Traditional Chinese botanical medicines and plant-derived bioactive compounds in amyotrophic lateral sclerosis: mechanistic evidence, translational challenges, and therapeutic opportunities.

INTRODUCTION: Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease with limited therapeutic options, highlighting the need for multi-target interventions. Traditional Chinese botanical medicine (TCBM) interventions have attracted increasing attention as potential multi-target pharmacological candidates for ALS. In addition, a separate body of evidence derived from selected plant-derived bioactive compounds directly investigated in ALS-specific models provides complementary mechanistic insights into ALS-related pathways. However, the pharmacological mechanisms and translational p

Carbon dot-based nanomaterials in neurodegeneration: addressing neurobiological barriers and targeting challenges.

Neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis, represent a major global health challenge due to their progressive nature and limited therapeutic options. One of the primary obstacles to treating these disorders is the blood-brain barrier (BBB), which restricts the delivery of most therapeutic agents into the central nervous system. In recent years, carbon-based nanomaterials, particularly carbon dot (CD) nanostructures, have emerged as promising platforms for neurological applications owing to their ultra

Fibrocystic disease within an axillary accessory breast (Kajava class IV): a case report.

INTRODUCTION: Accessory breast tissue is a congenital anomaly occurring in 0.4-6% of the population, most commonly in the axilla. While this tissue is subject to the same physiological and pathological changes as orthotopic breast tissue, fibrocystic disease presenting as a large axillary mass is rare, representing a diagnostic challenge that mimics lipomas and other conditions common in the axillary region. CASE PRESENTATION: We report a case of 33-year-old female who presented with a chronic painful mass in the left axilla that have been present for 10 years. Clinical examination revealed a

Genetic tumor syndromes in female cancer: insights into inherited cancer predisposition and clinical implications.

A relevant proportion of malignancies predominantly or exclusively affecting women, including breast and gynecologic cancers, is attributable to hereditary tumor syndromes, profoundly impacting cancer risk, prognosis, and therapeutic management. Today, the routine use of comprehensive germline panels has shifted the focus from solely pathogenic BRCA1/2 variants to include numerous pathogenic variants of other high- and moderate-risk genes. A broad spectrum of genetic alterations has been identified as causative for Hereditary Breast and Ovarian Cancer syndrome (HBOC), encompassing not only BRC

An epidemiological study estimating the burden of cancer risk in patients with Raynaud's phenomenon.

INTRODUCTION/OBJECTIVE: Raynaud's phenomenon (RP) is a common vasospastic condition that may develop secondary to cancer and/or in association with systemic autoimmune rheumatic diseases (SARDs). We aimed to estimate the risk of cancer in RP without known SARDs, phenotypically akin to 'primary' RP. METHODS: A cohort study using data from North American electronic healthcare organization records. RP was defined using ≥ 2 ICD (I73.0) codes, excluding SARDs. Comparators had ≥ 2 irritable bowel syndrome ICD (K58) codes: selected with similar epidemiology to RP, and without any known excess in can

Additional ultrasound in women with dense breasts in a breast cancer-screening program based on digital breast tomosynthesis: evidences on cancer detection, false positives and associated costs.

Breast density is associated with increased risk of breast cancer (BC) and reduces mammography performance. Supplemental ultrasound (US) improves the performance of mammography screening programs in women with dense breasts but only few evidences are available about its added value when digital breast tomosynthesis (DBT) screening is performed. Our study evaluated the contribution of US in detecting BC in women with dense breasts in a screening program based on DBT. From February 2022 to October 2023, asymptomatic women (age range, 50-69 years) attending BC Screening in Verona, Italy, were pro

Case report: Carcinoma en cuirasse-widespread cutaneous metastases from breast adenocarcinoma.

RATIONALE: Cutaneous metastatic carcinoma is a malignant tumor that originates outside the skin. Studies have found that the most common primary tumor in female patients with cutaneous metastatic carcinoma is breast cancer. Cutaneous metastatic carcinoma spreads to the adjacent skin through blood vessels, lymphatics, or interstitial spaces. It forms ulcerative or nodular lesions. Carcinoma en cuirasse is characterized by extensive induration and sclerosis of the skin, resembling armor, and is a rare form of cutaneous metastatic carcinoma. PATIENT CONCERNS: We report a rare case of a 59-year-o

In vivo CRISPR/Cas9 Screening Reveals that UBE2L3 Modulates Autophagic Flux through TSC2 Ubiquitination and Potentiates PD-1 Blockade in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC), a distinct breast cancer subtype, poses significant challenges to conventional therapeutic approaches, and effective targeted therapies are limited. CRISPR/Cas9 library screening has demonstrated unprecedented efficiency and revolutionary potential in the identification of therapeutic targets. In this study, we performed In vivo CRISPR/Cas9 library screening and identified the E2 ubiquitin-conjugating enzyme UBE2L3 as a critical regulatory factor in the progression of TNBC. Loss of UBE2L3 restricted tumor cell growth by modulating autophagy in TNBC cells.

Lymphocytic mastopathy mimicking carcinoma in a non-insulin-dependent diabetic: a case report.

BACKGROUND: Lymphocytic mastopathy (also known as diabetic mastopathy, fibrotic mastopathy, or sclerosing lymphocytic lobulitis) is a rare benign fibroinflammatory breast disease that clinically and radiologically mimics breast carcinoma. It is most commonly associated with long-standing diabetes mellitus and autoimmune disorders. CASE PRESENTATION: We report the case of a 63-year-old woman with a 6-month history of a gradually enlarging, painless right breast lump. The patient was a known type 2 diabetic for 15 years on oral hypoglycaemic agents. Mammography and ultrasonography revealed an i

Co-Pathogenic Role of BRCA1 and OBSCN Deletions in Chinese Familial Breast Cancer: A Case Report.

BACKGROUND The incidence of breast cancer is high among women, with a significant proportion of cases being familial. However, the driver genes for breast cancer can differ across families. CASE REPORT Our patient was a 37-year-old woman diagnosed with triple-negative breast cancer (TNBC) by pathology, revealing invasive ductal carcinoma of the outer upper quadrant of the breast, WHO grade 3. The maximum diameter of the microscopic invasive cancer was approximately 0.5 cm. No definite vascular tumor thrombus or nerve invasion was observed. Some (30-90%) of the tumor cells disappeared, and the

[ 18 F]Fluoroestradiol Uptake in Benign Nodular Sclerosing Adenosis of the Breast.

A 70-year-old woman with estrogen receptor-positive (ER+) invasive ductal carcinoma (IDC) of the right breast underwent 18 F-fluoroestradiol ( 18 F-FES) PET/CT demonstrating uptake in the IDC (SUVmax 2.5) and also extensive 18 F-FES uptake in bilateral breasts corresponding to marked bilateral background enhancement on MRI. MRI-guided biopsy of a representative enhancing 18 F-FES-avid region in the left breast (SUVmax 3.5) revealed nodular sclerosing adenosis without cellular atypia or malignancy, but with 30% ER positivity on immunohistochemistry. False-positive uptake of 18 F-FES is reported

Prognostic and Immunoinfiltration Analysis of Transcription Factor BTF3 in Pan-cancer.

INTRODUCTION: BTF3, also known as BETA-NAC, BTF3b, BTF3a, and NACB, is a transcription factor originally isolated from HeLa cell extracts. It forms stable complexes with RNA polymerase II and plays a critical role in transcription initiation. Although aberrant BTF3 expression has been reported in certain malignancies, its overall role across diverse tumor types remains poorly defined. This study aimed to elucidate the functional implications of BTF3 across various cancers. METHODS: BTF3 expression was analyzed using datasets from The Cancer Genome Atlas (TCGA), including TCGA_GTEx, TCGA unpai

Microglandular adenosis, triple negative breast carcinoma and DNA repair defects.

AIMS: A subset of microglandular adenosis (MGA) displays protein expression and molecular genetic alterations similar to those of synchronous triple-negative breast carcinoma (TNBC), supporting the hypothesis that MGA is a non-obligate precursor lesion to a subset of breast carcinomas. Here, we further explore this association in the context of genomic instability. METHODS: We use whole-genome sequencing to investigate the genetic landscape of two unusual cases of MGA associated with carcinoma in the setting of two distinct varieties of genomic instability. RESULTS: The first case describes

Attenuated to absent myoepithelial cells in benign and non-invasive breast lesions-potential diagnostic pitfalls.

Loss of the myoepithelial cell (MEC) layer at the epithelial-stromal interface is a key histological marker of invasion in malignant breast lesions. However, certain benign and non-invasive entities may exhibit diminished or absent MECs, creating significant diagnostic challenges and occasionally leading to erroneous interpretation as invasive carcinoma. Despite their bland cytology, some of these lesions, such as microglandular adenosis (MGA) and pleomorphic adenoma, display infiltrative-like growth patterns. Loss of MECs around proliferating epithelium-lined clefts accompanied by spindled st

Incidental [68Ga]Ga-DOTATATE uptake in bone metastases of breast carcinoma in a patient with a secondary carcinoid tumor.

BACKGROUND: Lobular breast cancer often shows only low [18F]FDG uptake, limiting the sensitivity to detect metastases. Alternative tracers, targeting tumor activated fibroblasts or estrogen receptors, can offer higher accuracy. Also, Somatostatin Receptor 2 (SSTR2) is significantly expressed in some breast cancers, particularly in estrogen receptor rich tumors. Typical pulmonary carcinoids also have low [18F]FDG accumulation, but approximately 63% of well-differentiated lesions express SSTR2. CASE PRESENTATION: We report the case of a 66-year-old female patient with lobular breast cancer and

Radiation-induced morphoea in the setting of previous breast cancer: A case report.

Radiation-induced morphoea is an under-recognised complication of radiotherapy that can cause significant pain, functional impairment, and disfigurement. It is frequently misdiagnosed due to its non-specific presentation. We report the case of a 70-year-old female with prior right breast invasive ductal carcinoma, status post-lumpectomy, who developed painful cutaneous thickening and atrophy within the irradiated field 18 months after adjuvant radiotherapy. Malignant recurrence was excluded, and biopsy revealed dermal sclerosis with collagen thickening consistent with radiation-induced morphoe

Multi‑scale immune and spatial profiling of sclerosing adenosis and surrounding breast tissue identifies an immune‑cold field phenotype associated with breast cancer risk.

BACKGROUND: Benign breast disease (BBD) is common and confers heterogeneous increases in breast cancer risk; however, risk prediction relies mainly on histopathology and clinical factors. Sclerosing adenosis (SA) is a proliferative BBD lesion associated with an approximately two-fold increase in risk, yet most women with SA never develop breast cancer. We hypothesize that the immune-stromal microenvironment of SA and its surrounding lobular field relates to subsequent invasive breast cancer. METHODS: In a nested case-control study within a BBD cohort, we profiled 24 sclerosing adenosis (SA) b
