Human adenovirus (HAdV) is one of the causative viral pathogens of acute gastroenteritis (AGE) in children worldwide, accounting for approximately 10% of diarrheal cases. To enhance understanding of the prevalence and epidemiology of HAdV infection, we conducted a 3-year molecular detection and characterization study of HAdV associated with AGE in Chiang Mai, Thailand, from 2022 to 2024. Among 1356 pediatric patients, HAdV was detected in 85 cases (6.3%). The highest HAdV infection rate was observed in 2023 (10.4%). HAdV infections were most frequently detected in children aged 3-4 years. Co-c

The virome plays a significant role in maintaining the gut health of the host. Fecal virome transplantation (FVT), a burgeoning therapeutic strategy, holds promise in regulating intestinal microecology and treating associated diseases. However, the presence of eukaryotic viruses in FVT poses potential risks, which may compromise its safety and efficacy. This study leverages the key distinction between bacteriophages and eukaryotic viruses-namely, the presence or absence of an envelope-to reduce the burden of eukaryotic viruses in samples via solvent/detergent (S/D) treatment, while preserving

Platforms based on adenoviral vectors have proven their efficiency in developing vaccines and therefore can be considered potential candidates for immunization strategies against CCHFV. In this study, four replication-deficient recombinant human adenovirus serotype 5 (AdV5) vectors were engineered to express CCHFV antigens: a dual-antigen NP-P2A-Gc construct and monocistronic constructs for nucleoprotein (NP), glycoprotein complex precursor (GPC), and mature glycoprotein (Gc). Following GMP-compatible production and purification, vaccine candidates were evaluated in BALB/c mice (n = 4 per grou

Human adenoviruses (HAdVs) are characterized by extensive type diversity and widespread preexisting immunity, yet whether natural HAdV infection induces immune imprinting, the tendency of the immune system to preferentially recall responses to the first encountered antigenic variant, remains unclear. In this cross-sectional study of 1150 adults, neutralizing antibody (NT50) profiles against HAdV-3, -5, and -7 were determined, and multi-positive sera were analyzed by cross-adsorption neutralization and the cross-antibody contribution index (CACI). Seroprevalences were 69.8% (HAdV-3), 63.0% (HAd

We conducted systematic surveillance of circulating viruses in two cities in Chile through regular wastewater analysis between 2018 and 2021. Samples were concentrated, nucleic acids were extracted, and the samples were analyzed by massive sequencing. After bioinformatic processing, we detected a significant number of Adenovirus F41 genomes in samples collected during a specific period (January-March 2020), particularly in treatment plants from Santiago de Chile. To confirm whether the virus was circulating in the city during that period, we analyzed stool pools collected at one of the city's

Human adenovirus type 55 (HAdV-55) is an emerging respiratory pathogen associated with severe pneumonia, for which no licensed vaccines are currently available. The lack of physiologically relevant small-animal models has limited preclinical evaluation of vaccine candidates. In this study, we generated a transgenic mouse model co-expressing human CD46 and desmoglein-2 (DSG-2), key entry receptors for HAdV-55, and evaluated its utility for vaccine efficacy testing. A bicistronic expression system enabling simultaneous expression of CD46 and DSG-2 was constructed and functionally validated in vi

Ebola virus is a highly virulent pathogen causing severe hemorrhagic fever with 25%-90% mortality. The 2014 outbreak infected over 28,000 people, causing 11,325 deaths. Although there is one FDA-approved vaccine available for limited use, additional vaccines are needed. We developed adenoviral-vectored vaccines expressing the ebolavirus glycoprotein (GP), the sole viral surface protein and primary immunogen. Vaccine constructs spanned three species and five human adenovirus subtypes, including low-seroprevalent serotypes to minimize pre-existing immunity. We compared single-dose and prime-boos

BACKGROUND: Excessive unnecessary protein accumulation in cardiomyocyte is a leading contributor for pathological cardiac hypertrophy and has been found closely regulated by the mTORC1 (mechanistic target of rapamycin complex 1) signaling and lysosome transmembrane proteins. However, the precise regulatory mechanism stratifying mTORC1 signaling and the specific functions of lysosomal proteins in protein homeostasis of cardiomyocytes remain largely unidentified.
METHODS: We screened lysosomal genes conserved in mice, rats, and humans. Adenoviral infection of rat cardiomyocytes was used to asse

Highly pathogenic avian influenza (HPAI) represents an enormous pandemic risk amid the ongoing H5N1 panzootic. HPAI, defined by its high lethality in domestic poultry, includes the influenza A virus (IAV) H5N1 and H7N9 subtypes and has a 40-50% case fatality rate in humans. To facilitate the development of efficacious HPAI vaccination regimens and isolation of HPAI-neutralizing monoclonal antibodies (nmAbs) for prophylactic and therapeutic use, we performed a proof-of-concept pilot study wherein we developed an array of mRNA and adenovirus-vectored vaccines encoding HPAI hemagglutinins (HAs) a

Background and objectives Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to Western Ghats in India, necessitates rapid and accurate field-based diagnostics for effective outbreak response and timely patient management. Traditional diagnostic methods require laboratory infrastructure, sometimes limiting timely case detection in resource limited settings. We evaluated the diagnostic performance, usability, and operational feasibility of a point of care (POC) assay for detection of KFD virus under field conditions. Methods We assessed a real-time PCR-based portable

Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to forested regions of South India, with case fatality rates ranging from 3% to 10% and seasonal outbreaks causing recurrent illness among at-risk populations such as forest workers and villagers. The historically used formalin-inactivated KFD vaccine showed limited effectiveness in field studies, providing only about 62% protection. This moderate level of protection highlights the need for improved vaccines that offer higher efficacy and longer-lasting immunity for populations living in endemic areas. A major chall

Ticks are arthropod vectors that transmit a wide range of emerging and re-emerging vector-borne diseases (VBDs), many of which are of considerable zoonotic importance. Recently, tick-borne diseases (TBDs), particularly those caused by arboviruses, have emerged as a significant public health concern in West Africa. The complex socio-ecological landscape in West Africa contributes to the circulation of several tick-borne arboviruses (TBAs), including Crimean-Congo hemorrhagic fever virus (CCHFV) and Dugbe virus (DUGV), both members of the genus Orthonairovirus, as well as other tick-borne flaviv

TRIpartite Motif (TRIM) protein 5 alpha (TRIM5α) is a well-characterized cellular inhibitor of lentivirus replication that limits transmission of related viruses between primates. We previously reported that TRIM5α derived from humans and rhesus macaques inhibits replication of orthoflaviviruses belonging to the tick-borne encephalitis virus (TBEV) serocomplex, including TBEV, Kyasanur forest disease virus and Langat virus (LGTV), but interestingly not the tick-borne Powassan virus (POWV). To further characterize the primate TRIM5α and orthoflavivirus interface, we screened TRIM5α variants fro

Ticks are vectors of a wide range of pathogens, relevant to both veterinary and public health. Although tick surveillance is frequently conducted in the Netherlands, data on tick-borne pathogens in free-living herbivores remains limited, leaving the impact of these pathogens on animal health poorly understood. This study aimed to characterise tick species and determine seroprevalence and infection prevalence of selected tick-borne pathogens in large herbivores at the Oostvaardersplassen nature reserve and questing ticks collected in the area. Between September 2023 and December 2024, blood was

Tick-borne diseases constitute a major group of emerging vector-borne infections and represent a growing public health concern in many European countries. In Europe, the most clinically significant agents transmitted by Ixodes ricinus are tick-borne encephalitis virus (TBEV) and Borrelia burgdorferi sensu lato, the causative agents of tick-borne encephalitis and Lyme borreliosis, respectively. Ixodes ricinus also serves as a vector for other pathogenic microorganisms, including louping-ill virus (LIV) and Anaplasma phagocytophilum. Various endosymbionts, such as Wolbachia pipientis and Midichl

Climate change is reshaping the epidemiology of vector-borne diseases in Europe by altering the ecological conditions that determine vector survival, seasonal activity and pathogen transmission. Rising temperatures, milder winters, prolonged warm seasons and changing precipitation patterns are increasing the suitability of parts of Europe for competent mosquito, tick and sandfly vectors. These changes, combined with human mobility and land-use change, increase the probability that imported pathogens encounter permissive conditions for local transmission. This Opinion article examines autochtho

Flaviviruses, encompassing notable pathogens, like Dengue, Zika, West Nile, and tick-borne encephalitis viruses, elicit complex cellular stress responses, involving pathways such as the unfolded protein response (UPR), integrated stress response (ISR), apoptosis, autophagy, and the antiviral immune response. These pathways regulate cell fate by either promoting survival to counteract virus-induced damage or triggering cell death programs under prolonged and irreparable stress. Therefore, the primary aim of flavivirus-induced cellular responses is to protect cells and hinder viral propagation.

The Powassan virus (POWV) is a neurovirulent tick-borne virus that causes age-associated lethality and long-term neurologic sequelae in 50% of survivors. In aged mice the POWV strain LI9 mirrors human lethality and neuropathology; however, an avirulent POWV mutant, LI9-D308N, fails to enter the CNS or cause lethal disease. The D308N mutation is present in an envelope protein domain associated with cell attachment, yet the role of D308N mutations in cell tropism and neuroinvasion remains to be resolved. Here, we engineered fluorescent mScarlet3 and mNeonGreen reporter genes into WT LI9, and avi
