Adrenergic Contraction of Human Prostate Tissues Is Maintained Under the Rab GTPase Inhibitors Nexhinib20 and CID1067700.

α1-Adrenoceptors are an important drug target in benign prostatic hyperplasia (BPH) and have been intensively studied at the expression level, but their posttranslational regulation in the prostate is widely unknown. Here we examined modulation of prostate smooth muscle contractions by inhibitors for RabGTPases, which are involved in intracellular transport of internalized receptors and in vesicle function. Tissues were obtained from laser enucleation for BPH and from radical prostatectomy without prior BPH surgery. Contractions were induced in an organ bath, with and without the Rab27 inhibit

Intrapancreatic Fat Deposition and Prostate Cancer Risk: A Primary Case-Control Study.

IntroductionVisceral fat is associated with cancer risk. Intrapancreatic fat deposition (IPFD) has been associated with pancreatic diseases, but its role in extra-pancreatic malignancies remains unclear. This case-control study aimed to investigate the association between IPFD and prostate cancer risk.MethodsA total of 143 patients with newly diagnosed, treatment-naïve prostate cancer and 286 age-matched controls without malignancy were enrolled in this retrospective study. IPFD was assessed using unenhanced computed tomography (CT) by two markers: pancreatic CT attenuation (PanCT) and pancrea

Evaluation of online dose-guided adaptive fractionation for intact prostate radiotherapy.

BACKGROUND: Dose-guided adaptive fractionation (DGAF) was proposed as a simpler alternative of adaptive radiation therapy, using organ variation simulation for proof of concept. However, its dosimetric benefit using actual daily anatomy changes in prostate cancer patients remains unclear. PURPOSE: The purpose of this study was to retrospectively evaluate the dosimetric benefit and potential risk of DGAF for prostate cancer patients treated with moderately hypofractionated radiotherapy and stereotactic body radiotherapy using daily cone-beam CT. METHODS: Patients assessed in this study were t

Loss of NCOA6 protein causes aggressive prostate cancer with EGFR overexpression and dependency.

Loss of tumor suppressors can drive carcinogenesis while simultaneously creating actionable therapeutic vulnerabilities. Epidermal growth factor receptor (EGFR) is frequently overexpressed in prostate cancer (PCa), yet EGFR inhibitors have shown limited clinical benefit, and the basis for this variability remains unclear. Here, analysis of 1,719 human PCa specimens reveals frequent downregulation of the nuclear receptor coactivator 6 (NCOA6), particularly in metastatic disease. Genetic loss of NCOA6 accelerates tumor growth in human PCa xenografts, induces prostate tumorigenesis in mice, and c

Efficacy and Safety of Tranexamic Acid in Endoscopic Surgery for Benign Prostatic Hyperplasia: A TURP-Dominated Systematic Review and Meta-Analysis.

BACKGROUND: Hemorrhaging continues to be a significant issue in endoscopic procedures for benign prostatic hyperplasia (BPH). Tranexamic acid (TXA) has garnered heightened scrutiny for its ability to mitigate bleeding-associated problems during transurethral prostate procedures. We performed a comprehensive meta-analysis to assess the effectiveness and safety of TXA in individuals having endoscopic procedures for BPH, acknowledging that the existing data primarily originated from transurethral resection of the prostate (TURP). METHODS: PubMed, Web of Science, Cochrane Library, Embase, CNKI, W

Tracking outcome changes in benign prostatic hyperplasia trials: a version history review of ClinicalTrials.gov records.

BACKGROUND: Benign prostatic hyperplasia (BPH) is a common condition with trials that frequently rely on subjective outcomes. While federal regulations require prospective outcome registration in ClinicalTrials.gov, modifications to these outcomes are permitted and often unjustified, potentially compromising transparency and interpretability. This study examined the frequency, timing, type, and transparency of prespecified outcome modifications in BPH trials. METHODS: We conducted a registry-based cross-sectional analysis of interventional BPH trials registered on ClinicalTrials.gov between J

Comparison of volumetric dynamic optical coherence tomography with biological methods for evaluation of radiation effects in prostate tumor spheroids.

SIGNIFICANCE: Three-dimensional (3D) tumor spheroids are more physiologically representative of in vivo patient tumors compared with two-dimensional monolayer culture. However, their 3D nature challenges the use of conventional biological techniques such as proliferation assays, fluorescence microscopy, and the clonogenic assay, which is the gold standard method for assessing cell survival following radiation. However, the clonogenic assay requires spheroid disaggregation. AIM: Non-invasive volumetric imaging with dynamic optical coherence tomography (dOCT) enables cellular activity to be vis

Dehydrated human amniotic membrane for early return of erectile function after robotic radical prostatectomy: a 10-year journey and lessons learned.

The first description of dehydrated human amniotic membrane (dHAM) use in robotic urologic surgery was by Razdan et al. in 2014 at the World Congress of Endourology. Since then, it has been utilized as a nerve and anastomotic wrap to enhance early return of erectile function and continence, respectively, during robotic radical prostatectomy (RALP). We describe our ten-year experience with dHAM as a nerve wrap. After obtaining hospital institutional review board (IRB) approval, a retrospective review of a prospectively maintained single center prostate cancer database was performed. All men und

Personalized genomic landscape of prostate cancer in African American men reveals novel drivers and therapeutic opportunities.

African American (AA) men experience disproportionately high prostate cancer incidence and mortality. Despite this disparity, current genomic repositories are predominantly derived from European populations, and studies using archived tissues are often limited by stromal contamination. We sought to characterize the high-resolution somatic mutational landscape of AA prostate cancer using a high-fidelity, patient-matched model system to identify population-specific drivers and therapeutic targets. We performed whole-genome sequencing (WGS) on 10 matched pairs of primary cell cultures derived fro

Immune biomarkers of response and resistance to ipilimumab and nivolumab in metastatic castration-resistant prostate cancer.

Immune checkpoint therapies have shown limited efficacy in metastatic castration-resistant prostate cancer (mCRPC). The AMADEUS prostate expansion cohort (NCT03651271) evaluated the efficacy, safety, and biomarkers of escalating ipilimumab doses combined with nivolumab. Patients received ipilimumab 1 milligram per kilogram (mg/kg) + nivolumab 360 mg (I1+N360), ipilimumab 3 mg/kg + nivolumab 1 mg/kg (I3+N1), or ipilimumab 5 mg/kg + nivolumab 1 mg/kg (I5+N1). Coprimary endpoints were 6-month disease control rate (DCR) and changes in tumoral CD8 percentage. Thirty-two patients were enrolled. DCRs

Citron kinase activity controls alternative splicing events that drive prostate cancer growth.

Prostate cancer (CaP) remains a major cause of cancer death in men, underscoring the need for mechanistically novel therapies. We previously identified the action of the mitotic kinase citron (CIT) kinase as a key driver of CaP progression and promising therapeutic target, but selective CIT inhibitors are not available. We therefore asked whether a better understanding of the action of CIT substrates, which execute CIT kinase action in CaP cells, can lead to novel approaches and/or alternative targets to indirectly interfere with CIT's activity during CaP progression. In view of the enrichment
