OBJECTIVE: To investigate the potential role of butyrylcholinesterase (BChE) enzyme activity and tentative association of BCHE gene single nucleotide polymorphisms (rs3495, rs1803274) with risk of male infertility. Male infertility is a highly prevalent multifactorial condition with complex heterogeneous phenotypic spectrum. A significant global variation has been observed in the prevalence of male infertility contributing to approximately 20%-70% of overall infertility. An interplay of genetic, hormonal, environmental, and lifestyle factors are involved in the development of male infertility.

Adenosine triphosphate (ATP) is essential for sperm motility. We previously found that optimized non-thermal dielectric barrier discharge (DBD) plasma treatment enhanced boar sperm quality by increasing ATP levels. However, the molecular mechanisms underlying this process, particularly the role of competing endogenous RNA (ceRNA) networks, remain unclear. In this study, a total of 266 mRNAs, 163 miRNAs, and 37 circRNAs were identified as differentially expressed in boar spermatozoa treated with optimized DBD plasma. Functional enrichment analysis revealed that ATP-related pathways, including A

Male infertility is a prevalent global health issue, with urological disorders representing some of the most common and correctable causes. Key conditions such as varicocele, obstructive azoospermia, erectile dysfunction and Peyronie's disease impair fertility through distinct pathophysiological mechanisms, including disrupted spermatogenesis, reproductive tract obstruction and failed sperm delivery. The effective management of these conditions hinges on a systematic diagnostic evaluation, which integrates clinical history, physical examination, semen analysis and specialized imaging. Modern m

Male infertility contributes to nearly half of the global infertility, yet nursing scholarship continues to marginalize men's experiences, reflecting entrenched assumptions that equate reproduction with femininity. This article reframes male infertility as a relational, socially embedded phenomenon and a site of disciplinary transformation. Drawing on intersectionality, masculinity theory, performativity, and stigma, this article shows how race, class, sexuality, disability, and cultural context intersect to produce compounded stigma, invisibility, and restricted access to care. For example, q

BACKGROUND: Normozoospermia does not guarantee fertility. Sperm DNA fragmentation (SDF) reflects intrinsic sperm quality and adverse reproductive outcomes, and double-strand breaks (DSBs) indicate more severe DNA damage.
METHODS: From July 2021 to February 2025, semen parameters, total motile sperm count (TMSC), and SDF from 534 normozoospermic men at National Cheng Kung University Hospital were retrospectively analyzed. Total DNA fragmentation (Total DFI) was measured using the sperm chromatin dispersion (SCD) test, and DSB DFI was assessed using the SDF Releasing (SDFR) assay.
RESULTS: Pat

CONTEXT: Oxidative stress has a detrimental effect on sperm quality, which can lead to infertility. Antioxidant enzymes, such as superoxide dismutases (SODs), have crucial role to protect sperms from oxidative stress. Variations in the SOD genes have thus been linked to male fertility.
AIMS: The primary aim of the study was to evaluate the genetic factors contributing to male reproductive health in the population of Jammu and Kashmir. Specifically, the study aimed to assess the impact of SOD1/rs4998557 and SOD2/rs4880 polymorphisms on risk of male infertility and to investigate the gene-gene

Identifying the potential oligogenic causes of rare diseases remains a challenge, notwithstanding the advancements made in the last decade. While a variety of predictive and ranking approaches have been proposed, their precision remains limited, as only a small number of high-quality training cases are available and it remains difficult to know which features may be most relevant for the design of new predictors. We hypothesize here that structured biological information, which provides an integration of various relevant biological networks and ontologies in a single heterogeneous knowledge gr

RATIONALE AND OBJECTIVES: Contrast-enhanced ultrasound (CEUS) is an effective method for assessing varicocele (VC) hemodynamics; however, its value in predicting improvements in semen parameters after varicocelectomy remains unclear. This study investigated whether CEUS-based hemodynamic patterns could predict surgical outcomes and developed and validated a novel predictive model.
MATERIALS AND METHODS: In this prospective cohort study, 187 patients with VC undergoing microscopic varicocelectomy were included. A total of 23 clinical, grayscale ultrasound, and CEUS-based hemodynamic parameters

The progression of spermatogenesis is under dynamic transcriptional regulation. As a subunit of the transcription-export complex 2 (TREX-2), PCI domain-containing protein 2 (PCID2), participates in RNA processing. However, the physiological functions of PCID2 in spermatogenesis remain poorly understood. Here, we generate germline conditional knockout (Pcid2-SKO) mice using Stra8-Cre, and it is found that Pcid2-SKO mice are infertile, exhibit extensive germ cell apoptosis, impaired spermatogonial differentiation, and failure of meiosis initiation. Single-cell transcriptome analysis reveals deve

Lead (Pb) exposure is a risk factor of male infertility, while the transcriptional and epigenetic changes associated with lead exposure in spermatozoa are poorly understood. Our previous findings revealed significant changes in DNA methylation of the calcium (Ca) homeostasis pathway of human spermatozoa in men with a blood Pb level over 5 µg dl -1 , which was associated with decreased sperm motility. In this study, we explored the effects of Pb exposure on expression of differentially methylated genes (DMGs) by analyzing semen samples from six healthy, non-smoking, and non-drinking men (aged 2

Chronic methamphetamine (Meth) exposure has been recognized as a critical risk factor for male reproductive dysfunction, yet its mechanistic underpinnings remain elusive. This study elucidates the molecular pathways through which Meth compromises spermatogenesis in a murine model. Male mice subjected to 15 days of chronic Meth administration presented severe testicular atrophy, characterized by seminiferous epithelial disorganization and diminished sperm reserves in both the testes and epididymides. Quantitative assessments revealed marked reductions in sperm motility and increased tail abnorm

Macrozoospermia is a rare cause of male infertility characterized by a high proportion of sperm with large, irregular heads. In this study, we identified a novel homozygous Aurora kinase C (AURKC) missense variant (c.253G > A (p.Glu85Lys)) in a Chinese infertile man using whole-exome sequencing. Segregation of the AURKC c.253G > A variant within the family was confirmed by Sanger sequencing, and In silico prediction tools suggested that the variant is pathogenic. Sperm ultrastructural abnormalities in the patient were further examined using transmission electron microscopy. Our findings identi

Oligoasthenozoospermia (OAT) is a major cause of declining male fertility worldwide, characterized by reduced sperm count and motility. Its pathogenesis involves multiple factors including genetics, hormones, environment, and lifestyle. Due to ethical and practical limitations in human studies, animal models have become essential tools for elucidating OAT mechanisms and evaluating therapeutic strategies. This review aims to systematically organize and evaluate existing OAT animal models, including those established through chemical agents, heavy metals, endocrine disruptors, physical stress, g

BACKGROUND: Non-obstructive azoospermia (NOA) is a multifactorial disease with high genetic and phenotypic heterogeneity. The main NOA pathogenesis is still unknown, which makes it challenging to find precise biomarkers for this condition. Long non-coding RNAs (lncRNAs) have been linked to many processes related to male reproductive health, such as spermatogenesis and sperm maturation. So, this study investigated a panel of seven lncRNAs from the seminal plasma of patients with NOA, which has the potential to be used as noninvasive prognostic biomarkers in NOA. METHODS: The differential gene e

Accurate assessment of sperm concentration and motility is critical for the diagnosis and management of male infertility. However, current methods, manual hemocytometer counting and commercial computer-aided sperm analysis (CASA) systems, are limited by labor intensity, human error, and variable performance under diverse sample conditions. Here, we present an artificial intelligence (AI)-driven computer vision tool for high-resolution, quantitative analysis of sperm motility and concentration. In a prospective study of 26 semen samples (22 patients, 4 donors), we benchmarked the AI model again

Infertility affects one in six couples worldwide, yet men’s reproductive health remains marginalized in nursing scholarship. This exclusion is not simply a gap in evidence; it reflects deeper inequities rooted in gendered histories of reproduction, where women’s bodies have been medicalized and men’s virility preserved as cultural capital. Such dynamics have produced epistemological silences that perpetuate inequitable care. Drawing on theories of traditional masculinity, stigma, and nursing epistemology, this commentary positions male infertility as a critical equity case that exposes how dis

Infertility affects 15% of couples worldwide, with male factors contributing to over half of the cases. Varicocele, present in 35% of men with primary infertility, impairs sperm quality. This study evaluates the effect of electroacupuncture (EA) on improving sperm motility and morphology in varicocele patients. A single-blind, randomized controlled trial was conducted with 14 participants divided into an EA group (n=7) and a control group (n=7). The intervention group received EA in combination with a herbal regimen and Tribulus Terrestris, while the control group received only the herbal regi

Male factor infertility accounts for roughly half of all infertility cases, yet receives far less clinical, research, and public attention than female infertility. Historical, cultural, and medical factors, particularly the absence of comparable therapeutic advances, contribute to this neglect. A diagnosis can threaten identity and masculinity, leading to depression, anxiety, and sexual dysfunction, while stigma and cultural taboos around semen collection compound distress. Men often feel excluded from care and remain socially invisible. Applying patient-centered frameworks such as the 4 "I" m

BACKGROUND: Azoospermia affects 1% of men and 10-20% of infertile males, yet the psychosocial mechanisms underlying self-esteem impairment remain poorly characterized. Guided by Connell's Masculinity Theory and Bury's Biographical Disruption Framework, this mixed methods study examined self-esteem experiences among Chinese men with azoospermia.
METHODS: An explanatory sequential design was employed. Phase 1 surveyed 216 men using Rosenberg's Self-Esteem Scale, with multiple regression identifying predictors. Phase 2 involved semi-structured interviews with 16 purposively sampled participants,

Perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA), two predominant per- and polyfluoroalkyl substances, are ubiquitous environmental contaminants that have been implicated in male infertility. However, the effects and underlying mechanisms of these compounds during human sperm capacitation remain unclear. In this study, human spermatozoa were exposed to PFOA and PFOS during capacitation. Acrosome reaction and sperm viability were assessed by flow cytometry. Sperm motility and hyperactivation were assessed using computer-assisted sperm analysis. Proteomic and phosphoproteomic p
