oxidative cues into functional changes in RNA metabolism and cellular behavior. Redox imbalance can recalibrate the activity, localization, and substrate selectivity of m6A writers, erasers, and readers, thereby reshaping transcriptomic programs that control inflammation, antioxidant defense, proteostasi

Oxidative stress is a central driver of renal ischemia-reperfusion (I/R) injury, and developing targeted antioxidant systems against it remains an unmet challenge. Here, we address this challenge by engineering a dual-functional metal-ion/L-serine nanoarchitecture integrating two key components: precise kidney targeting via L-serine's molecular recognition of the injury biomarker kidney injury molecule-1 (Kim-1), and bioinspired antioxidant catalysis through metal-ion/amino-acid-coordinated enzyme mimicry. Through systematic screening of metal ions (Mg2 +, Ca2 +, Mn2 +, Fe3+, Co2 +, Cu2 +, and

Metabolically-dysfunction-associated steatohepatitis (MASH) is characterised by embryonic Kupffer cell (emKC) loss and proinflammatory macrophage infiltration. While iron dysregulation is implicated in MASH, the role of labile iron (Fe2 + ) in mediating emKC survival and function remains unknown. In human and mouse MASLD/MASH livers, emKCs exhibit repressed iron metabolism, reduced labile iron pools, and mitochondrial dysfunction. KC-specific ferroportin knockout mice recapitulate these defects, accelerating Western diet-induced steatosis and fibrosis. Conversely, ferritin depletion restores e

Cadmium is a toxic heavy metal classified by the International Agency for Research on Cancer as a human carcinogen and recognized as an endocrine-disrupting chemical. The present study aimed to evaluate tissue-specific susceptibility to cadmium-induced oxidative damage to membrane lipids (lipid peroxidation, LPO) and to assess the antioxidative effects of melatonin in porcine tissue homogenates representing endocrine (the thyroid and the ovary) and non-endocrine (the liver, the kidney, and the brain) organs. Homogenates were incubated with cadmium chloride (CdCl2; 2.5-1000 µM) without/with mel

Chronic kidney disease (CKD) progresses to renal fibrosis and anemia, ultimately leading to chronic renal failure (CRF). Renal anemia, primarily caused by impaired erythropoietin (EPO) production and dysregulated iron metabolism, reduces QOL and increases cardiovascular risk. To establish an experimental model of CRF-associated anemia, male Wistar rats underwent 5/6 nephrectomy (Nx). In a subset of Nx rats, tacrolimus (TAC, 1 mg/kg, subcutaneously every other day for two weeks starting at week 4) was administered to exacerbate renal injury. Renal function, hematological parameters, iron-relate

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly termed non-alcoholic fatty liver disease (NAFLD), represents the most prevalent chronic liver condition worldwide, affecting approximately 25%-30% of the global adult population. Endocrine-disrupting chemicals (EDCs)-including phthalates, bisphenols, per- and polyfluoroalkyl substances (PFAS), persistent organic pollutants (POPs) and heavy metals-are ubiquitous environmental contaminants with established metabolic toxicity. However, the quantitative association between EDC exposure and MASLD prevale
