Piezo1 knockdown alleviates high‑altitude cerebral edema by improving brain microvascular endothelial cell tight junctions via blocking the calcium influx‑mediated Calpain‑1 activity.

BACKGROUND: Since lifestyle changes are main therapies for non-alcoholic fatty liver disease (NAFLD), changing dietary components (nutritional or bioactive) may play a parallel important role. Few studies have assessed the effects of curcumin on NAFLD (mainly antioxidant and anti-inflammatory effects). We aimed to determine the effects of nano-curcumin (NC) on overweight/obese NAFLD patients by assessing glucose, lipids, inflammation, insulin resistance, and liver function indices, especially through nesfatin. METHODS: This double-blind, randomized, placebo-controlled clinical trial was condu

Multiple gastrointestinal cancers in a single patient is a rare entity. In our study, we are showing the clinical presentation and management of these patients. A fifty-nine-year-old asthenic male (already treated case of metachronous colorectal cancer in 2008 and 2011) presented with complaints of generalized weakness and fatigue. Strong family history was present with two of his first-degree relatives having diagnosed with gastrointestinal cancer at the age < 50 years with one of them having stomach carcinoma and another with GEJ tumors. On evaluation, upper GI endoscopy revealed growth at c

Simultaneous optimization of film morphology and structure dimensionality in Cs3Sb2I9 through butylamine gas treatment.

Food safety is a critical public health issue worldwide as the consumption of unhygienic food causes millions of illnesses annually. The contaminated food is primarily unsafe due to the presence of microorganisms (i.e. bacteria and viruses), chemicals (i.e. pesticides and heavy metals), and physical hazards (i.e. glass, metal and plastic). Foodborne illnesses such as salmonellosis, campylobacteriosis, listeriosis, E. coli infection, botulism, cholera, norovirus infection, and others are commonly caused by the problems associated with food hygiene. From the modest beginnings rooted in basic hyg

Screening and Identification of Vibrio cholerae Bacteriophages VC3 and Its Role in the Inhibition and Removal of Biofilms in Seafood.

As biological control agents, bacteriophages can both inhibit the pathogenic bacteria and remove the bacterial biofilms from the seafood. Vibrio cholerae is the pathogen of cholera and the severe infection could lead watery diarrhea and even death. The double-layer agar plate method was used to isolate and screen the V. cholerae bacteriophages from the samples of aquaculture water and sewage. Purified bacteriophages were examined through genome sequencing, as well as morphological and biological characterizations. Among the isolated bacteriophages, bacteriophage VC3 was found to be a long-tail

Coeliac disease and the intestinal barrier: mechanisms of disruption and strategies for restoration.

Coeliac disease is characterised by immune-mediated damage to the small intestine in response to dietary gluten in genetically predisposed individuals. Increased intestinal permeability is a central component to its pathophysiology. This review explores the evidence for increased permeability in coeliac disease and the underlying mechanisms, including the roles of zonulin, inflammatory cytokines, microbial alterations and immune responses to gliadin peptides. We also review comprehensively the therapies targeting barrier integrity and normalising intestinal permeability, including particular d

Piloting a mobile early warning alert and response system for East and Central Darfur, Sudan.

Conflict in Sudan since April 2023 has disrupted health information systems, particularly in hard-to-reach areas like Darfur. In response to limited surveillance capacity and reports of suspected outbreaks (e.g. measles, cholera, hepatitis E), Sudan's Federal Ministry of Health, Health Cluster partners and the WHO piloted an emergency surveillance programme using the WHO-developed Early Warning, Alert, and Response System Mobile (EWARS Mobile) in Central and East Darfur states from September 2024 to January 2025. The approach enabled simplified, offline reporting of eight priority diseases and

Plasma fractionation in Latin America: a path towards sovereignty and sustainability in the supply of plasma-derived medicines.

PURPOSE: Increased reactive oxygen species (ROS) released by NADPH oxidase and inflammation are associated with arterial hypertension and eye diseases associated with high blood pressure, including glaucoma, retinopathies (e.g., age-related macular degeneration), and choroidopathies affecting ocular function; however, the mechanisms underlying these adverse outcomes remain undefined. The present study was designed to highlight the importance of oxidative stress in severe hypertension-related eye damage. METHODS: Male Wistar rats (n = 7, unless otherwise specified for specific experiments) wer

Alcaligenes lipid A as a sublingual adjuvant to augment protective immune responses in the respiratory and gastrointestinal tracts.

We previously identified Alcaligenes as symbiotic bacteria residing within Peyer's patches and demonstrated that their primary components, lipopolysaccharides, and their active center, lipid A, are excellent adjuvants for mucosal vaccination. Here, we evaluated the effectiveness of Alcaligenes-derived lipid A as an adjuvant for sublingual immunization, a novel vaccination route. Mice sublingually immunized with Alcaligenes lipid A and ovalbumin (OVA) showed enhanced production of OVA-specific IgA in both the respiratory and gastrointestinal tracts. In addition, increased serum levels of OVA-sp

CholeraSeq: a comprehensive genomic pipeline for cholera surveillance and near real-time outbreak investigation.

SUMMARY: Next Generation Sequencing is widely deployed in cholera-endemic regions, yet an end-to-end reproducible pipeline that unifies read QC, filtering, reference mapping, variant calling/annotation, recombination screening, and extraction of parsimony informative sites/variant codons, phylogenetic inference for downstream phylodynamic and epidemiological analyses have been lacking, slowing outbreak investigation and public health response. CholeraSeq is a high-throughput genomics pipeline for cholera genomic surveillance. It ingests consensus genomes, short read sequence data, draft assemb

Sodium butyrate augments the antibacterial activity of tetracycline against clinical isolates of multidrug-resistant Vibrio cholerae.

BACKGROUND: Antibiotic resistance poses a major challenge in treating Vibrio cholerae infections. One promising method to counter resistance is the co-administration of antibiotics with non-antibiotic adjuvants to enhance their efficacy. This study investigated the combined action of sodium butyrate (SB) and tetracycline on tetracycline-resistant V. cholerae strains. RESULTS: The combined activity of SB and antibiotics was assessed on eight V. cholerae clinical isolates using the Fractional Inhibitory Concentration Index (FICI), with SB-Tetracycline showing strong synergy (FICI: 0.09-0.5). [The impact of global change on infectious disease epidemiology].

Global change -including biodiversity loss, land-use alterations and climate change- is reshaping the epidemiology of infectious diseases. These factors facilitate the emergence of new pathogens, interspecies transmission, and the geographical expansion of vectors and reservoirs. Vector-borne diseases such as dengue, malaria and West Nile fever are increasing in both incidence and distribution. Similarly, water- and food-borne diseases (e.g. salmonellosis, cholera) are becoming more frequent, driven by extreme weather events, poor water and sanitation infrastructure, and changes in aquatic eco

Distinct PlzC mechanisms integrate chemotaxis and c-di-GMP signaling to regulate Vibrio cholerae motility and biofilm formation.

Bacterial motility and biofilm formation are essential for the adaptation and survival of Vibrio cholerae, the causative agent of cholera. In many bacterial species, the second messenger c-di-GMP regulates these processes through PilZ domain proteins, however the downstream mechanisms have remained poorly defined. Here, we identify the PilZ domain protein PlzC as a key positive regulator of motility and biofilm formation through distinct mechanisms. A suppressor screen for mutants restoring ΔplzC migration identified downstream regulators, including CheX, a CheY-3 phosphatase. Genetic and phen

Severe gastroenteritis caused by Vibrio cholerae serogroup O24 in an immunocompromised patient: A case report.

Non-O1 and non-O139 Vibrio cholerae (NOVC) strains rarely harbor the cholera toxin gene and are typically associated with mild, self-limiting gastroenteritis. However, an increasing number of reports in recent years have described severe gastroenteritis or extraintestinal infections with fatal outcomes, particularly in immunocompromised individuals, such as those with underlying conditions including cirrhosis. Here, we describe a case of severe gastroenteritis caused by V. cholerae serogroup O24, following the consumption of seafood during prolong prednisolone treatment for systemic lupus eryt
