Human adenoviruses (HAdVs) are double-stranded DNA viruses that can cause a wide range of infections, including respiratory, gastrointestinal, and ocular diseases, as well as less common conditions such as hepatitis, hemorrhagic cystitis, and encephalitis. While most infections in immunocompetent individuals remain clinically asymptomatic, immunocompromised patients are at a high risk of developing severe HAdV infections. Due to their exceptional transduction efficiency, HAdVs are also widely used as vaccine vectors, such as in vector-based COVID-19 vaccines. Although HAdVs are present as path

Human adenoviruses (HAdVs) cause a broad spectrum of diseases, ranging from mild respiratory and gastrointestinal infections to severe, sometimes even fatal illness in immunocompromised individuals. Species D adenoviruses represent the largest group, possibly driven by strong evolutionary pressure, particularly within the highly variable early transcription unit 3 (E3), which encodes immunomodulatory proteins. Among these, E3/49K is unique to species D and utilizes a particularly intriguing immunomodulatory strategy. It is a heavily glycosylated transmembrane protein that undergoes proteolytic

Hepatitis-hydropericardium syndrome (HHS) caused by fowl adenovirus serotype 4 (FAdV-4) has led to substantial economic losses in the poultry industry. Subunit vaccines based on the knob domains of Fiber1 and Fiber2 (F1F2K) are safe but exhibit limited immunogenicity. To address this, we sought to enhance the immune response by presenting F1F2K on self-assembling Mi3 nanoparticles (F1F2K:Mi3) and to compare its efficacy with the F1F2K monomer. F1F2K was conjugated to Mi3 via the SpyTag/SpyCatcher system. Chickens were immunized twice with F1F2K:Mi3 nanoparticles, F1F2K monomer, Mi3, or PBS, an

Adenoviral infection has been implicated in spontaneous abortion, but the relationship between adenovirus-associated pregnancy loss and systemic apoptotic activation remains incompletely understood. This study investigated serum caspase-3 (CASP3) concentrations and cleaved caspase-3 protein expression in women with adenovirus-associated spontaneous abortion and evaluated the diagnostic performance of serum CASP3. A comparative cross-sectional study was conducted including 90 pregnant women: 46 with adenovirus-associated spontaneous abortion and 44 healthy pregnant controls. Serum CASP3 concent

Equine adenovirus type 1 (EAdV-1) infection has been recognised among horses worldwide. The virus predominantly targets respiratory tract but has also been recovered from other body systems. The objective of the study was to determine the frequency of EAdV-1 infection among selected horse populations in Poland and to estimate genetic variability within the viruses obtained. A quantitative PCR assay targeting a partial hexon gene was used to screen 1150 nasal swabs collected from 1145 horses from 19 horse studs throughout Poland. The samples were collected opportunistically between 2012 and 202

Fowl adenovirus serotype 4 (FAdV-4) is the major pathogen responsible for avian hepatitis-hydropericardium syndrome (HHS), which poses a severe threat to the global poultry industry. This study aimed to systematically explore virus-host interactions and elucidate the transcriptomic alterations and underlying molecular mechanisms in LMH cells following FAdV-4 infection. LMH cells were infected with FAdV-4 at a multiplicity of infection (MOI) of 1 for 24 h, and whole-transcriptome sequencing was subsequently performed. Differential expression analysis was conducted between the FAdV-4-infected gr

Reliable evidence on COVID-19 vaccine effectiveness during the period of Omicron variant predominance was essential to inform global vaccination strategies. Our living systematic review, commissioned by the World Health Organization (WHO), was established to provide an ongoing synthesis and critical appraisal of comparative observational studies evaluating the effectiveness of vaccines against COVID-19/SARS-CoV-2 infection caused by the Omicron variant. Weekly searches covering seven databases up to 31 December 2022 sought evidence for WHO-approved vaccine platforms, including mRNA vaccines (B

Fowl adenovirus serotype 4 (FAdV-4) is a nonenveloped double-stranded DNA virus with a 43-45 kb genome, making it a promising viral vector for developing multivalent poultry vaccines. However, limited genomic studies have hindered the design of strategies for generating recombinant FAdV-4 capable of expressing multiple foreign proteins while maintaining high viral titers. In this study, three deletion mutants (rD14AB, rD2042, rD19A) were generated to assess their roles in viral replication and pathogenicity. Only rD2042 showed impaired replication and virulence compared with wild-type FAdV-4.

Rapid, sensitive, and RADIANT-deployable nucleic acid diagnostics are essential for the prevention and control of porcine adenovirus type 3 (PAdV-3). In this study, we developed RADIANT (RAA-Cas13d DIagnostic plATform for extractioN-free Testing), an extraction-free platform capable of delivering accurate and portable detection in resource-limited settings. By systematically optimizing the reaction buffer composition, recombinase-aided amplification (RAA), T7 transcription, and CRISPR/EsCas13d-mediated cleavage were integrated into a one-pot reaction, reducing the total time-to-result to withi

Human adenovirus type 55 (HAdV-55) is an emerging respiratory pathogen associated with severe pneumonia and acute respiratory disease, yet no licensed vaccine is currently available. Here, we evaluated the immunogenicity and protective efficacy of an inactivated HAdV-55 vaccine formulated with aluminum hydroxide (iHAdV-55 + AH) in cynomolgus macaques. Animals received a prime-boost intramuscular immunization, and HAdV-55-specific binding IgG and neutralizing antibody responses were monitored longitudinally for up to 52 weeks. Protective efficacy was assessed in two independent respiratory chal

UNLABELLED: High mobility group box 1 (HMGB1) is a host-derived proinflammatory molecule released during cell injury. Elevated extracellular HMGB1 levels have been associated with disease severity. HMGB1 contributes to host influenza responses by stimulating TLR4/MD-2 signaling, amplifying lung inflammation, and tissue damage via cytokine production and leukocyte infiltration. HMGB1 also plays a central role in other inflammatory diseases, including sepsis and liver toxicity, making it both a biomarker of disease severity and a potential therapeutic target. We previously reported development o

The emergence of diverse SARS-CoV-2 Omicron subvariants has reduced the efficacy of existing vaccines, highlighting the need for multivalent vaccines with broader neutralizing capacity. We developed a trivalent adenoviral vector-based vaccine (Ad5-CoV19-prototype/Delta/BF.7) encoding the spike proteins of the ancestral strain, Delta variant, and Omicron BF.7 strain, and evaluated its immunogenicity in BALB/c mice. Four- to 6-week-old female BALB/c mice were intramuscularly immunized with 10 µg of monovalent or trivalent vaccines using single-dose or two-dose (2-week interval) regimens. Neutral

Existing vectored immunoprophylaxis (VIP) approaches have primarily focused on IgG, which provides systemic protection but is less specialized in mucosal immunity. In contrast, secretory IgA (sIgA) plays a central role at epithelial surfaces, promoting pathogen neutralization while limiting inflammation. Although monoclonal IgA therapies are effective, their short half-life requires repeated dosing. Thus, VIP strategies enabling sustained sIgA expression at mucosal sites could transform mucosal infection prevention and treatment. This review outlines the key challenges associated with in vivo

In this study, a recombinant adenovirus vaccine candidate constructed and evaluated to addressed the widespread clinical problem of co-infections of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) and Porcine Circovirus Type 2 (PCV2). Epidemiological analysis revealed a PRRSV-positive rate of 40.8%, with prevalent strains belonging to Lineage 1 and Lineage 8. Analysis of key amino acid sites in the GP5 protein indicated the presence of highly virulent strain characteristics and widespread wild-type strain variation. Based on these findings and previous studies, we selected multiple

Adenoviruses (Adenoviridae) and their associated replication-deficient dependoparvoviruses (Parvoviridae) are recognized as significant components of the avian virome. While well-studied in domestic birds, these viruses remain underexplored in wild Antarctic birds. Information is limited on viruses circulating in penguins, particularly outside of the Antarctic Peninsula Region. Here we leverage a fecal sample collected for diet analyses to identify an aviadenovirus (37,731 nt) and two distinct dependoparvoviruses. The Adélie penguin (Pygoscelis adeliae) feces-derived adenovirus exhibits a geno

The E3 ubiquitin ligase TRIM23 is involved in diverse cellular processes, however, its function in antiviral defense against adenovirus remains unclear. Here, we identify a novel mechanism by which TRIM23 restricts human adenovirus type 5 (HAdV-C5) replication. TRIM23 expression was upregulated upon HAdV-C5 infection, and functional studies showed that its overexpression inhibited viral replication, while knockdown enhanced it. Mechanistically, TRIM23 interacts with the viral E1A protein and promotes its degradation through a mechanism dependent of its canonical E3 ligase activity. Moreover, T
