Multi-omics analysis identifies BUB1B as a cell cycle-related driver of bladder cancer progression.

Bladder cancer (BC) is characterized by a high incidence and frequent recurrence. Despite efforts to improve survival and reduce mortality among BC patients, significant progress remains elusive. The role of the cell cycle in BC has been increasingly studied, and our analysis revealed that BC exhibits elevated expression of BUB1 mitotic checkpoint serine/threonine kinase B (BUB1B). However, the role of BUB1B in BC remains not fully understood. A Multi-omics analysis was performed by integrating BUB1B expression with clinical data, and the results indicated that the upregulation of BUB1B was as

Repeat transurethral resection and contemporary oncologic outcomes among patients with non-muscle invasive urothelial carcinoma of the bladder.

PURPOSE: Repeat transurethral resection (re-TURBT) is recommended for all patients with high-risk bladder cancer to improve staging and oncologic outcomes. However, contemporary evidence on the role of re-TURBT is lacking. We therefore examined the contemporary rates of upstaging at re-TURBT and the association of re-TURBT with disease recurrence and progression. MATERIALS AND METHODS: We identified patients with an incident diagnosis of high-grade Ta, low-grade T1 and high-grade T1 urothelial carcinoma of the bladder between 2010 and 2022. The association of re-TURBT with disease recurrence

Circulating Transcriptome Analysis Finds Gene Signatures and Immune Cell Activation and Abundance Predict Response to Immunotherapy in Bladder Cancer.

PURPOSE: Immune checkpoint blockers (ICB) have revolutionized oncology by achieving durable tumor responses in advanced cancers. Nevertheless, currently approved biomarkers (PD-L1, microsatellite instability, and tumor mutational burden) have suboptimal positive and negative predictive values for tumor response and survival. EXPERIMENTAL DESIGN: We aimed to assess the value of RNA sequencing from whole blood to predict responses to ICB. We performed total paired-end RNA sequencing at 20 million reads and analyzed differential gene expression, signaling pathway activation, and immune cell abun

A high-throughput photothermal aptasensing platform using MnO₂@CuS nanoprobes for noninvasive detection of bladder cancer cells in simulated liquid biopsy.

A high-throughput aptamer-based photothermal (PT) sensing platform has been developed using MnO₂@CuS nanoprobes for the noninvasive detection of exfoliated bladder cancer cells (HT-1376) in simulated liquid biopsy samples. A specific aptamer (Apt1) targeting epithelial cell adhesion molecule (EpCAM) was immobilized on an amino-functionalized glass slide to selectively capture target cells, and a thiolated aptamer-conjugated MnO₂@CuS nanoprobe (Apt2) was employed for photothermal signal output. Upon 808 nm laser irradiation, infrared thermographic imaging was used to monitor temperature elevati

Translational Advances in Urothelial Carcinoma: From Bench to Bedside.

Urothelial carcinoma (UC), encompassing both bladder cancer and upper tract UC, is a biologically heterogeneous malignancy that has long complicated patient stratification and therapeutic prediction. Recent genomic and transcriptomic analyses, however, have delineated reproducible molecular subtypes across non-muscle-invasive and muscle-invasive disease. These classification frameworks have refined biological understanding and improved both risk stratification and treatment selection. Emerging evidence also indicates that genomic alterations, such as FGFR3 mutations, may confer distinct immuno

Beyond Sterility: the Urinary Microbiome in Bladder Cancer Carcinogenesis and Treatment.

PURPOSE OF REVIEW: Emerging evidence has challenged the paradigm of bladder sterility, opening new opportunities for understanding bladder cancer biology. RECENT FINDINGS: Disruption of commensal intravesical bacterial communities appears to increase risk of carcinogenesis, while the presence of specific bacterial strains has been linked to differential treatment responses. However, the interplay between urinary microbiome and bladder cancer remains incompletely defined. This review summarizes the current literature regarding the role of urinary microbiome in the pathogenesis and treatment ou

TSTA3 promotes the malignant progression of bladder cancer by regulating LAMP2 and MAPK signaling pathway-mediated epithelial-mesenchymal transition.

BACKGROUND: Bladder cancer (BCa) is the most frequently seen malignancy of the urinary tract. However, its molecular mechanisms and therapeutic targets are not well established. OBJECTIVE: This study aims to investigate the mechanism by which tissue specific transplantation antigen P35B (TSTA3) mediates the suppression of epithelial-mesenchymal transition (EMT) in BCa through targeted regulation of lysosome-associated membrane protein 2 (LAMP2) expression via the mitogen-activated protein kinase (MAPK) signaling pathway. METHODS: Public datasets were analyzed to predict TSTA3 expression and

Jujuboside A induces bladder cancer cell apoptosis by inhibiting ATP1A2-mediated mitochondrial energy metabolism regulation.

BACKGROUND: Conventional treatments for bladder cancer exhibit various limitations. Therefore, natural products, such as jujuboside A (JuA), have been explored for their multi-target effects and low toxicity. However, the specific effects of JuA in bladder cancer remain unclear. OBJECTIVE: To determine whether JuA affects mitochondrial energy metabolism and apoptosis in bladder cancer cells by regulating the ATPase Na+/K+ transporting subunit alpha 2 (ATP1A2) expression. METHODS: Differentially expressed genes (DEGs) in bladder cancer were analyzed using the GSE133624 dataset. ATP1A2 overexp

Microbiome-linked transcriptomic signatures in NMIBC: Toward personalized uro-oncology.

BACKGROUND: Nonmuscle Invasive Bladder Cancer (NMIBC) is a prevalent malignancy marked by high recurrence and progression rates. Emerging evidence suggests that demographic and environmental factors may alter the bladder's native oncobiome, influencing tumor behavior. This exploratory pilot study examined whether paired tumor and adjacent normal bladder mucosa exhibit distinct host transcriptomic and microbial signatures that may illuminate early tumor-microbiome interactions in NMIBC. METHODS: A meta-transcriptomic analysis was conducted on paired tumor and adjacent normal bladder mucosa fro

A multi-modal approach for decision making in bladder cancer.

Bladder cancer remains a major global health challenge, characterized by diagnostic uncertainty, substantial treatment costs and high recurrence rates. Current diagnostic and treatment modalities, including cystoscopy, transurethral resection of bladder tumour and standard histopathology, have limitations, including the inability to detect flat lesions, frequent understaging and interobserver variability, highlighting a crucial need for improved approaches. Advances in artificial intelligence (AI), blue-light cystoscopy, narrow-band imaging, cytology and urinary markers show promise in enhanci

Inhibiting autophagy enhances anti-cancer properties of sulforaphane.

Bladder cancer (BC) has an extremely low survival rate due to its tendency to metastasize. The antimalarial drug chloroquine (CQ) can inhibit BC progression and invasiveness by targeting basal autophagy. However, the mechanism by which CQ affects BC is not defined. Here, we revealed that although CQ showed an anticancer effect by reducing the migration and proliferation of the analyzed bladder cancer cells, it increased the expression of ICAM-1, a protein whose expression is associated with higher tumorigenic potential. Sulforaphane (SFN), a well-known ICAM-1 inhibitor, significantly contribut

Tissue polypeptide antigen as a non-invasive bladder Cancer biomarker.

Tissue Polypeptide Antigen (TPA), a non-invasive biomarker comprising fragments of cytokeratins 8, 18, and 19, has demonstrated potential for non-invasive surveillance of bladder cancer. However, challenges related to standardization impede its clinical implementation. This narrative review provides a focused overview of the relationship between TPA molecular biology and clinical laboratory practice, with special emphasis on analytical standardization, preanalytical harmonization, and evidence-based diagnostic algorithms. Urinary TPA has a sensitivity of 80-85% and specificity of 97% in optimi
