Botrytis cinerea Protoplasts Production and Inoculation.

The absence of a cell wall turns fungal protoplasts very receptive to the uptake of exogenous genetic material and, consequently, they can be used for genetic transformation or transfection experiments. Additionally, isolated monosporic fungal strains, naturally infected or previously transfected with a mycovirus, can be used in assays of fungal protoplasts fusion. This technology will overcome the barriers of vegetative incompatibility with other Botrytis cinerea strains, or even with other fungi, and successfully allow the transference of mycoviruses from an infected to a non-infected fungal

Virus-Induced Gene Silencing for Nodulation Analysis in Soybean.

Virus-induced gene silencing (VIGS) is a powerful reverse genetics tool that enables the transient knockdown of gene expression in plants by exploiting their natural antiviral defense mechanisms. This chapter presents a detailed protocol for applying a Cowpea severe mosaic virus (CPSMV)-based VIGS system to study gene function in soybean (Glycine max), with a focus on symbiotic root nodulation. VIGS allows systemic silencing of target genes, making it especially valuable to study pathways involving systemic signaling. The protocol includes Agrobacterium-mediated propagation of CPSMV variants i

WHO target product profiles for dengue therapeutics: an interdisciplinary analysis of ethical, regulatory, and implementation considerations.

Dengue, a mosquito-borne viral disease, is undergoing its largest surge in history, driven by climate and demographic changes. Expanding transmission has highlighted previously under-recognised populations at heightened risk of severe disease, including people with multimorbidity and immunosuppression, for whom live-attenuated vaccines are contraindicated or unlicensed. This evolving epidemiology underscores the urgent need for safe and effective treatments for dengue, particularly for those with complex risk profiles compounded by socioeconomic disadvantage. We examined ethical, regulatory, a

Accelerating the clinical development of treatments for dengue: WHO dengue therapeutics landscape analysis and target product profiles.

Dengue, an Aedes mosquito-borne viral infection, is on the rise with climate and demographic change. In 2023, WHO declared dengue its highest grade of emergency, following the largest number of cases and deaths in recorded history. This emergency highlighted the absence of safe and effective dengue-specific treatments as a gap in the product landscape for care for people with dengue. To accelerate development of dengue-specific treatments for wide implementation and access in all dengue-endemic countries, WHO conducted a landscape analysis of the dengue therapeutics pipeline and convened a glo

Cost-effectiveness of the hepatitis B birth-dose vaccine in South Africa: a mathematical modelling analysis.

BACKGROUND: In 2022, South Africa had an estimated hepatitis B prevalence of 4·7-6·0%. In an effort to eliminate viral hepatitis as a public health threat, a targeted (ie, selective) hepatitis B birth-dose vaccination (HepB-BD) policy was introduced in 2023. Under this policy, only infants born to mothers with confirmed hepatitis B positivity are eligible for HepB-BD, which is given in addition to peripartum antiviral prophylaxis (PAP) for mothers from the second or third trimester of pregnancy. This policy differs from the WHO recommendation that all newborns should receive a HepB-BD vaccine

Lentiviral Transduction of Embryonic Stem Cells.

Lentiviral vectors provide an efficient and reliable method for stable gene knockdown in embryonic stem cells (ESCs) through RNA interference. Here, we describe a detailed protocol for lentiviral transduction of mouse ESCs using lentiviral shRNA expression vectors. The protocol encompasses lentiviral particle production in HEK-293T packaging cells, determination of viral titer, transduction of ESCs cultured under feeder-free conditions, and selection of stably transduced cells. Additionally, we describe methods for evaluating transduction efficiency using fluorescence microscopy and flow cytom

Improving thermostability of Acinetobacter baumannii phages during cold storage through infecting naïve bacterial hosts.

Despite prior research on optimal temperatures for Acinetobacter baumannii phage storage, thermostability of phages produced from original versus naïve hosts was not addressed. Three A. baumannii phages harvested from three multidrug-resistant A. baumannii strains were used to infect the original hosts and two other naïve strains. For each phage, the quantity of phage production from the three hosts (i.e., one original and two naïve) was compared. The phages produced were stored at two different temperatures (i.e., -20 °C without glycerol and 4 °C) for 24 weeks with the survival probability as

Immunogenicity and Safety of Co-administration of Adjuvanted Respiratory Syncytial Virus Prefusion F Protein Vaccine With 20-Valent Pneumococcal Conjugate Vaccine in Adults Aged ≥60 Years: A Randomized, Non-inferiority Trial.

BACKGROUND: Respiratory syncytial virus (RSV) illness and pneumococcal disease present high disease burden and health risks to older adults. Vaccination against these 2 illnesses is a key strategy to reduce this burden, and co-administration of the 2 vaccines could enhance vaccine uptake. This study evaluated the immunogenicity, safety, and reactogenicity of co-administration of adjuvanted RSV prefusion F protein vaccine (adjuvanted RSVPreF3) with 20-valent pneumococcal conjugate vaccine (PCV20) in adults aged ≥60 years. METHODS: This phase III, open-label, randomized, non-inferiority trial w

Minimal small RNA precursors unlock virus-based precision RNAi.

Artificial small RNAs (art-sRNAs) mediate highly specific RNAi in plants, but their broader use has been constrained by long precursor architectures and dependence on transgenic delivery. Recent work shows that accurate and efficient art-sRNA biogenesis can be achieved using precursors of minimal size rather than full-length endogenous scaffolds, without compromising silencing efficacy. We discuss how minimal art-sRNA precursors represent a versatile platform for precision RNAi and, critically, unlock RNA viruses as stable, programmable platforms for the production of highly specific art-sRNAs

Automated Video Tracking to Phenotype Plant Resistance to Aphid-Transmitted Yellow Dwarf Viruses in Grass Seed Crops.

Grass seed crops are susceptible to yellow dwarf viruses transmitted by aphids. The Willamette Valley in Oregon, United States, is the leading producer of cool-season grass seed crops globally, and industry reports have attributed seed yield loss and shortened stand longevity to aphid-transmitted yellow dwarf viruses. Genetic resources are needed for effective and sustainable management of this pest, specifically the Rhopalosiphum padi-PAV pathosystem, in grass seed production to reduce foliar insecticide applications and maintain optimum seed yield potential. Type I interferon primes the alveolar epithelium to receive reparative signals from tissue-resident macrophages.

Lung repair in response to viral infection requires integrated communication between epithelial and immune compartments, yet the impact of antiviral mediators on epithelial regenerative capacity remains poorly defined. Here, we demonstrate that type I interferon (IFN-I) signaling primes the lung for alveolar renewal following viral challenge. IFN-I contributes to the induction of an interferon-stimulated gene-high (ISG)hi Sca-1Pos population of alveolar type II (ATII) epithelial cells. Sca-1Pos ATIIs exhibit enhanced proliferative capacity and increased organoid-forming efficiency compared wit

Human interferon stimulated genes target ancient features of animal and bacterial viral replication.

Animal and bacterial cells defend against viral infection by rapidly activating antiviral restriction factors. In human cells, antiviral immunity is initiated by interferon signaling that results in expression of hundreds of interferon-stimulated genes (ISGs)1,2,3,4,5,6. Complex regulatory networks and co-evolution of viral evasion strategies complicate analysis of immune proteins under native conditions and the function of most individual ISGs remains unknown7,8,9,10,11,12. Here we discover that heterologous expression of ISGs in bacteria is sufficient to protect against infection by diverse

A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

Whole genome sequence-based comparative genomics reveals preliminary genomic features of Salmonella enterica subsp. enterica serovar Enteritidis phage type 1 and phage type 4 strains from EnteroBase.

BACKGROUND: Whole Genome Sequencing (WGS) enables detailed characterization of circulating and emerging bacterial strains. Although tens of thousands of Salmonella genomes have been acquired over the years, analyses of the genomic differences between strains of different phage types are scarce. RESULTS: We compared two Salmonella enterica subsp. enterica serovar Enteritidis (SEn) phage types, namely phage types 1 and 4 from available databases, using bioinformatic tools and nanopore sequencing of a Chilean PT1 strain. Comparisons between the two phage types show very low genomic divergence an

The origins and evolution of the RNA virome in land flora.

The colonization of land by plants was a major transition in the history of life, fundamentally reshaping the evolution of terrestrial organisms and Earth's ecosystems. Plants are hosts to an enormous diversity of viruses, but the origins and evolution of these viruses are not thoroughly understood. In this study, we mined the transcriptomes and genomes of 2402 species sampled across the plant kingdom and discovered about 35 000 plant-associated RNA viruses, including several potential families not described previously, resulting in a 2.2-fold increase in the known plant-associated RNA virus d

Explaining the Link Between HIV Stigma and Treatment Outcomes: From Stigma Avoidance to Socially Motivated Nonadherence.

BACKGROUND: Stigma is an important factor in medication adherence, but the mechanisms linking stigma to adherence are not understood, despite having been examined across multiple populations using a variety of study designs. Guided by a stigma-coping framework, we tested a model in which enacted stigma predicts antiretroviral (ART) nonadherence through stigma avoidance strategies and socially motivated nonadherence. METHOD: All adults waiting to receive their ART prescriptions (N = 1979) during multiple days and times were recruited from two public clinics in Cape Town South Africa (2021-2024

Complete Genome Sequence of Halorubrum Pleomorphic Virus 8.

Pleomorphic archaeal viruses are widespread and abundant in hypersaline environments and beyond. Still, a relatively low number of isolates are available for this viral group. Here, we present the complete genome sequence of Halorubrum pleomorphic viruses 8 (HRPV-8), which was previously isolated from the Samut Sakhon solar saltern, Thailand, on a Halorubrum species. The HRPV-8 genome was assembled into a circular 10,824-bp-long contig with a GC content of 63.2%. The genome annotation resulted in 12 open reading frames, including a VP4-like spike protein-encoding gene, which is a hallmark of p

Genome sequence determination and analysis of a betabaculovirus from the moth Glena bisulca, a defoliating pest of pines.

A baculovirus isolated from the moth Glena bisulca was characterized by electron microscopy of its occlusion bodies (OBs) and sequencing of its genome. The OBs of this virus, Glena bisulca granulovirus isolate 1002 (GlbiGV-1002), were ovocylindrical with a single virion, as described for other betabaculoviruses. The 104,702 bp circular genome contained six direct repeat (dr) regions and 121 open reading frames. A core gene phylogeny placed GlbiGV-1002 in betabaculovirus clade b with Cnaphalocrocis medinalis granulovirus (CnmeGV) as the nearest relative. Kimura-2-parameter nucleotide distance e

Flavaglines as cross-kingdom translation inhibitors: Heavy halides at C6 potentiate antineoplastic, antiviral, and antifungal activities.

The growing prevalence of cancer, coupled with the rise of multidrug-resistant pathogens, presents a formidable challenge to modern medicine. The flavagline class of secondary metabolites has been shown to possess a unique capacity to target multiple conserved pathways across various human malignancies and microbial pathogens. While numerous structure-activity relationship (SAR) campaigns have established trends with respect to specific indications, cross-kingdom comparative analyses have not been reported to date. In this work, a structurally diverse library comprising 17 novel and 5 natural

Plasmid architecture determines the stability of inverted terminal repeats in adeno-associated virus vectors.

Recombinant vectors derived from adeno-associated viruses (rAAVs) are a mainstay of human gene therapy. rAAVs are produced from plasmids containing transgene cassettes flanked by inverted terminal repeats (ITRs), which form structured DNA elements that stabilize the ends of the single-stranded viral genome and are the only viral sequences required in cis for genome packaging. For decades, it has been recognized that propagation of ITR-containing plasmids can result in deletions and other mutations, prompting the use of specialized bacterial strains, modified growth conditions, and truncated or
