INTRODUCTION: Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease with limited therapeutic options, highlighting the need for multi-target interventions. Traditional Chinese botanical medicine (TCBM) interventions have attracted increasing attention as potential multi-target pharmacological candidates for ALS. In addition, a separate body of evidence derived from selected plant-derived bioactive compounds directly investigated in ALS-specific models provides complementary mechanistic insights into ALS-related pathways. However, the pharmacological mechanisms and translational p

Neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis, represent a major global health challenge due to their progressive nature and limited therapeutic options. One of the primary obstacles to treating these disorders is the blood-brain barrier (BBB), which restricts the delivery of most therapeutic agents into the central nervous system. In recent years, carbon-based nanomaterials, particularly carbon dot (CD) nanostructures, have emerged as promising platforms for neurological applications owing to their ultra

A relevant proportion of malignancies predominantly or exclusively affecting women, including breast and gynecologic cancers, is attributable to hereditary tumor syndromes, profoundly impacting cancer risk, prognosis, and therapeutic management. Today, the routine use of comprehensive germline panels has shifted the focus from solely pathogenic BRCA1/2 variants to include numerous pathogenic variants of other high- and moderate-risk genes. A broad spectrum of genetic alterations has been identified as causative for Hereditary Breast and Ovarian Cancer syndrome (HBOC), encompassing not only BRC

INTRODUCTION/OBJECTIVE: Raynaud's phenomenon (RP) is a common vasospastic condition that may develop secondary to cancer and/or in association with systemic autoimmune rheumatic diseases (SARDs). We aimed to estimate the risk of cancer in RP without known SARDs, phenotypically akin to 'primary' RP.
METHODS: A cohort study using data from North American electronic healthcare organization records. RP was defined using ≥ 2 ICD (I73.0) codes, excluding SARDs. Comparators had ≥ 2 irritable bowel syndrome ICD (K58) codes: selected with similar epidemiology to RP, and without any known excess in can

Background and Clinical Significance: Radiation-induced lumbosacral plexopathy (RILP) is a rare but potentially debilitating complication of radiotherapy, typically affecting patients treated for pelvic malignancies. We report the first documented case of asymmetric RILP following radiotherapy for breast cancer. Case Presentation: A 64-year-old woman developed progressive left lower limb weakness, foot drop, and sensory disturbances four years after receiving locoregional radiotherapy extending to the left thoracoabdominal and lumbar areas. Electrophysiological studies revealed an asymmetric s

Breast density is associated with increased risk of breast cancer (BC) and reduces mammography performance. Supplemental ultrasound (US) improves the performance of mammography screening programs in women with dense breasts but only few evidences are available about its added value when digital breast tomosynthesis (DBT) screening is performed. Our study evaluated the contribution of US in detecting BC in women with dense breasts in a screening program based on DBT. From February 2022 to October 2023, asymptomatic women (age range, 50-69 years) attending BC Screening in Verona, Italy, were pro

RATIONALE: Cutaneous metastatic carcinoma is a malignant tumor that originates outside the skin. Studies have found that the most common primary tumor in female patients with cutaneous metastatic carcinoma is breast cancer. Cutaneous metastatic carcinoma spreads to the adjacent skin through blood vessels, lymphatics, or interstitial spaces. It forms ulcerative or nodular lesions. Carcinoma en cuirasse is characterized by extensive induration and sclerosis of the skin, resembling armor, and is a rare form of cutaneous metastatic carcinoma.
PATIENT CONCERNS: We report a rare case of a 59-year-o

Triple-negative breast cancer (TNBC), a distinct breast cancer subtype, poses significant challenges to conventional therapeutic approaches, and effective targeted therapies are limited. CRISPR/Cas9 library screening has demonstrated unprecedented efficiency and revolutionary potential in the identification of therapeutic targets. In this study, we performed In vivo CRISPR/Cas9 library screening and identified the E2 ubiquitin-conjugating enzyme UBE2L3 as a critical regulatory factor in the progression of TNBC. Loss of UBE2L3 restricted tumor cell growth by modulating autophagy in TNBC cells.

BACKGROUND The incidence of breast cancer is high among women, with a significant proportion of cases being familial. However, the driver genes for breast cancer can differ across families. CASE REPORT Our patient was a 37-year-old woman diagnosed with triple-negative breast cancer (TNBC) by pathology, revealing invasive ductal carcinoma of the outer upper quadrant of the breast, WHO grade 3. The maximum diameter of the microscopic invasive cancer was approximately 0.5 cm. No definite vascular tumor thrombus or nerve invasion was observed. Some (30-90%) of the tumor cells disappeared, and the

INTRODUCTION: BTF3, also known as BETA-NAC, BTF3b, BTF3a, and NACB, is a transcription factor originally isolated from HeLa cell extracts. It forms stable complexes with RNA polymerase II and plays a critical role in transcription initiation. Although aberrant BTF3 expression has been reported in certain malignancies, its overall role across diverse tumor types remains poorly defined. This study aimed to elucidate the functional implications of BTF3 across various cancers.
METHODS: BTF3 expression was analyzed using datasets from The Cancer Genome Atlas (TCGA), including TCGA_GTEx, TCGA unpai

The present study aimed to evaluate the incidence of short-term complications and health-related quality of life (HRQoL) in patients with breast cancer undergoing breast-conserving surgery (BCS) with intraoperative radiotherapy (IORT), and to identify associated influencing factors. The study prospectively analyzed clinical data from women who underwent BCS with IORT at Tianjin Medical University Cancer Institute and Hospital (Tianjin, China) between March 2021 and June 2023. Telephone follow-up was conducted within 3 months post-surgery to assess surgery- and radiotherapy-related complication

BACKGROUND: Lobular breast cancer often shows only low [18F]FDG uptake, limiting the sensitivity to detect metastases. Alternative tracers, targeting tumor activated fibroblasts or estrogen receptors, can offer higher accuracy. Also, Somatostatin Receptor 2 (SSTR2) is significantly expressed in some breast cancers, particularly in estrogen receptor rich tumors. Typical pulmonary carcinoids also have low [18F]FDG accumulation, but approximately 63% of well-differentiated lesions express SSTR2.
CASE PRESENTATION: We report the case of a 66-year-old female patient with lobular breast cancer and

Radiation-induced morphoea is an under-recognised complication of radiotherapy that can cause significant pain, functional impairment, and disfigurement. It is frequently misdiagnosed due to its non-specific presentation. We report the case of a 70-year-old female with prior right breast invasive ductal carcinoma, status post-lumpectomy, who developed painful cutaneous thickening and atrophy within the irradiated field 18 months after adjuvant radiotherapy. Malignant recurrence was excluded, and biopsy revealed dermal sclerosis with collagen thickening consistent with radiation-induced morphoe

BACKGROUND: Sirolimus is a mammalian target of rapamycin inhibitor that has been used in solid organ transplantation and is now increasingly being administrated to treat various manifestations of Tuberous Sclerosis (TSC). Lack of data regarding its safety during breastfeeding has led to its contraindication by pediatric and breastfeeding guidelines.
METHODS: Levels of sirolimus were evaluated in breast milk samples collected during 24 h on the 10th and 15th day postpartum, by a lactating TSC patient chronically medicated with 4 mg of sirolimus per day. Controls were healthy lactating women. S

OBJECTIVES: Systemic sclerosis (SSc) is a heterogeneous systemic autoimmune disease with distinct subsets identified by specific autoantibodies. Some environmental agents might play a role in SSc pathogenesis, including silicone breast implants (SBI). This association has been controversial in previous literature and only few studies reported the auto-antibody status in these SSc women. The objective of this study was to evaluate the association of SBI with SSc in a large cohort of Italian patients, classified according to their SSc-related autoantibodies and to their history of breast cancer.

BACKGROUND: Localized scleroderma, or morphoea, is a rare autoimmune disease that affects the skin and underlying tissue. It is more common in girls and women than in boys and men. The incidence has two peaks: one in childhood and another in middle-age. Concomitant autoimmune diseases are frequently observed, whereas systemic sclerosis occurs rarely simultaneously.
OBJECTIVES: To assess the clinical features, comorbidities and treatments of localized scleroderma in Southwest Finland.
METHODS: Patients diagnosed with localized scleroderma (International Classification of Diseases, 10th Revisi

INTRODUCTION: Systemic sclerosis (SSc, scleroderma) is a rare autoimmune disease characterized by fibrosis of the skin and internal organs. Skin sclerosis, especially around the joints, may lead to functional impairment. Dysfunction of internal organs, particularly of the respiratory and cardiovascular systems, significantly affects the patient's physical performance.
AIM: To analyse hand joint function, strength, chest expansion, exercise tolerance using the 6-minute walk test (6MWT), and the range of temporomandibular joint (TMJ) movements in patients with systemic sclerosis treated at the

Autoimmune/inflammatory syndrome induced by adjuvants (ASIA) encompasses a spectrum of immune-mediated clinical conditions triggered by exposure to adjuvant substances such as aluminum, vaccine components, and silicone breast implants (SBIs). We present the case of a 68-year-old woman with a history of silicone breast augmentation, followed by implant rupture and reimplantation. Years later, she developed Raynaud's phenomenon, cutaneous calcinosis, and sicca symptoms. The patient also exhibited cognitive impairment and depressive symptoms. Laboratory investigations confirmed the presence of a

OBJECTIVE: SSc is associated with increased malignancy risk, a growing cause of mortality in SSc. However, standardised incidence ratios (SIR) vary widely across studies and different malignancy subtypes. This meta-analysis aimed to determine the SIR of different malignancy subtypes and the temporal relationship with SSc diagnosis.
METHODS: A systematic review and meta-analysis were conducted by searching Embase, MEDLINE and Cochrane Library databases from inception to 7 December 2024. Cohort studies reporting SIRs of malignancies in SSc patients were included. Studies where SIR was not repor
