Explaining the Link Between HIV Stigma and Treatment Outcomes: From Stigma Avoidance to Socially Motivated Nonadherence.

BACKGROUND: Stigma is an important factor in medication adherence, but the mechanisms linking stigma to adherence are not understood, despite having been examined across multiple populations using a variety of study designs. Guided by a stigma-coping framework, we tested a model in which enacted stigma predicts antiretroviral (ART) nonadherence through stigma avoidance strategies and socially motivated nonadherence. METHOD: All adults waiting to receive their ART prescriptions (N = 1979) during multiple days and times were recruited from two public clinics in Cape Town South Africa (2021-2024

Complete Genome Sequence of Halorubrum Pleomorphic Virus 8.

Pleomorphic archaeal viruses are widespread and abundant in hypersaline environments and beyond. Still, a relatively low number of isolates are available for this viral group. Here, we present the complete genome sequence of Halorubrum pleomorphic viruses 8 (HRPV-8), which was previously isolated from the Samut Sakhon solar saltern, Thailand, on a Halorubrum species. The HRPV-8 genome was assembled into a circular 10,824-bp-long contig with a GC content of 63.2%. The genome annotation resulted in 12 open reading frames, including a VP4-like spike protein-encoding gene, which is a hallmark of p

Genome sequence determination and analysis of a betabaculovirus from the moth Glena bisulca, a defoliating pest of pines.

A baculovirus isolated from the moth Glena bisulca was characterized by electron microscopy of its occlusion bodies (OBs) and sequencing of its genome. The OBs of this virus, Glena bisulca granulovirus isolate 1002 (GlbiGV-1002), were ovocylindrical with a single virion, as described for other betabaculoviruses. The 104,702 bp circular genome contained six direct repeat (dr) regions and 121 open reading frames. A core gene phylogeny placed GlbiGV-1002 in betabaculovirus clade b with Cnaphalocrocis medinalis granulovirus (CnmeGV) as the nearest relative. Kimura-2-parameter nucleotide distance e

Flavaglines as cross-kingdom translation inhibitors: Heavy halides at C6 potentiate antineoplastic, antiviral, and antifungal activities.

The growing prevalence of cancer, coupled with the rise of multidrug-resistant pathogens, presents a formidable challenge to modern medicine. The flavagline class of secondary metabolites has been shown to possess a unique capacity to target multiple conserved pathways across various human malignancies and microbial pathogens. While numerous structure-activity relationship (SAR) campaigns have established trends with respect to specific indications, cross-kingdom comparative analyses have not been reported to date. In this work, a structurally diverse library comprising 17 novel and 5 natural

Plasmid architecture determines the stability of inverted terminal repeats in adeno-associated virus vectors.

Recombinant vectors derived from adeno-associated viruses (rAAVs) are a mainstay of human gene therapy. rAAVs are produced from plasmids containing transgene cassettes flanked by inverted terminal repeats (ITRs), which form structured DNA elements that stabilize the ends of the single-stranded viral genome and are the only viral sequences required in cis for genome packaging. For decades, it has been recognized that propagation of ITR-containing plasmids can result in deletions and other mutations, prompting the use of specialized bacterial strains, modified growth conditions, and truncated or

Structural polymorphism and population-variable coding capacity of HERV-K(HML-2) in human pangenomes.

Approximately 8% of the human genome is derived from ancient retroviral infections. The most recently integrated of these endogenous retroviruses is the HERV-K(HML-2) clade, whose expression has been associated with cancer, amyotrophic lateral sclerosis, and embryogenesis. Studies of HERV expression, particularly HML-2, have relied predominantly on short-read sequencing. However, the high similarity among HML-2 proviruses prevents many short reads from being assigned uniquely to individual loci. We therefore compared haplotype-resolved long-read genome assemblies from 292 donors to resolve var

EsViritu is a sensitive and specific profiler for viral pathogen detection and genome recovery in metagenomes.

Metagenomic sequencing allows for detection of any number of pathogenic viruses from a single sample without a priori knowledge of its contents. This has created a substantial opportunity to characterize pathogens in humans and their environment, but the field is held back by a lack of bioinformatics tools for taxonomic profiling and reconstructing virus genomes. The ideal profiler would simultaneously do two things: report viruses at single-read sensitivity so that ultra-low-abundance pathogens are not missed and hold near-perfect specificity so that a pathogen is never falsely reported. Here

Development and application of a smartphone-based thermal contrast amplification reader for photothermal lateral flow immunoassays.

The high cost and poor portability of external infrared thermal imagers have limited the widespread adoption of photothermal lateral flow immunoassays (PT-LFIAs) in on-site testing. To address this, we developed a smartphone-based handheld Thermal Contrast Amplification (TCA) Reader, eliminating the need for costly external hardware. Compared with existing recent external thermal imager-based PT-LFIA readout, this reader integrates a selected thermal-sensor-equipped smartphone, a motorized laser module, Bluetooth-enabled control, and a fixed 3D-printed holder into a compact device. The reader

Molecular detection of biofilm-related genes and antimicrobial resistance patterns in Pseudomonas aeruginosa isolates from Northern Iran.

Pseudomonas aeruginosa is widely recognized as one of the primary opportunistic pathogens responsible for a wide range of nosocomial infections, notably in immunocompromised patients. Antibiotic resistance and biofilm formation, as two major virulence factors of P. aeruginosa, are related to the persistence of prolonged infections. This study was designed to (1) evaluate the frequency of multidrug-resistant (MDR) P. aeruginosa in northern Iran and (2) analyze the frequency of biofilm-associated genes and their correlation with antimicrobial patterns. Clinical samples (n = 80) were collected fr

Superinfection exclusion strategy of siphophage T5: analysis of the FhuA:Llp complex.

Superinfection exclusion is a widespread viral mechanism that protects the infected cell from overinfection by other identical or closely related viruses. Shortly after infection by bacteriophage T5, the bacterium Escherichia coli produces Llp, encoded by the newly injected viral genome. Llp is a small lipoprotein targeted to the inner leaflet of the host outer membrane. It binds to FhuA, an outer-membrane iron-ferrichrome transporter, which is the T5 receptor at the E. coli cell surface. The interaction between Llp and FhuA prevents any further bacteriophage binding and infection. Here, we de

Identification and fine mapping of a novel linear B-cell epitope within the S1-CTD of the QX-type infectious bronchitis virus.

Infectious bronchitis virus (IBV) causes severe economic losses to the poultry industry. The IBV S1-CTD mediates receptor binding and serves as a key target for antibody recognition. In this study, the S1-CTD protein of the QX-type IBV QXL87 strain was expressed in HEK293F cells. Four monoclonal antibodies (10H5, 11D11, 12H7, and 13H1) were generated via hybridoma technology and characterized by indirect ELISA, Western blotting, and immunofluorescence assay; variable-region sequencing showed that the four mAbs are clonally related and recognize the same antigenic region. Fecal Metabolomics-Based Dietary Profiling Reveals Food Markers of HIV Brain Health Disorders.

BACKGROUND: Despite widespread viral suppression, nearly half of people with HIV (PWH) experience brain health disorders, such as neurocognitive impairment (NCI) and depression. Dietary interventions can improve neuropsychiatric outcomes in other populations, but there is limited research in PWH. OBJECTIVE: Recent advances allow the detection of food molecules from fecal samples to provide semi-quantitative, high-resolution measures of dietary exposure. We applied a metabolomics-based method to identify food items associated with brain health outcomes in PWH. METHODS: Participants were 170 P

Community Connectedness and Its Association with Stigma and Viral Suppression Among MSM with HIV: Findings from HPTN 078.

Gay, bisexual, and other men who have sex with men (MSM) continue to be disproportionately affected by HIV in the United States, with viral suppression rates among MSM across varied racial and ethnic groups remaining below national targets. Stigma is a major barrier to viral suppression, while community connectedness may serve as a protective factor. However, few studies have examined whether different forms of community connectedness are associated with viral suppression or moderate the negative effects of stigma. Data were drawn from 490 MSM with HIV enrolled in the HIV Prevention Trials Net

Bidirectional Synergistic Interaction Between Influenza Virus and SARS-CoV-2 in the Postpandemic Era: A Modeling Study.

INTRODUCTION: Influenza and COVID-19 are major respiratory diseases whose co-circulation dynamics are not fully understood. Surveillance data from the postpandemic period, free from the confounding effects of nonpharmaceutical interventions (NPIs), provide a unique opportunity to study their interactions. This study aimed to (1) estimate the direction and strength of the epidemiological interactions between influenza virus and SARS-CoV-2 and (2) quantify the impact of these interactions on the cumulative disease burden. METHODS: We developed a deterministic influenza virus × SARS-CoV-2 12-com

Evaluating the potential of treatment options for chronic hepatitis B.

INTRODUCTION: Chronic hepatitis B (CHB) remains a leading cause of cirrhosis and hepatocellular carcinoma worldwide, but long-term or even lifelong viral suppression with nucleos(t)ide analogues (NAs) seldom clears hepatitis B surface antigen (HBsAg), so-called 'functional cure.' In recent years, a shift in treatment strategy from viral suppression toward cure has emerged, catalyzed by three revised international hepatitis B guidelines (2025-2026) and the first-in-class phase 3 evidence that a finite antiviral therapy, the antisense oligonucleotide bepirovirsen, can achieve a functional cure i
