Iron and ferroptosis in kidney disease: molecular and metabolic mechanisms.

Maintaining iron homeostasis is necessary for kidney functioning. There is more and more research indicating that kidney disease is often caused by iron imbalance. Over the past decade, ferroptosis' role in mediating the development and progression of renal disorders, such as acute kidney injury (renal ischemia-reperfusion injury, drug-induced acute kidney injury, severe acute pancreatitis induced acute kidney injury and sepsis-associated acute kidney injury), chronic kidney disease (diabetic nephropathy, renal fibrosis, autosomal dominant polycystic kidney disease) and renal cell carcinoma, h

An Ultrasmall Self-Assembled Gallic-Acid-Based Natural Multifunctional Defense Networks for Therapeutic Application in Calcium Oxalate Nephropathies.

Kidney stones, which have high prevalence and recurrence rates, often cause severe oxidative damage and inflammation. The ultrasmall hydrodynamic diameter of nanoparticles is crucial for their enrichment in the kidneys to exert biological activity. However, integrating crystallization inhibition and therapeutic functions into a single ultrasmall nanoparticle is challenging. A novel ultrasmall iron (Fe)-gallic acid (Ga) metal-phenolic networks (Fe-Ga MPNs) is developed for treating calcium oxalate (CaOx) nephropathies. These MPNs can specifically adsorb on the high-energy ( 1 ¯ 01 $\bar{1}01$

Iron in Chronic Kidney Disease and End-Stage Kidney Disease-Current Trends and Future Direction.

Anaemia is a frequent and serious complication in chronic kidney disease (CKD), affecting both non-dialysis-dependent (NDD) and dialysis-dependent (DD) patients. While erythropoietin (EPO) deficiency is the primary cause, iron deficiency (ID) also plays a crucial role. ID in CKD can be classified as either absolute, resulting from blood loss, or functional, driven by inflammation and elevated hepcidin levels, which trap iron in macrophages and hepatocytes, preventing its use in erythropoiesis. Elevated hepcidin also reduces dietary iron absorption in the gut, making oral iron supplements ineff

Engineered molybdenum disulfide nanosheets as scavengers against oxidative stress inhibit ferroptosis to alleviate acute kidney injury.

Acute kidney injury (AKI) is a common clinical kidney dysfunction associated with high morbidity, elevated mortality, and poor prognosis. It results from redox imbalance caused by abnormal excess production of endogenous reactive oxygen species (ROS) at the renal tubules, which in turn initiates a series of pathological processes, such as cellular apoptosis, necrosis, and ferroptosis, eventually leading to structural and functional impairment of the kidney. Thereinto, ferroptosis induced by the lethal accumulation of lipid peroxidation is extensively involved in renal damage. Nanotechnology-me

Insulin like growth factor binding protein 7 activate JNK/ERK signaling to aggravate uranium-induced renal cell cytotoxicity.

Acute kidney injury (AKI) can occur primarily by exposing kidneys to uranium (U). Insulin-like growth factor binding protein 7 (IGFBP7) can regulate sepsis-induced AKI and epithelial-mesenchymal transition through ERK1/2 signaling. In vitro, the IGFBP7's role and mechanism of action in uranium-induced NRK-52E cells, however, remains unknown. To evaluate the effect of U exposure on kidneys, rat kidney proximal cell line NRK-52E was treated with different concentrations (200, 400, and 800 µmol/L) of it. Subsequently, three siRNAs targeting IGFBP7 were transfected with the HiPerFect reagent. The

Amino-Acid-Engineered Bionanozyme Selectivity for Colorimetric Detection of Human Serum Albumin.

Nanozymes are emerging nanomaterials owing to their superior stability and enzyme-mimicking catalytic functions. However, unlike natural enzymes with inherent amino-acid-based recognition motifs for target interactions, manipulating nanozyme selectivity toward specific targets remains a major challenge. In this study, we introduce the de novo strategy using the supramolecular assembly of l-tryptophan (l-Trp) as the recognition amino acid with copper (Cu) ions for creating a human serum albumin (HSA)-responsive bionanozyme. This amino-acid-engineered bionanozyme enables selective colorimetric d

Ferroptosis Activation Contributes to Kidney Aging in Mice by Promoting Tubular Cell Senescence.

Kidney aging is the strongest independent risk factor for chronic kidney disease. Ferroptosis, a recently discovered form of cell death mediated by iron overload and lipid peroxidation accumulation, has an unclear role in kidney aging. To determine the pathophysiological role of ferroptosis during kidney aging, we employed immunofluorescence, Western blotting, and qRT-PCR to analyze renal cells and tissues in natural aging and accelerated aging mouse models. We investigated the activation of ferroptosis during aging and examined the effects of the ferroptosis inhibitor ferrostatin-1 and the fe

Hexavalent chromium induces ferroptosis in small intestinal tissue of broilers through GPX4/HMGB1/p38-MAPK pathway.

As a major environmental heavy metal pollutant, hexavalent chromium (Cr(VI)) causes irreversible damage to animals and humans. Nevertheless, how Cr(VI) exposure causes intestinal damage in broilers remains inadequately explored. This study explores Cr(VI)-induced poisoning using potassium dichromate to build a Cr(VI) poisoning model. The results indicate that Cr(VI) exposure evidently reduced the body weight and the functions of liver and kidney in broilers. Histopathological analysis revealed different degrees of structural damage in all three segments of the small intestines by Cr(VI) exposu

Study on HIF-PHI combined with iron supplement in treatment of renal anemia in rats.

BACKGROUND: Roxadustat is a novel hypoxia- inducible factor-prolyl hydroxylase inhibitor(HIF-PHI) used to treat anemia in chronic kidney disease (CKD) patients. It has been reported that roxadustat can slow down kidney damage and delay the development of kidney fibrosis. Anemia and iron deficiency are often associated with the vast majority CKD patients, and insufficient available iron or total iron storage is often the most common cause of anemia and ESAs resistance in CKD patients. The role of iron availability in the pathogenesis of anemia in chronic kidney disease has received increasing a

Ferroptosis Mediates the Progression of Hyperuricemic Nephropathy by Activating RAGE Signaling.

Aims: Hyperuricemic nephropathy (HN) represents a prevalent complication of hyperuricemia, typified by tubular dysfunction, inflammation, and progressive renal fibrosis with unclear mechanisms. Ferroptosis, an iron-dependent regulated cell death, is implicated in multiple diseases, but has rarely been linked to HN. In this study, we aim to explore the possible role of ferroptosis in HN and its underlying mechanisms. Results: We showed that urate oxidase knockout mice, a model of hyperuricemia, exhibited renal impairment with elevated uric acid, creatinine, and blood urea nitrogen levels, accom

Polydatin Mitigates Lead-Induced Nephropathy by Modulating Oxidative Stress, Inflammation, and the AMPK/AKT/Nrf2 Pathway in Rats.

This study investigated the molecular mechanisms underlying lead (Pb)-induced nephropathy and assessed the nephroprotective potential of Polydatin (PD). Forty male Wistar rats were divided into five groups (n = 8/group): negative control (NC), normal rats treated with 200 mg/kg/day of PD (NPD200), positive control (PC) receiving Pb only (30 mg/kg/day), and two groups co-administered Pb with PD (100 or 200 mg/kg/day). Serum and urine Pb levels were determined by an atomic absorption spectrophotometer. Markers of renal tissue damage (TGF-β/iNOS/NGLA/KIM-1) and renoprotective molecules (Nrf2/AMKα

Dansylated Amino Acid-Modified Long-Acting PSMA Derivatives 68Ga/177Lu-LNC1011 as Prostate Cancer Theranostics.

Prostate-specific membrane antigen (PSMA)-targeted radiopharmaceutical therapy has demonstrated promising potential for treating metastatic castration-resistant prostate cancer. Recently, albumin-binding motif-modified PSMA radioligands with prolonged blood circulation were developed to improve tumor uptake and therapeutic effectiveness, properties which, however, were associated with an increased risk of bone marrow toxicity. This study presents new PSMA-targeted radioligands incorporating dansylated amino acids as relatively weak and preferable albumin binders to achieve a fine balance betwe

Lentinan alleviates cadmium-induced kidney injury by reducing cadmium accumulation via promoting cadmium excretion and metallothionein synthesis and regulating silencing information regulator1/nuclear factor erythroid 2-related factor 2/nuclear factor kappa-B signaling pathway.

Cadmium (Cd) is a widely distributed environmental heavy metal pollutant. It is extremely toxic to the kidney. This study investigated the potential mechanisms of action of lentinan (LNT), a fungal polysaccharide, on protecting against Cd-induced kidney injury in mice. Male Kunming mice were administered with CdCl2 (2.5 mg/kg/b.w.) by intragastric gavage and LNT in drinking water (1 mg/mL) for 10 weeks. Histological examination revealed that LNT reduced the glomerular atrophy, lymphocyte infiltration, tubular congestion, and collagen accumulation caused by Cd exposure. However, oral administra

mTOR-Mediated Autophagy Regulates Cadmium-Induced Kidney Injury via Pyroptosis.

The heavy metal cadmium (Cd) affects the global livestock production economy mainly through the contamination of feed raw materials and secondary contamination in feed processing, and it also poses a serious threat to food safety and human health. The nucleotide-binding oligomerization domain-like pyrin-domain-containing protein 3 (NLRP3) inflammasome is a key regulatory element of pyroptosis, which is engaged in kidney injury. Meanwhile, autophagy is also involved in renal inflammation. Mammalian target of rapamycin (mTOR) plays an important role in pyroptosis and autophagy, but its function

Micro- and Nano-Plastic-Induced Adverse Health Effects on Lungs and Kidneys Linked to Oxidative Stress and Inflammation.

Micro- and nano-plastics (MNPs) are small plastic particles that result from the breakdown of larger plastics. They are widely dispersed in the environment and pose a threat to wildlife and humans. MNPs are present in almost all everyday items, including food, drinks, and household products. Air inhalation can also lead to exposure to MNPs. Research in animals indicates that once MNPs are absorbed, they can spread to various organs, including the liver, spleen, heart, lungs, thymus, reproductive organs, kidneys, and even the brain by crossing the blood-brain barrier. Furthermore, MPs can trans

Iron Metabolism and Ferroptosis in Diabetic Kidney Disease.

Diabetic kidney disease (DKD) is a major diabetic microvascular complication that still lacks effective therapeutic drugs. Ferroptosis is a recently identified form of programmed cell death that is triggered by iron overload. It is characterized by unrestricted lipid peroxidation and subsequent membrane damage and is found in various diseases. Accumulating evidence has highlighted the crucial roles of iron overload and ferroptosis in DKD. Here, we review iron metabolism and the biology of ferroptosis. The role of aberrant ferroptosis in inducing diverse renal intrinsic cell death, oxidative st

The association between low-concentration heavy metal exposure and chronic kidney disease risk through α-klotho.

Although the association between pollution exposure and chronic kidney disease (CKD) has been explored, previous studies have focused on specific effects observed via in vitro or animal experiments. We first conducted a priority screening of pollutants for population CKD risk by using machine learning approaches. We then used the National Health and Nutrition Examination Survey (NHANES) 2007-2016 data from 2415 adults aged 40 years and over to study the joint effects of low-concentration metal exposure and the mediating effects of α-klotho by using Bayesian kernel machine regression (BKMR) and

Novel native probiotics with protective effects against nickel-induced kidney injury: a strategy for heavy metal toxicity.

Recent studies have shown that exposure to heavy metals, particularly Nickel, and exacerbates kidney diseases by increasing oxidative stress. Probiotics, recognized for their antioxidant properties, may provide a beneficial approach to mitigate this oxidative damage. This research aims to explore the efficacy of native probiotics in reducing oxidative stress in kidney tissues, potentially leading to improved protective strategies against the toxicity caused by heavy metals. In this study, forty male NMRI mice were exposed to Nickel to induce oxidative stress, followed by probiotic treatment to

Drug-Induced Membranous Nephropathy: Piecing Together Clues to Understand Disease Mechanisms.

There is a resurgence of interest in drug-induced membranous nephropathy because of the widespread availability of recently discovered culprit medications, such as lipoic acid supplements, mercury in skin-lightening creams, and nonsteroidal anti-inflammatory drugs, and the relationship between these drugs and newly described target antigens. The clinical syndromes associated with drug-induced membranous nephropathy are similar in that proteinuria ranges from low grade to nephrotic range and generally remits within months of drug cessation. Histology is notable for subepithelial deposits that a

Habitat analysis of iron deposition in the basal ganglia for diagnosing cognitive impairment in chronic kidney disease: evidence from a case-control study.

BACKGROUND: Chronic kidney disease induces alterations in the heterogeneity of iron deposition within the basal ganglia. Quantitative analysis of the heterogeneity of iron deposition within the basal ganglia may be valuable for diagnosing chronic kidney disease-related cognitive impairment. METHODS: In this prospective observational cohort study, quantitative susceptibility mapping (QSM) was performed in chronic kidney disease patients. Susceptibility values of each nucleus within the basal ganglia were measured. Radiomic features were extracted from habitats of the basal ganglia on QSM image
