A Senescence-Related Signature for Risk Stratification and Predicted Immunotherapy Response in Breast Cancer: Integrative Pathology and Multi-Omics Analysis.

Cellular senescence exerts context-dependent effects on breast cancer progression and therapeutic response through dynamic interactions with the tumor microenvironment; however, its pathological manifestations, cellular organization, and clinical significance remain incompletely understood. In this study, we integrated whole-slide pathology images, bulk transcriptomic profiles, single-cell ribonucleic acid (RNA) sequencing, spatial transcriptomics, and clinical data to systematically characterize senescence-associated features in breast cancer. Quantitative pathology and deep-learning features

PTHrP induces breast cancer bone metastases via microbial-dependent expansion of TNF/IL-17-producing T cells.

Bone metastases are a frequent complication of breast cancer, leading to reduced quality of life and increased morbidity. By intratibially injecting PyMT breast cancer cells in mice, we showed that the presence in the gut microbiome of segmented filamentous bacteria (SFB), a taxon that induces IL-17-producing Th17 cells and TNF-α (TNF)-producing T cells (TNF+ T cells), accelerated intraosseous tumor growth. SFB increased the activity of parathyroid hormone-related protein (PTHrP), a known driver of breast cancer bone metastasis produced by breast cancer cells. SFB promoted the PTHrP-induced ex

Septins buffer actomyosin forces to protect the nucleus from genotoxic mechanical stress.

Invasively migrating cells thread their nucleus through confined interstitial spaces. How cells protect the nucleus from intracellular forces is poorly understood. Here, we show that the septin cytoskeleton buffers the actomyosin forces that have been shown to power nuclear movement. Septin filaments comprising SEPT9, a septin amplified in breast cancer, align with perinuclear actomyosin cables, which exhibit higher tensile stress during 3D confined migration through narrower pores. SEPT9 depletion amplifies actin stress during confined migration and after myosin II hyperactivation in nonmigra

Mitochondria-Targeted Prodrug Nanoassemblies Elicit Antitumor Immunity via Mitophagy-Boosted Ferroptosis in Triple-Negative Breast Cancer.

Ferroptosis, an immunogenic iron-dependent cell death driven by lipid peroxidation, holds promise for treating triple-negative breast cancer (TNBC). However, its efficacy is severely compromised by restricted iron availability and the potent GPX4/GSH defense system. Herein, we developed a mitochondria-targeted prodrug by conjugating sulfasalazine (SAS) to triphenylphosphonium (TPP) via a disulfide-containing linker, which self-assembles into carrier-free nanoassemblies (ASSP). After cellular internalization and lysosomal release, SAS-SS-TPP accumulates in mitochondria, where thiol-disulfide ex

High-Affinity Y1 Receptor Theranostics: First-in-Class Stabilized Nonapeptides for Breast Cancer Imaging and Therapy.

Theranostic agents integrate diagnostic and therapeutic capabilities into a single molecular platform, offering an advanced approach to personalized medicine. The neuropeptide Y1 receptor (Y1R) is a promising target for breast cancer theranostics given its overexpression across multiple disease stages. Building on the Y1R-selective antagonist [Lys2, Arg4]BVD-15, we report the systematic development of stabilized nonapeptide analogues through strategic modifications. Lead candidates were radiolabeled in >95% radiochemical purity and showed Y1R binding affinities up to 60-fold higher than the pa

Developing a Ferric(III) Agent for Targeted Inhibition of Growth and Metastasis of Triple-Negative Breast Cancer Based on Human Serum Albumin by Chemodynamic Therapy-Driven Synergistic Therapy.

Triple-negative breast cancer (TNBC) remains challenging due to its aggressiveness and the lack of targeted therapies. In this study, we proposed designing a ferric (Fe) agent based on human serum albumin (HSA) for chemodynamic therapy-driven synergistic therapy. To this end, we not only optimized a series of Fe(III) compounds to obtain Fe4, which has remarkable toxicity against TNBC cells, but we also constructed the HSA-Fe4 NPs delivery system. Structural analysis revealed that Fe4 binds within the IB subdomain of HSA, with His-146 coordinating the iron center. Importantly, Fe4/HSA-Fe4 NPs s

Electrogenic HfO2/TiO2 Living Biointerface Dismantles Metabolic Radioresistance for Amplified Radio-Immunotherapy.

Radiotherapy (RT) efficacy in aggressive solid tumors is hindered by lactate-driven metabolic radioresistance and an immunosuppressive microenvironment. Here, we engineer a living biointerface (SHT) via in situ biomineralization of HfO2/TiO2 onto an electrogenic Shewanella oneidensis chassis to address this multifaceted challenge. This structural integration bridges the bacterial metabolism with interfacial radiocatalysis. The TiO2 shell acts as an efficient electron acceptor to accelerate bacterial extracellular electron transfer, driving rapid intratumoral lactate depletion, while the high-Z

Magnetic Particle Imaging Reveals Heterogeneous Retention, Leakage, and Redistribution of Nanoparticles Following Intratumoral Injection.

Intratumoral (i.t.) delivery of nanoparticles (NPs) is widely used to achieve high local NP concentrations. However, the temporal and spatial fate of i.t.-injected NPs remains poorly understood. We present a quantitative approach using whole-body in vivo magnetic particle imaging (MPI) to track magnetic NPs (MNPs) following i.t. injection. Using fiducial-calibrated imaging, we quantified MNP mass over time in subcutaneous 4T1 breast tumors. Longitudinal imaging revealed progressive loss of i.t. MNP content and heterogeneous off-target redistribution across animals despite standardized delivery

"Imaging Triplets on Two Wheels": 61Cu-, 64Cu-, and 68Ga-labeled Bicyclic Peptides Targeting Nectin-4 Enable High-Contrast PET Imaging of Urothelial Carcinoma and Triple-Negative Breast Cancer.

Nectin-4 is a validated target for theranostic approaches toward urothelial carcinoma, and radiolabeled ligands for PET and SPECT imaging are gaining increasing interest for noninvasive quantification of Nectin-4 prior to a targeted therapy. The goal of this study was to develop bicyclic peptides that allow radiolabeling with three different radionuclides (61Cu, 64Cu, and 68Ga) and to evaluate their suitability for high-contrast PET imaging in urothelial carcinoma (UC) and triple-negative breast cancer (TNBC). For this purpose, NECT-330, NECT-334, and NECT-346 were designed based on our previo

IL-1β/IL-6 inflammatory crosstalk drives DNA methylation and histone remodeling associated with cytokine induced senescence in luminal breast cancer.

BACKGROUND: Chronic inflammation exerts paradoxical effects in breast cancer, inducing growth arrest while sustaining tumor progression. However, how autocrine and paracrine inflammatory cytokine induced senescence integrates with tumor-promoting behaviour remains unclear. Here, we identify a reciprocal IL-1β/IL-6 circuit that links inflammatory cues, epigenetic remodeling and cytokine induced senescence in luminal breast cancer cells. METHODS: Following treatment with IL-1β and IL-6, MCF-7 cells were examined using cell-cycle, apoptosis, reactive oxygen species flow cytometry assays, and sen

Aggregation-Induced Emission Nanoassembly with Enhanced Radiosensitization for Boosting Breast Cancer Radiotherapy.

Radiosensitizers are crucial in augmenting the radiosensitivity of tumors, with organic variants showing significant promise because of their enhanced biocompatibility. Nonetheless, their clinical application has been impeded by two primary limitations: the inadequate generation of reactive oxygen species (ROS) upon X-ray irradiation and insufficient penetration into the tumor tissues. Herein, we developed an aggregation-induced emission nanoassembly (TTVP NPs) specifically engineered for enhanced radiotherapy in breast cancer. Benefiting from their ultrasmall particle size and positive surfac

Design, Synthesis and Biological Evaluation of New TRPM7 Channel Inhibitors as Potential Therapeutics for Breast Cancer.

TRPM7 channel is a critical regulator of cellular divalent cation homeostasis and a validated driver of cancer progression. Despite its therapeutic relevance, currently available TRPM7 inhibitors are limited in number and modest in potency with no TRPM7-targeted agents having advanced to clinical application. Herein, we report the structure-guided design and synthesis of a series of 2-aminobenzimidazole analogues based on NS8593, leading to the identification of two potent TRPM7 inhibitors, SDUY212 and SDUY225, that demonstrated robust electrophysiological inhibitory activity against TRPM7. Discovery of Dual PARP/GPX4 Inhibitors for the Treatment of Triple-Negative Breast Cancer.

Poly(ADP-ribose) polymerase (PARP) inhibitors have achieved clinical success in BRCA-mutated triple-negative breast cancer (TNBC), while their clinical utility remains limited in BRCA wild-type (WT) tumors. Co-targeting of PARP and other oncogenic signaling pathways represents an effective strategy to broaden the therapeutic applicability of PARP inhibitors in TNBC patients. Herein, we identified dual PARP/GPX4 inhibitors via a structure-based hybridization strategy and explored the dual-target antitumor effect in blocking the progression of TNBC. We found that compound 12 exhibited potent bio

Intratumoral Microbiome-Tumor Microenvironment Crosstalk in Breast Cancer Aggressiveness.

Breast cancer (BC) remains the most common malignancy among women worldwide, with incidence and mortality rates continuing to rise. Alongside tumor heterogeneity and diverse clinical outcomes, increasing attention has been directed toward the role of the intratumoral microbiome as a regulator of breast tumorigenesis. Recent evidence demonstrates that breast tissue harbors one of the most diverse microbial communities compared with other cancer types, with marked differences between normobiota and malignant tissue. Intratumoral microorganisms are potentially associated with BC progression throu

Integrating UHPLC-MS and MALDI-MSI for Spatial Nucleoside Profiling in FFPE Breast Cancer: A Multimodal Molecular Pathology Framework.

Formalin-fixed, paraffin-embedded (FFPE) tissues constitute the primary material for diagnostic pathology and retrospective clinical research, yet their use in metabolomics remains limited due to molecular cross-linking and analyte degradation. Here, we establish a cost-efficient molecular pathology workflow that integrates ultra-high-performance liquid chromatography mass spectrometry (UHPLC-MS) with matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to quantify and spatially map nucleosides in FFPE breast cancer tissues. Optimized extraction using methanol yiel

OpenDVP: an experimental and computational framework for community-empowered deep visual proteomics.

Deep visual proteomics (DVP) is an emerging approach for cell-type-specific and spatially resolved proteomics. However, its broad adoption has been constrained by the lack of an open-source end-to-end workflow within a community-driven ecosystem. Here we introduce openDVP, an experimental and computational framework for accessible and scalable DVP. OpenDVP combines open-source image analysis tools (MCMICRO, QuPath and napari) with scverse formats (AnnData and SpatialData) for multiomics integration. It provides two workflows: a fast-track pipeline requiring no image analysis expertise, and an

Neutrophils alter the metabolic profile of a coculture model of triple-negative breast cancer and stromal cells.

BACKGROUND: Breast cancer is one of the most common cancer types. Triple-negative breast cancer (TNBC), one of the most lethal breast cancer subtypes, is characterized by substantial neutrophil accumulation in the tumor microenvironment (TME), although the impact of neutrophils on disease progression remains unclear. METHODS: To model interactions between cancer cells and stromal cells, we used a coculture system combining a TNB Ccell line with breast fibroblasts and exposed these cultures to neutrophils isolated from healthy donors. RESULTS: Coculture generated a pro-inflammatory environmen

A Metabolic Nanoplatform Potentiates Triple-Negative Breast Cancer Radiosensitivity via Disrupting Carbonic Anhydrase IX/Pentose Phosphate Pathway Crosstalk.

Radiotherapy resistance in tumors driven by metabolic adaptations within acidic microenvironments involves the upregulation of both carbonic anhydrase IX (CA IX) and the pentose phosphate pathway (PPP) as well as the functional crosstalk between them. To overcome this, we developed a polyvinylpyrrolidone (PVP)-modified gadolinium acetazolamide (PGA) nanoradiosensitizer. PGA simultaneously inhibits CA IX to trigger intracellular acidification and tumor cell acidosis, suppresses PPP through regulation of glucose-6-phosphate dehydrogenase (G6PD), and depletes key radioresistance-sustaining metabo
