Endocrine-disrupting chemicals in male reproductive health: exposure risks, reproductive toxicity and emerging perspectives.

Male reproductive health represents a growing global health challenge, and increasing evidence highlights the contribution of environmental exposures to declining male reproductive function. Endocrine-disrupting chemicals are widespread environmental contaminants that interfere with endocrine regulation and have been associated with impaired semen quality, hormonal imbalance and testicular dysfunction. Here, we summarize current advances in understanding EDC-induced male reproductive toxicity, focusing on major chemical classes, including bisphenols, phthalates, pesticides, per- and polyfluoro

General versus site specific quantile random forest models for virtual patient-specific quality assurance of VMAT plans.

BACKGROUND: Modern radiotherapy techniques like volumetric-modulated arc therapy (VMAT) require labor-intensive patient-specific quality assurance (PSQA) measurements, which for busy clinics pose a substantial workflow burden. Most of these measurements simply confirm acceptable plans. This inefficiency has motivated the development of machine learning (ML) models for virtual PSQA to predict the gamma passing rate (GPR) and safely reduce the need for pre-treatment measurements. Plan complexity, often quantified by metrics like the small aperture score (SAS) and average leaf pair opening (ALPO)

Successful management of synchronous prostate adenocarcinoma and lung squamous cell carcinoma with sintilimab and ADT in a frail elderly patient with bronchiectasis: a case report.

BACKGROUND: The management of elderly patients with multiple primary malignant neoplasms (MPMNs) presents a significant clinical challenge, requiring a balance between oncological efficacy and treatment tolerability. This case report details an elderly, non-verbal male with significant cardiopulmonary comorbidities who presented with synchronous prostate adenocarcinoma and lung squamous cell carcinoma (LUSC). The coexistence of two primary malignancies complicated by recurrent pulmonary infection is exceedingly rare in both clinical guidelines and real-world practice, making the proposed treat

The molecular heterogeneity and clinical impact of genomic driver alterations in prostate cancer.

Prostate cancer is a common malignancy with a heterogeneous disease course. Prior studies have identified a paucity of point mutations in protein-coding genes and only a few clinically relevant driver alterations. Here, using the Pan Prostate Cancer Group whole genome sequencing dataset from 1001 men, we perform an integrative analysis of cancer driver genes by quantifying recurrent genomic alterations among small mutations, structural variants, and copy number alterations in 961 primary prostate tumour samples. We identify at least one recurrent somatic or germline alteration in 97% of tumour

A Combination of Ursolic Acid and Oleanolic Acid Disrupts Bioenergetic Homeostasis in Prostate Cancer and Inhibits Cancer Growth by Cytotoxic Autophagy.

Oleanolic acid (OA) and ursolic acid (UA) are bioactive pentacyclic triterpenoids (PTTs) and potential therapeutics for cancers. If tested individually, their therapeutic efficacy is limited. The antitumor potential of OA, UA, and their combination was tested against castration-resistant prostate cancer cells (CRPCs) in vitro and in vivo. UA, OA, and UA + OA (1:1) inhibited the cell proliferation and clonogenic growth of CRPC cells. Compared to the potency of individual OA and UA on these cells, their potency was synergistically enhanced when tested as an OA + UA mix. Neither of these compound

The Enemy of My Enemy: Two Cases of De Novo Metastatic Small-Cell Neuroendocrine Prostate Cancer with BRCA2 Alterations and Brain Metastases.

De novo small-cell neuroendocrine prostate cancer (SCNE PC) is a rare and highly aggressive subtype of prostate cancer with limited treatment options. BRCA2 alterations may increase sensitivity to platinum-based chemotherapy and PARP inhibition; however, evidence regarding their clinical relevance in SCNE PC is limited. We present two cases of de novo SCNE PC with distinct BRCA2 alterations. Both patients presented with a similarly extensive disease burden and initially received carboplatin and etoposide (CE), but their clinical courses differed markedly. Patient 1 developed brain metastases t

Gut Microbiota Signatures Associated with Metastatic Prostatic Cancer Treated with Androgen Receptor Pathway Inhibitor (ARPI) Therapy.

Metastatic prostate cancer (MPCa) remains an aggressive disease despite the introduction of androgen receptor pathway inhibitors (ARPIs), and biomarkers predicting progression are still lacking. The aim of this study was to investigate gut microbiota (GM) composition and functional potential associated with advanced disease and progression under ARPI therapy. The GM of 15 patients with metastatic PCa receiving ARPIs, including 5 patients with disease progression, was compared with that of 39 patients with untreated (UT) PCa using shallow shotgun metagenomic sequencing. The metastatic PCa cohor

Synthesis of Curcumin Analogs Targeting Androgen Receptor for the Selective Inhibition of Castration-Resistant Prostate Cancer Cells.

In the present study, a series of curcumin analogs (A1-P3) were designed and synthesized for the inhibition of prostate cancer cells by targeting androgen receptor (AR). All synthesized compounds were structurally characterized by 1H NMR, 13C NMR, and high-resolution mass spectrometry (HRMS). In vitro cytotoxicity assays demonstrated that multiple compounds exhibited comparable or improved cytotoxicity against AR-positive 22Rv1 cells as compared to dimethylcurcumin (DMC, ASC-J9). Among these compounds, compounds A2, A3, B1-B3, D1, F1, F3, G1, G3, H1, and P3 possessed higher selectivity toward

Homoharringtonine Suppresses Prostate Cancer Progression Through Modulation of the TNF/NF-κB and MAPK Signaling Pathways.

Prostate cancer (PCa) represents one of the leading causes of cancer-related mortality among men. The efficacy of its treatment is often compromised by chemotherapeutic toxicity and multidrug resistance, underscoring the urgent need to develop efficient and low-toxicity alternative therapeutic strategies. Homoharringtonine (HHT), a bioactive compound, has demonstrated clinical anticancer efficacy in hematologic malignancies. However, its role and underlying mechanisms in prostate cancer remain largely unclear. This study provides evidence that HHT suppresses prostate cancer cell proliferation

Advances in Urinary Liquid Biopsy: A Review of Emerging Sensing Platforms for Early Cancer Screening and Diagnosis.

Early screening of many types of cancers remains limited by the invasiveness of gold-standard tissue biopsies and the poor specificity of traditional clinical biomarkers like prostate-specific antigen (PSA). Non-invasive liquid biopsies are the emerging alternatives. Urine is one of the preferred body fluids for liquid biopsy as it offers a rich, easily accessible and large-volume source of cancer-specific metabolites, proteins, nucleic acids, extracellular vesicles, and cancer cells. This review reports the recent advancements in urinary cancer (bio)sensing technologies, including mainly opti

Adrenergic Contraction of Human Prostate Tissues Is Maintained Under the Rab GTPase Inhibitors Nexhinib20 and CID1067700.

α1-Adrenoceptors are an important drug target in benign prostatic hyperplasia (BPH) and have been intensively studied at the expression level, but their posttranslational regulation in the prostate is widely unknown. Here we examined modulation of prostate smooth muscle contractions by inhibitors for RabGTPases, which are involved in intracellular transport of internalized receptors and in vesicle function. Tissues were obtained from laser enucleation for BPH and from radical prostatectomy without prior BPH surgery. Contractions were induced in an organ bath, with and without the Rab27 inhibit
