Mycobacterium tuberculosis Rv2074 is a coenzyme F420-dependent mycobilin reductase.

Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis, scavenges iron from its human host to support survival and pathogenesis. While siderophore-mediated iron acquisition in Mtb is well characterized, the mechanisms of heme‑iron acquisition are less understood. In the Mtb cytosol, the IsdG-type heme-degrading enzyme MhuD converts heme into iron and mycobilin isomers; however, the fate of MhuD-generated mycobilin is unknown. Recent studies have identified four Mtb flavin/deazaflavin oxidoreductases (FDORs), which are predicted to be non-canonical coenzyme F420-dependent biliver

Mutational profiling of ESX/type VII secretion system-associated genes in drug-resistant Mycobacterium tuberculosis from Khyber Pakhtunkhwa, Pakistan.

Tuberculosis (TB) remains a major global public health challenge, and the increasing burden of drug-resistant Mycobacterium tuberculosis (MTB) strains poses a significant obstacle to the success of TB control programs. This study aimed to characterize the phenotypic and genotypic drug-resistance profiles and to investigate mutations in ESX/type VII secretion system-associated secretory genes. Whole-genome sequencing (WGS) was performed to detect mutations behind resistance to rifampicin (rpoB, rpoC), isoniazid (katG, inhA), fluoroquinolones (gyrA, gyrB), and bedaquiline (Rv0678, Rv2535c (pepQ)

Construction, expression, and immunogenicity analysis of a fusion gene based on the PE/PPE family in combination with adjuvants.

Tuberculosis (TB) remains the leading global cause of death from a single infectious agent and a major fatal infectious disease worldwide. However, the Bacillus Calmette-Guérin (BCG) vaccine-currently the only licensed TB vaccine in clinical practice-has critical limitations in preventing latent infection and disease reactivation, failing to deliver comprehensive protective immunity. Given these gaps, targeting Mycobacterium tuberculosis (M.tb) antigens with strong immunogenic potential is a key strategy for next-generation TB vaccine development. The pathogenic M.tb harbors the Pro-Glu (PE) a

Identifying overlooked factors associated with multidrug-resistant tuberculosis: an ecological study across 201 countries and regions, 1990-2021.

BACKGROUND: Multidrug-resistant tuberculosis (MDR-TB) is a critical global health challenge. Whether environmental, nutritional, and socioeconomic factors are associated with MDR-TB remains insufficiently investigated. METHODS: We analyzed outcome and exposure data from the Global Burden of Disease (GBD) Study and four additional databases covering 201 countries and territories from 1990 to 2021. Sex ratio, diabetes prevalence, and HIV/AIDS burden were regressed out of each outcome before analysis. Every candidate exposure was then both regressed against the confounder-adjusted outcome and co

FDG-PET in Infection Imaging of Immuno-compromised Patients.

Infectious syndromes represent a monumental challenge in the management of immunocompromised patients, directly driving high rates of morbidity and critical mortality. This clinical cohort comprised oncological populations undergoing aggressive cytoreductive chemotherapy, end-stage renal disease patients dependent on specialized long-term dialysis access networks, and individuals undergoing treatment with advanced antiretroviral regimens. Because these individuals suffer from blunted or suppressed cellular immune mechanisms, standard structural imaging methodologies (such as standard digital r

Subnational spatial co-distribution of tuberculosis and diabetes mellitus across Ghana's 16 regions following administrative reorganisation: An ecological analysis.

BACKGROUND: National administrative boundary reorganisations, increasingly common across sub-Saharan Africa, create discontinuities in surveillance data alignment that complicate subnational burden estimation for infectious and non-communicable diseases alike. Ghana's expansion from 10 to 16 administrative regions in 2019 provides a tractable case for demonstrating how publicly available surveillance and survey data can be used to characterise subnational co-distribution of tuberculosis and diabetes mellitus (DM) under a revised boundary structure. DM is an established risk factor for active T

Outcomes of non-tuberculous mycobacterial infection of the bone, joint and soft tissue: a seven-year multicentre retrospective study.

PURPOSE: Extrapulmonary nontuberculous mycobacterial (EPNTM) infections of bone, joint, and soft tissue are difficult to treat and lack species-specific outcome data. We sought to describe antimicrobial regimens and surgical management by Mycobacterium spp. and to identify predictors of a clinical cure within 12 months in this adult cohort with EPNTM. METHODS: We conducted a multicentre retrospective cohort study across five tertiary hospitals in Australia and New Zealand 2014-2020. Adults with culture-confirmed EPNTM infection from sterile or intraoperative specimens were included. Primary o

Unified hybrid RNA-regulated CRISPR diagnostic for concurrent Mycobacterium tuberculosis detection and rifampicin resistance profiling.

CRISPR-based nucleic acid detection is challenged by protospacer adjacent motif (PAM) dependency, amplicon consumption and single-tube multiplexing barriers, and poor discrimination of single-nucleotide variations (SNVs). These limitations are particularly acute in detecting Mycobacterium tuberculosis (MTB) and rifampicin (RIF) resistance mutations for tuberculosis (TB) diagnostics. Herein, we devised TB-SCANER, a broad-spectrum, one-pot CRISPR-Cas12a diagnostic platform for MTB detection and RIF resistance profiling. The assay employs two unlocking single-stranded RNAs (uLsRNAs) transcribed f

Scalable cultured gut microbiota-derived nanovesicles for oral augmentation of checkpoint blockade therapy.

Microbiota-targeted strategies, such as fecal microbiota transplantation (FMT), hold promise for enhancing immunotherapy but face clinical challenges including safety risks, batch-to-batch variability, and dependence on substantial quantities of donor fecal material. To overcome these limitations, we established a standardized in vitro expansion platform for responder patient-derived gut microbiota. Through optimization of culture conditions, we achieved stable viability and retention of key immunotherapy-enhancing strains over serial passages, enabling a scalable 6.4 × 107-fold expansion. Lev

Application and prospects of veterinary diagnostic techniques in laboratory monkeys.

As the non-human primate models with the closest genetic relationship to humans, laboratory monkeys are indispensable in life sciences research, drug development, and biological product evaluation. However, infectious diseases either endemic in laboratory monkey colonies or acquired during breeding and husbandry-for example, B virus and Mycobacterium tuberculosis-threaten animal welfare and colony security, while also elevating zoonotic transmission risks and compromising the reliability of research data. The diversity, complexity, and clinically insidious nature of diseases in laboratory monk

Identification of mitochondrial metabolism-associated tuberculosis key genes and construction of a diagnostic model based on WGCNA.

Despite recent advances, tuberculosis (TB) remains a complex global health challenge, with its causative bacterium, Mtb, actively manipulating host cellular metabolism to survive. Therefore, studying mitochondrial metabolism-related genes is crucial, as they offer a promising avenue for identifying diagnostic biomarkers and developing new host-directed therapies to combat the disease. This study integrated transcriptomic datasets from three TB cohorts in the Gene Expression Omnibus (GEO) database (training set: GSE83456; validation sets: GSE42826 and GSE19444). Differential analysis was perfor
