Viral infections of the central nervous system (CNS) pose a significant risk for seizures, and in some cases, long-term epileptogenesis. Seizure development following viral infection involves diverse CNS mechanisms, with emerging evidence that peripheral factors, including gut microbiome composition and immune-inflammatory responses, also contribute. The gut microbiome is a multifaceted ecosystem with important roles in CNS health, and advances in sequencing have enabled investigation of host and microbial genomic changes associated with seizure development. While both animal and human studies

INTRODUCTION: Herpes simplex virus (HSV) infection is a lifelong viral infection with high global prevalence. Emerging evidence suggests that chronic viral infections, including HSV, may contribute to the development of cardiometabolic diseases through persistent inflammation, endothelial dysfunction and immune dysregulation. However, findings on the association between HSV infection and cardiometabolic outcomes remain inconsistent. This systematic review and meta-analysis will synthesise the available evidence on the relationship between HSV infection and cardiometabolic diseases.

Arthropod-borne, positive-sense RNA viruses encompass many human pathogens posing significant health threats worldwide. Understanding intracellular dynamics and host-factor interactions of the viral genome is essential for devising effective antiviral strategies. Here, we engineer dual-labeled Zika and chikungunya viruses incorporating a Mango-II aptamer for RNA pulldown and real-time imaging, achieving high-resolution imaging of flavivirus RNA in live cells. Tandem imaging with viral replication proteins revealed intracellular dynamics of replication complexes and RNA molecules of these virus

Human noroviruses (HuNoVs) are the leading global cause of acute gastroenteritis, yet no vaccines or antiviral therapies are currently approved. The non-structural protein NS3 is a membrane-bound AAA+ ATPase of superfamily 3 (SF3) with multiple proposed roles in the norovirus replication cycle. However, the structure of NS3, and the mechanisms by which it contributes to genome replication and membrane remodeling, have remained unknown. We engineered a soluble, hexameric, and catalytically active form of NS3 and determined its cryo-EM structure in the presence of a nucleotide analogue at 2.9 Å

Adeno-associated virus (AAV) is widely used for liver-directed gene therapy, with AAV serotype 3B (AAV3B) showing particularly high tropism for human hepatocytes. However, pre-existing neutralizing antibodies (NAbs) against AAV represent a major barrier to efficient gene transfer and patient eligibility. We have recently demonstrated that subjects with intermediate NAb levels (1:5-1:45), that would be excluded from clinical trials, can be converted in eligible patients by the administration of IgG-cleaving enzymes. It is thus crucial to assess not only the seroprevalence but also titer distrib

Hantaviruses are globally distributed zoonotic pathogens belonging to the family Hantaviridae and are responsible for two major human diseases, hemorrhagic fever with renal syndrome (HFRS) and hantavirus cardiopulmonary syndrome (HCPS). Despite substantial progress in hantavirus research, important challenges remain in understanding the interactions among viral evolution, reservoir ecology, environmental change, transmission dynamics, and host immunopathology. This review provides an integrated One Health perspective that connects molecular virology and host-virus interactions with transmissio

Horses are essential for transportation and livelihoods in Indigenous communities of Costa Rica. However, information on their exposure to viral pathogens remains scarce. We conducted a retrospective cross-sectional serosurvey using samples collected from horses from four Indigenous communities (Alto Pacuare, Amubre, Paso Marcos, and Vereh) sampled between March and October 2011 (n = 278). Sera were tested in 2014 using commercial immunodiffusion or ELISA assays for antibodies against West Nile virus (WNV), equine herpesviruses 1 and 4 (EHV-1 and EHV-4), influenza A virus, equine arteritis vir

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) is a life-threatening infectious disease with high mortality. Although sarcopenia and myosteatosis are associated with adverse outcomes in multiple acute illnesses, their prognostic impact on SFTS remains unknown. This study aimed to evaluate the association between body composition parameters and 28-day mortality in SFTS patients, and to identify independent prognostic factors.
METHODS: In this retrospective observational single-center study, 234 SFTS patients with complete clinical data and available computed tomography (CT) ima

Dual adeno-associated virus (AAV) vector reconstitution has become an indispensable technique for delivering large genes in neuroscience and gene therapy. A novel technology termed StitchR addresses the challenge of low efficiency in dual-AAV reconstruction, yet related tools enabling targeted gene delivery to specific neural cell populations remain insufficiently developed. Herein, we developed four StitchR-based dual-viral reconstitution strategies by combining high-efficiency AAV serotypes with neuron- or glia-specific promoters. Specifically, the AAV11-hSyn-driven StitchR strategy achieved

BACKGROUND: Newly positive hepatitis B virus (HBV) serologic results after allogeneic hematopoietic cell transplantation (HCT) may reflect passive antibody transfer from blood-derived products rather than HBV infection. We investigated the incidence and temporal patterns of hepatitis B surface antibody (anti-HBs) and antibody to hepatitis B core antibody (anti-HBc) acquisition after allogeneic HCT.
STUDY DESIGN AND METHODS: We retrospectively evaluated 193 adults who underwent allogeneic HCT for hematologic diseases between May 2012 and March 2025 at a single institution in Japan. Anti-HBs, a

Hyalomma marginatum (Ixodida: Ixodidae) is the principal vector of Crimean-Congo hemorrhagic fever virus (CCHFV) and an emerging public health concern because climate change may facilitate its establishment in regions where it is currently absent. We used ecological niche modeling to characterize the current climatic suitability of H. marginatum and project its future redistribution across the Old World under CMIP6 climate scenarios. Models were calibrated using a comprehensive dataset of spatially filtered occurrence records and bioclimatic predictors within a biologically informed accessible

BACKGROUND: Dengue fever demonstrates a broad spectrum of clinical severity, ranging from self-limiting illness to life-threatening complications. Early identification of patients at risk of progression remains a clinical challenge, particularly in high-burden settings. Although platelet count and World Health Organization (WHO) dengue classification are routinely used in clinical practice, reliance on individual parameters may not adequately capture the multifactorial nature of disease severity. Development of multivariable predictive models integrating clinical and laboratory markers availab

Obesity is a complex and chronic public health challenge that reflects deep-rooted social, economic, and environmental determinants rather than solely individual choices. Its global prevalence has risen sharply over recent decades, affecting more than one in eight people worldwide, with a growing burden in low- and middle-income countries. This paper explores obesity as a complex and growing public health challenge that extends beyond individual behavior. It highlights the social, economic, and environmental factors driving the epidemic, with a focus on the Indian context where undernutrition

Overweight increases dyspnea and cough. Overweight patients with asthma experience more symptoms, frequent exacerbations and have poorer effects of asthma medications. In COPD, light and moderate overweight are associated with a better prognosis, whereas severe overweight is not. Similar protective effects of overweight have been reported in severe bacterial pneumonia and ARDS, but not in COVID-19 or severe influenza. This review finds that the beneficial effects of weight loss are best documented in asthma and in respiratory sleep-related disorders.

Infectious diseases result from complex interactions between a host, a pathogen and a vector within a given environment. Over recent decades, climate change, population movements and international trade have contributed to the emergence of arboviruses in temperate regions by facilitating the establishment of their vectors and pathogens. The presence of Aedes and Culex mosquitoes in Europe enables local transmission of viruses such as dengue (DENV), chikungunya (CHIKV) and West Nile virus (WNV). Clinicians should therefore consider these infections in the differential diagnosis of febrile illne

INTRODUCTION: Chikungunya virus (CHIKV) is primarily transmitted to humans through bites of infected Aedes mosquitoes, causing febrile illness as well as acute or chronic arthritis. With increasing reports in previously non-endemic regions, it poses an emerging threat to public health and a growing risk to travelers. Despite the recent approval of live-attenuated and virus-like particle-based vaccines, a safe, efficacious, and cost-effective vaccine remains urgently needed. The envelope glycoproteins E1 and E2, which play crucial roles in viral attachment and entry, are the major immunodominan

INTRODUCTION: Marburg virus (MARV) is a hemorrhagic fever virus, and its glycoprotein (GP) represents an important target for vaccine research. MHC class I (MHC-I)-restricted T cell epitopes are critical for inducing cytotoxic CD8+ T cell responses. In this study, we aimed to identify MHC-I-restricted T cell epitopes derived from the MARV Angola strain GP through an integrated bioinformatics and experimental approach.
METHODS: We retrieved the MARV Angola strain GP sequence and divided it into 673 overlapping 9-mer peptides. By integrating five computational prediction algorithms, immunogenic

BACKGROUND: In China, hand, foot, and mouth disease (HFMD) persists as a prevalent infectious disease with high transmissibility. Nevertheless, the link between environmental determinants and HFMD incidence has not been fully elucidated. The study assessed associations between air pollution, meteorological conditions, and HFMD incidence in Shenzhen, China, while also evaluating shifts in these associations before and after the Enterovirus 71 vaccine and during the COVID-19 epidemic.
METHODS: Daily data on HFMD cases, air pollutants (such as PM2.5, PM10, SO2, NO2, CO, and O3), and meteorologic

BACKGROUND: Health-related quality of life (HRQoL) is a key patient-centered outcome in HIV care, particularly in the current treatment era in which virological control is increasingly achievable but residual physical, mental, and social burden may persist. However, the clinical applicability of HIV-specific HRQoL instruments in Turkish-speaking people living with HIV (PLWH) remains insufficiently established. We aimed to characterize HRQoL in a multicenter Turkish sample of PLWH and to evaluate whether the Turkish version of the Medical Outcomes Study HIV Health Survey (MOS-HIV) provides reli
