Gendered ageism among women undergoing mastectomy across two life stages: early adulthood and later life in Pakistan.

This qualitative study examines how gender and age shape breast cancer and mastectomy experiences among women in Pakistan. Using critical phenomenology and drawing on 12 in-depth interviews with women across two distinct age groups (early adulthood and later life), the study identifies three key themes regarding age- and gender-based inequalities that shape women's access to care. These inequalities are produced not only within the healthcare system but also through social and familial structures: [1] From Surveillance to Neglect: Age-Stratified Gatekeeping and Delayed Diagnosis, [2] Sacrifici

Phytochemical modulation of DNMT1-mediated DNA methylation: computational insights into breast cancer.

CONTEXT: Aberrant DNA methylation, particularly DNMT1-mediated hypermethylation of tumor suppressor and DNA repair genes, contributes to breast cancer progression. The limitations associated with existing DNMT inhibitors highlight the need for alternative and potentially safer epigenetic modulators. Here, we developed a computational workflow to identify phytochemicals targeting the catalytic methyltransferase domain of DNMT1. Screening of the SelleckChem phytochemical library identified Echinacoside (-9.56 kcal/mol), 1,2,3,4,6-O-Pentagalloylglucose (-9.49 kcal/mol), and Complanatoside A (-9.2

Investigation of DNA methylation levels before and after treatment in breast cancer patients receiving neoadjuvant chemotherapy.

PURPOSE: Drug resistance is a major cause of treatment failure in breast cancer patients. Numerous genetic and epigenetic markers have been proposed to influence Neoadjuvant Chemotherapy (NACT) response, and genes that regulate the effectiveness of chemotherapeutic agents undergo expression regulation via methylation. In this study, we aimed to investigate the methylation levels of KDM5A, KDM5B, and KDM5C, as well as their associated genes, EZH2, TFAP2C, ZMYND8, and SIN3B (Exon 1-2 and 19-20), in blood samples collected from patients before and after NACT treatment. Besides, we also examined p

Vacuum-Assisted Breast Biopsy and Excision for Breast Lesions: A Systematic Review and Meta-analysis of Malignant Upgrade in B3 Lesions.

Vacuum-assisted breast biopsy (VAB/VABB) and vacuum-assisted excision (VAE) serve distinct diagnostic and therapeutic roles, requiring pathway-specific evaluation of their outcomes. This systematic review reconstructed a report-level and outcome-level dataset from 26 publications, of which 18 were reviewed in full text and 8 remained incompletely retrieved. Reports without complete full text were retained for narrative context but excluded from quantitative synthesis. Five full-text studies contributed 98 malignant upgrades among 844 B3 lesions after diagnostic VAB/VABB. The pooled proportion

Breast Carcinoma With Features of Both Solid Papillary Carcinoma and Invasive Lobular Carcinoma: Diagnostic Challenges and Considerations.

Solid papillary carcinomas (SPCs) are uncommon breast tumors, and they are listed as a separate entity of papillary neoplasm, with no clear etiology and pathophysiology recognized so far. SPC without invasion is treated and managed as a form of ductal carcinoma in situ; when invasion is present, both ductal and lobular carcinoma can be present as the invasive component. Here we present a breast tumor showing features of both SPC and invasive lobular carcinoma (ILC). For either diagnosis, this tumor has unusual morphologic and immunophenotypic characteristics that have not been reported in lite

[Challenges and opportunities in breast cancer prevention in Mendoza].

INTRODUCTION: Mendoza has a medium-high breast cancer mortality rate, the fifth highest in Argentina. Awareness and prevention are essential to reduce its mortality, both closely linked to knowledge. Primary prevention aims to avoid risk factors, while secondary prevention focuses on early detection. MATERIALS AND METHODS: This study assessed the level of knowledge about breast cancer and its preventive strategies through a survey conducted via a Google Form, addressed to women over 18 years of age, answered by 760 women. RESULTS: Only 37.4% identified mammography as the most effective detec

What Matters in Breast Cancer Survivorship Care? Survivors' and Caregivers' Perspectives on Specialist and Potential Primary Care Involvement in Singapore.

Introduction/ObjectivesIn Singapore, breast cancer survivorship follow-up is largely specialist-led, creating sustainability challenges and leaving psychosocial needs unmet. Although primary care and shared-care models are adopted internationally, they remain underdeveloped locally. This study explored breast cancer survivors' and caregivers' perspectives on the current oncologist-led model and potential primary care involvement in survivorship care.MethodsA qualitative descriptive study was conducted using semi-structured interviews with 37 breast cancer survivors and caregivers recruited pur

Bridging gaps in breast cancer clinical trial enrollment: insights from a community-based research registry initiative.

OBJECTIVE: Cancer research registries are critical for ensuring representative participation in clinical trials. However, registries based primarily in hospitals may under-enroll women and other populations who experience structural, logistical, cultural, and informational barriers to care and research participation. Community-based organizations (CBOs) may help address this gap by developing registries that engage diverse populations of women at increased risk for and surviving with breast cancer more directly. The purpose of this study was to examine demographic, clinical, and organizational

Surface-Enhanced Raman Scattering pH Sensing in 3D-Printed Breast Cancer Models In Vitro.

With the increasing complexity of in vitro models for tissues, cancer, and other diseases, there is a need to develop robust biosensors capable of addressing these dynamic systems. Herein, we demonstrate the use of surface-enhanced Raman scattering (SERS)-based biosensors to monitor pH in 3D cellular models. We load breast cancer cells with pH-sensitive SERS nanotags comprising polymer-encapsulated gold nanostars functionalized with the pH-sensitive Raman reporter molecule 4-mercaptobenzoic acid, we then seed the labeled cells onto a 3D-printed hydrogel scaffold and expose the model to differe

A Selective ERα Photodegrader for Precise and Efficient Breast Cancer Treatment.

Breast cancer is the most prevalent malignancy in women, with over 70% of cases being estrogen receptor α (ERα)-positive. Photodynamic therapy (PDT) is spatially precise yet limited by tumor-selective photosensitizers. Herein, we report the first co-crystal structure of 2,2'-dihydroxytriphenylamine bound to ERα, which provided a structural blueprint for designing EP5, an innovative ERα-targeted photodegrader. EP5 exhibits high binding affinity toward ERα (KD = 56.7 nM), enabling its selective accumulation in ERα-positive breast cancer cells. Upon white-light irradiation, EP5 triggers selective

A Bead-Based DNA Classifier for Multi-Input Cancer Diagnosis.

DNA computation offers a promising route for multi-input cancer diagnosis, yet its translation into practical classifiers is often impeded by complex probe design and operational intricacies. Here, we introduce a bead-based DNA classifier (BDC) platform designed for multimiRNA-based diagnostic applications. This platform performs weighted molecular computation through a combination of magnetic bead capture and polymerase-mediated strand displacement. An in silico diagnostic model for breast cancer was built using an SVM algorithm trained on TCGA miRNA-seq data, which revealed a three-miRNA sig

Discovery of Tetrasubstituted Quinazolin-4(3H)-ones as Wild-type-sparing, Highly Selective PI3KαH1047R Inhibitors.

Inhibiting PI3Kα has demonstrated promising therapeutic effects for patients with HR+/HER2- and PIK3CA-mutated advanced or metastatic breast cancer. However, hyperglycemia and other notable adverse events (AE) associated with concomitant wild-type inhibition remain a challenge and limit dosing. Selectively targeting the mutant PI3KαH1047R could reduce wild-type-driven AEs while retaining clinical efficacy. Herein, we report the design and identification of tetrasubstituted quinazolin-4(3H)-ones that can selectively inhibit PI3KαH1047R through a proposed slow-on and slow-off mechanism. Compound

Breast cancer screening for persons experiencing homelessness.

BACKGROUND: Primary care providers (PCPs) care for their patients in part by helping them to follow cancer screening guidelines, including screening mammography. However, completing screening and follow-up due to abnormal results can be difficult for PCPs caring for patients with barriers to care, such as persons experiencing homelessness (PEH). The large population of PEH in the US have many unmet healthcare needs, high cancer prevalence and mortality, and therefore a need for cancer screening interventions. Hence we created and evaluated a specialized breast cancer screening program for PEH.

Do new guidelines enable a more accurate comparison of [18F]FDG PET/CT features with histopathology in primary breast cancer?

BACKGROUND: Recent EANM-SNMMI guidelines suggest that quantitative [18F]FDG PET parameters, including standardised uptake values (SUV), metabolic tumour volume (MTV) and total lesion glycolysis (TLG), provide valuable prognostic information in breast cancer. We aim to evaluate the association between [18F]FDG PET parameters in a large real-world cohort of high-risk breast cancer patients and the four main intrinsic subtypes of breast cancer defined by the EANM-SNMMI guidelines. METHODS: All women with newly diagnosed high-risk breast cancer undergoing [18F]FDG PET/CT between 1/8/22  and 17/4/

Age-specific survival after breast conservation versus mastectomy for ductal carcinoma in situ of the breast.

PURPOSE: Breast conservation for invasive breast cancer is associated with better survival and fewer complications than mastectomy, which is especially relevant for older patients. It is unclear whether this can be extrapolated to ductal carcinoma in situ (DCIS). METHODS: This population-based cohort study used data from the Breast Cancer Database Sweden (BCBaSe 3.0) to compare overall survival (OS), breast cancer-specific survival (BCSS) and local recurrence (LR) probabilities in three different age groups (< 60, 60-69, 70 + years) of women with primary DCIS undergoing breast-conserving surg

Preoperative prediction of positive margins in breast-conserving surgery using an ensemble model based on DCE-MRI: a multi-center retrospective and prospective study.

BACKGROUND: Breast-conserving surgery (BCS) is the standard care for early breast cancer. However, positive surgical margins lead to high recurrence and require reoperation, remaining a clinical challenge. This study aims to develop an ensemble model to preoperatively predict positive margins using dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI). METHODS: A total of 887 patients from three Chinese medical centers were included. Patients from the Yunnan (581 patients) and Fujian (51 patients) provinces were used for model development, with external testing on 188 patients from G

Macrophage-Targeted Quantum Dots with Short-Wave Infrared Emission for In Vivo Breast Cancer Imaging.

In vivo characterization and visualization of tumors often relies on radioactive tracers and expensive, complex equipment, which limits the frequency of analysis, throughput, and accessibility. Short-wave infrared (SWIR) fluorescence is a new nonradioisotopic imaging modality with flexible readouts at high depth and high signal-to-background contrast. Here, we report semiconductor quantum dots (QDs) with SWIR fluorescence engineered to selectively target tumor-associated macrophages (TAMs) by functionalization with the polysaccharide dextran (Dex). TAMs are often abundant in the tumor microenv
