The preservation of sperm genetic integrity is pivotal for successful assisted reproduction. This study aimed to evaluate the effect of polyvinylpyrrolidone (PVP) on sperm DNA integrity by quantifying the mean number of DNA breakpoints (MDB). A total of 98 sperm samples were collected and subjected to comprehensive assessment of conventional sperm parameters, including concentration, progressive motility (PR), and sperm morphology rate. Furthermore, the MDB was assessed utilizing the TdT-SD Fluo-biosensor. The ultrastructure of the spermatozoa was meticulously examined using scanning electron

Human spermatogonial stem cells (SSCs) are crucial for spermatogenesis and male reproduction. Although abnormal pyroptosis caused by inflammation impacts male fertility, the molecular mechanisms underlying the pyroptosis of human SSCs are elusive. To induce pyroptosis, lipopolysaccharide (LPS) was introduced into the vas deferens of mice. RNA sequencing (RNA-Seq) of human SSCs was employed to identify NOD-like receptor thermal protein domain associated protein 3 (NLRP3), and RNA interference (RNAi) was used to determine the function and mechanism of NLRP3 in controlling pyroptosis of human SSC

Varicocele-induced decline in sperm quality is a common cause of male infertility. While varicocelectomy improves semen parameters, the mechanisms remain unclear. Recent studies suggested that small noncoding RNAs (sncRNAs) in sperm play a significant role in male reproductive health. This study investigated whether varicoceles and surgical treatment influence the expression profiles of sperm sncRNAs and explored the related epigenetic mechanisms. This study included 63 infertile males undergoing varicocelectomy. Semen analysis was performed, followed by high-throughput deep sequencing of sper

PURPOSE: Mammalian sperm require a large amount of ATP to sustain motility. While glycolysis and oxidative phosphorylation (OXPHOS) are established ATP sources, they cannot fully explain motility maintenance under glucose-deprived conditions. This study examined whether gluconeogenesis functions as a third metabolic pathway supporting sperm energy production.
METHODS: Sperm from mice, humans, cattle, and horses were comparatively analyzed under glucose-containing and glucose-free conditions with inhibitors of OXPHOS (FCCP) and fructose-1,6-bisphosphatase 1 (FBPase1). Motility and ATP levels w

Non-obstructive azoospermia (NOA) and cryptozoospermia are two significant conditions contributing to male infertility. However, the underlying genetic factors in most cases remain unknown. In our study, whole exome sequencing identified novel biallelic variants in TBC1 domain family member 8 (TBC1D8) in two patients. Patient P1 with NOA harbored c.890C>T (p.A297V) and c.2461G>A (p.V821I), and patient P2 with cryptozoospermia carried c.854C>T (p.P285L) and c.1912G>A (p.D638N). Bioinformatic analyses predicted that all identified TBC1D8 variants were likely pathogenic. Compared with a normal co

Male infertility is one of the most prevalent disorders affecting the male reproductive system, with the majority of cases originating from sperm abnormalities. Autophagy, a highly conserved cellular process, plays critical roles in spermatogenesis, sperm maturation, and functional maintenance. Aberrant autophagic activity is closely associated with various sperm abnormalities, including oligozoospermia, asthenozoospermia, teratozoospermia, and azoospermia. In this narrative review, we provide a comprehensive overview of the current knowledge on the role of autophagy in sperm abnormality-relat

BACKGROUND: Male infertility represents a major global health challenge. Heat shock protein A1A (HSPA1A), a stress-inducible molecular chaperone, shows potential importance in spermatogenesis, though its precise mechanistic role remains undefined.
METHODS: Analysis of human sperm transcriptome data (GSE6969) revealed HSPA1A expression in fertile versus infertile samples. Functional characterization involved the overexpression of HSPA1A in spermatogonia (GC-1 spg) and its knockdown in spermatocytes (GC-2 spd(ts)), assessing apoptosis, proliferation, and cell cycle progression. HSPA1A-interacti

The maintenance of mammalian spermatogenesis depends on the intricate molecular and cellular interactions between spermatogonial stem cells and their cognate niche in the seminiferous epithelium of the testis. To sustain the continuous production of sperm, spermatogonia proliferate and differentiate under the control of various niche factors, promoting either self-renewal or commitment to spermatogonial differentiation. Single-cell RNA sequencing analyses have identified different subpopulations of spermatogonia in primates based on the expression of specific marker genes (PIWIL4, GFRA1, NANOS

Sperm DNA fragmentation index (DFI) is a biomarker of male infertility, influenced primarily by oxidative stress. While specific nutrients and antioxidants have been studied, evidence on overall dietary patterns and sperm DNA integrity, particularly in idiopathic infertile men, is limited. This case-control study aimed to investigate the association between empirically derived dietary patterns and sperm DNA fragmentation in men with idiopathic infertility. In this case-control study conducted between May 2024 and March 2025 at a fertility clinic in Tehran, Iran, 310 idiopathic infertile men ag

This study investigates the protective effects of nicotinamide mononucleotide (NMN) against aluminum-induced testicular dysfunction in mice. While previous research has demonstrated the general protective role of NMN in testicular function and highlighted the interaction between aluminum and the NLRP3 inflammasome, the precise mechanisms through which NMN mitigates aluminum-induced reproductive toxicity remain unclear. Our findings show that NMN protects Sertoli cells by inhibiting NLRP3-mediated pyroptosis, which subsequently improves spermatogenesis and testosterone synthesis. We also identi

Obesity-related spermatogenic dysfunction has attracted growing attention. Concurrently, gonadotropin-inhibitory hor-mone (GnIH) has been recognized as a pivotal node bridging metabolic dysregulation and testicular dysfunction. Therefore, this study investigates the mechanistic role of the GnIH/G protein-coupled receptor 147 (GPR147) signaling pathway in spermatogenic dysfunction and metabolic syndrome-associated male infertility. This study employed Gene Expression Omnibus (GEO) database analysis and molecular biology techniques to examine the effects of GnIH/GPR147 signaling ablation on test

Obesity increases the risk of male infertility, primarily attributable to reduced testosterone levels. Exosomes, which facilitate intercellular communication during reproduction, may influence this process. However, the relation between exosomal cargo changes and high-fat diet (HFD)-induced testosterone decrease remains unclear. Here, we show that exosomes influence testosterone synthesis and spermatogenesis in HFD mice. Transferring exosomes derived from the serum of HFD mice to mice fed a normal diet decreases testosterone levels and sperm counts. Treatment with inhibitors of exosomes (GW486
