Altered sncRNA Signatures in Semen Extracellular Vesicles Between Patients with Benign and Malignant Prostate Disease as Potential Non-Invasive Biomarkers in the PSA Grey Zone.

PSA testing plays an important role in the diagnostic workup of prostate cancer; despite this, its cancer specificity is a well-recognised limitation. Consequently, more reliable, non-invasive diagnostic tools that are capable of improving specificity while maintaining sensitivity are needed, thereby enabling better risk stratification, personalised patient management, and reduction in unnecessary invasive procedures. In this study, we characterised the small RNA profile-including miRNAs and tsRNAs-in seminal small extracellular vesicles (sEVs) using high-throughput sequencing to expand biomar

Seminal fluid cytokines in reproductive health and fertility of men.

Seminal fluid is more than a vehicle for sperm transport. It contains a complex mix of immune-regulatory cytokines that, together with other bioactive factors, influence sperm maturation and survival, and interact with female reproductive tract cells and tissues to facilitate conception and stimulate female receptivity to pregnancy. Cytokines synthesized in the seminal vesicles, prostate, and other male accessory glands come into contact with sperm at ejaculation, and upon intromission are delivered to the cervix and uterus in soluble form, as cargo of seminal extracellular vesicles, or in phy

Mechanisms and Current Therapeutic Targets of Podocyte Damage in Diabetic Kidney Disease.

INTRODUCTION: Diabetic Kidney Disease (DKD) is a leading cause of end-stage kidney disease, and podocyte injury is a pivotal driver of albuminuria, glomerulosclerosis, and progressive loss of kidney function. METHODS: A targeted narrative search of PubMed/MEDLINE, Scopus, and Web of Science was performed, primarily covering 2015-2025 and supplemented by seminal earlier studies. Inclusion criteria prioritized human studies, randomized and translational studies, and mechanistic work directly addressing podocyte injury or podocyte-relevant therapeutic effects in DKD. Studies with limited mechani

Zinner syndrome in adult male: a rare case report.

BACKGROUND: Zinner syndrome (ZS) is an extremely rare congenital genitourinary disease. Its typical triad consists of seminal vesicle cysts, ejaculatory duct obstruction, and ipsilateral renal agenesis or hypoplasia. At present, the exact pathogenesis of ZS remains a mystery. It is hypothesized that ZS might be associated with aberrations that occur during the intricate process of embryonic development. CASE DESCRIPTION: A 40-year-old male presented to the hospital with a chief complaint of experiencing dysuria for 5 consecutive days, accompanied by frequent urination and urgency. Through com

Human Herpes Virus Genotype and Immunological Gene Expression Profile in Prostate Cancer with Prominent Inflammation.

We aim to investigate the role of the Herpesviridae family (HHV) in the onset and progression of prostate cancer (PCa) and to profile the local PCa immunological status. A total of 116 "tru-cut" biopsies (58 PCa and 58 benign prostatic hyperplasia [BPH]) and 49 formalin-fixed paraffin-embedded (FFPE) instances of PCa were analysed using real-time qPCR and histological examination. Infection with CMV, EBV, HHV6, and HHV7 was detected in 11.5% of the "tru-cut" biopsies (25.9% in BPH and 6.9% in the PCa group). In the formalin-fixed paraffin-embedded (FFPE) samples, infection was detected in 69.4

Candidate Transcript Panel in Semen Extracellular Vesicles Can Improve Prediction of Aggressiveness of Prostate Cancer.

The need for prostate cancer (PCa)-specific biomarkers that enable more accurate detection of the disease and better prediction of tumor aggressiveness remains ongoing due to the low cancer specificity of PSA screening. Several potential mRNA markers for diagnosing PCa, in tissue and urine, have been reported in the literature. In this study, we aim to explore the potential of selected prostate-specific molecules and transcripts contained in small extracellular vesicles (sEVs) in semen to predict PCa risk reclassification for patients with moderately elevated PSA levels-a clinical scenario whe

Non-Vesicular Extracellular Particle (NVEP) Proteomes from Diverse Biological Sources Reveal Specific Marker Composition with Varying Enrichment Levels.

Extracellular particles (EPs), an umbrella term encompassing membrane-enclosed extracellular vesicles (EVs) and non-vesicular extracellular particles ([NVEPs], previously described as extracellular condensates [ECs]) contain a complex cargo of biomolecules, including DNA, RNA, proteins, and lipids, reflecting the physiological state of their cell of origin. Identifying proteins associated with EPs that regulate host responses to physiological and pathophysiological processes is of critical importance. Here, we report the findings of our study to gain insight into the proteins associated with N

Atypical Presentation of IgG4-Related Disease: A Case Report of Obstructive Seminal Vesiculitis.

Immunoglobulin G4-related disease (IgG4-RD) is a systemic fibroinflammatory condition known to form "pseudotumors" that mimic malignancy. We previously reported the case of a 59-year-old male who presented with acute kidney injury from a unilateral ureteric obstruction, diagnosed as right-sided seminal vesiculitis. We now report the crucial clinical follow-up. During surveillance, the patient developed a new, contralateral pelvic mass and presacral thickening. A PET-CT scan confirmed high metabolic activity, raising suspicion for malignancy. Serological testing was definitive, revealing a mark

Prostogrit mitigates testosterone/estradiol induced prostatic enlargement/remodeling in rat model of benign prostatic hyperplasia and fine-tunes prostatic expression of Adra1a and Il6 genes.

BACKGROUND: Benign prostate hyperplasia (BPH), the pathological basis of benign prostatic syndrome (BPS), is an ageing-associated, androgen-driven, non-malignant growth of the prostate, with an inflammatory etiology. It is characterized by hyperproliferation of prostate cells and bothersome lower urinary tract symptoms. Current therapies for BPH provide both symptomatic relief and are disease modifying. However, they are associated with treatment limiting adverse effects. Accordingly, safer and effective alternatives to conventional treatments are required. Prostogrit is a novel, Ayurvedic med

Adjuvant-induced arthritis induces epithelial proliferation and differential expression of SVS2 and SVS3 in the seminal vesicles.

BACKGROUND: Rheumatoid arthritis (RA) is an inflammatory disease triggered by chronic and systemic activation of the immune system, with consequences for male fertility. However, the influence of RA on sexual accessory glands remains poorly investigated. OBJECTIVES: This study evaluated the late impact of adjuvant-induced arthritis (AIA) on the morphophysiology of the seminal vesicles and verified whether these effects can be influenced by androgen deprivation. MATERIALS AND METHODS: Adult male Wistar rats were allocated into four experimental groups and subjected to induction of AIA (via in

Semen quality and pathological lesions in rams naturally infected with Brucella ovis during an outbreak in Saskatchewan.

This report documents clinical signs, semen parameters, and reproductive organ lesions in rams during a natural Brucella ovis outbreak. Eleven rams from 1 flock underwent 2 breeding soundness evaluations and necropsies. Brucella ovis was isolated from all semen ejaculates and confirmed via whole-genome sequencing. Despite above-threshold scrotal circumferences, all rams were categorized as unsatisfactory due to poor semen quality and leukocytes in semen. Semen motility values in 9 of 11 infected rams and morphologically normal sperm in all infected rams were below the recommended threshold val

Knockout of phosphatidylethanolamine binding protein4 (PEBP4) promotes chronic non-bacterial prostatitis by mediating the activation of NF-κB signaling.

BACKGROUND: The etiology of chronic prostatitis remains unclear; consequently, this disease is associated with recurrence and ineffective clinical therapy. Therefore, there is an urgent need to investigate the underlying pathogenesis of chronic prostatitis in order to develop more efficacious treatments. OBJECTIVE: The previous study found that knocking out of PEBP4 leads to chronic prostatitis in the male mice. This research aimed to identify the role of PEBP4 in prostatitis, determine the molecular pathogenic mechanisms associated with chronic prostatitis, and provide guidelines for the dev

Seminal vesicles in focus: An illustrated overview.

Seminal vesicles play a crucial role in the male reproductive system, as they are responsible for secreting a fluid that forms most of the ejaculate. Seminal vesicles' pathology can present with non-specific symptoms, making imaging diagnosis essential for proper patient management. Various imaging modalities can be used to evaluate these glands, with MRI beneficial in illustrating the spectrum of seminal vesicle disease. Typical seminal vesicles appear as elongated fluid-containing structures, but congenital anomalies, inflammatory conditions, and neoplastic disorders can alter their appearan

Early-life gut inflammation drives sex-dependent shifts in the microbiome-endocrine-brain axis.

Despite recent advances in understanding the connection between the gut microbiota and the adult brain, significant knowledge gaps remain regarding how gut inflammation affects brain development. We hypothesized that gut inflammation during early life would negatively affect neurodevelopment by disrupting microbiota communication to the brain. We therefore developed a novel pediatric chemical model of inflammatory bowel disease (IBD), an incurable condition affecting millions of people worldwide. IBD is characterized by chronic intestinal inflammation, and is associated with comorbid symptoms

Sperm acquire epididymis-derived proteins through epididymosomes.

STUDY QUESTION: Are epididymosomes implicated in protein transfer from the epididymis to spermatozoa? SUMMARY ANSWER: We characterized the contribution of epididymal secretions to the sperm proteome and demonstrated that sperm acquire epididymal proteins through epididymosomes. WHAT IS KNOWN ALREADY: Testicular sperm are immature cells unable to fertilize an oocyte. After leaving the testis, sperm transit along the epididymis to acquire motility and fertilizing abilities. It is well known that marked changes in the sperm proteome profile occur during epididymal maturation. Since the sperm is
