Aeromonas in South Asia: genomic insights into an environmental pathogen and reservoir of antimicrobial resistance.

Aeromonads are an ecologically versatile group of bacteria that cause infections in aquatic animals and are recognised as emerging human pathogens. Despite this, our understanding of Aeromonas diversity, especially the relationship between clinical and environmental strains, remains limited. Here, we present a genomic analysis of the Aeromonas genus, comprising 1853 genomes, and a detailed comparison of clinical and environmental strains from South Asia, including 996 newly sequenced genomes from Bangladesh and India. Phylogenetic analyses revealed that Aeromonas is a highly diverse genus, wit

In-silico identification of anti-cholera phytochemicals from Indian medicinal plants.

Cholera is a severe diarrheal disease caused by ingestion of food or water contaminated with pathogenic Vibrio cholerae. Treatment for cholera includes rehydration therapy and antibiotics to avert death and reduce bacterial burden to prevent rapid transmission of the disease. In addition, in Indian subcontinent, there is historical evidence of using plants for treating cholera. This study was designed to investigate the cholera toxin-inhibitory properties of phytochemicals sourced from Indian medicinal plants. For this, three reported genotypes of cholera toxin subunit B (ctxB) associated with

Fractional nonlinear dynamics and forward bifurcation in a memory-based cholera model.

This study explores the use of fractional-order epidemic models to capture the memory-dependent and nonlinear behaviors inherent in cholera transmission. We present a fractional-order susceptible-infected-recovered-individuals adopting preventive measures-bacteria model that integrates preventive behavior and environmental feedback. By applying the Caputo derivative, we demonstrate the existence, uniqueness, and boundedness of the model's solutions and derive an analytical expression for the basic reproduction number R0. Our stability and bifurcation analyses show how memory influences the sys

TseVF-TsiVF, a novel bacteriolytic effector-immunity pair of Vibrio fluvialis VflT6SS2, provides a fitness advantage in microbial competition.

Vibrio fluvialis (V. fluvialis) is a halophilic Gram-negative bacterium regarded as an emerging pathogen that can cause severe cholera-like diarrhea and various extraintestinal infections. Type VI secretion systems (T6SSs) are nano-weapons used by bacteria to deliver toxic effectors into host cells or antagonize competitors. VflT6SS2 of V. fluvialis is involved in bacterial pathogenicity and competitive environmental survival. Previously, we have identified some regulatory factors of VflT6SS2 and a pesticin domain-containing effector, TssI2. Here we reported a novel effector-immunity pair of V

A Vibrio-susceptibility class of antimicrobial peptide Ajapocin via membranolytic pattern to combat "non-cholera" pathogens in vivo infection models.

Pathogenic "non-cholera" Vibrio species of Vibrio parahaemolyticus (V. parahaemolyticus) and Vibrio vulnificus (V. vulnificus) frequently pose a serious threat to aquaculture security and public health by causing infectious diseases. In this study, we reported the discovery of a marine-sourced antimicrobial peptide (AMP) called Ajapocin, which identified through a sequence optimization strategy. Ajapocin exhibited potent activity against V. parahaemolyticus and V. vulnificus pathogens, with minimum inhibitory concentrations (MICs) of 6-12 μM-comparable to the clinical agent Polymyxin B (PMB).

Antimicrobial resistance and genomic characteristics of avian Pasteurella multocida.

BACKGROUND: Pasteurella multocida, the cause of fowl cholera, poses a significant economic threat to poultry. The escalation of antimicrobial resistance (AMR) in this pathogen, driven by drug misuse, is a serious concern. Crucially, a comprehensive understanding of AMR patterns, molecular epidemiology, and virulence in Chinese avian strains is still lacking. This study aimed to elucidate the phenotypic and genomic characteristics of P. multocida strains isolated from avian in Zhejiang Province, China. RESULT: Twenty avian-origin P. multocida strains isolated in Zhejiang Province were subjecte

Co-administration of oral killed whole-cell recombinant cholera toxin B-subunit vaccine (WC-rCTB) and live Salmonella Typhi Ty21a vaccine: a prospective randomized open-label trial.

BACKGROUND: Cholera and typhoid fever are often co-endemic, making vaccine co-administration practical. However, due to lack of immunogenicity data, current guidelines advise against co-administration of the oral inactivated whole-cell recombinant cholera toxin B-subunit vaccine (WC-rCTB) and the oral live Salmonella Typhi Ty21a vaccine. METHODS: Healthy adults (18-65 years) were randomized 1:1:1 to receive WC-rCTB with Ty21a (group Ch + Ty), WC-rCTB alone (group Ch) or Ty21a alone (group Ty). Peripheral blood mononuclear cells (PBMCs) were isolated on Days 0, 5 and 7 from all, plus on Days 1

Event-based surveillance in the WHO African region: baseline implementation before the COVID-19 pandemic.

Event-based surveillance (EBS) is a critical component of epidemic intelligence, supporting the timely detection of unusual public health events. However, prior to the COVID-19 pandemic, the status of EBS implementation across the WHO African region was not well documented. This study establishes a regional baseline of EBS modalities in place before the pandemic to inform future surveillance system evaluation and strengthening. We conducted a cross-sectional survey across all 47 WHO Member States in the African region between December 2019 and February 2020. A structured questionnaire was diss

EPICERTIN, an engineered variant of cholera toxin B subunit, promotes survival and a pro-remodeling macrophage phenotype for mucosal healing in colitis.

EPICERTIN, a modified cholera toxin B subunit (CTB), facilitates mucosal healing in preclinical colitis models, but its anti-inflammatory mechanisms remain unclear. Here, we investigated EPICERTIN's effects on macrophages. In a dextran sulfate sodium-induced colitis mouse model, oral administration of EPICERTIN reduced neutrophil infiltration and increased CX3CR1+MHCIIlo/- (M2-like) over CX3CR1+MHCIIhi (M1-like) macrophages in the colon lamina propria. This was concurrent with upregulation of colony-stimulating factor 2 (Csf2) and growth factors (Egf, TgfA, Fgf, Pdgf) involved in mucosal remod

The gendered health impacts of climate change on smallholder women farmers in Mhondoro Ngezi, Zimbabwe.

BACKGROUND: Climate change poses significant threats to the livelihoods, health, and overall well-being of smallholder women farmers globally. In Mhondoro Ngezi, where agricultural production is predominantly rain-fed, high temperatures, recurrent droughts, and erratic rainfall patterns have intensified women's vulnerability to climate-induced health risks. OBJECTIVE: This study examined the gendered health impacts of climate change on women farmers and explored how these challenges intersect with broader socio-economic inequalities. METHODOLOGY: The research employed the Q methodology to id

Cloning, Expression and Purification of the Beta Subunit of Cholera Toxin Using Escherichia coli as an Expression Host and pET-24a(+) as a Cloning Vector.

Cholera toxin has a biologically active A subunit and a binding B subunit. Being non-toxigenic, the Cholera toxin B (CTB) subunit has multiple applications in immunology and rapid diagnostics. A recombinant form of CTB (rCTB) is available for commercial use. However, the COVID-19 pandemic led to a severe global shortage in the production and distribution of commercial CTB, which posed a significant problem for researchers worldwide. Due to the non-availability of commercial rCTB post the COVID-19 pandemic, we attempted and successfully produced rCTB in a pET-based expression system in Escheric

Long-Term Antibody Responses to Vibrio cholerae Across Natural Infection, Vaccination, and Challenge: A Systematic Review.

BACKGROUND: Cholera is a critical global health threat, with significant impact on low- and middle-income countries. Clinical surveillance is limited by non-specific symptoms, dependence on care-seeking, and availability of microbiological diagnostics, making burden estimation difficult. Serosurveillance is a potential solution, but the design and interpretation of cross-sectional seroepidemiological studies are currently limited by a lack of knowledge of longitudinal antibody trajectories. METHODS: We systematically reviewed literature from 1980 to 2025 containing longitudinal antibody measu

Motor Neuron Size-Dependent Differences in mRNA Expression of Tropomyosin-Related Kinase Receptor B in Adult Rats.

Motor neuron plasticity and survival depend on several trophic factors, with signaling via the tropomyosin receptor kinase B (TrkB) receptor being particularly important beyond development and across the lifespan. We hypothesized that differences in motor neuron properties may reflect differential trophic influences across motor units within a single motor unit pool. Based on extensive previous characterization of rat phrenic motor neurons (PhMN), somal surface area was used as an indicator of motor unit type (smaller neurons innervating slow-twitch, fatigue-resistant units versus larger neuro

Atypical El Tor Vibrio cholerae from the second major global seventh-pandemic cholera wave is endemic in Sabah, Malaysia.

Cholera, caused by Vibrio cholerae, remains a significant diarrheal disease, especially in coastal regions of developing countries. In Malaysia, cholera is largely non-endemic except in Sabah, which has had recurrent outbreaks accounting for ∼75% of national cases between 2004 and 2014. To understand the origin and transmission of the disease, we sequenced the genomes of clinical isolates of V. cholerae O1 collected during an outbreak in 2019 and 2020. Genotypic analyses revealed that all Sabah isolates were atypical El Tor biotype harboring Classical CTX prophage elements. In particular, the

Maternal expression and breast milk transfer of an mRNA- encoded monoclonal antibody in a murine model of cholera.

Breast milk confers infants with immunity to a multitude of pathogens reflective of prior maternal infections and vaccinations. However, in outbreak situations where infants may be vulnerable to lethal infections due to gaps in the maternal immune repertoire, a case can be made for supplementing breast milk with one or more pathogen-specific monoclonal antibodies (mAbs) with known prophylactic or therapeutic activity. As oral delivery of recombinant IgG and IgA mAbs to infants has proven challenging, we investigated the use of mRNA-lipid nanoparticle (LNP) technology to stimulate pathogen-spec

Multicountry genomic analysis underscores regional cholera spread in Africa.

Cholera remains a significant public health burden in many countries in sub-Saharan Africa, though the exact mechanisms of bacterial emergence and spread remain largely undefined. We generated genomic data from 763 Vibrio cholerae O1 isolates predominantly collected between 2019-2024 to create the largest dataset of V. cholerae genomes sequenced locally in Africa. This dataset enabled us to interrogate recent patterns of spread, including the rapid circulation of the AFR15 lineage associated with unusually large outbreaks in Southern Africa. We provide evidence for the movement of the AFR15 li

Nighttime lights as a proxy for conflict intensity and infrastructure recovery in Yemen and Ukraine.

INTRODUCTION: Quantifying the impacts of armed conflict on civilians and infrastructure remains a major challenge, particularly where reporting is limited. Most conflict measurement tools require affected populations to report events and are limited by short time series, under-reporting, and varying methods. These tools do not capture infrastructural rebuilding, which has important health implications. Given this, we demonstrate the utility of nighttime lights (NTL) as a complementary tool for measuring conflict dynamics and infrastructure recovery with an epidemiological application. METHODS

The GlyGly-CTERM domain functions as an independent motif that targets proteins to rhombosortase in Vibrio cholerae.

Vibrio cholerae secretes a variety of effector proteins that are freely released into the extracellular space via its Type II Secretion System (T2SS) including cholera toxin, the causative agent of the disease cholera. In contrast to cholera toxin, a growing number of T2SS effectors is increasingly understood to remain associated with the cell surface. The serine protease VesB from V. cholerae is a surface protein that is produced with a short C-terminal motif, called GlyGly-CTERM. This motif is linked to the rest of VesB via a predicted unstructured linker. In addition to VesB, V. cholerae en

Synthesis, Biological Evaluation, and Computational Analysis of 1,4-Naphthoquinone Derivatives as Inhibitors of the Sodium-Dependent NADH:Ubiquinone Oxidoreductase (NQR) in Vibrio cholerae.

The therapeutic efficacy of antibiotics has been significant in extending human life expectancy by combating virulent bacterial infections. Nevertheless, multidrug-resistant (MDR) microorganisms remain a global crisis as these bacteria have developed resistance to conventional antibacterial agents. An unexplored antibiotic target found exclusively in bacteria is the Na+-translocating NADH:ubiquinone oxidoreductase (NQR), which is an indispensable membrane-bound bacterial enzyme complex that enables cellular functionality and is present in many infectious bacterial species, including Vibrio cho
